A method using automated on-line solid-phase extraction (SPE) liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS) for the determination of 8-iso-PGF2α in human exhaled breath condensate (EBC) was developed and validated. A C18 SPE column with an affinity sorbent was used for on-line extraction. A C18 column was employed for LC separation and ESI-MS/MS was utilized for detection. 8-iso-PGF2α-d4 was used as an internal standard for quantitative determination. The extraction, cleanup and analysis procedures were controlled by a fully automated six-port switch valve. Identification and quantification were based on the following transitions: m/z 353 → 193 for 8-iso-PGF2α and m/z 357 → 197 for 8-iso-PGF2α-d4, respectively. Good recoveries from 98.94 to 99.86% were measured and satisfactory linear ranges for these analytical compounds were determined. Intra-day and inter-day precision showed that coefficients of variance (CV) ranged from 6.5 to 8.0% and 5.2 to 6.3%, respectively. The applicability of this newly developed method was demonstrated by analyzing human EBC samples for an evaluation of the future risk of human exposure to nanoparticles.  相似文献

Contamination of the exterior surface of vials of cytostatic drugs by the drugs themselves is a potential hazard to human health. This study developed a validated method using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) for the determination of contamination of the exteriors of vials of cisplatin and carboplatin. Large Alpha® sampling swabs were employed to wipe the vial exterior. Cisplatin or carboplatin and gold(III) as an internal standard were derivatized by N,N-diethyldithiocarbamate (DDTC). Pt(DDTC)3+ and Au(DDTC)2+ were monitored by the respective transitions of m/z 639.3-490.9 and 493.0-345.0, respectively. Each separation was completed within 9 min using a 3 μm particle ODS-column. Calibration curves for cisplatin and carboplatin were linear over concentration ranges of 30-10,000 and 30-30,000 pg vial−1, respectively. The accuracies and precisions were 96.1-102.5% and within 8.2% for intra-assay and 99.6-103.3% and within 7.6% for inter-assay, respectively. Their lower limit of quantification was 30 pg vial−1. Amounts of 0.17-17.0 ng vial−1 as cisplatin and 0.48-794 ng vial−1 as carboplatin were detected from the exterior surface of the vials. This validated method using LC-ESI-MS/MS for the determination of platinum anticancer drugs is helpful for monitoring contamination of the exterior surface of drug vials.  相似文献

A new simple, sensitive and selective liquid chromatography coupled with mass spectrometry (LC/MS) method for quantification of captopril after precolumn derivatization with p-bromo-phenacyl-bromide in human plasma was validated. Plasma samples were analysed on a monolithic column (Cromolith Performance-RP 18e, 100 mm × 4.6 mm I.D., 3 μm) under isocratic conditions using a mobile phase of a 40:60 (v/v) mixture of acetonitrile and 0.1% (v/v) formic acid in water. The flow rate was 1 mL/min at the column temperature of 30 °C. In these chromatographic conditions, the retention time was 4.4 min for captopril derivative. The detection of the analyte was in MRM mode using an ion trap mass spectrometer with electrospray positive ionisation. The monitored ions were 216, 253, 255, 268, 270 m/z derived from 415 m/z for derivatized captopril. The sample preparation was very simple and consisted in plasma protein precipitation from 0.2 mL plasma using 0.3 mL methanol after the derivatization reaction was completed. Calibration curves were generated over the range of 10-3000 ng/mL with values for coefficient of correlation greater than 0.993 and by using a weighted (1/y2) quadratic regression. The values for precision (CV %) and accuracy (relative error %) at quantification limit were less than 9.9% and 3.9%, for within- and between-run, respectively. The mean recovery of the analyte was 99%. Derivatized samples demonstrated good short-term, long-term, post-preparative and freeze-thaw stability. This is the first reported LC-MS/MS method for analysis of captopril in human plasma that uses protein precipitation as sample processing procedure. The method is very simple and allows obtaining a very good recovery of the analyte. The validated LC-MS/MS method has been applied to a pharmacokinetic study of 50 mg captopril tablets on healthy volunteers.  相似文献

