The total oil content and composition of fatty acids and phytosterols of five Transylvanian (Romania) pomaces of wild and cultivated blueberries (Vaccinium myrtillus), wild cowberry (Vaccinium vitis-idaea) and raspberry (Rubus idaeus), and cultivated black chokeberry (Aronia melanocarpa), were determined by capillary gas chromatography. Out of the five pomace oils, the percentages of polyunsaturated fatty acids (PUFAs) ranged from 37 % to 69 %. The lipid classes analysed (PLs ?? polar lipids, TGs ?? triacylglycerols, SEs ?? sterol esters) were separated and identified using thin-layer chromatography. TGs showed the highest PUFAs content (ranging from 41.9 % to 72.5 %) and PUFAs/SFAs (saturated fatty acids) ratios (in the range of 5.8?C33.1 %). In the case of PL and SE fractions, the levels of SFA were significantly higher than in TGs. The total amount of sterols was in the range of 101.6?C168.2 mg per 100 g of lipids of the pomaces analysed. The predominant phytosterols were ??-sitosterol, stigmastanol + isofucosterol, and campesterol. The results indicated that the investigated pomace oils, due to their good balance between n-6 and n-3 fatty acids (except for chokeberry) and high ??-sitosterol content, could be excellent sources of PUFAs and phytosterols, thus suggesting potential value-added utilisation of berry waste oils for preparing functional foods or food supplements.  相似文献

Microalgae is a rich source of polyunsaturated fatty acid. This study was conducted to identify and isolate microalgal strain with the potentials for producing polyunsaturated fatty acids (PUFAs) and determine its cytotoxic effect on some cancer cells. The algal strain (Chlorella sp. S14) was cultivated using modified BG-11 media, and algal biomass obtained was used for fatty acid extraction. Gas chromatographic–mass spectrometry was used to identify and quantify the levels of the fatty acid constituents. The total content of monounsaturated fatty acids (1.12%) was low compared to polyunsaturated fatty acids (PUFAs) (52.87%). Furthermore, n-3 PUFAs accounted for (12.37%) of total PUFAs with the presence of α-linolenic acid (2.16%) and cis-11,14,17-eicosatrienoic acid (2.16%). The PUFA-rich extract did not exhibit a cytotoxic effect on normal cells. Treatment with the PUFA-rich extract (150 µg/mL) significantly reduced cell viability in MCF-7 (31.58%) and A549 (62.56%) cells after the 48 h treatment. Furthermore, treatment of MCF-7 with fatty acid extracts (125 and 150 µg/mL) showed a significant reduction in MDA levels, increase in catalase activities and decrease in GSH level compared to untreated cells. However, a slight decrease in MDA level was observed in A549 cells after the 48 h treatment. There are no significant changes in catalase activities and GSH level in treated A549 cells. However, a slight reduction of NO levels was observed in treated MCF-7 and A549 cells. These results indicate the potentials of PUFA-rich extracts from Chlorella sp. S14 to reduce viability and modulate redox status in A549 and MCF-7 cells.  相似文献

To understand more fully the effect of polyunsaturated fatty acids (PUFAs) on lipid bilayers, we investigated the effects of treatment with fatty acids on the properties of a model membrane. Three kinds of liposomes comprising dipalmitoylphosphatidylcholine (DPPC), dioleylphosphatidylcholine (DOPC), and cholesterol (Ch) were used as the model membrane, and the fluorescence anisotropy of 1,6-diphenyl-1,3,5-hexatriene (DPH) and detergent insolubility were determined. Characterization of the liposomes clarified that DPPC, DPPC/Ch, and DPPC/DOPC/Ch existed as solid-ordered phase (L beta), liquid-ordered phase (l o), and a mixture of l o and liquid-disordered phase (L alpha) membranes at room temperature. Treatment with unsaturated fatty acids such as oleic acid (OA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) markedly decreased the fluorescence anisotropy value and detergent insolubility. PUFAs and OA had different effects on the model membranes. In DPPC liposomes, the most prominent change was induced by PUFAs, whereas, in DPPC/Ch and DPPC/DOPC/Ch liposomes, OA had a stronger effect than PUFAs. The effect of PUFAs was strongly affected by the amount of Ch in the membrane, which confirmed a specific effect of PUFAs on the Ch-poor membrane domain. We further explored the effect of fatty acids dispersed in a water-in-oil-in-water multiple emulsion and found that unsaturated fatty acids acted on the membranes even when incorporated in emulsion form. These findings suggest that treatment with PUFAs increases the segregation of ordered and disordered phase domains in membranes.  相似文献

