High-performance liquid chromatographic methods were developed for the separation of the enantiomers of thirteen unusual beta-3-homo-amino acids and three of its ethyl esters on a chiral stationary phase containing (+)-(18-crown-6)-2,3,11,12-tetracarboxylic acid as chiral selector. The effects of the mobile phase composition and the acidic modifiers on the separation were investigated. The structures of the substituents in beta-position substantially influenced the retention and enantioseparation. The influence of ionic strength on the enantioseparation was established experimentally. The elution sequence was determined in all cases.  相似文献

合成了三-(4-甲基苯甲酸)纤维素酯(MCTB)手性固定相,用反相高效液相色谱法在该手性固定相上对2-(4-羟基苯氧基)丙酸酯对映体进行拆分.实验结果表明,在三-(4-甲基苯甲酸)纤维素酯手性固定相上,以甲醇与水的体积分数为75:25做流动相能较好的拆分2-(4-羟基苯氧基)丙酸甲酯、乙酯和丁酯对映体,其分离因子分别为1.38、1.49、0.98;同时还发现2-(4-羟基苯氧基)丙酸酯中酯基团的大小对其对映体的分离也有明显的影响,其中以乙酯的拆分效果最佳。  相似文献

Summary The liquid-chromatographic separation of the enantiomers of pyrethroic acids and their esters has been investigated on a polysaccharide-derived chiral stationary phase (CSP), Chiralpak AS. A new type of chiral surfactant, 3-[(3-dehydroabietamidopropyl) dimethylammonio]-1-propanesulfonate (DHAMAP) has been synthesized. The ability of this compound to perform chiral separation of D/L-amino acids derivatized with naphthalene-2, 3-dicarboxaldehyde (NDA-D/L-amino acids) has been investigated by capillary electrophoresis (CE). Enantiomeric separation of NDA-D/L-amino acids was achieved with a running buffer consisting of 50 mM borate (pH 9.75) and 25 mM DHADMP. Under the conditions selected, 6 pairs of tested amino acids enantiomers including NDA-D/L-tryptophan, NDA-D/L-phenylalanine, D/L-D/L-kynurenine, NDA-D/L-beta-phenylalanine, NDA-D/L-4-methylphenylalanine and NDA-D/L-arginine were well resolved. The resolution values were in the range of 1.56-5.40.  相似文献

采用高效液相色谱法在手性柱上分离芳氧基苯氧基丙酸类除草剂中间体(±)-2-(4-羟基苯氧基)丙酸酯的对映体。实验结果表明,在三苯甲酸纤维素酯的手性柱上,以无水乙醇为流动相能较好地分离(±)-2-(4-羟基苯氧基)丙酸乙酯和甲酯,其分离因子α值分别为1.44和1.29;同时还发现在三苯甲酸纤维素酯的手性柱上2-取代芳氧基或芳基丙酸酯结构中的酯基团的大小对其对映体的分离有明显的影响,其中以乙基为最佳。并通过对照试验证实了2-(4-羟基苯氧基)丙酸乙酯的右旋体先流出,其左旋体后流出。  相似文献

Direct reversed-phase high-performance liquid chromatographic methods were developed for the separation of enantiomers of eighteen unnatural β-amino acids, including several β-3-homo-amino acids. The direct separations of the underivatized analytes were performed on chiral stationary phases containing macrocyclic glycopeptide antibiotics such as teicoplanin (Chirobiotic T and T2), teicoplanin aglycone (Chirobiotic TAG), vancomycin (Chirobiotic V and V2), and ristocetin A (Chirobiotic R) as chiral selectors. The effects of the organic modifier, mobile phase composition and pH on the separations were investigated. A comparison of the separation performances of the macrocyclic glycopeptide stationary phases revealed that the Chirobiotic T2 column exhibited better selectivity than the Chirobiotic T column for the separation of β-3-homo-amino acid enantiomers; vancomycin or ristocetin A exhibited lower selectivity. The elution sequence was determined in some cases: the S enantiomers eluted before the R enantiomers, with the exception of the Chirobiotic R column, where the elution sequence R < S was observed.  相似文献

A parallel library of 108 4-aryl-1,4-dihydropyrimidine (DHPM) enantiomers, which are potential selectors for chiral HPLC separations, was synthesized using the single-step Biginelli multicomponent condensation. The individual compounds were screened by observing the enantioselectivity for resolution on a "brush-type" L-(3,5-dinitrobenzoyl)leucine-based chiral stationary phase, and separation factors alpha up to 12 were achieved. The best candidates from the library contained an ortho-substituted aromatic group at C4 carbon atom of the pyrimidine ring and an alkyl substituent at N1 nitrogen atom. Resolution of the enantiomers of the lead compound, 4-(9-phenanthryl)-DHPM 8, using semipreparative chiral HPLC followed by attachment to monodisperse macroporous aminomethacrylate beads, provided the novel polymer based chiral stationary phase with good enantioselectivities in the resolution of several pi-acidic aryl-dihydropyrimidines and derivatized profens. In addition, 3,5-dinitrobenzamido derivatives of alpha-amino acids could be resolved under normal phase HPLC conditions with separation factors up to 8.  相似文献

