The highly selective, fast and effective sample pretreatment technique molecularly imprinted solid-phase extraction (MISPE) can overcome the low sensitivity of the highly efficient capillary electrophoresis-UV method (CE-UV). In this work, narrowly dispersible bisphenol A (BPA)-imprinted polymeric microspheres with a high capacity factor of k′ = 6.8 and an imprinted factor of I = 6.53 were investigated as selective solid-phase extraction (SPE) sorbents for use in extraction of BPA from different sample matrices (tap water, wastewater, Yangtze River water, soil from the Yangtze River, shrimp and human urine). Washing and eluting protocols of MISPE were optimized. Under optimal conditions, recoveries of MISPE were investigated. Recoveries were basically constant and the relative standard deviation (RSD) was lower than 5.8% when loading volumes changed from 1 to 50 mL. Recoveries ranged from 71.20% to 86.23% for different sample matrices. Compared with C18 SPE, MISPE had higher selectivity and recovery for BPA. BPA was determined with good accuracy and precision in different complex samples using CE-UV coupled with MISPE. Spiked recoveries ranged from 95.20% to 105.40%, and the RSD was less than 7.2%. Because a large loading volume was achieved, the enrichment efficiency of pretreatment and the sensitivity of this method were improved. The limits of detection of this MISPE-CE-UV method for BPA in tap water, wastewater, Yangtze River water, soil from the Yangtze River, shrimp and human urine were 3.0 μg L− 1, 5.4 μg L− 1, 6.9 μg L− 1, 2.1 μg L− 1, 1.8 μg L− 1 and 84 μg L− 1, respectively.  相似文献

Dispersive liquid–liquid microextraction (DLLME) coupled with high-performance liquid chromatography (HPLC)-UV detection was applied for the extraction and determination of bisphenol A (BPA) in water samples. An appropriate mixture of acetone (disperser solvent) and chloroform (extraction solvent) was injected rapidly into a water sample containing BPA. After extraction, sedimented phase was analyzed by HPLC-UV. Under the optimum conditions (extractant solvent: 142 μL of chloroform, disperser solvent: 2.0 mL of acetone, and without salt addition), the calibration graph was linear in the range of 0.5–100 μg L−1 with the detection limit of 0.07 μg L−1 for BPA. The relative standard deviation (RSD, n = 5) for the extraction and determination of 100 μg L−1 of BPA in the aqueous samples was 6.0%. The results showed that DLLME is a very simple, rapid, sensitive and efficient analytical method for the determination of trace amount of BPA in water samples and suitable results were obtained.  相似文献

Biosensor immunoassays for the detection of bisphenol A   总被引:1,自引:0,他引:1
Bisphenol A (BPA) is a xenoestrogen found in the environment, in consequence, for the biosensor detection of BPA we raised antibodies (polyclonal (PAbs) and monoclonal (MAbs)) against a structural analogue of BPA, 4,4 bis-(4-hydroxyphenyl) valeric acid (BVA). The kinetics of the MAb-BPA interaction were evaluated and the MAb providing the highest affinity was directly immobilized onto the sensor chip surface to evaluate a direct assay. Afterwards, the performance of the MAbs and the PAbs was compared in an inhibition assay using a BVA-coated chip. The highest sensitivity (limit of detection (LOD) of 0.4 μg L−1) was obtained with MAb 12 in the direct assay. However, the inhibition assay was the most robust and the PAbs showed the highest sensitivity (LOD of 0.5-1 μg L−1). The antibodies were specific for BVA and BPA as only minor cross-reactivities were found toward structurally related compounds or other endocrine disruptors. In the inhibition assay (with a run time of 6 min), water samples spiked with BPA at different levels (0.5-50 μg L−1) resulted in recoveries varying between 68% and 121%. The sensitivity of the inhibition assay could be improved 40 times (LOD of 0.03 μg L−1 with the Mab 12-based assay) using solid phase extraction (SPE).  相似文献

