采用噻唑蓝(MTT)法、碱性磷酸酶(ALP)比活性测定、油红O染色和茜素红染色及定量分析,研究了不同浓度的Fe3+和Fe2+对原代培养的成骨细胞增殖、分化及矿化功能的影响.结果表明:浓度为1×10-9~1×10-4 mol·L-1的Fe3+和Fe2+促进成骨细胞增殖,但是在较高浓度1×10-3 mol·L-1时,它们则抑制成骨细胞增殖.与成骨细胞作用48 h,浓度为1×10-8~1×10-4 mol·L-1的Fe3+和Fe2+抑制其分化,但在较低的浓度1×10-9 mol·L-1时则对其分化没有影响:进一步延长作用时间为72 h,Fe3+对成骨细胞分化没有影响,除1×10-6mol·L-1浓度的Fe2+促进成骨细胞分化外,其他浓度的Fe2+则抑制其分化;测试浓度下的Fe3+对成骨细胞向脂肪细胞的横向分化表现为抑制或没有影响,而Fe2+的影响则依赖于浓度和作用时间.在1×10-8~1×10-5mol·L-1浓度范围内,Fe3+和Fe2+对矿化结节的影响表现出相反的效应.在较高浓度(1×10-4mol·L-1)下,它们促进矿化节结的形成,而在较低浓度(1×10-9mol·L-1)下,Fe3+抑制矿化节结的形成,Fe2+则没有影响.结果提示:浓度.作用时间和铁离子的价态都是影响Fe3+和Fe2+生物效应(从毒性到活性,从损伤到保护,从上调到下调)转变的关键因素.  相似文献

Pyrene has been found to form ground and excited electronic state complexes of 1:1 stoichi-ometry with GMP, CMP, TMP and AMP. The values of their ground state association constants are 45 M-1, 13M-1, 14 M-1, and 52 M-1 respectively. The fluorescence of pyrene is strongly quenched by GMP, CMP, and TMP but only slightly by AMP. Fluorescence quenching analysis has yielded the values 87M-1, 73 M-1, and 154 M-1 for the excited state association constants with GMP, CMP, and TMP, respectively. The corresponding values for the excited state second-order rate constant for complex formation are: 3.3 times 109M-1 s-1 4.1 times 109M-1 s-1, and 4.0 times 109M-1 s-1. The probabilities of complex formation per collision between an excited pyrene molecule and a nucleotide are: 0.52, 0.64, and 0.63. The values for the excited state rate constant for dissociation of the complex are: 3.8 times 107s-1 5.6 times 107s-1, and 2.6 times 107s-1. The possibility is discussed that partial transfer of charge from pyrene to nucleotide may be playing a role in the complex formation process.  相似文献

The effect of halides and different buffer anions on the quenching of the fluorescence of the new probe 10,10′-bis(3-sulfopropyl)-9,9′-biacridine (SPBA) has been studied using fluorescence and decay time measurements. The linearity of the Stern-Volmer plot indicates that fluorescence quenching by halides can be described reasonably well by a single-exponential decay with a K of 4.06 times 106M-1s-1for chloride, 7.83 times 106M-1s-1for bromide and 1.12 times 107M-1s-1for iodide. We have found that SPBA is collisionally quenched also by the buffers 3-(N-mor-pholino)propanesulfonic acid (MOPS) and N-2-hydroxy-ethylpiperazine-N′-ethansulfonic acid (HEPES). The bi-molecular rate constants are 1.67 × 106M-ls-1for HEPES and 1.44 times 106M-1s-1for MOPS.  相似文献

The 1D- and 2D-6Li, 6Li-INADEQUATE experiments are described as new tools for the detection of scalar coupled nonequivalent 6Li nuclei in organolithium clusters. Practical applications of these sequences are demonstrated for the 6Li-NMR spectra of (E)-1-lithio-2-(2-lithiophenyl)-1-phenylhex-1-ene ( 1 ) and (E)-2-lithio-1-(2-lithiophenyl)-1-phenylpent-1-ene ( 2 ), where signals due to dimers and monomers can be distinguished. The performance of the 2D-6Li, 6Li-INADEQUATE and the 6Li, 6Li-COSY-45-LR experiment are compared. The 6Li chemical shifts of 1 and 2 are discussed.  相似文献

A radiochemical neutron activation method for the simultaneous determination of arsenic, cadmium, cobalt, mercury, molybdenum, and zinc in fresh water is described. The method is based on anion-exchange separation in hydrochloric acid media followed by simple precipitations. The determination limits, based on analysis of a 5-ml sample without preconcentration, and with a well-type NaI(Tl) detector, are as follows: As, 10-3 μg l-1 ; Cd, 6 × 10-2 μg l-1 ; Co, 4 × 10-3 μg l-1 ; Hg, 7 × 10-3 μg l-1 ; Mo, 10-1 μg l-1 ; Zn, 2 × 10-1 μg l-1. The method is adequate for the analysis of natural fresh waters.  相似文献