An alternative method for the enantiomer specific determination of hexabromocyclododecanes (HBCD) by LC-ESI-MS/MS using an ion trap analyser is proposed. The method is based on the formation of a chlorine adduct (m/z 676.6) of the (±)α-, (±)β-, and (±)γ-HBCD enantiomers and their further fragmentation into their stable quasi-molecular ion (m/z 640.6). In this way, problems related to the ion trap low mass cutoff and variable amounts of other adduct peaks in the samples are solved. Parameters affecting separation, ionisation and MS/MS detection were studied. Method performance was also evaluated: calibration curves were found linear from 20 to 400 pg μL−1 for each enantiomer; detection limits ranged between 1.5 and 4.3 pg μL−1; repeatability and reproducibility, expressed as relative standard deviation, were lower than 6% and 13%, respectively. The application to different types of spiked samples (pork meat, lean fish, and butter) pointed out the occurrence of matrix effects that could be solved by using labelled standards.  相似文献

A simple, sensitive LC-ESI-MS method was optimized for quantitative analysis of octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX) in environmental samples. Under negative ionization mode, HMX can form adduct ions with various organic acids and salts, including acetic acid, formic acid, propionic acid, ammonium nitrate, ammonium chloride, sodium nitrite, and sodium nitrate. Acetic acid was chosen as additive and the ion, [M + CH3COO] with m/z = 355 was used for selective ion monitoring (SIM) in this study. Good sensitivity was achieved with low acetic acid concentration in the mobile phase and relatively low capillary temperature. The method detection limit was 0.78 pg for HMX in standard solution. Linearity (R2 > 0.9998) was obtained at low concentrations (0.5-50 μg/L). This method has been used to determine HMX concentrations in water samples and lizard egg samples from an animal exposure study.  相似文献

A new IC-ESI-MS/MS method, with simple sample preparation procedure, has been developed for quantification and confirmation of perchlorate (ClO4) anions in water, fresh and canned food, wine and beer samples at low part-per-trillion (ng l−1) levels. To the best of our knowledge, this is the first time an analytical method is used for determination of perchlorate in wine and beer samples. The IC-ESI-MS/MS instrumentation consisted of an ICS-2500 ion chromatography (IC) system coupled to either an API 2000™ or an API 3200™ mass spectrometer. The IC-ESI-MS/MS system was optimized to monitor two pairs of precursor and fragment ion transitions, i.e., multiple reaction monitoring (MRM). All samples had oxygen-18 isotope labeled perchlorate internal standard (ISTD) added prior to extraction. Chlorine isotope ratio (35Cl/37Cl) was used as a confirmation tool. The transition of 35Cl16O4 (m/z 98.9) into 35Cl16O3 (m/z 82.9) was monitored for quantifying the main analyte; the transition of 37Cl16O4 (m/z 100.9) into 37Cl16O3 (m/z 84.9) was monitored for examining a proper isotopic abundance ratio of 35Cl/37Cl; and the transition of 35Cl18O4 (m/z 107.0) into 35Cl18O3 (m/z 89.0) was monitored for quantifying the internal standard. The minimum detection limit (MDL) for this method in de-ionized water is 5 ng l−1 (ppt) using the API 2000™ mass spectrometer and 0.5 ng l−1 using the API 3200™ mass spectrometer. Over 350 food and beverage samples were analyzed mostly in triplicate. Except for four, all samples were found to contain measurable amounts of perchlorate. The levels found ranged from 5 ng l−1 to 463.5 ± 6.36 μg kg−1 using MRM 98.9 → 82.9 and 100 μl injection.  相似文献

This paper describes a novel method that combines dansyl chloride (DNS-CL) derivatization with high-performance liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI/MS/MS) for the sensitive and selective determination of piperazine phosphate in human plasma. After addition of ondansetron hydrochloride as internal standard (IS), piperazine phosphate was derivatized and then extracted with ethyl acetate. After being evaporated and reconstituted, the sample was analyzed using LC-ESI/MS/MS. Separation was achieved using an Agilent ZORBAX SB-C18 (150 mm × 2.1 mm I.D., 3.5 μm) column and isocratic elution with 10 mM ammonium acetate solution (pH 3.0)-methanol (50: 50, v/v). Detection was performed on a triple-quadrupole mass spectrometer utilizing electrospray ionization (ESI) interface operating in positive ion and selected reaction monitoring (SRM) mode with the precursor to product ion transitions m/z 320 → 171 for DNS-CL-piperazine phosphate and m/z 294 → 170 for the IS. The method was fully validated for its selectivity, sensitivity, linearity, precision, accuracy, recovery, matrix effect and stability. The coefficient (r) of piperazine phosphate with a linear range of 0.1-15 μg mL−1 was 0.9974-0.9995. The limit of detection and lower limit of quantification in human plasma were 0.01 and 0.1 μg mL−1, respectively. The validated LC-ESI/MS/MS method has been successfully applied to a bioequivalence study of piperazine phosphate trochiscus in Chinese healthy male volunteers.  相似文献