Polychlorotrifluoroethylene (PCTFE) in the form of beads was applied, as packing material for flow injection on-line column preconcentration and separation systems coupled with flame atomic absorption spectrometry (FAAS). Its performance characteristics were evaluated for trace copper determination in environmental samples. The on-line formed complex of metal with diethyldithiophosphate (DDPA) was sorbed on the PCTFE surface. Isobutyl methyl ketone (IBMK) at a flow rate of 2.8 mL min−1 was used to elute the analyte complex directly into the nebulizer-burner system of spectrophotometer. The proposed sorbent material reveal, excellent chemical and mechanical resistance, fast adsorption kinetics permitting the use of high sample flow rates up to 15 mL min−1 without loss of retention efficiency. For copper determination, with 90 s preconcentration time the sample frequency was 30 h−1, the enhancement factor was 250, which could be further improved by increasing the loading (preconcentration) time. The detection limit (3s) was cL = 0.07 μg L−1, and the precision (R.S.D.) was 1.8%, at the 2.0 μg L−1 Cu(II) level. For lead determination, the detection limit was cL = 2.7 μg L−1, and the precision (R.S.D.) 2.2%, at the 40.0 μg L−1 Pb(II) level. The accuracy of the developed method was evaluated by analyzing certified reference materials and by recovery measurements on spiked natural water samples.  相似文献

A general and broad class-specific enzyme-linked immunosorbent assay was developed for the O,O-dimethyl organophosphorus pesticides, including malathion, dimethoate, phenthoate, phosmet, methidathion, fenitrothion, methyl parathion and fenthion. Three haptens with different spacer-arms were synthesized. The haptens were conjugated to bovine serum albumin (BSA) for immunogens and to ovalbumin (OVA) for coating antigens. Rabbits were immunized with the immunogens and six polyclonal antisera were produced and screened against each of the coating antigens using competitive indirect enzyme-linked immunosorbent assay for selecting the proper antiserum. The effect of hapten heterology on immunoassay sensitivity was also studied. The antibody-antigen combination with the most selectivity for malathion was further optimized and tested for tolerance to co-solvent, pH and ionic strength changes. The IC50 values, under optimum conditions, were estimated to be 30.1 μg L−1for malathion, 28.9 μg L−1 for dimethoate, 88.3 μg L−1 for phenthoate, 159.7 μg L−1 for phosmet, 191.7 μg L−1 for methidathion, 324.0 μg L−1 for fenitrothion, 483.9 μg L−1 for methyl parathion, and 788.9 μg L−1 for fenthion. Recoveries of malathion, dimethoate, phenthoate, phosmet and methidathion from fortified Chinese cabbage samples ranged between 77.1% and 104.7%. This assay can be used in monitoring studies for the multi-residue determination of O,O-dimethyl organophosphorus pesticides.  相似文献

A highly sensitive flow analysis system has been developed for the trace determination of reactive phosphate in natural waters, which uses a polymer inclusion membrane (PIM) with Aliquat 336 as the carrier for on-line analyte separation and preconcentration. The system operates under flow injection (FI) and continuous flow (CF) conditions. Under optimal FI conditions the system is characterised by a linear concentration range between 0.5 and 1000 μg L−1 P, a sampling rate of 10 h−1, a limit of detection of 0.5 μg L−1 P and RSDs of 3.2% (n = 10, 100 μg L−1) and 7.7% (n = 10, 10 μg L−1). Under CF conditions with 10 min stop-flow time and sample solution flow rate of 1.32 mL min−1 the flow system offers a limit of detection of 0.04 μg L−1 P, a sampling rate of 5 h−1 and an RSD of 3.4% (n = 5, 2.0 μg L−1). Interference studies revealed that anions commonly found in natural waters did not interfere when in excess of at least one order of magnitude. The flow system, operating under CF conditions, was successfully applied to the analysis of natural water samples containing concentrations of phosphate in the low μg L−1 P range, using the multipoint standard addition method.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. The methods precision ranged from 8% to 14% for spiked river water and 7% to 14% for spiked tap water.  相似文献