Free radicals with reactive chemical properties can fight tumors without causing drug resistance. Reactive oxygen species (ROS) has been widely used for cancer treatment, but regrettably, the common O2 and H2O2 deficiency in tumors sets a severe barrier for sufficient ROS production, leading to unsatisfactory anticancer outcomes. Here, we construct a chlorine radical (.Cl) nano-generator with SiO2-coated upconversion nanoparticles (UCNPs) on the inside and Ag0/AgCl hetero-dots on the outside. Upon near-infrared (NIR) light irradiation, the short-wavelength emission UCNP catalyzes .Cl generation from Ag0/AgCl with no dependence on O2/H2O2. .Cl with strong oxidizing capacity and nucleophilicity can attack biomolecules in cancer cells more effectively than ROS. This .Cl stress treatment will no doubt broaden the family of oxidative stress-induced antitumor strategies by using non-oxygen free radicals, which is significant in the development of new anticancer agents.  相似文献

Hens?? eggs represent a rich source of important nutrients, including lipids and carotenoids. The lipid composition of hens?? eggs is influenced by genetic factors, age, and diet. The aim of this study was to compare the fatty acids, cholesterol, and carotenoids content of the egg yolk of ISA Brown and Araucana hens grown in free-range housing systems. Fatty acids and cholesterol were analysed by GC-FID and GC-MS and carotenoids were quantified by RP-HPLC-PDA. The Araucana egg yolk has a higher lipid content and higher egg-to-albumen ratio than the ISA Brown yolk, while the total cholesterol, carotenoids content and profile are not significantly different. The lipids of the Araucana egg yolk have a higher content of mono-unsaturated fatty acids (MUFAs), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA) and a better n-6/n-3 ratio than the ISA Brown egg yolk lipids. The major carotenoids were lutein and zeaxanthin, which account for more than 83 % in egg yolk. Eggs of both breeds, when raised organically, represent very good sources of highly bio-available lutein and zeaxanthin, pigments which are related to lower risk of age-related macular degeneration. We report for the first time on the fatty acids composition in lipid fractions and the profile and content of carotenoids of the Araucana egg yolk.  相似文献

This work reports for the first time the nutritional profile and antioxidant potential of the edible sea cucumber Holothuria arguinensis from the North-eastern Atlantic. H. arguinensis has high levels of protein, with the amino acids profile dominated by alanine, glycine and proline and low lysine/arginine ratios. Its carbohydrate and energetic contents are also low as well as the total lipid levels, although its lipid profile is rich in polyunsaturated fatty acids (PUFA), especially arachidonic, eicosapentaenoic and docosahexaenoic acids. In addition, H. arguinensis has high levels of calcium. The water and ethanol extracts show ability to scavenge free radicals and to chelate copper and iron ions. Our results indicate that H. arguinensis has a balanced nutritional quality suitable for human consumption. In addition, it contains compounds with antioxidant potential; thus its intake can contribute for a healthy and well-balanced diet.  相似文献

A proton‐coupled electron transfer (PCET) process plays an important role in the initial step of lipoxygenases to produce lipid radicals which can be oxygenated by reaction with O2 to yield the hydroperoxides stereoselectively. The EPR spectroscopic detection of free lipid radicals and the oxygenated radicals (peroxyl radicals) together with the analysis of the EPR spectra has revealed the origin of the stereo‐ and regiochemistry of the reaction between O2 and linoleyl (= (2Z)‐10‐carboxy‐1‐[(1Z)‐hept‐1‐enyl]dec‐2‐enyl) radical in lipoxygenases. The direct determination of the absolute rates of H‐atom‐transfer reactions from a series of unsaturated fatty acids to the cumylperoxyl (= (1‐methyl‐1‐phenylethyl)dioxy) radical by use of time‐resolved EPR at low temperatures together with detailed kinetic investigations on both photoinduced and thermal electron‐transfer oxidation of unsaturated fatty acids provides the solid energetic basis for the postulated PCET process in lipoxygenases. A strong interaction between linoleic acid (= (9Z,12Z)‐octadeca‐9,12‐dienoic acid) and the reactive center of the lipoxygenases (FeIII? OH) is suggested to be involved to make a PCET process to occur efficiently, when an inner‐sphere electron transfer from linoleic acid to the FeIII state is strongly coupled with the proton transfer to the OH group.  相似文献

