An analytical method based on separation by high performance liquid chromatography (HPLC) and detection by optical fiber (OF) coated with an enzyme (laccase), has been developed for separation and quantification of catecholamines, namely epinephrine, dopamine and norepinephrine. The application of OF as a detector in this analytical system relies on the variation of the reflected optical power detected when the catecholamines eluted from the HPLC column act as the substrate of the laccase immobilized on a tip of a single-mode OF. The developed method shows a high linearity in a range between 5 and 125 pg/mL and detection limits of 3.5, 2.9 and 3.3 pg/mL for epinephrine, dopamine and norepinephrine, respectively. The analytical performance of the proposed method was compared with a classical analytical method, namely high performance liquid chromatography-electrochemical detector (HPLC-ED) regarding catecholamines detection, showing great analytical advantages such as low cost of equipment. Additionally, the proposed method was applied to catecholamines determination in actual samples of plasma and human urine.  相似文献

A simple, efficient, innovative and environmentally friendly analytical technique was successfully applied for the first time for the extraction and preconcentration of polybrominated diphenyl ethers (PBDEs) from water samples. The PBDEs selected for this work were those most commonly found in the literature in natural water samples: 2,2′,4,4′-tetraBDE (BDE-47), 2,2′,4,4,5-pentaBDE (BDE-99), 2,2′,4,4,6-pentaBDE (BDE-100) and 2,2,4,4′,5,5′-hexaBDE (BDE-153). The extracted PBDEs were separated and determined by gas chromatography–mass spectrometry (GC–MS). The extraction/preconcentration technique is based on ultrasound-assisted emulsification-microextraction (USAEME) of a water-immiscible solvent in an aqueous medium. Several variables including, solvent type, extraction time, extraction temperature and matrix modifiers were studied and optimized over the relative response the target analytes. Chloroform was used as extraction solvent in the USAEME technique. Under optimum conditions, the target analytes were quantitatively extracted achieving enrichment factors (EF) higher than 319. The detection limits (LODs) of the analytes for the preconcentration of 10 mL sample volume were within the range 1–2 pg mL−1. The relative standard deviations (RSD) for five replicates at 10 pg mL−1 concentration level were <10.3%. The calibration graphs were linear within the concentration range of 5–5000 pg mL−1 for BDE-47 and BDE-100; and 5–10,000 pg mL−1 for BDE-99 and BDE-153, respectively. The coefficients of estimation were ≥0.9985. Validation of the methodology was performed by standard addition method at two concentration levels (10 and 50 pg mL−1). Recovery values were ≥96%, which showed a successful robustness of the analytical methodology for determination of picogram per milliliter of PBDEs in water samples. Significant quantities of PBDEs were not found in the analyzed samples.  相似文献

A fully automated cold fiber solid phase microextraction device has been developed by coupling to a GERSTEL multipurpose (MPS 2) autosampler and applied to the analysis of volatiles and semi-volatiles in aqueous and solid matrices. The proposed device was thoroughly evaluated for its extraction performance, robustness, reproducibility and reliability by gas chromatograph/mass spectrometer (GC/MS). With the use of a septumless head injector, the entire automated setup was capable of analyzing over 200 samples without any GC injector leakages. Evaluation of the automated cold fiber device was carried out using a group of compounds characterized by different volatilities and polarities. Extraction efficiency as well as analytical figures of merit was compared to commercial solid phase microextraction fibers. The automated cold fiber device showed significantly improved extraction efficiency compared to the commercial polydimethylsiloxane (PDMS) and cold fiber without cooling for the analysis of aqueous standard samples due to the low temperature of the coating. Comparing results obtained from cold fiber and commercial divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fiber temperature profile demonstrated that the temperature gap between the sample matrix and the coating improved the distribution coefficient and therefore the extraction amount. The linear dynamic range of the cold fiber device was 0.5 ng mL−1 to 100 ng mL−1 with a linear regression coefficient ≥0.9963 for all compounds. The limit of detection for all analytes ranged from 1.0 ng mL−1 to 9.4 ng mL−1. The newly automated cold fiber device presents a platform for headspace analysis of volatiles and semi-volatiles for large number of samples with improved throughput and sensitivity.  相似文献