A fast, sensitive and selective method for the determination of aluminium based on the reaction of the metal with pyrogallol red (PR) in the presence of tetrabutylammonium tetrafluoroborate (TBATFB) to form an Al(PR)3x9TBATFB complex which is adsorbed on the mercury electrode is presented. Under these conditions complexation of aluminium is rapid and no waiting period or heating of the sample is required. The reduction current of the accumulated complex is measured by scanning the potential in the cathodic direction. The variation of peak current with pH, adsorption time, adsorption potential, ligand and quaternary ammonium salt concentration, and some instrumental parameters, such as stirring rate in the accumulation stage, and step amplitude, pulse amplitude and step duration while obtaining the square wave voltamperograms were optimized. The best experimental parameters were pH 8.5, (NH4Ac-NH3 buffer), CPR = 25 μmol L−1, CTBATFB over 75 μmol L−1, tads = 60 s, and Eads = −0.60 V versus Ag/AgCl. A linear response is observed over the 0.0-30.0 μg L−1 concentration range, with a detection limit of 1.0 μg L−1. Reproducibility for 9.0 μg L−1 aluminium solution was 2.3% (n = 6). Synthetic sea water and sea water reference material CRM-SW were used for validation measurements. Aluminium in urine samples of a volunteer who ingested 800 mg of Al(OH)3 was analyzed.  相似文献

A novel technique, high temperature headspace liquid-phase microextraction (HS-LPME) with room temperature ionic liquid (RTIL), 1-butyl-3-methylimidazolium hexafluorophosphate ([C4MIM][PF6]) as extractant, was developed for the analysis of dichlorodiphenyltrichloroethane (p,p′-DDT and o,p′-DDT) and its metabolites including 4,4′-dichlorodiphenyldichloroethylene (p,p′-DDE) and 4,4′-dichlorodiphenyldichloroethane (p,p′-DDD) in water samples by high performance liquid chromatography with ultraviolet detection. The parameters such as salt content, sample pH and temperature, stirring rate, extraction time, microdrop volume, and sample volume, were found to have significant influence on the HS-LPME. The conditions optimized for extraction of target compounds were as follows: 35% NaCl (w/v), neutral pH condition, 70 °C, 800 rpm, 30 min, 10 μL [C4MIM][PF6], and 25 mL sample solutions. Under the optimized conditions, the linear range, detection limit (S/N = 3), and precision (R.S.D., n = 6) were 0.3-30 μg L−1, 0.07 μg L−1, and 8.0% for p,p′-DDD, 0.3-30 μg L−1, 0.08 μg L−1, and 7.1% for p,p′-DDT, 0.3-30 μg L−1, 0.08 μg L−1, and 7.2% for o,p′-DDT, and 0.2-30 μg L−1, 0.05 μg L−1, and 6.8% for p,p′-DDE, respectively. Water samples including tap water, well water, snow water, reservoir water, and wastewater were analyzed by the proposed procedure and the recoveries at 5 μg L−1 spiked level were in the range of 86.8-102.6%.  相似文献