Lipids being hydrophobic or amphiphilic can be encapsulated by cyclodextrin complexation. Among the various groups of lipids cholesterol, fatty acids, phospholipids and sphingolipids are overviewed concerning the structural requirements for both the lipid and the cyclodextrin component of the complexes. The chain length and the number and position of the double bonds in the fatty acids, the polarity of the head-group in the phospholipids and sphingolipids are important factors. Concerning the cyclodextrins, in addition to the most crucial cavity size also the chemical microenvironment of cavity entrances determine the interaction with lipids. While fatty acids, phospholipids and sphingolipids prefer the alpha-cyclodextrin cavity, cholesterol is complexed first of all by the beta-cyclodextrin and its derivatives. Methylated beta-cyclodextrin has extreme affinity to all of these lipids, which are common constituents of cell membranes. Based on the knowledge on the specific cyclodextrin-lipid interactions, cyclodextrin derivatives are able to selectively remove certain lipid components from model and biological membranes and can be selected making possible to modulate the lipid profile in such membranes.

The in vivo metabolism of plasma lipids generates lipid hydroperoxides that, upon one‐electron reduction, give rise to a wide spectrum of genotoxic unsaturated aldehydes and epoxides. These metabolites react with cellular DNA to form a variety of pre‐mutagenic DNA lesions. The mechanisms of action of the radical precursors of these genotoxic electrophiles are poorly understood. In this work we investigated the nature of DNA products formed by a one‐electron reduction of (13S)‐hydroperoxy‐(9Z,11E)‐octadecadienoic acid (13S‐HPODE), a typical lipid molecule, and the reactions of the free radicals thus generated with neutral guanine radicals, G(?H).. A novel approach was devised to generate these intermediates in solution. The two‐photon‐induced ionization of 2‐aminopurine (2AP) within the 2′‐deoxyoligonucleotide 5′‐d(CC[2AP]TCGCTACC) by intense nanosecond 308 nm excimer laser pulses was employed to simultaneously generate hydrated electrons and radical cations 2AP.+. The latter radicals either in cationic or neutral forms, rapidly oxidize the nearby G base to form G(?H).. In deoxygenated buffer solutions (pH 7.5), the hydrated electrons rapidly reduce 13S‐HPODE and the highly unstable alkoxyl radicals formed undergo a prompt β‐scission to pentyl radicals that readily combine with G(?H).. Two novel guanine products in these oligonucleotides, 8‐pentyl‐ and N2‐pentylguanine, were identified. It is shown that the DNA secondary structure significantly affects the ratio of 8‐pentyl‐ and N2‐pentylguanine lesions that changes from 0.9:1 in single‐stranded, to 1:0.2 in double‐stranded oligonucleotides. The alkylation of guanine by alkyl radicals derived from lipid hydroperoxides might contribute to the genotoxic modification of cellular DNA under hypoxic conditions. Thus, further research is warranted on the detection of pentylguanine lesions and other alkylguanines in vivo.  相似文献