A high sensitive method for determination of cyclamate in foods by ion-pair high-performance liquid chromatography-electrospray ionization mass spectrometry was developed and validated. The separation was achieved on a C8 column with 5 mM tris(hydroxymethyl)aminomethane aqueous solution (pH 4.5, adjusted by acetic acid) as mobile phase with an isocratic mode. The quantification of target compound was completed using a selected ion recording (SIR) at m/z 178 obtained from ESI-mode. Tiopronin was used as internal standard for the quantification of cyclamate. The correlation coefficient of the calibration curve were better than 0.996, in the range of 50-5000 ng/mL. The limit of detection is 5 ng/mL, the limit of quantification is 20 ng/mL. The inter- and intra-day accuracy, precision were investigated in detail. The method can be used to monitor effectively the content of the artificial sweetener in foods. The method has obvious merits such as high sensitivity, specificity and simply versus other methods reported.  相似文献

Recently, cobalt (Co) is reported to be taken as a supplement by athletes for improving anaerobic performance. For the diagnosis of abuse, the limit of detection (LOD) of Co in the analysis should be lower than the concentrations of Co in plasma and urine of normal persons. A simple, rapid and sensitive method has been developed for the determination of Co in urine. Co was complexed with diethyldithiocarbamate (DDC) and extracted with isoamyl alcohol in the presence of citric acid. The detection of Co was achieved by injecting a 1-μL aliquot of isoamyl alcohol containing Co-DDC complex directly into an electrospray ionization tandem mass spectrometric (ESI-MS-MS) instrument without chromatographic separation. The quantification was performed using selected reaction monitoring at m/z 291 of the product ion Co(C4H10NCS)2+ which was produced by collision-induced dissociation from the precursor ion Co(DDC)2+ at m/z 355. ESI-MS-MS data were obtained in less than 10 min with an LOD of 0.05 μg L−1 and a linear calibration range of 0.1-100 μg L−1 using 10 μL of urine. The procedure was validated with certified reference materials (SRM 2670a and SRM 1643e). This method is suitable for the analysis of Co in the laboratories already equipped with an ESI-MS-MS instrument.  相似文献

19-Nortestosterone (nandrolone) major metabolites in human urine are excreted as sulfoconjugated and glucuroconjugated forms. A sensitive and selective liquid chromatography/tandem mass spectrometry (LC/MS/MS) method in negative ESI mode was developed for direct quantification of 19-norandrosterone sulfate (19-NAS) and 19-noretiocholanolone sulfate (19-NES). For both sulfoconjugates, the [M−H] ion at m/z 355 and the fragment ion at m/z 97 were used as the precursor and product ions, respectively. The purification method involved a complete and rapid separation of sulfates and glucuronides in two extracts after loading the sample on a weak anion exchange solid phase extraction support (SPE Oasis® WAX). Then, sulfates were separated by LC (Uptisphere® ODB, 150 mm × 3.0 mm, 5 μm) and analyzed on a linear trap and a triple quadrupole mass spectrometer. The lower limit of detection (LLOD) and lowest limit of quantification (LLOQ) were of 100 pg mL−1 and 1 ng mL−1, respectively. Assay validation demonstrated good performances in terms of trueness (92.0-104.9%), repeatability (0.6-7.2%) and intermediate precision (1.3-10.8%) over the range of 1-2500 ng mL−1. Finally, 19-NAS and 19-NES in urine samples collected after intake of 19-norandrostenedione (nandrolone precursor) were quantified. This assay may be easily implemented to separate glucuronide and sulfate steroids from urine specimens prior to quantification by LC/MS/MS.  相似文献

