Pseudo-SMB, often called “J-O process”, is a modified SMB process to completely separate a ternary mixture with two discrete steps per one cycle. For improved separation, two new design parameters, the position of step 1 (χS1) and the number of port switches during step 2 (nSMB), were introduced. A multi-objective optimization method was used to optimize the operating conditions of the pseudo-SMB process with four average zone flow-rate ratios for one cycle. Nadolol isomers were selected for the model solutes and the global objective for the design of the pseudo-SMB was to collect 99% of the intermediate retained solute. The separation was optimized for 8-column pseudo-SMB system with three column lengths (2.5, 5.0, and 10 cm) and three feed composition ratios (1/1/1, 1/2/1, and 2/1/2). The simulation results showed that productivity was increased 4.3 times (nSMB = 20, χS1 = 0.5, 1/1/1) and desorbent to feed ratio D/F was decreased 45% (nSMB = 16, χS1 = 0.5, 1/1/1) compared to normal operation (nSMB = 8, χS1 = 0.5, 1/1/1). Productivity and D/F were significantly improved when short columns were used in the pseudo-SMB process. The pseudo-SMB was compared with recycle chromatography and SMB cascades for the same total amount of adsorbent. Recycle chromatography and 8-column SMB cascades using 20 cm and 40 cm of total column lengths were not able to separate the intermediate component with the target purity and the same feed rate of the pseudo-SMB process.  相似文献

The application of gradients in simulated moving bed (SMB) chromatography has recently attracted interest as a method for further improving the performance of this continuous separation process. One possible implementation of gradients consists in setting the solvent strength in the desorbent stream higher than that in the feed stream. As a result, the components to be separated are more retained in the zones upstream of the feed position and more easily eluted in the zones downstream of the feed position. If a liquid mobile phase is used, gradients can be created by dosing different solvents into the feed and desorbent ports. In a closed-loop gradient SMB arrangement the solvent strength within the unit will depend on the two feed compositions and on the characteristic flow-rates of the process. In this work an equilibrium stage model describing a true moving bed process is used to analyze numerically the main features of a two-step gradient SMB process. The adsorption isotherms are assumed to be always linear under isocratic conditions. The relevant Henry constants depend in a nonlinear manner on the composition of the solvent. Based on numerical simulations the impact of the two inlet solvent compositions is demonstrated in terms of the size and shape of regions of applicable flow-rates. Different strategies of designing the process are discussed and compared with respect to maximizing productivities and minimizing desorbent requirements.  相似文献

A gas phase simulated moving bed technology using improved 13X zeolite beads and isobutane as desorbent is assessed for the separation of propane/propylene. Adsorption equilibrium (via gravimetric method) and dynamics (via breakthrough curves) were determined in order to validate the mathematical model used to describe the adsorption process. Simulation results have shown that it is possible to separate propylene from a mixture with propane using gas phase SMB technology. The results indicate that high purity propylene (99.99 % desorbent free basis) can be recovered up to 99.96 % with a productivity of 17.6 mol kg?1 h?1, with propane being also recovered at high levels (99.98 %) and high purity (99.89 % desorbent free basis). Comparing the SMB simulation results obtained for this new 13X zeolite beads with those obtained with a commercial 13X zeolite characterized elsewhere, it was found that the productivity of the process was raised by 25 %, with half desorbent consumption.  相似文献

We report on a numerical and experimental study of two-column versions of streamlined, multicolumn, semi-continuous chromatography for binary separation. The systems combine a flexible node design, cyclic flow-rate modulation, and relayed operation of the inlet/outlet ports to extend the mass-transfer zone over the largest possible length, while keeping it inside the system at all times. One advantage of these streamlined designs is the simplicity of their physical realization: regardless of the number of columns, they only require two pumps to supply feed and desorbent into the system, while the flow rates of liquid withdrawn from the system are controlled by material balance using simple two-way valves. In one case, an extra pump is needed to recirculate the fluid in closed-loop. A rigorous model-based optimization approach is employed in the optimal design of the cycles to generate solutions that are physically realizable in the experimental set-ups. The optimized schemes for two-column operation supply fresh feed into the middle of the system where the composition of the circulating fluid is closest to that of the feedstock fluid, and recover the purified products, extract and raffinate, alternately at the downstream end of the unit, while desorbent is continuously supplied into the upstream end of the system. By internally recycling part of the non-pure cut fraction, the scheme with a step of closed-loop recycling significantly reduces its solvent consumption. The feasibility and effectiveness of the reported two-column processes have been verified experimentally on the linear separation of nucleosides by reversed phase subject to 99% purity constraints on both products. It is shown that our processes compare favorably against single-column batch chromatography, steady-state recycling, and four-column, open-loop SMB, for the same amount of adsorbent; they are also better than the four-column, closed-loop SMB at high feed throughputs.  相似文献

