A method is proposed for the comprehensive characterization and comparison of columns in the high-performance liquid chromatographic (HPLC) and capillary electrochromatographic (CEC) modes. Using this approach, column parameters such as the number of plates, the eddy-diffusion and mass-transfer contributions to peak broadening, the permeability, and the analysis time are incorporated in a single graph and a comparison in terms of efficiency and speed is obtained. The chromatographic performance of silica-based and polymer-based monolithic capillary columns is discussed and a comparison is made with the performance of packed columns. Also, the potential of ultra-high-pressure liquid chromatography is discussed in this context. In the HPLC mode, the best results were obtained with silica monoliths; in the CEC mode, the low-density methacrylate-ester-based monoliths showed the best performance.  相似文献

In this study, a comparative investigation was performed of HPLC Ascentis® (2.7 μm particles) columns based on fused-core particle technology and Acquity® (1.7 μm particles) columns requiring UPLC instruments, in comparison with Chromolith™ RP-18e columns. The study was carried out on mother and vegetal tinctures of Passiflora incarnata L. on one single or two coupled columns. The fundamental attributions of the chromatographic profiles are evaluated using a chemometric procedure, based on the AutoCovariance Function (ACVF). Different chromatographic systems are compared in terms of their separation parameters, i.e., number of total chemical components (mtot), separation efficiency (σ), peak capacity (nc), overlap degree of peaks and peak purity. The obtained results show the improvements achieved by HPLC columns with narrow size particles in terms of total analysis time and chromatographic efficiency: comparable performance are achieved by Ascentis® (2.7 μm particle) column and Acquity® (1.7 μm particle) column requiring UPLC instruments. The ACVF plot is proposed as a simplified tool describing the chromatographic fingerprint to be used for evaluating and comparing chemical composition of plant extracts by using the parameters D% – relative abundance of the deterministic component – and cEACF – similarity index computed on ACVF.  相似文献

Functionalized monolithic columns were prepared via ring-opening metathesis polymerization (ROMP) within silanized fused silica capillaries with an internal diameter of 200 μm by in situ grafting. This procedure is conducted in two steps, the first of which is the formation of the basic monolithic structure by polymerization of norborn-2-ene (NBE) and 1,4,4a,5,8,8a-hexahydro-1,4,5,8-exo,endo-dimethanonaphthalene (DMN-H6) in a porogenic system (toluene and 2-propanol) using RuCl2(PCy3)2(CHPh) as ROMP initiator. In the second step the still active initiator sites located on the surface of the structure-forming microglobules were used as receptor groups for the attachment (“grafting”) of functional groups onto the monolithic backbone by flushing the monolith with 7-oxanorborn-2-ene-5,6-carboxylic anhydride (ONDCA). Functionalization conditions were first defined that did not damage the backbone of low polymer content (20%) monoliths allowing high-throughput chromatographic separations. Variation of the functionalization conditions was then shown to provide a means of controlling the degree of functionalization and resulting ion-exchange capacity. The maximum level of in situ ONDCA grafting was obtained by a 3 h polymerization in toluene at 40 °C. The weak cation-exchange monoliths obtained provided good separation of a standard peptide mixture comprising four synthetic peptides designed specifically for the evaluation of cation-exchange columns. An equivalent separation was also achieved using the lowest capacity column studied, indicative of a high degree of robustness of the functionalization procedure. As well as demonstrably bearing ionic functional groups enabling analyte separation in the cation-exchange mode, the columns exhibited additional hydrophobic characteristics which influenced the separation process. The functionalized monoliths thus represent useful tools for mixed-mode separations.  相似文献

Several imidazolium-based ionic liquids (ILs) with varying cation alkyl chain length (C4–C10) and anion type (tetrafluoroborate ([BF4]), hexafluorophosphate ([PF6]) and bis(trifluoromethylsulfonyl)imide ([Tf2N])) were used as reaction media in the microwave polymerization of methacrylate-based stationary phases. Scanning electron micrographs and backpressures of poly(butyl methacrylate-ethylene dimethacrylate) (poly(BMA-EDMA)) monoliths synthesized in the presence of these ionic liquids demonstrated that porosity and permeability decreased when cation alkyl chain length and anion hydrophobicity were increased. Performance of these monoliths was assessed for their ability to separate parabens by capillary electrochromatography (CEC). Intra-batch precision (n = 3 columns) for retention time and peak area ranged was 0.80–1.13% and 3.71–4.58%, respectively. In addition, a good repeatability of RSDRetention time = <0.30% and ∼1.0%, RSDPeak area = <1.30% and <4.3%, and RSDEfficiency = <0.6% and <11.5% for intra-day and inter-day, respectively exemplify monolith performance reliability for poly(BMA-EDMA) fabricated using 1-hexyl-3-methylimidazolium tetrafluoroborate ([C6mim][BF4]) porogen. This monolith was also tested for its potential in nanoLC to separate protein digests in gradient mode. ILs as porogens also fabricated different alkyl methacrylate (AMA) (C4–C18) monoliths. Furthermore, employing binary IL porogen mixture such as 1-butyl-3-methylimidazolium tetrafluoroborate ([C4mim][BF4]) and 1-butyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide ([C4mim][Tf2N]) successfully decreased the denseness of the monolith, than when using [C4mim][Tf2N] IL alone, enabling a chromatographic run to be performed with 1:1 ratio produced baseline separation for the analytes. The combination of ILs and microwave irradiation made polymer synthesis very fast (∼10 min), entirely green (organic solvent-free) and energy saving process.  相似文献

