以手性 H’4-NOBIN为原料合成了新型手性氨基酚2和N,P配体5, 并将化合物2应用于催化二乙基锌对醛的不对称加成反应, 产率达90%, 对映体过量最高为45.9% ee. 化合物5用于钯催化的1,3-二苯基-2-烯丙基乙酸酯的不对称烯丙基烷基化反应, 产率为89%, 对映体过量最高为81.6% ee. 结果表明氨基酚2手性诱导作用弱于未氢化的NOBIN, 但却高于其八氢衍生物, 而N,P配体5给出相反的结果.  相似文献

The synthesis of 2-(2-diphenylphosphinophenyl)-oxazolines from o-fluoroben-zonitrile and (-)-norephedrine or (+)-endo-2-hydroxy-endo-3-aminobornane is described. The Pa-catalyzed alkylation of (E)-1,3-diphenylallyl acetate with the sodium salt of dimethylmalonate using these chiral ligands occurs with an 82–87% yield and 88–93% ee.  相似文献

The enantioselective alkylation of aryl aldehydes by diethylzinc in the presence of catalytic amounts of several chiral pyrolidine- and piperidine-based amino alcohol ligands, synthesized from the reaction of (R)-2-amino-1-butanol and (S)-2-amino-3-phenylpropan-1-ol with appropriate dibromoalkanes, was studied. The influence of temperature and ligand structure has been investigated. Addition of diethylzinc to aromatic aldehydes under the optimized condition gave the corresponding products in excellent yields with ee values of up to 77%.  相似文献

The asymmetric allylic alkylation of racemic 1,3-diphenyl-2-propenyl acetate 3 with dimethyl malonate proceeded smoothly in the presence of lithium acetate, BSA (N,O-bis(trimethylsilyl)acetamide), [Pd(η3-C3H5)Cl]2, and chiral tert-butanesulfinylphosphine ligand 2c to give the allylic alkylation product in good yield and high enantiomeric excess (up to 93% ee), while the enantioselectivities of allylic amination of 3 with various amines were moderate (up to 76% ee).  相似文献

Substituted acetic acids and formamides react in the presence of phosphorus oxychloride to yield the vinamidinium hexafluorophosphate salts 5a-d, 6a-d, and 7 in moderate to good unoptimized recrystallized yields (40-67%) as easily handled nonhygroscopic solids. The 1,3-differentially substituted vinamidinium salts 8 was prepared by amine exchange in 81% yield as are the cyclic diazapinium salts 9 and 10 in > 76% yield. The symmetrical 2-chlorovinamidinium 11 was prepared by displacement of 3 in 71% yield. The 2-chlorovinamidinium salts are cleanly reduced to the parent vinamidinium salts 12-16 using HI or PPh3/pTSA in up to 99% assay yield.  相似文献

The asymmetric allylic alkylation of rac-1,3-diphenyl-2-propenyl acetate 1 with dimethyl malonate 2a proceeded smoothly in the presence of lithium acetate, BSA (N,O-bis(trimethylsilyl)acetamide), [Pd(η3-C3H5)Cl]2, and the chiral ligand (R)-i-Pr2N-PHEST (R)-5a to give the allylic alkylation product (R)-3a in 89% yield with 99% ee. Furthermore, the asymmetric allylic amination of 1 with potassium phthalimide 2c has been carried out using the same ligand to give the allylic amination product (S)-3c in 10% yield with 66% ee.  相似文献

[structure: see text] Various N-benzylcinchonidinium salts were prepared to study electronic factors in the catalytic enantioselective phase-transfer alkylation of glycine anion equivalent. An ortho-fluoro substituent on the benzyl group in the quaternary ammonium salt dramatically increased the enantioselectivity in the alkylation. O(9)-Allyl-N-2',3',4'-trifluorobenzylhydrocinchonidinium bromide (27), which gave the highest enantioselectivity of the catalysts studied, was used to prepare 12 alpha-alkylated amino acid derivatives in 94 approximately >99% ee.  相似文献

