Described herein is an effective and practical modular flow design for the meta-selective C−H arylation of anilines. The design consists of four continuous-flow modules (i.e., diaryliodonium salt synthesis, meta-selective C−H arylation, inline copper extraction, and aniline deprotection) which can be operated either individually or consecutively to provide direct access to meta-arylated anilines. With a total residence time of 1 hour, the desired product could be obtained in high yield and excellent purity without the need for column chromatography, and the residual copper content meets the standards for parenterally administered pharmaceutical substances.  相似文献

The meta ‐C−H arylation of free phenylacetic acid was realized using 2‐carbomethoxynorbornene (NBE‐CO2Me) as a transient mediator. Both the modified norbornene and the mono‐protected 3‐amino‐2‐hydroxypyridine type ligand are crucial for this auxiliary‐free meta ‐C−H arylation reaction. A series of phenylacetic acids, including mandelic acid and phenylglycine, react smoothly with various aryl iodides to provide the meta ‐arylated products in high yields.  相似文献

Ruthenium(II)‐catalyzed meta ‐selective C−H (di)fluoromethylation was accomplished by phosphine and carboxylate cooperation. The remote C−H functionalization was characterized by ample substrate scope, thereby setting the stage for meta ‐(di)fluoromethylation through facile C−H cleavage.  相似文献

A copper‐mediated decarboxylative coupling of benzamides with ortho ‐nitrobenzoic acids by 8‐aminoquinoline‐directed C−H cleavage has been developed. This reaction proceeds smoothly with only a copper salt to produce the corresponding biaryl compounds in good yields. The products can be easily transformed into various nitrogen‐containing heterocyclic compounds. Moreover, the combination of copper and a suitable base promotes a decarboxylative C−H arylation and cyclization sequence to deliver phenanthridinone derivatives in one pot.  相似文献

Methods for positionally selective remote C−H functionalizations are in high demand. Herein, we disclose the first heterogeneous ruthenium catalyst for meta ‐selective C−H functionalizations, which enabled remote halogenations with excellent site selectivity and ample scope. The versatile heterogeneous Ru@SiO2 catalyst was broadly applicable and could be easily recovered and reused, which set the stage for the direct fluorescent labeling of purines. In contrast to palladium, rhodium, iridium, or cobalt complexes, solely the ruthenium catalysis manifold provided access to meta ‐halogenated purine derivatives, illustrating the unique power of ruthenium C−H activation catalysis.  相似文献

The para ‐selective C−H alkylation of aniline derivatives furnished with a pyrimidine auxiliary is herein reported. This reaction is proposed to take place via an N−H‐activated cyclometalate formed in situ. Experimental and DFT mechanistic studies elucidate a dual role of the ruthenium catalyst. Here the ruthenium catalyst can undergo cyclometalation by N−H metalation (as opposed to C−H metalation in meta ‐selective processes) and form a redox active ruthenium species, to enable site‐selective radical addition at the para position.  相似文献

Palladium(II)‐catalyzed meta ‐C−H arylation and alkylation of benzylsulfonamide using 2‐carbomethoxynorbornene (NBE‐CO2Me) as a transient mediator are realized by using a newly developed electron‐deficient directing group and isoquinoline as a ligand. This protocol features broad substrate scope and excellent functional‐group tolerance. The meta ‐substituted benyzlsulfonamides can be readily transformed into sodium sulfonates, sulfonate esters, and sulfonamides, as well as styrenes by Julia‐type olefination. The unique impact of the isoquinoline ligand underscores the importance of subtle matching between ligands and the directing groups.  相似文献

Ligand development for rhodium(III)‐catalyzed C−H activation reactions has largely been limited to cyclopentadienyl (Cp) based scaffolds. 2‐Methylquinoline has now been identified as a feasible ligand that can coordinate to the metal center of Cp*RhCl to accelerate the cleavage of the C−H bond of N ‐pentafluorophenylbenzamides, providing a new structural lead for ligand design. The compatibility of this reaction with secondary free amines and anilines also overcomes the limitations of palladium(II)‐catalyzed C−H amination reactions.  相似文献

C−H activation is a versatile tool for appending aryl groups to aromatic systems. However, heavy demands on multiple catalytic cycle operations and site‐selectivity have limited its use for graphene segment synthesis. A Pd‐catal‐ yzed one‐step synthesis of functionalized triphenylene frameworks is disclosed, which proceeds by 2‐ or 4‐fold C−H arylation of unactivated benzene derivatives. A Pd2(dibenzylideneacetone)3 catalytic system, using cyclic diaryliodonium salts as π‐extending agents, leads to site‐selective inter‐ and intramolecular tandem arylation sequences. Moreover, N ‐substituted triphenylenes are applied to a field‐effect transistor sensor for rapid, sensitive, and reversible alcohol vapor detection.  相似文献