We have developed a method of independently tailoring the macro- and mesoporous structures in titania (TiO2) monoliths in order to achieve liquid chromatographic separations of phosphorous-containing compounds. Anatase TiO2 monolithic gels with well-defined bicontinuous macropores and microstructured skeletons are obtained via the sol–gel process in strongly acidic conditions using poly(ethylene oxide) as a phase separator and N-methylformamide as a proton scavenger. Aging treatment of the wet gels in the mother liquor at temperatures of 100–200 °C and subsequent heat treatment at 400 °C allow the formation and control of mesoporous structures with uniform pore size distributions in the gel skeletons, without disturbing the preformed macroporous morphology. The monolithic TiO2 rod columns with bimodal macro–mesoporous structures possess the phospho-sensitivity and exhibit excellent chromatographic separations of phosphorus-containing compounds.  相似文献

Methacrylate-ester-based monoliths containing quaternary ammonium groups were prepared in situ in capillary columns and in simultaneous experiments in vials, employing thermal initiation. The chromatographic properties of the monoliths were determined with capillary electrochromatography (CEC), and their morphology was studied with mercury-intrusion porosimetry on the bulk materials. Materials with different, well repeatable pore-size distributions could be prepared. A satisfactory column-to-column and run-to-run repeatability was obtained for the electro-osmotic mobility, the retention characteristics (k-values) and the efficiency on the columns prepared and tested in the CEC mode. A relatively high electro-osmotic flow was observed in the direction of the positive electrode. The electro-osmotic mobility was found to be influenced only marginally by mobile-phase parameters such as the pH, ionic strength, and acetonitrile content. The retention behavior of the monolithic columns was similar to that of columns packed with C18-modified silica particles. Columns could be prepared with optimum plate heights ranging from 6 microm for unretained compounds to 20 microm for well retained (k=2.5) polyaromatic hydrocarbons. However, for specific analytes a - still unexplained - lower chromatographic column efficiency was observed.  相似文献

The preparation of hexyl methacrylate (HMA) monolithic columns for CEC separations has been investigated with two initiation systems: (i) ammonium peroxodisulphate and TEMED to activate the polymerization reaction, and (ii) by thermal initiation with AIBN. For both initiators, the influence of composition of porogenic solvent on morphological and chromatographic properties of monoliths was investigated. Two porogenic solvent systems, aqueous and non-aqueous media, were also studied for monolithic beds polymerized with AIBN. Under optimal conditions, low minimum plate heights (9.6 mum for peroxodisulphate, 8.4 and 10.0 mum for AIBN in aqueous and non-aqueous porogenic solvents, respectively) were obtained. A comparison in terms of chromatographic performance of HMA monoliths with butyl methacrylate columns polymerized with both initiators was also performed. The resulting HMA-based stationary phases also exhibited a good repeatability and column-to-column reproducibility, with RSD values below 5.6% in the studied electrochromatographic parameters. The potential of HMA-based columns was demonstrated by the analysis of complex mixtures of polyaromatic hydrocarbons and anabolic steroids.  相似文献

Capillary electrochromatography (CEC) has been performed with a series of C14, methlyacrylamide based monolithic columns. These monoliths with different porosities were prepared by in-situ copolymerization in fused-silica capillaries. The porous properties of monoliths were further observed using scanning electron micrographs (SEM) and measured using a mercury porosimeter. The effect of various alcohols as porogens on porous structural properties and chromatographic behaviors were also investigated. The effects of organic additive, pH value and ionic strength in mobile phase on electroosmosis flow (EOF) and separation were further discussed. Meanwhile, the baseline separation of 6 neutral compounds can be well obtained. The distribution ratio of U(IV),D U(IV), between undiluted tri-n-butyl phosphate and aqueous nitric acid has been determined by means of extraction chromatographic columns. The separation factor, β, of U(VI) and U)IV) (defined as the ratio of the distribution ratios,D, \(\beta = \frac{{D_{U(VI)} }}{{D_{U(IV)} }}\) ) was obtained experimentally via the retention volumes.D U(VI) is known from the literature andD U(IV) was calculated. TheD U(IV)-values agree with those obtained by batch experiments, the reproducibility was even better. The stability of U(IV) in the chromatographic columns is good. In the very rare cases, in which U(IV) was oxidized to U(VI) during the experiment, this can be recognized from the shape of the elution curve, because the U(IV) peak shows a tailing. As long as the U(IV) peak is visible, even these experiments can be evaluated which was not possible in batch experiments.  相似文献