Fish oil is considered a healthy food due to the presence of large amounts of polyunsaturated fatty acids (PUFAs), especially in the form of n-3 fatty acids 5,8,11,14,17-eicosapentaenoic acid (20:5n-3; EPA) and 4,7,10,13,16,19-docosahexaenoic acid (22:6n-3; DHA). However, fish oil is known to contain many other PUFAs, some of which are uncommon and whose bioactivity is scarcely investigated. In this study, we isolated the rare PUFA 6,9,12,15-hexadecatetraenoic fatty acid (16:4n-1) which bears a double bond on the terminal carbon from fish oil in form of its methyl ester. We used high-speed counter-current chromatography (HSCCC) for the fractionation of 500 mg-portions of fatty acid methyl esters prepared from a fish oil capsule and investigated the fractions by GC/MS. Twenty-eight 13-mL fractions were collected and fatty acid methyl esters were detected in fractions 11–23. The elution was carried out in normal phase mode, providing the long-chained saturated and monoenoic fatty acids first. More than 100 fatty acids ranging from 10:0 to 26:0 could be identified in the HSCCC fractions, and most of them were polyunsaturated. The reproducibility of the HSCCC method was shown by repeated injection of the fish oil and the fractions containing 6,9,12,15-hexadecatetraenoic fatty acid (16:4n-1). The late eluting 16:4n-1 methyl ester was isolated in pure form and its structure was verified.

Combining desorption ionization with tandem mass spectrometry overcomes the disadvantage of limited fragmentation accompanying desorption and permits mixtures of closely related substances to be investigated directly. These features of the combination are illustrated by completing the structure-proof of a minor component of an ornithine-containing lipid mixture isolated from Thiobacillus thiooxidans. The minor component is a homolog of the major constituent and differs from the principal component owing to the presence of a double bond in lieu of a cyclopropyl ring in one of the constituent fatty acids. Another feature of the combined method is the potentially complementary nature of collision-activated dissociation spectra of protonated and cationized biomolecules. This is illustrated by the differences in the collision-activated dissociations of the [M + Na]+ of sucrose, desorbed by field desorption, and [M + H]+, desorbed by fast atom bombardment. A third illustration is the application of field desorption and tandem mass spectrometry to an organometallic compound. The combined approach allows the ligands to be identified and the relative ligand binding energies to be approximated.  相似文献

In order to better understand the metabolic fate of n-3 polyunsaturated fatty acids (PUFAs), an efficient access to symmetrical and unsymmetrical triacylglycerols (TGs), esterified with PUFAs, with known high purity, is required. In this context, we optimized the esterification of a mixture of glycerols protected as dioxane and dioxolane with PUFAs. The kinetics of this reaction depends on various factors, such as the fatty acid chain length and the stereochemistry of the dioxane. Then, one-pot acetal hydrolysis and esterification of hydroxyl groups led to the desired structured TGs without either double bond isomerization or acyl migration (except when symmetrical TGs are acylated with long-chain saturated fatty acids in external positions). PUFAs location on the glycerol backbone was assayed by NMR, HPLC and pancreatic lipase hydrolysis.  相似文献

The decomposition of unsaturated fatty acids and tocopherols in 10 commercial edible oils during deep frying was investigated. The dominant tocopherol in oils rich in polyunsaturated fatty acids (PUFAs) was γ-tocopherol, except for natural perilla oil (δ-tocopherol dominant), and the main tocopherol in oleic acid-rich oils was α-tocopherol. The PUFA-rich oils had higher tocopherol contents than the oleic acid-rich oils. Both the reduction rate of total unsaturated fatty acid (TUFA) and total tocopherol (TToc) were linear with frying time (t). The decomposition rate of TToc is faster than that of TUFA since the slope values obtained from fitting equations (Y?=?k t) kTToc (1.520–14.483) were obviously larger than for kTUFA (0.155–0.270). By establishing a dynamic decomposition index, unsaturated fatty acids and tocopherol in oils showed dynamic decomposition over multiple frying cycles. The obtained results showed that decomposition characteristics of oils are related to their fatty acid compositions.  相似文献