A mass spectrometric (MS) method for the identification of iron protoporphyrin (IX) (FePTP, heme b) in marine particulate material and phytoplankton is described. Electrospray ionisation of FePTP produced the molecular Fe(III)PTP+ ion (m/z = 616) or the pseudomolecular [Fe(II)PTP + H]+ ion (m/= 617), depending on the oxidation state of the central iron ion. Collision induced dissociation (CID) in the ion trap mass spectrometer resulted in a single detected product ion (m/z = 557) indicative of loss of ethanoic acid from a carboxylic acid side chain. Widening the isolation width to 616 ± 3 resulted in production of a mass spectrum demonstrating the distinctive isotopic ratio of the iron containing fragment, further increasing the specificity of the analysis. Selective reactant monitoring (SRM) of the fragment ion (m/z = 557) was applied to the detection of FePTP after chromatography of ammoniacal OGP extracts of marine samples. The detection limit for FePTP analysed by SRM after chromatography was 1.2 ± 0.5 fmol. For phytoplankton samples, reasonably good agreement was achieved between results obtained with SRM and those obtained by monitoring absorbance at λ = 400 nm using a diode array detector (DAD). Use of SRM for analysis of particulate material obtained from the high latitude North Atlantic allowed for the analysis of FePTP in the presence of a co-eluting compound that interfered with detection by DAD. Simultaneous collection of mass spectra from m/z = 300 to 1500 resulted in identification of the pseudomolecular ion for the interfering compound. The CID fragmentation pattern and UV–visible mass spectra indicated that the interfering compound was a previously unidentified chlorin type compound. Comparison of FePTP determined by SRM and DAD on samples where this compound could not be detected showed that results collected using the two methods correlated. The use of both MS and DAD results in a powerful tool for quantifying this important biogenic component of the particulate iron pool.  相似文献

A simple, rapid, and sensitive high-performance liquid chromatography (HPLC)-electrospray ionization (ESI) tandem mass spectrometric method (LC-MS/MS) has been developed for simultaneous determination of cilazapril levels and its active metabolite, cilazaprilat, in human plasma using enalapril as internal standard. The acquisition was performed in the multiple reaction monitoring mode; monitoring the transitions: m/z 418.4 > 211.1 for cilazapril and m/z 390.3 > 211.1 for cilazaprilat. The method involves a simple single-step liquid-liquid extraction with ethyl acetate. The analyte was chromatographed on an YMC C8 reversed-phase chromatographic column by isocratic elution with 10 mM ammonium formate buffer-methanol (10:90, v/v; pH 3.2 with formic acid). Numerous compounds did not interfere with specific multiple reaction monitoring in tandem mass spectrometric detection following C8 reversed-phase chromatographic separation under conditions that eluted cilazapril, cilazaprilat, and enalapril within 2 min. This method was validated over 0.1-500 ng ml−1 of cilazapril and 0.5-50 ng ml−1 of cilazaprilat. Cilazapril and cilazaprilat were stable in standard solution and in plasma samples under typical storage and processing conditions. The assay was successfully applied to a pharmacokinetic study of cilazapril given as a single oral dose (5 mg) to healthy volunteers.  相似文献

An analytical method for the determination of US EPA priority pollutant 16 polycyclic aromatic hydrocarbons (PAHs) in edible oil was developed by an isotope dilution gas chromatography–mass spectrometry (GC–MS). Extraction was performed with ultrasonication mode using acetonitrile as solvent, and subsequent clean-up was applied using narrow gel permeation chromatographic column. Three deuterated PAHs surrogate standards were used as internal standards for quantification and analytical quality control. The limits of quantification (LOQs) were globally below 0.5 ng/g, the recoveries were in the range of 81–96%, and the relative standard deviations (RSDs) were lower than 20%. Further trueness assessment of the method was also verified through participation in international cocoa butter proficiency test (T0638) organised by the FAPAS with excellent results in 2008. The results obtained with the described method were satisfying (z ≤ 2). The method has been applied to determine PAH in real edible oil samples.  相似文献