Porous polymer monoliths have been prepared in capillaries with circular or square cross-sections and lateral dimensions of 50, 75, 100 μm as well as in a rectangular 38 μm × 95 μm capillary. These capillaries have been used to determine the effect of the size and shape of their cross-section on the porous and hydrodynamic properties of poly(butyl methacrylate-co-ethylene dimethacrylate) monoliths. The capillaries were studied by scanning electron microscopy and evaluated for their permeability to flow and their performance in the liquid chromatographic separation of a protein mixture comprising ribonuclease A, cytochrome c, myoglobin, and ovalbumin using a linear gradient of acetonitrile in the mobile phase. No differences resulting from channel geometry were found for the various capillary columns. These results demonstrate that standard capillaries with circular geometry are a good and affordable alternative conduit for modeling the processes carried out in microfluidic chips with a variety of geometries.  相似文献

A theoretical study on the retention behaviour and chromatographic performance of neutral solutes using a lauryl methacrylate‐based monolithic column under voltage gradient mode in CEC was carried out. Through a flexible mathematical function based on a modified Gaussian model, the peak shape of compounds was firstly fitted under constant and gradient voltage. Using the peak shape parameters and retention time, the estimation of global chromatographic performance, efficiency and peak capacity under several voltage conditions was performed. The influence of voltage gradient on the separation efficiency is discussed and simple equations are presented to calculate retention and peak widths under voltage gradient conditions. A comparison in terms of chromatographic performance of a test mixture of neutral solutes under constant and gradient voltage modes was also carried out. The experiments carried out under gradient voltage showed better efficiencies (172 000 plates/m) and lower peak widths than those obtained under constant voltage (52 000 plates/m).  相似文献

A novel porous polymer monolith was prepared in situ in a fused-silica capillary using photoinitiated polymerization. Bisphenol A dimethacrylate (BPADMA) was selected as a crosslinker, copolymerized with benzyl methacrylate (BMA) in the presence of a binary porogenic solvent consisting of cyclohexanol and 1-decanol in ≤10 min. The resulting poly(BMA-co-BPADMA) monoliths exhibited good permeability and mechanical stability. Mixtures of alkylbenzenes, polycyclic aromatic hydrocarbons (PAHs) or phenolic compounds were successfully separated by CEC. A similar monolith was also prepared with ethylene dimethacrylate (EDMA) as the crosslinker instead of BPADMA to compare the separation ability of the resulting monoliths. The results indicated that poly(BMA-co-BPADMA) monoliths have better selectivity for aromatic analytes and greater chromatographic stability in higher aqueous mobile phase.  相似文献

In this study, high-efficiency LC–MS/MS separations of complex proteolytic digests are demonstrated using 50 mm, 250 mm, and 1 m long poly(styrene-co-divinylbenzene) monolithic capillary columns. The chromatographic performance of the 50 and 250 mm monoliths was compared at the same gradient steepness for gradient durations between 5 and 150 min. The maximum peak capacity of 400 obtained with a 50 mm column, increased to 485 when using the 250 mm long column and scaling the gradient duration according column length. With a 5-fold increase in column length only a 20% increase in peak capacity was observed, which could be explained by the larger macropore size of the 250 mm long monolith. When taking into account the total analysis time, including the dwell time, gradient time and column equilibration time, the 50 mm long monolith yielded better peptide separations than the 250 mm long monolithic column for gradient times below 80 min (nc = 370). For more demanding separation the 250 mm long monolith provided the highest peak production rate and consequently higher sequence coverage. For the analysis of a proteolytic digest of Escherichia coli proteins a monolithic capillary column of 1 m in length was used, yielding a peak capacity of 1038 when applying a 600 min gradient.  相似文献