The enantiomers of thirty-nine O-ethyl O-phenyl N-isopropyl phosphoroamidothioates have been separated by high performance liquid chromatography on a Pirkle model chiral stationary phase using ten mobile phase. Chromatographic data are presented for the separation of these organic phosphorus enantiomers on the chiral phase. 22R-布地奈德的药物活性比22S-布地奈德的强2~3倍,开发布地奈德对映体拆分和定量分析方法,可为其药物研发及质量控制提供重要依据。目前,主要以反相C18固定相对布地奈德对映体进行拆分,而采用手性固定相对其进行拆分少有报道。通过考察固定相、流动相和柱温对布地奈德对映体拆分的影响,建立了基于直链淀粉-三[(S)-1-苯乙基氨基甲酸酯]手性固定相快速拆分和检测布地奈德对映体的高效液相色谱方法,其色谱条件如下:色谱柱为Chiralpak AS-RH色谱柱(150 mm×4.6 mm, 5.0 μm),流动相为乙腈-水(45∶55, v/v),柱温40 ℃,流速1.0 mL/min,二极管阵列检测器(DAD),检测波长246 nm,进样量10 μL。在该色谱条件下,布地奈德的两个对映体得到较好拆分,22R-布地奈德和22S-布地奈德的保留时间分别6.40 min和7.77 min,分离度为4.64; 22R-布地奈德和22S-布地奈德分别在各自范围内线性关系良好,相关系数(R2)均为0.9999,检出限分别为0.05 μg/mL和0.07 μg/mL,定量限分别为0.16 μg/mL和0.20 μg/mL; 4个添加水平的样品加标回收率为102.63%~104.17%,相对标准偏差(RSD)为0.08%~0.57%(n=6)。将该方法应用于1批次4个吸入用布地奈德混悬液实际样品进行检测,22R-布地奈德和22S-布地奈德的含量分别为283.15~284.63 μg/mL和259.86~261.51 μg/mL。该方法操作简便,分析时间短,重复性好,准确度高,可用于布地奈德对映体的拆分及其制剂的质量控制。  相似文献

The simultaneous enantiomeric separation of 30 racemic acids including 24 hydroxy acids in a single analysis is described for the determination of their absolute configurations. It involves the conversion of each enantiomer into diastereomeric O-trifluoroacetylated (-)-menthyl ester for the direct separation by gas chromatography on achiral dual-capillary columns of different polarities, with subsequent identification and chiral discrimination by retention index (I) library matching. Among the acids studied, the enantiomers of 27 acids were discriminatively resolved on both non-polar DB-5 and the intermediate-polar DB-17 columns with resolution factors in the range of 0.7-7.7 and separation factors in the range of 1.002-1.021. Enantiomers of 3-hydroxybutyric and alpha-methoxyphenylacetic acids were partially resolved on DB-5 (resolution factor of 0.9), but not resolved on DB-17, while the baseline resolution for 3-hydroxydecanoic acid and the minimal separation on the peak top (resolution factor less than 0.7) for 2-hydroxyglutaric acid were achieved on DB-17 but not on DB-5. The temperature-programmed I values measured on both columns were characteristic of each enantiomer and thus simple I matching with the reference values was useful in cross-checking for their chemical identification and the chiral discrimination as well. When applied to a clinical urine sample, the present method allowed positive identification of endogenous (S)-lactic acid and (S)-2-hydroxybutyric acid along with (R)-3-hydroxybutyric acid.  相似文献

Summary A new chiral derivatizing agent (CDA), (1S,2S)-1,3-diacetoxy-1-(4-nitrophenyl)-2-propylisothiocyanate, (S,S)-DANI, was applied to the separation of the enantiomers of unusual amino acids containing two chiral centers. Different β-methyl-α-amino acids (β-MePhe, β-MeTyr and β-MeTrp) and β-amino acids with cycloalkane skeletons (2-aminocyclopentanecarboxylic acid and 2-aminocyclohexanecarboxylic acid) were derivatized and the thiourea derivatives produced were separated by reversed-phase high-performance liquid chromatography. The applicability of this new CDA in the separation of unusual amino acids is demonstrated. The four stereoisomers of the investigated amino acids (except β-MePhe) could be separated in one chromatographic run. The enantiomers of eight unusual beta(2)- and beta(3)-homoamino acids were separated on chiral stationary phases containing the macrocyclic glycopeptide antibiotic teicoplanin (Chirobiotic T or T2) or teicoplanin aglycone (Chirobiotic TAG) as chiral selectors. The effects of the organic modifier, the mobile phase composition, and temperature on the separations were investigated. Linear van't Hoff plots were observed in the studied temperature range, 280-318 K, and the changes in enthalpy, Delta(DeltaH(o)), entropy, Delta(DeltaS(o)), and free energy, Delta(DeltaG(o)) were calculated. The values of the thermodynamic parameters depended on the nature of the selectors, the structures of the analytes, and especially the positions of the substituents on the analytes. A comparison of the separation performances of the macrocyclic glycopeptide stationary phases revealed that the Chirobiotic TAG column exhibited much better selectivity for beta(2)-homoamino acids, while the separation of beta(3)-homoamino acid enantiomers was better on Chirobiotic T or T2. The elution sequence was determined in some cases and was observed to be R < S.  相似文献