A novel analytical method was proposed for the rapidly simultaneous determination of nine mono-chlorophenols (MCPs) and di-chlorophenols (DCPs) in water samples using eluent generator ion chromatography (IC) coupled with an atmospheric pressure chemical ionization mass spectrometry (APCI-MS) in the negative mode. The IC separation was carried out on an IonPac® AS11 analytical column (250 mm × 4.0 mm) using gradient KOH containing 15% acetonitrile as organic modifier at a constant flow rate of 1.0 mL/min. The molecular ions m/z [M − H] 127 and 161 were selected for the quantification in selected ion monitoring (SIM) mode for MCPs and DCPs, respectively. The average recoveries were between 80.6% and 92.6%. Within-day and day-to-day relative standard deviations were less than 12.1% and 13.3%, respectively. The method allowed the nine objective compounds in water samples to be determined at μg/L levels. It was confirmed that this method could be used in routine analysis.  相似文献

A rapid method for the simultaneous identification and quantification of pesticide residues in edible seaweed has been developed. Target analytes were three pyrethroid, a carbamate and two organophosphorus pesticides. The procedure consists of a pressurized liquid extraction (PLE) with integrated clean-up, followed by gas chromatography coupled to tandem mass spectrometry. Five PLE parameters were investigated using a screening design: temperature, static extraction time, number of cycles, percent of flush volume and quantitative composition of the n-hexane/ethyl acetate extraction solvent. The effect of the in-cell clean-up with Florisil® and graphitized carbon black adsorbents was investigated using a Doehlert response surface design. Large volumes of sample extracts were injected using a programmed-temperature vaporizer (PTV-LVI) to improve both sensitivity and selectivity of measurements. Quantification was carried by the internal standard method with surrogate deuterated standards. The method showed excellent linearity (R2 > 0.999) and precision (relative standard deviation, RSD ≤ 8%) for all compounds, with detection limits ranging from 0.3 pg g−1 for chlorpyrifos-ethyl, to 3.0 pg g−1 for carbaryl (23.1 pg g−1 for deltamethrin). Recoveries in real seaweed samples were within the range 82–108%. The method was satisfactory validated for the analysis of wild and cultivated edible seaweeds. The presence of pyrethroid and organophosphorus pesticides in some of the samples was evidenced.  相似文献

Gas chromatography coupled to mass spectrometry in negative chemical ionization mode (GC-(NCI)MS) has been applied to the quantification and reliable identification of polybrominated diphenyl ethers (PBDEs) in animal and vegetable samples from aquaculture activities. Matrices analyzed included fish fillet, fish feed, fish oil and linseed oil, their fat content ranged from 5% to 100%. Solid-phase extraction (SPE) (using Florisil and silica cartridges) and normal-phase high performance liquid chromatography were tested for an efficient clean-up in order to obtain sample extracts free of interfering compounds. Combining sulphuric acid digestion and SPE with Florisil led to the highest efficiency in the elimination of interferences from the extracts. The sample procedure developed, together with the application of GC-(NCI)MS for measurement, led to the satisfactory determination of PBDEs at μg kg −1 levels in complex aquaculture matrices with high lipid content. The use of a short and thin film-thickness fused-silica capillary column allowed to determine the problematic BDE 209 with satisfactory results. Three m/z ions were acquired for each analyte, which ensured a reliable identification of compounds detected in samples.  相似文献

A simple method using a one-step liquid-liquid extraction (LLE) with butyl acetate followed by high-performance liquid chromatography (HPLC) with positive ion electrospray ionization tandem mass spectrometric (ESI-MS/MS) detection was developed for the determination of itopride in human plasma, using sulpiride as an internal standard (IS). Acquisition was performed in multiple reaction monitoring (MRM) mode, by monitoring the transitions: m/z 359.5 > 166.1 for itopride and m/z 342.3 > 111.6 for IS, respectively. Analytes were chromatographed on an YMC C18 reverse-phase chromatographic column by isocratic elution with 1 mM ammonium acetate buffer-methanol (20: 80, v/v; pH 4.0 adjusted with acetic acid). Results were linear (r2 = 0.9999) over the studied range (0.5-1000 ng mL−1) with a total analysis time per run of 2 min for LC-MS/MS. The developed method was validated and successfully applied to bioequivalence studies of itopride hydrochloride in healthy male volunteers.  相似文献