Bismuth and Sb were evaluated as internal standards (IS) to minimize matrix effects on the direct and simultaneous determination of As, Cu, and Pb in cachaça by graphite furnace atomic absorption spectrometry using W-coated platform plus Pd-Mg(NO3)2 as modifier. For 20 μL injected sample, calibration within the 0.5-10 μg L−1 As, 100-1000 μg L−1 Cu and 0.5-30 μg L−1 Pb intervals were established using the ratios As absorbance to Sb absorbance, Cu absorbance to Bi absorbance and Pb absorbance to Bi absorbance versus analytes concentration, respectively. Typical linear correlations of 0.998, 0.999 and 0.999 were, respectively, obtained. The proposed method was applied for direct determination of As, Cu and Pb in 10 commercial cachaça samples and results were in agreement with those obtained by inductively coupled plasma mass spectrometry at 95% confidence level. The found characteristic masses were 30 pg As, 274 pg Cu and 39 pg Pb. The useful lifetime of the graphite tube was around 760 firings. Recoveries of As, Cu and Pb added to cachaça samples varied, respectively, from 98% to 109%, 97% to 108% and 98% to 104% with internal standards and from 48% to 54%, 53% to 92% and 62% to 97% without internal standards. The limits of detection were 0.13 μg L−1 As, 22 μg L−1 Cu and 0.05 μg L−1 Pb. The relative standard deviations (n = 12) for a spiked sample containing 20 μg L−1 As, Pb and 500 μg L−1 Cu were 1.6%, 1.0%, and 1.8% with IS and 4.3%, 5.2%, and 5.5% without IS.  相似文献

As a first attempt, cloud point extraction (CPE) was developed to preconcentrate bisphenol A (BPA), α-naphthol and β-naphthol prior to performing capillary zone electrophoresis (CZE) analysis. The parameters influencing the CPE efficiency, such as Triton X-114 concentrations, pH value, extraction time and temperature were systematically evaluated. After diluting with acetonitrile, the surfactant-rich phase of CPE can be injected directly into the CE instrument. The CZE baseline separation was achieved with running buffer (pH 9.5) composed of 50 mM sodium tetraborate in 30% (v/v) methanol, and an applied voltage of 25 kV. Under the optimized CPE and CZE conditions, an preconcentration factor of 50 times could be obtained and the limit of quantification for the three analytes were found to be 1.67 μg L−1, 0.80 μg L−1 and 0.67 μg L−1 for BPA, α-naphthol and β-naphthol, respectively. The proposed methods have shown to be a green, rapid and effective approach for determination of three analytes present in river water samples.  相似文献

A sensitive procedure is presented for the voltammetric determination of nickel. The procedure involves an adsorptive accumulation of nickel 1-nitroso-2-napthol (NN) complex on a bismuth film electrode prepared ex situ by electrodeposition. The most suitable operating conditions and parameters such as pH, ligand concentration (CNN), adsorptive potential (Eads), adsorptive time (tads), scan rate and others were selected and the determination of nickel in aqueous solutions using the standard addition method was possible. The adsorbed Ni-NN complex gives a well defined cathodic stripping peak current at −0.70 V, which was used for the determination of nickel in the concentration range of 10.0-70.0 μg L−1 (pH 7.5; CNN 6.5 μmol L−1; Eads −0.30 V; tads 60 s) with a detection limit of 0.1 μg L−1. The relative standard deviation for a solution containing 10.0 μg L−1 of Ni(II) was 3.5% (n = 4). The proposed method was validated determining Ni(II) in certified reference waste water (SPS-WW1) and Certified Reference Water for Trace Elements (TMDA 51.3) with satisfactory results. Then lake water samples were analyzed.  相似文献

A gas chromatography–mass spectrometric (GC–MS) method has been established for the determination of hydrazine in drinking water and surface water. This method is based on the derivatization of hydrazine with ortho-phthalaldehyde (OPA) in water. The following optimum reaction conditions were established: reagent dosage, 40 mg mL−1 of OPA; pH 2; reaction for 20 min at 70 °C. The organic derivative was extracted with methylene chloride and then measured by GC–MS. Under the established condition, the detection and the quantification limits were 0.002 μg L−1 and 0.007 μg L−1 by using 5.0-mL of surface water or drinking water, respectively. The calibration curve showed good linearity with r2 = 0.9991 (for working range of 0.05–100 μg L−1) and the accuracy was in a range of 95–106%, and the precision of the assay was less than 13% in water. Hydrazine was detected in a concentration range of 0.05–0.14 μg L−1 in 2 samples of 10 raw drinking water samples and in a concentration range of 0.09–0.55 μg L−1 in 4 samples of 10 treated drinking water samples.  相似文献