A novel modification of simulated moving bed (SMB) technology, referred to as fractionation and feedback SMB (FF-SMB), has been introduced recently. This concept is based on fractionating one or both outlet streams and feeding the off-spec fractions back into the unit alternatingly with the original feed mixture. In this paper, the optimization problem of FF-SMB realizing one outlet fractionation is considered. A mathematical optimization framework based on a detailed process model is presented which allows to evaluate quantitatively the potential of this operating scheme. Detailed optimization studies have been carried out for a difficult separation characterized by small selectivity and low column efficiency. The results reveal that the proposed fractionation and feedback regime can be significantly superior to the classical SMB chromatography, in terms of both feed throughput and desorbent consumption. The effect of the feeding sequence on the performance of FF-SMB is also examined.  相似文献

The separation of dilute binary mixtures of proteins by salt aided ion-exchange simulated moving bed (SMB) chromatography is optimized with respect to throughput, desorbent consumption and salt consumption. The optimal flow-rate ratios are analytically determined via an adopted "triangle theory". Azeotropic phenomena are included in this procedure. The salt concentrations in the feed and recycled liquid are subsequently determined by numerical optimization. The azeotropic separation of bovine serum albumin and a yeast protein is used to illustrate the procedure. Gradient operation of the SMB is generally preferred over isocratic operation. A feed of azeotropic salt concentration can only be separated in a gradient SMB. Desorbent and salt consumption are always lower in gradient than in isocratic SMB chromatography.  相似文献

An optimization procedure for simulated moving bed (SMB) plants with low efficient stationary phases is presented. The new aspect is that the desorbent consumption can be cut by 70% by running the plant with lower internal liquid flows and a corresponding larger switch time while the productivity is kept constant. This concept was validated by the separation of fructose and glucose in water on a calcium resin with an eight-column SMB plant. The separation can be predicted well by a true moving bed (TMB) and a simulated moving bed simulation. Adsorption isotherms were determined up to 300 kg/m3 for glucose and 500 kg/m3 for fructose from 25 to 80 degrees C. Experimental SMB runs were performed over a wide range of feed concentrations (10-350 kg/m3) and temperatures (25-80 degrees C). The strong influence of the delay volume is pointed out. For an experimental run with high feed concentration a complete set of data is presented. To reduce biological growth separation at 80 degrees C is recommended.  相似文献

A bi-level optimizing control scheme originally proposed for a simulated moving bed (SMB) with linear isotherms has been extended to an SMB with nonlinear isotherms. Cyclic steady state optimization is performed in the upper level to determine the optimum switching period and time-varying feed/desorbent flow rates, and repetitive model predictive control is run in the lower level for purity regulation, taking the decision variables from the upper level as feed-forward information. Experimental as well as numerical study for an SMB process separating a high-concentration mixture of aqueous L-ribose and L-arabinose solutions showed that the proposed scheme performs satisfactorily against various disturbances. In contrast, an alternative scheme based on an SMB model with linear isotherms showed a limitation in the control performance; this scheme was apt to fail in purity regulation.  相似文献

Simulated moving-bed (SMB) chromatography is attractive for reducing sorbent and solvent consumption relative to fixed-bed systems. In this contribution, we describe a novel and versatile method for further reducing solvent consumption in the case of reversed-phase chromatography. The method is based on the variation of the distribution coefficients of solutes to be separated upon varying the composition of a multi-component mobile phase. If the solvent strength of the desorbent is set higher than the solvent strength of the feed, the components will have smaller distribution coefficients in the extraction section of the SMB and hence will be more easily eluted. This will result in a lower desorbent flow and possibly also in a shorter desorbent zone, and, ultimately, in more concentrated products. The so-called "Triangle-method" by Storti et al. [AIChE J., 39 (1993) 471] to obtain the region of complete separation, is extended for this novel SMB method. Theoretical evaluation of the proposed methodology supports the anticipated solvent reduction relative to fixed-bed RP-HPLC for the cases of the purification of the polyketide antibiotic nystatin and the separation of bovine insulin from porcine insulin.  相似文献

The solvent composition was adjusted in a theoretical study in order to maximize the efficiency of a simulated moving bed (SMB) process. The isocratic realization of the process as well as the solvent gradient mode were considered. The solvent composition and the flow rates were used as decision variables in a random search optimization algorithm known to be a reliable tool for nonlinear programming problems. The results of the optimization indicate that the optimal composition of the mobile phase depends strongly on the feed concentration. The asymmetry of the internal concentration profiles, which has a negative effect on the separation efficiency, can be partly damped by an increase of the solvent strength. In the cases studied the optimal solvent strength determined for concentrated feed streams is higher than that for diluted ones. Moreover, the optimum is strongly influenced by the value of the selectivity factor and its dependency on the mobile phase composition. Different results were obtained for cases, in which the separation factor increases with increasing the modifier concentration, than for cases, in which the separation factor decreases with increasing the modifier concentration. A similar analysis was performed for a solvent gradient SMB process, in which different solvents are used at the two inlet ports: a weak solvent in the feed stream and a strong solvent in the desorbent stream. Again the optimal mobile phase composition was strongly affected by the type of the isotherms and their non-linearity. The potential of a gradient SMB process in terms of increasing the productivity and reducing the eluent consumption is exemplified.  相似文献