Palladium nanoparticles (4 nm, fcc) were prepared through decomposition of [Pd2(dba)3] by H2 in the presence of a chiral xylofuranoside diphosphite. These particles catalyze the allylic alkylation of rac-3-acetoxy-1,3-diphenyl-1-propene with dimethyl malonate leading to an almost total conversion of the (R) enantiomer and almost no reaction with the (S). This gives rise to 97% ee for the alkylation product and a kinetic resolution of the substrate recovered with ca. 90% ee. This behavior was compared to that of a molecular catalyst at various dilutions, and the differences between the two systems are discussed. This is the first colloidal system shown to display such a high enantioselectivity besides the well-known Pt/cinchonidine system.  相似文献

Catalytic enantioselective alkylations of three classes of ketoimines are reported. Reactions are promoted in the presence 0.5-10 mol % of a Zr salt and a chiral ligand that contains two inexpensive amino acids (valine and phenylalanine) and involve Me2Zn or Et2Zn as alkylating agents. Requisite aryl- and alkyl-substituted alpha-ketoimine esters, accessed readily and in >80% yield on gram scale through a two-step sequence from the corresponding ketones, undergo alkylation to afford quaternary alpha-amino esters in 79-97% ee. Aryl-substituted trifluoroketoimines are converted to the corresponding amines by reactions with Me2Zn, catalyzed by a chiral complex that bears a modified N-terminus. The utility of the catalytic asymmetric protocols is illustrated through conversion of the enantiomerically enriched alkylation products to a range of cyclic and acyclic compounds bearing an N-substituted quaternary carbon stereogenic center.  相似文献

C1-Symmetric phosphino-oxazolines, (S,S)- and (S,R)-2-[4-(isopropyl)oxazol-2-yl]-2′-diphenylphosphino-1,1′-binaphthyl, which possess both phosphine and oxazoline moieties, were prepared from racemic binaphthol and enantiomerically pure (S)-(+)-2-amino-3-methyl-1-butanol in high yields. Reaction of 1,3-diphenyl-2-propenyl acetate with dimethyl sodiomalonate in the presence of 2 mol% of palladium catalysts bearing the new chiral ligands proceeded with high enantioselectivity to give allylic alkylation products of up to 91% ee.  相似文献

The reaction of 2-(1-naphthyl)-3-methylpyridine with olefins in the presence of [RhCl(coe)2]2 and PCy3 as the catalyst resulted in the alkylation of the naphthyl ring at the 2-position in good yield. The replacement of PCy3 with the chiral ferrocenyl phosphine, (R),(S)-PPFOMe, as the ligand resulted in atropselective alkylation of the naphthylpyridine derivatives. Ethylene reacted with the biaryl compounds to give the corresponding addition products in moderate yields with fair to good ee's (up to 49% ee).  相似文献

Two new synthetic methods were established for the efficient synthesis of optically active cyclohexene antisepsis agent, ethyl (6R)-6-[N-(2-chloro-4-fluorophenyl)sulfamoyl]cyclohex-1-ene-1-carboxylate [(R)-1: TAK-242)]. The first method involved recrystallization from methanol of the diastereomeric mixture (6RS,1'R)-7, obtained by esterification of carboxylic acid 3 with (S)-1-(4-nitrophenyl)ethanol [(S)-5)] to give the desired isomer (6R,1'R)-7 with 99% de in 32% yield. Subsequent catalytic hydrogenolysis and esterification gave (R)-1 with >99% ee. The second method employed enantioselective hydrolysis of acetoxymethyl ester 9a (prepared by alkylation of 3 with bromomethyl acetate) with Lipase PS-D to give the eutomeric enantiomer (R)-9a with excellent enantioselectivity (>99% ee) and high yield (48%). The desired (R)-1 was then obtained by transesterification with ethanol in the presence of concentrated sulfuric acid without loss of ee. Of these, the procedure employing enzymatic kinetic resolution using Lipase PS-D is the more efficient and practical preparation of (R)-1.  相似文献