To address people's concerns of exogenous oxytocin (OT) administration to lactating bovines, a study was undertaken to (a) establish an enzyme immunoassay (EIA) for OT determination in milk, (b) quantify OT in milk of cows administered OT, and (c) study influence of pasteurization on OT stability in milk. A sensitive EIA validated according to the criteria of European Union—Decision 2002/657/EC was developed for OT in skim milk in an analytical range of 10-250 pg mL−1 with a decision limit (CCα) of 30 pg mL−1 and detection capability (CCβ) of 41.5 pg mL−1. Milk samples collected from cows (n = 38) administered either 25 or 50 IU OT prior to milking were investigated for the presence of OT. There was no significant difference among both groups with the mean concentrations of OT being 15.8 and 14.9 pg mL−1 for cows subjected to 25 and 50 IU OT administration, respectively. The OT levels in skim milk of control cows (n = 30; untreated) were basal (around 10 pg mL−1). All the analyzed milk samples were below the CCα value of 30 pg mL−1. Pasteurization of OT spiked milk samples at different temperature and sample holding conditions reduced the immunological activity of OT to 43% at 110 °C. However, no further decline occurred in the immunological activity with increased pasteurization temperature and time. It was concluded that the milk OT concentrations after OT administrations were minimal and below the assay decision limit. However, OT was quite stable to pasteurization in OT spiked milk.  相似文献

A carbon monolith was synthesized via a polymerization–carbonization method, styrene and divinylbenzene being adopted as precursors and dodecanol as a porogen during polymerization. The resultant monolith had bimodal porous substructure, narrowly distributed nano skeleton pores and uniform textural pores or throughpores. The carbon monolith was directly used as an extracting fiber, taking place of the coated silica fibers in commercially available solid-phase microextraction device, for the extraction of phenols followed by gas chromatography–mass spectrometry. Under the studied conditions, the calibration curves were linear from 0.5 to 50 ng mL−1 for phenol, o-nitrophenol, 2,4-dichlorophenol and p-chlorophenol. The limits of detection were between 0.04 and 0.43 ng mL−1. The recoveries of the phenols spiked in real water samples at 10 ng mL−1 were between 85% and 98% with the relative standard deviations below 10%. Compared with the commercial coated ones (e.g. PDMS, CW/DVB and DVB/CAR/PDMS), the carbon monolith-based fiber had advantages of faster extraction equilibrium and higher extraction capacity due to the superior pore connectivity and pore openness resulting from its bimodal porous substructure.  相似文献

The simultaneous determination of catecholamines - epinephrine and norepinephrine by square wave voltammetry (SWV) at physiological pH 7.2 is reported using multi-walled carbon nanotubes modified edge plane pyrolytic graphite electrode (MWNT/EPPGE). A broad bump at ∼250 mV is appeared for the oxidation of epinephrine (EP) and norepinephrine (NE) at bare EPPGE whereas at MWNT/EPPGE two well-separated peaks at ∼150 and ∼215 mV are appeared for the oxidation of EP and NE, respectively. The oxidation peak current of both the neurotransmitters also increased significantly along with the negative shift of peak potentials using MWNT/EPPGE. The oxidation of both compounds occurred in a pH dependent, 2e and 2H+ process and the electrode reaction followed diffusion controlled pathway. Linear calibration curves were obtained for epinephrine and norepinephrine in the range 0.5-100 nM with limits of detection 0.15 × 10−9 and 0.90 × 10−10 M, respectively. The developed protocol is implemented for the simultaneous determination of epinephrine and norepinephrine in blood plasma and urine samples of smokers as well as in athletes.  相似文献