This paper presents an analysis of a hybrid process consisting of simulated moving bed (SMB) chromatography and crystallization and studies its performance for the separation of the Tr?ger's base enantiomers. The SMB is simulated using a detailed model including column efficiency, thus, implying a proper evaluation of the effect of column size on column efficiency and separation performance. The crystallization operations are accounted for through material balances, assuming equilibrium between enantiopure crystals and mother liquor. A genetic algorithm is used to optimize the combined process, using proper definitions of objective functions. Multi-objective optimization of this hybrid process for productivity and evaporation cost in terms of operating parameters, column length, and SMB feed concentration shows an optimum SMB purity value as a trade off between increased SMB performance and recycle of the mother liquor.  相似文献

In simulated moving-bed (SMB) applications, throughput and desorbent consumption are two key factors that control process cost. For a given adsorbent volume and product purity requirements, throughput and desorbent consumption depend on desorbent composition, column configuration, column length to diameter ratio, and adsorbent particle size. In this study, these design parameters are systematically examined for paclitaxel purification. The results show that if adsorbent particle size, column dimensions and column configuration are fixed, the higher the product purity required, the lower the throughput. If product purity and yield are fixed, the larger the solute migration speed ratio, the higher the throughput, and the lower the desorbent consumption. If total bed volume and product purities are fixed, the longer the separation zones, the higher the throughput, but the higher the desorbent flow-rate. An intermediate configuration gives the minimum desorbent consumption. If there are no limits on pressure drop or zone flow-rate, the larger the column length to diameter ratio, the smaller the adsorbent particle size, the higher the throughput, and the lower the desorbent consumption. If the maximum zone flow-rate is controlled by the pressure drop limit and not by the standing waves requirement, the longer the columns, the lower the zone flow-rates and the lower the throughput. For 150 microns adsorbent particles and a maximum zone flow-rate of 300 ml/min, a design with optimal throughput and desorbent consumption is found for paclitaxel purification.  相似文献

Simulated moving bed (SMB) chromatography is an important technique for the continuous separation of valuable products. Recently suggested process modifications have shown the potential for further improvement. In this work, a concept is presented that combines non-permanent product withdrawal at one or both outlet ports (leading periodically to a "product" and a "non-product" fraction) with an internal recycle and re-feeding of the "non-product" fraction in alternation to the original feed mixture. Using simulation studies for linear and non-linear isotherms, it was shown that in terms of process performance and product recovery, this fractionation and feed-back approach (FF-SMB) is superior to both the conventional SMB process as well as to a previously reported fractionation and discard strategy.  相似文献

Modeling and simulation are presented of the separation of a ternary mixture by pseudo-simulated moving bed chromatography, according to the JO process of Japan Organo Co. The process cycle is divided into two steps. In step 1, feed and eluent streams are introduced into the system, equivalent to a series of preparative chromatographic columns, and the intermediate component is produced. In step 2, similar to a simulated moving bed (SMB), there is no feed and the less adsorbed species is collected in the raffinate while the more retained species is collected in the extract; this step is described by an equivalent TMB model. A parametric study is presented in order to analyze the sensitivity of the model to variations on the duration of step 1 and step 2 and on the intraparticle mass transfer coefficient.  相似文献

二十碳五烯酸和二十二碳六烯酸是两种非常重要的ω-3多不饱和脂肪酸,广泛用于膳食补充剂和药品,同时它们的生理作用并不完全相同,因此分离制备它们的高纯度单体十分必要。首先以聚苯乙烯/二乙烯基苯(PS/DVB)聚合物为固定相,在液相色谱上分离二十碳五烯酸乙酯(EPA-EE)和二十二碳六烯酸乙酯(DHA-EE),考察了流动相、填料粒径、温度对分离的影响。然后采用粒径20 μ m、孔径10 nm的PS/DVB填料装填了8根150 mm×10 mm的半制备色谱柱,测定了半制备柱装填的均一性。最后尝试在模拟移动床(SMB)色谱上分离EPA-EE和DHA-EE的混合物,探究了Ⅱ区和Ⅲ区的流量、进料流量、进料浓度对分离的影响,结果表明SMB制备的EPA-EE和DHA-EE的相对纯度分别为91.6%和93.6%,回收率分别为97.0%和91.6%,固定相生产率为5.97 g/(L\5h),溶剂消耗为1.52 L/g。SMB制备EPA-EE和DHA-EE具有较大的应用潜力。  相似文献