Phase transition behavior of polyesters derived from 2,7-phenanthrene dicarbonic acid diethylester and alkanediols with even methylene carbon number was investigated by the simultaneous DSC-XRD method. The smectic A phase was observed on cooling from the molten state. The transition entropy from the isotropic state to the smectic A phase was about 9.0 J mol-1 K-1, which depended on the methylene carbon number. The linear expansion coefficients, based on the (001) spacing of the crystalline phase at room temperature, were 1.3·10-4 K-1 (crystalline phase), 5.7·10-4 K-1 (crystallization region), 1.7·10-3 K-1 (smectic A phase) during cooling, and 1.5·10-4 K-1 (crystalline phase), and 1.0·10-3K-1(melting region) on heating. Abstract —α-Tocopherol scavenges singlet oxygen (produced by methylene blue photosensitization in methanol) by a combination of chemical reaction (4.6 times 107M-1s-1) and quenching (6.2 times 108M-1 s-1). The total rate of scavenging (6.7 times 108 M-1s-1) makes it an effective protective agent against photooxidation mediated by singlet oxygen.  相似文献

Six Schiff-bases HL1-HL4, L5 and L6 [HL1 = 2,6-bis[1-(2-aminoethyl)pyrolidine-iminomethyl]-4-methyl-phenol, HL2 = 2,6-bis[1-(2-aminoethyl)piperidine-iminomethyl]-4-methyl-phenol, HL3 = N-{1-(2-aminoethyl)pyrolidine}salicylideneimine, HL4 = N-{1-(2-aminoethyl)piperidine}salicylideneimine, L5 = 2-benzoyl pyridine-N-{1-(2-aminoethyl)pyrolidine}, L6 = 2-benzoylpyridine-N-{1-(2-aminoethyl)piperidine}] have been synthesized and characterized. Zn(II) complexes of those ligands have been prepared by conventional sequential route as well as by template synthesis. The same complexes are obtained from the two routes as evident from routine physicochemical characterizations. All the Schiff-bases exhibit photoluminescence originating from intraligand (π–π*) transitions. Metal mediated fluorescence enhancement is observed on complexation of HL1-HL4 with Zn(II), whereas metal mediated fluorescence quenching occurs in Zn(II) complexes of L5 and L6.  相似文献

Incubation of the geraniols (R)-(8-2H1)[8-3H1]- 1 and (S)-(8-2H1)[8-3H1]- 1 with microsomal cytochrome P-450Cath. from the subtropical plant Catharanthus roseus (L.)G. DON resulted in the formation of the chiral 8-hydroxygeraniols (S)-(8-2H1)[8-3H1]- 2 and (R)-(8-2H1)[8-3H1]- 2 . Their absolute configuration was assigned on the basis of the 1H-decoupled 3H-NMR Spectra of the corresponding dicamphanates (S)-(8-2H1)[8-3H1]- 9 and (R)-(8-2H1)[8-3H1]- 9 , of which the configurations are established in relation to the synthetic reference samples. The results clearly indicate retention of configuration during the allylic oxidation of 1 .  相似文献

4-1BB, a member of the tumor necrosis factor receptor (TNFR) superfamily, is a major costimulatory receptor that is rapidly expressed on the surface of CD4+ and CD8+ T cells after antigen- or mitogen-induced activation. The interaction of 4-1BB with 4-1BBL regulates immunity and promotes the survival and expansion of activated T cells. In this study, the expression of 4-1BB and 4-1BBL was examined during regeneration of the murine thymus following acute cyclophosphamide-induced involution. Four-color flow cytometry showed that 4-1BB and 4-1BBL were present in the normal thymus and were preferentially expressed in the regenerating thymus, mainly in CD4+CD8+ double-positive (DP) thymocytes. Furthermore, the CD4loCD8lo, CD4+CD8lo and CD4loCD8+ thymocyte subsets, representing stages of thymocyte differentiation intermediate between DP and single-positive (SP) thymocytes, also expressed 4-1BB and 4-1BBL during thymus regeneration but to a lesser degree. Interestingly, the 4-1BB and 4-1BBL positive cells among the CD4+CD8+ DP thymocytes present during thymus regeneration were TCRhi and CD69+ unlike the corresponding controls. Moreover, the 4-1BB and 4-1BBL positive cells among the intermediate subsets present during thymus regeneration also exhibited TCRhi/int and CD69+/int phenotypes, indicating that 4-1BB and 4-1BBL are predominantly expressed by the positively selected population of the CD4+CD8+ DP and the intermediate thymocytes during thymus regeneration. RT-PCR and Western blot analyses confirmed the presence and elevated levels of 4-1BB and 4-1BBL mRNA and protein in thymocytes during thymus regeneration. We also found that the interaction of 4-1BB with 4-1BBL promoted thymocyte adhesion to thymic epithelial cells. Our results suggest that 4-1BB and 4-1BBL participate in T lymphopoiesis associated with positive selection during recovery from acute thymic involution.  相似文献