Fragrance suspected allergens including those regulated by the EU Directive 76/768/EEC have been determined in different types of waters using solid-phase microextraction (SPME) and gas chromatography-mass spectrometry (GC-MS). The procedure was based on headspace sampling (HS-SPME) using polydimethylsiloxane/divinylbenzene (PDMS/DVB) fibers and has been optimized by an experimental design approach. The method performance has been studied showing good linearity (R ≥ 0.994) as well as good intra-day and inter-day precision (RSD ≤ 12%). Detection limits (S/N = 3) ranged from 0.001 to 0.3 ng mL−1. Reliability was demonstrated through the quantitative recoveries of the compounds in real water samples, including baby bathwaters, swimming pool waters, and wastewaters. The absence of matrix effects allowed quantification of the compounds by external aqueous calibration. The analysis of 35 samples of different types of waters showed the presence of suspected allergens in all the analyzed samples. All targets were found in the samples, with the exception of methyl eugenol and amyl cinnamic alcohol. Highest concentrations of suspected allergens were present in baby bathwaters, containing from 5 to 15 of the compounds at concentrations ranging from few pg mL−1 to several hundreds of ng mL−1.  相似文献

A method based on solid-phase microextraction (SPME) followed by gas chromatography with microwave-induced plasma atomic emission detection for determining dimethylselenide (DMSe) and dimethyldiselenide (DMDSe) in milk and milk by-products is proposed. Parameters affecting the SPME, such as sample volume or mass, ionic strength, adsorption and desorption times and temperatures were optimized in the headspace mode. The matrix effect was evaluated for the different samples studied, concluding that standard additions calibration was required for quantification purposes. The detection limits ranged from 70 to 110 pg mL−1 for DMSe and from 80 to 400 pg mL−1 for DMDSe, depending on the sample under analysis. None of the twenty-three samples analyzed contained the studied compounds at concentrations above the corresponding detection limits.  相似文献

An orthogonal array design (OAD) was applied to optimize microwave-assisted derivatization (MAD) for analysis of trace amphetamine (AM) and methamphetamine (MA) by negative chemical ionization gas chromatography–mass spectrometry (NCI GC–MS). The 2,3,4,5,6-pentafluorobenzoyl chloride (PFBC) was used as a derivatization reagent. Experimental factors including solvent, microwave power, and irradiation time at four-levels were studied in 16 trials by OAD16 (44). The significance of these factors was investigated using analysis of variance (ANOVA) and percent contribution (PC). Solvent is statistically demonstrated a chief factor; microwave power and irradiation time are secondary factors. Under the optimum condition, calibration curve of AM is linear over a range from 0.01 to 100 ng mL−1 with correlation coefficient 0.9988, and MA from 0.1 to 1000 ng mL−1 with correlation coefficient 0.9951. The limit of detection (LOD) is 1.20 pg mL−1 for AM and 13.04 pg mL−1 for MA. An applicability of the method was tested by analyzing urine samples from amphetamine-type stimulants (ATS)-abusing suspects. Consequently, the OAD method not only optimizes the MAD condition for determination of trace AM and MA, but identifies the effects of factor solvent, microwave power and irradiation time on the MAD performance.  相似文献