During the last decade, silica monolithic capillaries have focused more and more attention on miniaturized separation techniques like CEC, nano-LC, and chip electrochromatography owing to their unique chromatographic properties and to their possible in situ synthesis. Nevertheless, the preparation of conventional silica-based individual monolithic columns is time consuming, owing to the individual steps involved, including the synthesis of the silica matrix and its subsequent on-column chemical grafting. The hybrid organic-inorganic monoliths, whose synthesis is based on the polycondensation of siloxane with organosiloxane precursors, seems to be an attractive alternative since their direct synthesis leads to silica monoliths with organic moieties covalently linked to the inorganic silica matrix through hydrolytically stable Si-C bonds. This study describes the synthesis of hybrid monoliths using propyltrimethoxysilane (C3-TriMOS) as a new kind of silica coprecursor to subsequently increase the hydrophobicity of the stationary phase. The influence of several experimental parameters (pH, gelation temperature, relative proportion of the precursors) on the textural (skeleton and macropore size) and chromatographic properties (efficiency, retention, and electroosmotic mobility) of the obtained monoliths are discussed. The results show that the optimal coprecursor incorporation is obtained after a postgelation step during which the condensation of the C3-TriMOS coprecursor is favored by an increase in the pH medium. Thermal hydrolysis of urea previously added to the polymerization mixture allows this in situ pH increase. These hybrid monoliths present hydrophobic properties and allow the separation of test mixtures in the RP mode without any further modification. Moreover, they present excellent efficiencies since reduced plate height as low as 5 and 15 microm are obtained in the electrodriven mode (CEC) and in the hydrodynamic one (nano-LC), respectively.  相似文献

In this paper we report the synthesis of monolithic molecularly imprinted polymers (monolithic MIPs) for the chromatographic separation of Z-phenylalanine by “in situ” polymerisation. Four different functional monomers were used for the preparation and the columns were tested with a standard mobile phase of 1% acetic acid in acetonitrile. 4-Vinylpyridine has shown the best separation performance with a resolution factor R>1 and was chosen for a further improvement of the imprinted stationary phase, which leads to a resolution factor R=2.06. All monoliths investigated have surface areas of around 400 m2 g−1. The columns containing 4-vinylpyridine as functional monomer were characterised by frontal analysis and peak fitting method. These methods allow the determination of the adsorption isotherm which quantify the separation potential of the imprinted monoliths.  相似文献

A gradient kinetic plot method is used for theoretical characterisation of the performance of polymeric particulate anion exchange columns for gradient separations of small inorganic anions. The method employed requires only information obtained from a series of isocratic column performance measurements and in silico predictions of retention time and peak width under gradient conditions. Results obtained under practically constrained conditions provide parameters for the generation of high peak capacities and rapid peak production for fast analysis to be determined. Using this prediction method, a maximum theoretical peak capacity of 84 could be used to achieve separation of 26 components using a 120 min gradient (Rs > 1). This approach provides a highly convenient tool for development of both mono- and multidimensional ion chromatography (IC) methodologies as it yields comprehensive understanding of the influence of gradient slope, analysis time, column length and temperature upon kinetically optimised gradient performance.  相似文献

Peak broadening and skewness are fundamental parameters in chromatography, since they affect the resolution capability of a chromatographic column. A common practice to characterise chromatographic columns is to estimate the efficiency and asymmetry factor for the peaks of one or more solutes eluted at selected experimental conditions. This has the drawback that the extra-column contributions to the peak variance and skewness make the peak shape parameters depend on the retention time. We propose and discuss here the use of several approaches that allow the estimation of global parameters (non-dependent on the retention time) to describe the column performance. The global parameters arise from different linear relationships that can be established between the peak variance, standard deviation, or half-widths with the retention time. Some of them describe exclusively the column contribution to the peak broadening, whereas others consider the extra-column effects also. The estimation of peak skewness was also possible for the approaches based on the half-widths. The proposed approaches were applied to the characterisation of different columns (Spherisorb, Zorbax SB, Zorbax Eclipse, Kromasil, Chromolith, X-Terra and Inertsil), using the chromatographic data obtained for several diuretics and basic drugs (β-blockers).  相似文献

Chiral-modified silica-based monoliths have become well-established stationary phases for both high performance liquid chromatography (HPLC) and capillary electrochromatography (CEC). The silica-based monoliths were fabricated either in situ in the capillaries for nano-HPLC and CEC or in a mould for “conventional” HPLC. The present review summarizes the chiral modification of silica monoliths and the recent development in the field of enantioselective separations by nano-HPLC and CEC.  相似文献

A novel approach is introduced and evaluated for the preparation of silica-based monoliths by a sol–gel technique where in situ polymerization was carried out by γ-ray irradiation within the capillary. The γ-radiation-initiated synthesis generated radicals directly on the monomer, thereby avoiding use of any initiator. The chromatographic behavior of the capillary monolithic columns was studied in the modes of CEC, p-CEC and low pressure-driven separation, all of which exhibited reversed-phase character. Various operational parameters, such as column temperature, separation voltage, acetonitrile content and buffer pH, were varied to assess their influence on column performance in the separation of a series of neutral compounds including thiourea, benzene, toluene, ethyl benzene, biphenyl and naphthalene. A scanning electron micrograph of a cross-section of the capillary column showed that the gel took the form of a spherical particle aggregate and adhered to the column inner wall. It provided a viable alternative to either thermally initiated or photo polymerization for the preparation of monolithic columns.  相似文献