A cost-effective sequential injection system incorporating with an in-line UV digestion for breakdown of organic matter prior to voltammetric determination of Zn(II), Cd(II), Pb(II) and Cu(II) by anodic stripping voltammetry (ASV) on a hanging mercury drop electrode (HMDE) of a small scale voltammetric cell was developed. A low-cost small scale voltammetric cell was fabricated from disposable pipet tip and microcentrifuge tube with volume of about 3 mL for conveniently incorporated with the SI system. A home-made UV digestion unit was fabricated employing a small size and low wattage UV lamps and flow reactor made from PTFE tubing coiled around the UV lamp. An in-line single standard calibration or a standard addition procedure was developed employing a monosegmented flow technique. Performance of the proposed system was tested for in-line digestion of model water samples containing metal ions and some organic ligands such as strong organic ligand (EDTA) or intermediate organic ligand (humic acid). The wet acid digestion method (USEPA 3010a) was used as a standard digestion method for comparison. Under the optimum conditions, with deposition time of 180 s, linear calibration graphs in range of 10-300 μg L−1 Zn(II), 5-200 μg L−1 Cd(II), 10-200 μg L−1 Pb(II), 20-400 μg L−1 Cu(II) were obtained with detection limit of 3.6, 0.1, 0.7 and 4.3 μg L−1, respectively. Relative standard deviation were 4.2, 2.6, 3.1 and 4.7% for seven replicate analyses of 27 μg L−1 Zn(II), 13 μg L−1 Cd(II), 13 μg L−1 Pb(II) and 27 μg L−1 Cu(II), respectively. The system was validated by certified reference material of trace metals in natural water (SRM 1640 NIST). The developed system was successfully applied for speciation of Cd(II) Pb(II) and Cu(II) in ground water samples collected from nearby zinc mining area.  相似文献

Present study described a simple, sensitive, and viable method for the determination of bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol in water samples using temperature-controlled ionic liquid dispersive liquid-phase microextraction coupled to high performance liquid chromatography-fluorescence detector. In this experiment, 1-octyl-3-methylimidazolium hexafluorophosphate ([C8MIM][PF6]) was used as the extraction solvent, and bisphenol A, 4-n-nonylphenol and 4-tert-octylphenol were selected as the model analytes. Parameters affecting the extraction efficiency such as the volume of [C8MIM][PF6], dissolving temperature, extraction time, sample pH, centrifuging time and salting-out effect have been investigated in detail. Under the optimized conditions, good linear relationship was found in the concentration range of 1.0-100 μg L−1 for BPA, 1.5-150 μg L−1 for 4-NP, and 3-300 μg L−1 for 4-OP, respectively. Limits of detection (LOD, S/N = 3) were in the range of 0.23-0.48 μg L−1. Intra day and inter day precisions (RSDs, n = 6) were in the range of 4.6-5.5% and 8.5-13.3%, respectively. This method has been also successfully applied to analyze the real water samples at two different spiked concentrations and excellent results were obtained.  相似文献

A simple, sensitive and inexpensive flow injection solid phase extraction (SPE) system was developed for automatic determination of trace level concentrations of metals. The potentials of this novel scheme, coupled to flame atomic absorption spectrometry (FAAS), were demonstrated for trace cadmium and lead determination in environmental water samples. The method was based on on-line chelate complex formation of target species with ammonium diethyldithiophosphate (DDTP), retention onto the surface of reversed-phase poly(divinylbenzene-N-vinylpyrrolidone) co-polymeric beads (Oasis HLB) and elution with methanol prior to atomization. A special PVC adapter was designed for easy and rapid replacement of the commercially available SPE cartridge. All main chemical and hydrodynamic parameters affecting the complex formation, sorption and elution of the analyte were optimized thoroughly. Moreover, the effect of potential interfering species occurring in environmental samples was also explored. For 90 s preconcentration time, enhancement factors of 155 and 180, detection limits (3s) of 0.09 μg L− 1 and 0.9 μg L− 1 and relative standard deviations (R.S.D.) of 2.9% (at 4.0 μg L− 1) and 2.6% (at 20.0 μg L− 1) were obtained for cadmium and lead, respectively, with a sample throughput of 24 h− 1. The measurement trueness of the developed method was evaluated by analyzing a certified reference material and spiked environmental water samples. The proposed method is well suited to detect the target elements at concentration levels below the maximum allowed concentrations endorsed by the European Framework Directive (2008/105/EC) in inland and coastal waters.  相似文献