The radical arylation of the para‐substituted anilines under three different conditions (A: arylhydrazines as the radical precursors and MnO2 as the oxidant in acetonitrile; B: arylhydrazine hydrochlorides as the radical precursors and O2 as the oxidant in aqueous sodium hydroxide solution; C: arenediazonium salts as the radical precursors and TiCl3 as the reductant in aqueous hydrochloric acid solution) has been theoretically studied and the origins of the ortho/meta regioselectivity have been explored. The arylation process is suggested to contain three steps: radical generation, radical addition, and rearomatization. Calculations show that the arylation of the neutral anilines is kinetically controlled under conditions A and B, and the regioselectivity is determined by the radical addition. As a directing group, ? NH2 plays an important role in these cases with the assistance of Mn(OH)2 (the reduced product of MnO2 under condition A) and Na+ (condition B). As for the arylation of the protonated anilines under condition C, the regioselectivity is affected by the substituents in the para‐position of anilines. Electron‐donating groups support meta‐addition and the selectivity is decided by the radical addition. Conversely, electron‐withdrawing groups favor ortho‐addition, and in this situation the arylation process is thermodynamically controlled and the regioselectivity is determined by the radical addition and rearomatization. © 2015 Wiley Periodicals, Inc.  相似文献

A novel copper‐catalyzed C?H arylation of heteroarenes with anilines by an in situ diazonium reaction is established by using silver nitrite (AgONO) as an unconventional nitrosating reagent under acid‐free conditions. It provides a complementary approach for the C?H arylation of electron‐rich heteroarenes with aromatic amines affording a variety of heterobiaryls in moderate to good yields.  相似文献

Mono‐, di‐, tri‐, and tetraarylated thieno[3,2‐b ]thiophenes were synthesized by direct site‐selective Pd‐catalyzed C−H activation reactions with various aryl bromides in the presence of a phosphine‐free Pd(OAc)2/KOAc catalyst system in N ,N ‐dimethylacetamide (DMAc). The arylation of 2‐arylthieno[3,2‐b ]thiophene took place at the C3 position if the 2‐aryl substituents possessed electron‐withdrawing groups and at the C5 position if they were bulky and possessed electron‐donating groups.  相似文献

An easily removable pyrimidine‐based auxiliary has been employed for the remote meta ‐C−H cyanation of arenes. The scope of this Pd‐catalyzed cyanation reaction using copper(I) cyanide as the cyanating agent was demonstrated with benzylsilanes, benzylsulfonates, benzylphophonates, phenethylsulfonates, and phenethyl ether derivatives. The method was utilized for the synthesis of pharmaceutically valuable precursors.  相似文献

Palladium‐catalyzed activation of remote meta ‐C−H bonds in arenes containing tethered alcohols was achieved with high regioselectivity by using a nitrile template. Computational studies on the macrocyclic transition state of the regioselectivity‐determining C−H activation steps revealed that both the C‐N‐Ag angles and gauche comformations of phenyl ether play an extremely important role in the meta selectivity.  相似文献

Fluorine is known to promote ortho‐C?H metalation. Based upon this reactivity, we employed an activated norbornene that traps the ortho‐palladation intermediate and is then relayed to the meta position, leading to meta‐selective C?H arylation of fluoroarenes. Deuterium experiment suggests that this meta‐arylation is initiated by ortho C?H activation and the catalytic cycle is terminated by C‐2 protonation. A dual‐ligand system is crucial for the observed high reactivity and site selectivity. Applying this approach to simple benzene or other arenes also affords arylation products with good yield and site selectivity.  相似文献

The arylation of pyrazole and derivatives can be achieved by coupling arenediazonium species (formed in situ from anilines) by using a catalytic system that employs low‐toxicity and inexpensive copper metal under very mild and ligand‐free conditions (T=20 °C). From other nitrogen heterocycles, the presence of an additive (NBu4I) significantly improves the efficiency of the catalytic system. These results represent the first examples of C?N bond formation from arenediazonium species.  相似文献

Rhodium‐catalyzed ortho ‐C−H functionalization is well known in the literature. Described herein is the Xphos‐supported rhodium catalysis of meta ‐C−H olefination of benzylsulfonic acid and phenyl acetic acid frameworks with the assistance of a para ‐methoxy‐substituted cyano phenol as the directing group. Complete mono‐selectivity is observed for both scaffolds. A wide range of olefins and functional groups attached to arene are tolerated in this protocol.  相似文献

Biaryl compounds are extremely important structural motifs in natural products, biologically active components and pharmaceuticals. Selective synthesis of biaryls by distinguishing the subtle reactivity difference of distal arene C−H bonds are significantly challenging. Herein, we describe para-selective C−H arylation, which is acheived by a unique combination of a meta-directing group and norbornene as a transient mediator. Upon direct meta-C−H palladation, one-bond relay palladation occurs in presence of norbornene and subsequently para-C−H arylation is achieved for sulfonates, phosphonates and phenols bearing 2,6-disubstitution patterns. The protocol is amenable to electron-deficient aryl iodides. Multisubstituted arenes and phenols are obtained by postsynthetic modification of the products. The protocol allows the synthesis of hexa-substituted benzene by sequential selective distal C−H functionalization.  相似文献

A rare example of C(sp3)?H functionalization of simple alkanes with unactivated alkenes is presented. In the presence of a copper salt and di‐tert‐butyl peroxide (DTBP), N‐allyl anilines underwent exo‐selective alkylation/cyclization cascade with unactivated alkenic bonds as radical acceptors and simple alkanes as radical precursors, providing a direct access to 3‐alkyl indolines. The present protocol features simple operation, broad substrate scope and great exo selectivity.  相似文献

A highly site‐selective, heteroatom‐guided, palladium‐catalyzed direct arylation of 4H‐chromenes is reported. The C?H functionalization is driven not only by the substituents and structure of the substrate but also by the coupling partner being used. The diastereoselective assembly of the core structure of Myristinin B has been achieved by using a dual C?H functionalization strategy for regioselective direct arylation.  相似文献