For the first time, electrospun composite nanofibers comprising polymeric crown ether with polystyrene (PCE‐PS) have been used for the selective extraction of catecholamines – dopamine (DA), norepinephrine (NE) and epinephrine (E) – prior to their analysis by high‐performance liquid chromatography–electrochemical detection. Using a minicartridge packed with PCE‐PS composite nanofibers, the target compounds were extracted effectively from urine samples to which diphenylborinic acid 2‐aminoethyl ester was added as a complexing reagent. The extracted catecholamines could be liberated from the fiber by the addition of acetic acid. A good linearity was observed for catecholamines in the range of 2.0–200 ng mL?1 (NE, E and DA). The detection limits of catecholamines (signal‐to‐noise ratio = 3) were 0.5 ng mL?1 (NE), 0.2 ng mL?1 (E) and 0.2 ng mL?1 (DA), respectively. Under the optimized conditions, the absolute recoveries of the above three catecholamines were 90.6% (NE), 88.5% (E) and 94.5% (DA). The repeatability of extraction performance was from 5.4 to 9.2% (expressed as relative standard deviation). A novel and efficient analytical methodology is proposed for extracting and preconcentrating polybrominated diphenyl ethers (PBDEs) from samples of environmental interest prior gas chromatography–mass spectrometry (GC–MS) analysis. It is based on the induction of micellar organized medium by using a non-ionic surfactant (Triton X-114) to extract the target PBDEs. To enable coupling the efficient extracting technique with GC analysis, ultrasound-assisted back-extraction (UABE) into an organic solvent was required. Several factors, including surfactant type and concentration, equilibration temperature and time, ionic strength, pH and buffers nature and concentration were studied and optimized over the extraction efficiency of the proposed technique. Under optimal experimental conditions, the target analytes were quantitatively extracted achieving an enrichment factor of 250 when 10 mL aliquot of ultrapure water spiked with PBDE-standard mixture (10 pg mL−1 each PBDE) was extracted. Method detection limits (MDLs) calculated with aqueous PBDEs solutions as three times the signal-to-noise ratio (S/N), ranged from 1 to 2 pg mL−1 with RSDs values ≤8.5% (n = 5). The coefficients of estimation of the calibration curves obtained following the proposed methodology were ≥0.9987 and linear range of all PBDEs was 4–150 pg mL−1. The proposed methodology was validated by carrying out a recovery study by spiking the samples at two different concentration levels of PBDEs (10 and 50 pg mL−1 for waters samples). Recoveries values in the range of 96–106% for water samples were obtained showing satisfactory robustness of the method for analyzing PBDEs in water samples. The proposed methodology was applied for the analysis of PBDEs: 2,2′,4,4′-tetraBDE (BDE-47), 2,2′,4,4,5-pentaBDE (BDE-99), 2,2′,4,4,6-pentaBDE (BDE-100) and 2,2,4,4′,5,5′-hexaBDE (BDE-153) in water samples, including drinking, lake, river water and soil samples. In the presented work, a disposable immunosensor for the detection of testosterone, an endogenous steroid hormone, in bovine urine has been developed using screen-printed electrodes (SPEs). Due to concerns over the use of steroid hormones as growth promoters, the EU prohibits their use in food producing animals. Consequently, rigorous screening procedures have been implemented in all member states to detect the illegal administration of such compounds. Competitive immunoassays were developed, initially by enzyme linked immunosorbent assay (ELISA), and subsequently transferred to an electrochemical immunosensor format using disposable screen-printed carbon electrodes. Horseradish peroxidase (HRP) was the enzyme label of choice and chronoamperometric detection was carried out using a tetramethylbenzidine/hydrogen peroxide (TMB/H2O2) substrate system, at +100 mV. The EC50 values obtained for the assay in buffer and urine gave relatively comparable results, 710 pg mL−1 and 960 pg mL−1, respectively. The linear range obtained for the assay in buffer extended from 0.03 ng mL−1 to 40 ng mL−1; while that in urine ranged from 0.03 ng mL−1 to 1.6 ng mL−1. The corresponding limits of detection (LOD) in buffer and urine were 26 pg mL−1 and 1.8 pg mL−1. Cross reactivity profiles of the antibody have been examined, with notable cross reactivities with 19-nortestosterone (11.6%) and boldenone (9.86%). Precision studies for the sensor demonstrated adequate reproducibility (CV < 13%, n = 3) and repeatability (CV < 9%, n = 3). Recovery data obtained showed good agreement between spiking studies and known concentrations of analyte. Sensors showed stability for 4 days at +4 °C. A sensitive, highly specific, inexpensive, disposable immunosensor, showing excellent overall performance for the detection of testosterone in bovine urine, has been developed.  相似文献