A method for the simultaneous determination of Pb(II), Cd(II), and Zn(II) at low μg L−1 concentration levels by sequential injection analysis-anodic stripping voltammetry (SIA-ASV) using screen-printed carbon nanotubes electrodes (SPCNTE) was developed. A bismuth film was prepared by in situ plating of bismuth on the screen-printed carbon nanotubes electrode. Operational parameters such as ratio of carbon nanotubes to carbon ink, bismuth concentration, deposition time and flow rate during preconcentration step were optimized. Under the optimal conditions, the linear ranges were found to be 2-100 μg L−1 for Pb(II) and Cd(II), and 12-100 μg L−1 for Zn(II). The limits of detection (Sbl/S = 3) were 0.2 μg L−1 for Pb(II), 0.8 μg L−1 for Cd(II) and 11 μg L−1 for Zn(II). The measurement frequency was found to be 10-15 stripping cycle h−1. The present method offers high sensitivity and high throughput for on-line monitoring of trace heavy metals. The practical utility of our method was also demonstrated with the determination of Pb(II), Cd(II), and Zn(II) by spiking procedure in herb samples. Our methodology produced results that were correlated with ICP-AES data. Therefore, we propose a method that can be used for the automatic and sensitive evaluation of heavy metals contaminated in herb items.  相似文献

A novel electrochemical hydride generator has been developed for the determination of As and Sb. This newly devised hydride generator is constructed from a flowing electrolytic cell, in which the tungsten wire is selected as cathode. Compared with some cathode material usually used in electrochemical hydride generator, the tungsten cathode is of better interference tolerance, corrosion-resistant and longer working time. The characteristics of the cathode material, hydride generating efficiency and interferences of concomitant have been studied in detail. The detection limits (3σ) of As and Sb in sample solution were 0.10 μg L−1 and 0.15 μg L−1, the precisions for 11 replicate measurements of 20 μg L−1 As and Sb were 1.3% and 1.7%. The electrochemical hydride generator coupled with atomic fluorescence spectrometry has been applied to the determination of total As and Sb in tobacco samples.  相似文献

The determination of tetracycline by fluorescence spectrophotometry in complex matrices has some difficulties, because the presence of other compounds in the matrix affects the analytical signal. In this work, the effect of some inorganic species that are present in whey milk on the fluorescence signal of tetracycline is studied using a D-optimal experimental design. Next, an experimental strategy is proposed in conjunction with Parallel Factor Analysis, PARAFAC, modeling that leads to suitably modeling the severe matrix effect in the determination of tetracycline in whey milk. A specific design is performed in such a way that the lack of trilinearity due to the effect of the presence of interferents on the signal is obviated. Then, ten test samples from three brands of milk, spiked with different quantities of tetracycline and measured in 2 days were analysed using this methodology (mean of the absolute value of the relative errors: 5.1%). The developed analytical method fulfils the property of trueness, the relative errors being, both in calibration and prediction, inside the interval set by Commission Decision 2002/657/EC at these concentration levels. Decision limits (CCα) at x0 = 0 μg L−1 and at x0 = 100 μg L−1 were 13.2 and 112.4 μg L−1 respectively, for α = 0.05; whereas detection capabilities (CCβ) were 25.9 μg L−1 and 124.4 μg L−1 respectively for α = β = 0.05.  相似文献