A procedure based on QuEChERS extraction and a simultaneous liquid–liquid partition clean-up was developed. The procedure involved extraction of hydrated soil samples using acetonitrile and clean-up by liquid–liquid partition into n-hexane. The hexane extracts produced were clean and suitable for determination using gas chromatography–tandem mass spectrometry (GC–MS/MS). The method was validated by analysis of soil samples, spiked at five levels between 1 and 200 μg kg−1. The recovery values were generally between 70 and 100% and the relative standard deviation values (%RSDs) were at or below 20%. The procedure was validated for determination of 19 organochlorine (OC) pesticides. These were hexachlorobenzene (HCB), α-HCH, β-HCH, γ-HCH, heptachlor, heptachlor epoxide (trans), aldrin, dieldrin, chlordane (trans), chlordane (cis), oxychlordane, α-endosulfan, β-endosulfan, endosulfan sulfate, endrin, p,p′-DDT, o,p′-DDT, p,p′-DDD and p,p′-DDE. The method achieved low limits of detection (LOD; typically 0.3 μg kg−1) and low limits of quantification (LOQ; typically 1.0 μg kg−1). The method performance was also assessed using five fortified soil samples with different physico-chemical properties and the method performance was consistent for the different types of soil samples. The proposed method was compared with an established procedure based on Soxtec extraction. This comparison was carried out using six soil samples collected from regions of Pakistan with a history of intensive pesticide use. The results of this comparison showed that the two procedures produced results with good agreement. The proposed method produced cleaner extracts and therefore led to lower limits of quantification. The proposed method was less time consuming and safer to use. The six samples tested during this comparison showed that soils from cotton growing regions contained a number of persistent OC residues at relatively low levels (<10 μg kg−1). These residues were α-HCH, γ-HCH, heptachlor, chlordane (trans), p,p′-DDT, o,p′-DDT, p,p′-DDD, p,p′-DDE, β-endosulfan and endosulfan sulfate.  相似文献

A sensitive, selective and rapid liquid chromatography tandem mass spectrometry (LC‐MS/MS) method was developed for the simultaneous determination of bupropion (BUP) and its major active metabolite hydroxybupropion (HBUP) in human plasma. Separation of both the analytes and venlafaxine as internal standard (IS) from 50 μL human plasma was carried out by solid‐phase extraction. The chromatographic separation of the analytes was achieved on a Zorbax Eclipse XDB C18 (150 × 4.6 mm, 5 µm) analytical column using isocratic mobile phase consisting of 20 mm ammonium acetate–methanol (10:90, v/v), with a resolution factor of 3.5. The method was validated over a wide dynamic concentration range of 0.1–350 ng/mL for BUP and 0.1–600 ng/mL for HBUP. The matrix effect was assessed by post‐column infusion and the mean process efficiency was 96.08 and 94.40% for BUP and HBUP, respectively. The method was successfully applied to a bioequivalence study of 150 mg BUP (test and reference) extended release tablet formulation in 12 healthy Indian male subjects under fed conditions. A rapid and specific liquid chromatographic-mass spectrometric method has been developed and validated for the determination of the carboxylic acid metabolite of clopidogrel in human plasma. Sulphafurazole was used as internal standard. The samples were subjected to a solid phase extraction procedure using Hypercarb cartridges. The chromatographic separation was performed on a reversed phase porous graphitized carbon column using a mobile phase consisting of 70% methanol in water containing 0.1% (v/v) trifluoroacetic acid, pumped at a flow rate of 0.25 ml min−1. The analytes were detected after positive electrospray ionization using the selected ion monitoring mode of the species at m/z 308 for the carboxylic acid metabolite of clopidogrel, m/z 322 for clopidogrel and m/z 268 for sulphafurazole. Calibration graphs were linear (r>0.9994, n=6), in the range 100-1000 ng ml−1 for the carboxylic acid metabolite of clopidogrel. The intra- and inter-day R.S.D. values were <3.1%, while the relative error Er was less than −9.6% (n=6). The limits of detection (3.3σ) and quantitation (10σ) for the carboxylic acid metabolite of clopidogrel were found to be 28 and 93 ng ml−1, respectively. The efficiency of the solid phase extraction procedure for the carboxylic acid metabolite of clopidogrel averaged 74.6%.  相似文献