Continuous counter-current chromatographic separation has been carried out in a simulated moving bed system (SMB). We have worked with a SMB pilot plant (8 columns, 4.4 litres of resin each) which allows the continuous withdrawal of two different fractions. A mixture of glucose-fructose has been separated. To calculate the concentration profile within the separator an axial dispersed plug flow model and an equilibrium stage model have been employed; software has been created to simulate the behaviour of the separator. The necessary parameters of the mode: the adsorption equilibrium constant, the height equivalent to a theoretical plate and the bed voidage, have been acquired experimentally from elution chromatography measurements. The results calculated by simulation give a good representation of the experimental concentration profiles; other separations like xylitol-arabitol have been simulated. The influence of some factors like desorbent flow-rate, feed flow-rate and the bed voidage have been studied using the software. Once the system has worked in a two withdrawal way, an extension of the pilot plant has been constructed so as to obtain a third one. The necessary parameters of the three withdrawal model will be studied.  相似文献

The resolution of racemic gas mixtures by simulated moving bed (SMB) and pressure swing adsorption (PSA) is investigated by dynamic simulation and optimization. Enantiomer separation of inhalation anesthetics is important because there is evidence that the purified enantiomers may have different pharmacological properties than the racemate. The model parameters reported in an experimental investigation performed elsewhere are used to study the feasibility of this separation using SMB and PSA configurations. Both processes were modeled in gPROMS® as systems of differential algebraic equations. Operating conditions are optimized such that the feed throughput and product recovery for each process were maximized subject to equal constraints on the pressures and superficial gas velocities. SMB was found to be capable of resolving racemic feed mixtures with purity and recovery exceeding 99%. On the other hand, PSA was also able to provide a single purified enantiomer with low recovery of about 30% which may limit its application to enantiomer separation. Nevertheless, PSA consumes less desorbent, and achieves higher throughput at the sacrifice of lower recovery.  相似文献

The simulated moving bed (SMB) technology is a proven tool for efficient separation of binary mixtures. However, relying on isocratic conditions limits the applicability of the classical SMB approach when considering the field of bioseparations. Here, the use of gradients opens up new possibilities. A gradient in a SMB process can be established by using different solvent strengths in the incoming feed and desorbent streams, resulting in two internal plateaus of elution strength. Thus, compared to the conventional process, the overall amount of solvent needed can be reduced, productivity can be increased and more concentrated product streams can be obtained. In this contribution, two case studies will be presented. At first, the separation of bovine IgG from lysozyme will be analyzed as a model system. Antibodies are a common target substance in bio-chromatography, as therapeutic monoclonal antibodies are among the most promising biopharmaceuticals. Using adsorption data obtained from single-column experiments, an appropriate SMB process was designed and implemented. The second target component is the active dimeric form of the bone morphogenetic protein-2 (BMP-2). This protein was isolated from a renaturation solution, which also contained its inactive monomeric form as well as other undefined proteins from the bacterial production strain. A 3-zone open-loop gradient-SMB approach was used successfully for both separations.  相似文献

For a dilute feed, a four-zone SMB with temperature gradient (4-zone TSMB-FC) can fractionate and simultaneously concentrate two solutes. Optimization of the operating parameters for this system, which include four zone flow rates and port switching time, is a major challenge because of the five variables. Initial choice of the variables using the triangle theory followed by a systematic search using rate model simulations is time-consuming. In this study, the SWD developed previously for isothermal systems is modified for the first time for a 4-zone TSMB-FC. The optimum operating parameters are determined with the new method. Only linear isotherm systems with significant mass transfer resistances are considered. For a dilute feed, a 4-zone SMB-FC can produce pure desorbent, in addition to extract and raffinate products with high purities. For the separation of p-xylene and toluene with silica gel in the temperature range from 0 to 80 °C, the enrichment factor in the linear region can be as high as 80 fold for an extract purity of 99.99 % and tenfold for raffinate purity of 99.8 %, while withdrawing the desorbent at a desorbent to feed flow rate ratio of 0.53 and 0.78, respectively.  相似文献

A novel pseudo simulated moving bed was suggested to separate a ternary mixture. A solvent gradient was created to make the solvent strength decreasing from zone II to zone III. Under suitable conditions, the least retained solute A moved forward and the most retained solute C moved backward in zones II and III whereas the medium retained solute B moved forward in zone II but backward in zone III to be trapped in the two zones consequently. Once the columns in zones II and III were saturated with solute B, the solvent dissolving the feed was introduced at the feed port to remove solute A from the raffinate-port and solute C from the extract-port. Finally, solute B was recovered from the extract port by stopping the liquid flow in zone II. This scheme was validated by the successful separation of dihydrocapsaicin from capsaicinoids.  相似文献