Three-dimensional macroporous gold nanoparticles/graphene composites (3D-AuNPs/GN) were synthesized through a simple two-step process, and were used to modify working electrode sensing platform, based on which a facile electrochemical immunoassay for sensitive detection of carcinoembryonic antigen (CEA) in human serum was developed. In the proposed 3D-AuNPs/GN, AuNPs were distributed not just on the surface, but also on the inside of graphene. And this distribution property increased the area of sensing surface, resulting in capturing more primary antibodies as well as improving the electronic transmission rate. In the presence of CEA, a sandwich-type immune composite was formed on the sensing platform, and the horseradish peroxidase-labeled anti-CEA antibody (HRP-Ab2)/thionine/nanoporous silver (HRP-Ab2/TH/NPS) signal label was captured. Under optimal conditions, the electrochemical immunosensor exhibited excellent analytical performance: the detection range of CEA is from 0.001 to 10 ng mL−1 with low detection limit of 0.35 pg mL−1 and low limit of quantitation (LOQ) of 0.85 pg mL−1. The electrochemical immunosensor showed good precision, acceptable stability and reproducibility, and could be used for the detection of CEA in real samples. Two simple, rapid and specific analytical methods for 13 catecholamines and their metabolites have been developed based on liquid chromatography tandem mass spectrometry in a multiple reaction monitoring mode. Tyrosine, dopamine, dihydroxyphenylalanine, epinephrine, norepinephrine, 3-methoxytyramine, normetanephrine, metanephrine and isoproterenol (internal standard) were separated on a Kromasil™ Cyano analytical column by a mobile phase consisting of 60% (v/v) acetonitrile and 40% (v/v) water adjusted with formic acid to pH 3.0, and detected by positive ionization electrospray tandem mass spectrometry. While vanillymandelic acid, 3,4-dihydroxymandelic acid, homovanillic acid, 3,4-dihydroxyphenylacetic acid, 4-hydroxy-3-methoxyphenylglycol and 5-hydroxy-2-indolecarboxylic acid (internal standard) were separated on a reversed-phase Shim-Pak VP-ODS column with the mobile phase of 60% (v/v) acetonitrile, and 40% (v/v) water adjusted with formic acid to pH 4.5 and detected in the negative ionization electrospray tandem mass spectrometry. The influence of various parameters such as column type and mobile phase composition on separation and sensitivity were investigated. The limits of detection were in the range of 0.5-20 ng mL−1. The mean recoveries determined from three different concentrations of each analyte were above 85.4%. The precision of the method calculated as relative standard deviation was lower than 5.3%. Deduced from the results of real sample analysis, adrenal gland synthesizes and stores the catecholamine hormones norepinephrine and epinephrine.  相似文献

A new kind of signal amplification strategy based on ferrocene (Fc) incorporated polystyrene spheres (PS-Fc) was proposed. The synthesized PS-Fc displayed narrow size distribution and good stability. PS-Fc was applied as label to develop immunosensors for prostate specific antigen (PSA) after the typical sandwich immunoreaction by linking anti-PSA antibody (Ab2) onto PS-Fc. After the fabrication of the immunosensor, tetrahydrofuran (THF) was dropped to dissolve PS and release the contained Fc for the following stripping voltammetric detection. PS-Fc as a new electrochemical label prevented the leakage of Fc and greatly amplified the immunosensor signal. In addition, the good biocompatibility of PS could maintain the bioactivity of the antibodies. The response current was linear to the logarithm of PSA concentration in the range from 0.01 ng mL−1 to 20 ng mL−1 with a detection limit of 1 pg mL−1. The immunosensor results were validated through the detection of PSA in serum samples with satisfactory results.  相似文献

In this study, detection of staphylococcal enterotoxin A (SEA) in multi-matrices using a highly sensitive and specific microplate chemiluminescence enzyme immunoassay (CLEIA) has been established. A pair of monoclonal antibodies (mAbs) was selected from 37 anti-SEA mAbs by pairwise analysis, and the experimental conditions of the CLEIA were optimized. This CLEIA exhibited high performance with a wide dynamic range from 6.4 pg mL−1 to 1600 pg mL−1, and the measured low limit of detection (LOD) was 3.2 pg mL−1. No cross-reactivity was observed when this method was applied to test SEB, SEC1, and SED. It has also been successfully applied for analyzing SEA in a variety of environmental, biological, and clinical matrices, such as sewage, tap water, river water, roast beef, peanut butter, cured ham, 10% nonfat dry milk, milk, orange juice, human urine, and serum. Thus, the highly sensitive and SEA-specific CLEIA should make it attractive for quantifying SEA in public health and diagnosis in near future.  相似文献

A novel technique for derivatization in a gas chromatograph injection port after a one-step extraction of trace perfluorocarboxylic acids (PFCAs) in water with ion pair formation during dispersive liquid–liquid microextraction (DLLME) was investigated. Tetrabutylammonium hydrogen sulfate (TBAHS) was used as the ion pair reagent. PFCA butyl ester derivatives were formed in the GC injection port and then analyzed using gas chromatography coupled to tandem mass spectrometry with negative chemical ionization. According to our analysis, the operative linear range for PFCA detection from 250 pg mL−1 to 2 μg mL−1 with a relative standard derivation (RSD) below 13%. Detection limits were achieved at the level of 37–51 pg mL−1. This method was successfully applied for the analyzing of PFCAs in river water samples from urban and industrial areas without tedious pretreatment. The concentration range over which PFCAs were detected is from 0.6 ng mL−1 to 604.9 ng mL−1.  相似文献