A liquid chromatographic ligand exchange chiral stationary phase (CSP) derived from (S)-leucinol was applied in the separation of the enantiomers of 12 beta-amino acids. The resolution was quite successful especially for the enantiomers of beta-amino acids containing aromatic functional group in the side chain. The chromatographic resolution behaviors were dependent on the organic modifier and Cu(II) concentration in aqueous mobile phase and the column temperature.  相似文献

A new chiral stationary phase (CSP) for the liquid chromatographic separation of enantiomers was prepared by bonding a novel enantiopure (diphenyl-substituted 1,1′-binaphthyl) crown ether to 5 μm silica gel. The resulting CSP was applied to the separation of the enantiomers of various natural and unnatural α-amino acids. All α-amino acids tested were resolved very well on the new CSP, with the exception of proline, which does not contain a primary amino group. The resolution of α-amino acid enantiomers on this new CSP was found to be dependent on the type and amounts of organic and acidic modifiers, and on column temperature.  相似文献

将C18柱与手性冠醚柱串联,建立了一种反相高效液相色谱法用于3种芳香族氨基酸对映体同时拆分的方法.考察了反相色谱流动相的组成、pH值、柱温、流速对对映体拆分的影响.实验结果表明,当流动相为HClO4-乙睛溶液(86:14,V/V,pH 2.0)、柱温20℃、流速0.4 mL/min时,3种氨基酸对映体可获得基线分离.进一步对比了C18柱、冠醚手性柱和串联顺序不同的4种分离模式,结果表明,C18柱不能拆分氨基酸对映体,仅能分离不同种类氨基酸;冠醚手性柱可分离氨基酸映体,但不同种类氨基酸色谱峰出现重叠;串联模式能实现3种氨基酸对映体的基线分离,实现双柱优势互补,而串联顺序对分离影响不大,仅影响色谱峰的峰形.  相似文献

建立了以直链淀粉衍生物为手性固定相的高效液相色谱-串联质谱(HPLC-MS/MS)直接拆分普萘洛尔、美托洛尔、阿罗洛尔和卡维地洛4种β-受体阻滞剂对映体的方法。考察了手性固定相的种类、流动相改性剂和添加剂的体积分数、柱温和流速等对4种药物对映体分离的影响。结果表明:在Chiralpak AD-H手性色谱柱上,在正己烷-乙醇-二乙胺(20∶80∶0.03,v/v/v)为流动相、流速0.550 mL/min、柱温40℃的条件下,普萘洛尔、美托洛尔、阿罗洛尔和卡维地洛对映体均达到基线分离,分离度分别为1.37、1.80、2.09和4.70。通过热力学研究及对映体结构分析对拆分机理进行了探讨,发现4种药物对映体的手性拆分均为焓驱动过程,而固定相的手性空腔对不同药物的拆分影响较大。研究结果为β-受体阻滞剂的深入研究提供了参考方法。  相似文献

A quinine-derived chiral anion-exchanger stationary phase was applied for the direct high-performance liquid chromatographic separation of the enantiomers of N-protected unusual beta-amino acids, i.e. 3-aminobutanoic acid, 3-aminopentanoic acid, 3-amino-4-methylpentanoic acid, 3-amino-4,4-dimethylpentanoic acid, 3-amino-4-methylhexanoic acid, 3-amino-4-ethylhexanoic acid, 3-amino-3-cyclohexylpropanoic acid, 3-amino-3-(3-cyclohexen-1-yl)propanoic acid and 3-amino-3-phenylpropanoic acid. The readily prepared N-2,4-dinitrophenyl derivatives were well separable, with good efficiency and high resolution. The chromatographic conditions (eluent composition, pH and buffer concentration) were varied to achieve optimal separation. In some cases, the elution sequences of the enantiomers of the derivatives were determined.  相似文献

A mixed chiral sorbent based on silica with immobilized macrocyclic antibiotics eremomycin and vancomycin was synthesized. A possibility of the separation of enantiomers of β-blockers (metoprolol, pindolol, alprenolol, oxprenolol, labetalol, and atenolol) and amino acids (tryptophan, phenylalanine, DOPA, methionine, and acetyl glutamic acid) on this chiral sorbent by HPLC was studied. The influence of the composition of the mobile phase (pH of buffer solution, its concentration, content of organic modifier, and its nature) on the retention times of β-blocker and amino acid enantiomers, selectivity, and resolution of peaks was studied. It was shown that the mixed chiral sorbent has enantioselectivity to both classes of compounds, while silica modified with vancomycin has no ability to the separation of enantiomers of non-derivatized amino acids, and silica modified with eremomycin has no ability to the separation of β-blocker enantiomers. High values of resolution for amino acids (max Rs > 4) and β-blockers (max Rs > 1) were obtained.  相似文献