Summary Relatively short-lived radioactive nuclides of cosmogenic origin, such as7Be (T1/2=53.3 d),32P (14.3 d),33P (25.3 d),35S (87.4 d) as well as22Na (2.6 y) have come to be recognized as potential tools to trace environmental processes, such as precipitation, washout (precipitation scavenging), resuspension, atmospheric particle deposition and deposition patterns of airborne contaminants, aerosol deposition and aerosol trapping by above ground vegetation (air-to-vegetation transfer). Of the above radionuclides,7Be as well as210Pb of terrestrial origin and137Cs, a fission product radionuclide, are commonly used for determining the atmospheric particle deposition parameters. It was found that the deposition velocity of7Be-associated aerosol particles varied from 0.3 to 0.8 cm. s-1(average 0.5 cm. s-1), the washout ratio for7Be varied from 103 to 375 (average 144), the resuspension factor for the7Be-associated particles varied from 1.6. 10-4to 4.2. 10-4m-1(average 2.3. 10-4m-1) and the air-to-vegetation transfer rate for7Be varied from 5258 to 16180 m3.kg-1(average 9856 m3.kg-1) for grass, gramineae or poaceae the species. For the137Cs-associated aerosol particles the deposition velocity varied from 1.3 to 6.3 cm. s-1(average 3.4 cm. s-1), the washout ratio for137Cs varied from 284 to 3810 (average 1295), the resuspension factor for the137Cs-associated aerosol particles varied from 0.1. 10-4to 1.2. 10-4m-1(average 0.6. 10-4m-1) and the air-to-vegetation transfer rate for137Cs varied from 39638 to 345279 m3.kg-1(average 181197 m3.kg-1) for grass, gramineae or poaceaethe species.  相似文献

Four 1:1, two-component salts combining the [Ni(dmit)2] anion (dmit2− = 2-thioxo-1,3-dithiole-4,5-dithiolato) and chiral stilbazolium-based countercations (HPMS+ = 4′-[2-(hydroxymethyl)pyrrolidinyl]-1-methylstilbazolium and MPMS+ = 4′-[2-(methoxy-methyl)pyrrolidinyl]-1-methylstilbazolium), or chiral ferrocenyl-based countercations (2+ = (E)-1-((R)-2-methylferrocenyl)-2-(1-methyl-4-pyridiniumyl)ethene; 3+ = (E)-1-((S)-2-trimethylsilylferrocenyl)-2-(1-methyl-4-pyridiniumyl)ethene) were prepared. Semiconducting behaviour (2·10−4 S·cm−1 measured on compressed pellets for [Ni(dmit)2] (MPMS), for example) is secured by the presence of the [Ni(dmit)2] anions. The chiral nature of the countercations ensures non-centrosymmetry of the structures (space group P1 for [Ni(dmit)2](2) and [Ni(dmit)2](3), for example). A ubiquitous antiparallel arrangement of the cations, which are thus packed in a pseudo-centrosymmetrical environment, results in almost vanishing second-order susceptibilities χ(2), and therefore zero efficiencies in second harmonic generation.  相似文献

A simple, precise and selective procedure for the spectrophotometric determination of carbon disulphide in water is described. The method is based on the quantitative formation of the 1,2,3,4-thiatriazol-5-thiolate ion from carbon disulphide and azide. The specific rate for this reaction at ionic strength 4.0 (NaN3) and 25°C is 8.4·10-3 mol-1 s-1; the kinetic parameters D3H and D3S are 16.7 kcal mol-1 and —14 cal mol-1deg-1, respectively. At the analytical wavelength of 313 nm, the molar absorptivity is 7.27 ·103 l mol-1cm-1. The system obeys Beer's law for 2.3–28.6 g ml-1 of carbon disulphide. The effects of foreign ions are discussed.  相似文献

Os3(μ-CO)(CO)93-Me3SiC2Me) alkyne complexes react with ferrocenylacetylene in hot benzene to form Os3(CO)931122-C(SiMe3)C(Me)C(H)C(Fe)} and a small amount of the isomeric Os3(CO)9(μ-η114-C(SiMe3)C(Me)C(Fc)C(H)} complex. The structure of the major isomer was confirmed by X-ray structural analysis of the single crystal. Thermolysis of this complex in refluxing benzene affords the Os3(μ-H)(CO)831141-C(SiMe3)C(Me)C(H)(C5H3FeC5H5)} complex with theortho-metallated ferrocene moiety. The spectral characteristics of clusters with the μ31122 and μ-η114 coordinations of the metallacyclopentadiene fragment have been established, which made it possible to choose between the alternative modes of bonding of diene with the trimetallic core.  相似文献