In the investigation of peroxyl radicals the pulse radiolysis technique can be used with some advantage to determine the rate of their unimolecular or bimolecular decay. If the identities of the products of the peroxyl radical reactions are known, pulse radiolysis often provides evidence for mechanistic details. The absorptions of the peroxyl radicals are neither very specific nor strong and optical detection is usually of little help. However, there are many peroxyl radical reactions which result in the formation of HO 2 . /H+O 2 . (pKa(HO 2 . )=4.7) or other acids. Thus in neutral and alkaline solutions such species can be monitored even quantitatively by the pulse conductometric method. Furthermore, O 2 . can be detected by its rapid reaction with tetranitromethane which yields the strongly absorbing nitroform anion. Since O 2 . is only a short-lived intermediate in neutral solutions, it can be distinguished from permanent acids which are often formed in peroxyl radical reactions. In alkaline solutions, where O 2 . is more stable, superoxide dismutase might be used with advantage to reduce its lifetime and to determine the yield of permanent acids. Some details of the fate of the peroxyl radicals derived from acetate, the -hydroxyethyl-peroxyl radicals, and the cyclopentylperoxyl radicals will be reviewed.  相似文献

The gallium ion (Ga3+) has long been believed to disrupt ferric homeostasis in the body by competing with iron cofactors in metalloproteins, ultimately leading to cell death. This study revealed that through an indirect pathway, gallium can trigger ferroptosis, a type of non-apoptotic cell death regulated by iron. This is exemplified by the gallium complex of the salen ligand ( Ga-1 ); we found that Ga-1 acts as an effective anion transporter that can affect the pH gradient and change membrane permeability, leading to mitochondrial dysfunction and the release of ferrous iron from the electron transfer chain (ETC). In addition, Ga-1 also targeted protein disulfide isomerases (PDIs) located in the endoplasmic reticulum (ER) membrane, preventing the repair of the antioxidant glutathione (GSH) system and thus enforcing ferroptosis. Finally, a combination treatment of Ga-1 and dietary polyunsaturated fatty acids (PUFAs), which enhances lipid peroxidation during ferroptosis, showed a synergistic therapeutic effect both in vitro and in vivo. This study provided us with a strategy to synergistically induce Ferroptosis in tumor cells, thereby enhancing the anti-neoplastic effect.  相似文献

Wild edible mushrooms are considered as a cheap food source, but rich in bioactive compounds, especially in phenolic compounds. The purpose of the study was to determine the antioxidant and antimicrobial activity and the content of polyphenolcarboxylic acids in 10 species of mushrooms commonly used (consumed) in Romania and two controls. The effect against free radicals, lipid peroxidation and reducing power were determined. The antimicrobial effect was revealed on some strains with pathogenic effect by disk diffusion assay. The antioxidant capacity expressed in vitro was correlated both with the presence of the main polyphenolcarboxylic acids, and also with the presence of other bioactive molecules (flavonoids, carotenoid compounds, etc.).  相似文献

Densities of solutions of oleic, linoleic, and linolenic acids in n-hexane and n-heptane were measured using a vibrating-tube densimeter at 298.15 K in a concentration range of 0–0.012 molar fractions of solute. The measurement error does not exceed ±5·10−6 g cm−3. The limiting partial molar volumes of fatty acids of the studied series in n-hexane and n-heptane and the excess volume properties of binary mixtures were calculated. On going from oleic to linolenic acid, the number of double bonds (>C=C<) in a solute molecule increases, the hydrocarbon chain length in a solvent molecule decreases, and compactness of the structure packing of the resulting solution increases. This is caused, as a whole, by the enhancement of the n-alkane—acid intermolecular interaction. Published in Russian in Izvestiya Akademii Nauk. A new electrochemical assay has been developed which is based on quantifying the attack of OH. radicals on a self‐assembled thiol monolayer on electrodes in the presence and absence of antioxidants. The OH. radicals were produced in Fenton solutions. The radical scavengers diminish the concentration of the OH. radicals in the Fenton solution, and the extent of thiol layer destruction is monitored with the help of a redox probe in solution, the signal of which is only generated at SAM‐free electrode surface. The results of the electrochemical assay were compared with those of the standard DPPH. assay. In case of each plant extract, an excellent linear correlation was observed between the radical scavenging activity and the concentration of plant extracts; however, the scavenging properties of different plants strongly differ in the electrochemical and the DPPH. assay. Since the proposed new method probes the reactivity of radical scavengers towards OH., one of the most important reactive oxygen radicals in tissues, it is supposed to provide much more relevant information than the DPPH. or other assays.  相似文献