This work describes the preparation of an electrochemical immunosensor for estradiol based on the surface modification of a screen printed carbon electrode with grafted p-aminobenzoic acid followed by covalent binding of streptavidin (Strept) and immobilization of biotinylated anti-estradiol (anti-estradiol-Biotin). The hormone determination was performed by applying a competitive immunoassay with peroxidase-labelled estradiol (HRP–estradiol) and measurement of the amperometric response at −200 mV using hydroquinone (HQ) as redox mediator. The calibration curve for estradiol exhibited a linear range between 1 and 250 pg mL−1 (r = 0.990) and a detection limit of 0.77 pg mL−1 was achieved. Cross-reactivity studies with other hormones related with estradiol at physiological concentration levels revealed the practical specificity of the developed method for estradiol. A good reproducibility, with RSD = 5.9% (n = 8) was also observed. The operating stability of a single bioelectrode modified with anti-estradiol-Biotin-Strept was nine days when it was stored at 8 °C under humid conditions between measurements. The developed immunosensor was applied to the analysis of certified serum and spiked urine samples with good results.  相似文献

Solid-phase microextraction (SPME) coupled with high-performance liquid chromatography (HPLC) for the determination of triazine is described. Carbowax/templated resin (CW/TPR, 50 μm), polydimethylsiloxane/divinylbenzene (PDMS/DVB, 60 μm), polydimethylsiloxane (PDMS, 100 μm), and polyacrylate (PA, 85 μm) fibers were evaluated for extraction of the triazines. CW/TPR and PDMS/DVB fibers were selected for further study. Several parameters of the extraction and desorption procedure were studied and optimized (such as types of fibers, desorption mode, desorption time, compositions of solvent for desorption, soaking periods and the flow rate during desorption period, extraction time, temperature, pH, and ionic strength of samples). Both CW/TPR and PDMS/DVB fibers are acceptable; a simple calibration-curve method based on simple aqueous standards can be used. The linearity of this method for analyzing standard solution has been investigated over the range 5-1000 ng mL−1 for both PDMS/DVB and CW/TPR fibers. All the correlation coefficients in the range 5-1000 ng mL−1 were better than 0.995 except Simazine and Atratone by CW/TPR fiber. The R.S.D.s range from 4.4% to 8.8 % (PDMS/DVB fiber) and from 2.4% to 7.2% (CW/TPR fiber). Method-detection limits (MDL) are in the range 1.2-2.6 and 2.8-3.4 ng mL−1 for the two fibers. These methods were applied to the determination of trazines in environmental water samples (lake water).  相似文献

In this work, we reported a sandwiched luminol electrochemiluminescence (ECL) immunosensor using ZnO nanoparticles (ZnONPs) and glucose oxidase (GOD) decorated graphene as labels and in situ generated hydrogen peroxide as coreactant. In order to construct the base of the immunosensor, a hybrid architecture of Au nanoparticles and graphene by reduction of HAuCl4 and graphene oxide (GO) with ascorbic acid was prepared. The resulted hybrid architecture modified electrode provided an excellent platform for immobilization of antibody with good bioactivity and stability. Then, ZnONPs and GOD functionalized graphene labeled secondary antibody was designed for fabricating a novel sandwiched ECL immunosensor. Enhanced sensitivity was obtained by in situ generating hydrogen peroxide with glucose oxidase and the catalysis of ZnONPs to the ECL reaction of luminol–H2O2 system. The as-prepared ECL immunosensor exhibited excellent analytical property for the detection of carcinoembryonic antigen (CEA) in the range from 10 pg mL−1 to 80 ng mL−1 and with a detection limit of 3.3 pg mL−1 (S N−1 = 3). The amplification strategy performed good promise for clinical application of screening of cancer biomarkers.  相似文献