Summary The sorption of Eu(III) ions onto PAN loaded PUF has been optimized and investigated under the influence of various temperatures, i.e., 303, 313 and 323 K. Maximum retention (>96%) of Eu(III) ions (1.79. 10-5M) onto PAN loaded PUF (7.75 mg. ml-1) was achieved after 30-minute equilibration time from pH 7 buffer solution. The variation of sorption with temperature yields the thermodynamic parameters ΔH=79±2 kJ. mol-1, ΔS=276±7 kJ. mol-1. K-1and ΔG=-1.4±0.1 kJ. mol-1at 298 K. The positive value of enthalpy and negative free energy show endothermic and spontaneous nature of sorption, respectively. The sorption data followed Freundlich, Dubinin-Radushkevich (D-R) and Langmuir isotherms at all the three given temperatures. The Freundlich constant 1/n=0.70, 0.62 and 0.55 and sorption capacities Cm=10.8 mmol. g-1, 6.1 mmol. g-1and 4.4 mmol. g-1, respectively, decreased with increasing temperature. Similarly, the sorption capacities of D-R isotherm Xm=197.6 μmol. g-1, 201.2 μmol. g-1and 137.4 μmol. g-1, also decreased with temperature. However, the sorption free energy E=10.2 kJ. mol-1, 11.2 kJ. mol-1and 12.7 kJ. mol-1, increased with temperature. The monolayer coverage (Q) computed from Langmuir isotherm was 11.1±0.6 μmol. g-1and remains constant at all the three temperatures investigated. However, the binding energy constant bincreased with temperature. The relationship of bwith temperature and differential heat of adsorption (ΔHdiff) have been evaluated and discussed.  相似文献

Rate constants for the reaction of superoxide O- 2 with various substrates were obtained through stationary electrode polarography theory and technique. In solvent acetonitrile, the substrate and the rate constants of the reaction O- 2 + AH- k2Product, are, AH = isopropanol (k2 < 0.01 M-1 s-1); ethanol (k2 = 1.42 × 102 M-1 s-1); methanol (k2 = 1.1 × 107 M-1 s-1), H2O (k2 = 1.0 × 105 M-1 s-1). In MeCN, O-2 was found to be rather unreactive towards glucose and acetone but it reacts with fructose and sucrose catalytically. However, in DMF2, O- 2reacts with glucose and fructose with k2 order of 105 M-1 s-1. The mechanism of the reaction of O- 2 with the substrates (AH) is proposed as O- 3 + AH k2O, AHk2 k-1 k [O2H + AH]-, k-2O2H + A- with k1 = 109 M-1 s-1 and k-1 = 108 -109 s-1. With these values of k-1 and k1, k k2(obs). The reversible E1/2 for O2 + e O- 2 in various solvents: MeCN, acetone, isopropanol, methanol, H2O were obtained either directly from the reversible voltammogram or from experimental voltammograms and the rate constants obtained (as above) using stationary electrode polargraphy theory; E1/2 being -0.82 (MeCN),-0.85 (acetone),-0.72 (isopropanol);-0.66 (MeOH),-0.56 (H2O) vs SCE.  相似文献

Pb, Zn, Cd, Ni, Mn, Fe, V and Cu in sea water are determined by extraction of their complexes with sodium diethyldithiocarbamate into chloroform, decomposition of the chelates and inductively-coupled plasma emission spectrometry. When 1-l water samples are used, the lowest determinable concentrations are: 0.063 μg Mn l-1, 0.13 μg Zn l-1, 0.25 μg Cd l-1, 0.25 μg Fe l-1, 0.38 μg V l-1, 0.5 μg Ni l-1, 0.5 μg Cu l-1, and 2.5 μg Pb l-1. Above these levels, the relative standard deviations are better than 12% for the complete procedure.  相似文献

Summary The gross alpha- gross beta-activity, 137Cs and 90Sr in the water and sediment samples taken from the Keban Dam Lake, Uluova Region, was investigated in 2003 and 2004. We have found that in spring the concentration of 137Cs in the surface water and deep sediment changed between 0.001-0.01 Bq/l (0.4-2.5 Bq.kg-1) that of 90Sr between 0.0001-0.009 Bq/l (0.1-9 Bq.kg-1) and in autumn the values were 0.0001-0.009 Bq/l (0.4-8 Bq.kg-1) for 137Cs and 0.0002-0.005 Bq/l (0.6-4 Bq.kg-1) for 90Sr, respectively.