Different chiral cyclic amino acid alkali metal and ammonium salts were used as catalysts for the asymmetric Michael addition of aldehydes to nitrostyrene. The reaction yield and stereoselectivity depend slightly on the salt cation. The highest yield of the reaction (up to 100%) was obtained with (S)-morpholine-3-carboxylic acid salts, which gave moderate to good enantioselectivities (up to 72% ee) and diastereoselectivities (dr up to 89:11) whereas the highest selectivity was obtained with proline sodium salt (ee 88%).  相似文献

Asymmetric glycolate alkylation using a protected acetophenone surrogate under solid-liquid phase-transfer conditions is a new approach to the synthesis of 2-hydroxy esters and acids. Diphenylmethyloxy-2,5-dimethoxyacetophenone 1 with a trifluorobenzyl cinchonidinium bromide catalyst 9 (10 mol %) and cesium hydroxide provided S-alkylation products 2 at -35 degrees C in high yield (80-99%) and with excellent enantioselectivities using a wide range of electrophiles (80-90% ee). Alkylated products were elaborated to useful alpha-hydroxy intermediates 3 using bis-TMS peroxide Baeyer-Villiger conditions and selective transesterification reactions. The ester products have been enantioenriched by simple recrystallization from ether to give a single isomer (99% ee). A tight ion-pair model is proposed for the observed S-stereoinduction that includes van der Waals contacts between the extended enolate and the isoquinoline of the catalyst. To demonstrate the utility of the new methodology, the anti-diabetes drug (-)-ragaglitazar 24 was synthesized in six steps from a key 2-alkoxy-3-p-phenoxypropionic acid 26 that was made using PTC glycolate alkylation.  相似文献

Chiral diaminophosphines 4 were prepared from (S)-prolinol-derived aminophosphine oxide 5 by bromination with ring expansion followed by amination with ring contraction and reduction, using trichlorosilane. In the presence of 4 as a ligand, palladium-catalyzed asymmetric allylic alkylation of 1,3-diphenyl-2-propenyl acetate (11) with a dialkyl malonate-BSA-LiOAc system was successfully carried out with good enantioselectivities (up to 98% ee).  相似文献

Herein we present a novel route to enantiomerically enriched chiral alpha-substituted carboxylic acids by crystallization-induced dynamic resolution (CIDR) of their diastereomeric salts with chiral amines. Thus, the racemic alpha-bromo acid 3 is converted reliably with (1R,2S)-2-amino-1,2-diphenylethanol in the presence of a catalytic amount of tetrabutylammonium bromide into its R-enantiomer 4 in 90% yield with 88% ee. Similarly, the racemic alpha-thiobenzoyl acid 5 could be resolved to 90% ee in 74% yield. Further enrichment to enantiomeric homogeneity could be achieved in both cases by crystallization. In a telescoped, two-step process, S-alpha-thiobenzoyl acid 6 (>or=99.6% ee) was prepared from the racemic bromide 3 in 63% yield. State-of-the-art parallel experimentation enabled rapid screening for suitable dynamic resolution conditions. Kinetic studies defined the influence of temperature, tetrabutylammonium bromide concentration, molarity, and solvent polarity on the resolution rate, product yield, and enantiomeric excess.  相似文献

The regioselectivity of the alkylation of alkali metal salts of 2 - methyl - cyclopentane - 1,3 - dione with alkyl halides depends significantly on the structure of the alkylating reagent and the type of solvent used in the reaction. Unsubstituted saturated primary alkyl halides with the exception of methyl iodide preferentially yield products of O-alkylation. Alkyl halides with an sp2- or sp-hybridized carbon atom in a β-position preferentially yield products of C-alkylation. Dipolar aprotic solvents favour O-alkylation; water favours C-alkylation. On the basis of these results procedures for the prepartion of 2-allyl-2-methyl-, 2-allyl-2-ethyl-, 2-methyl-2-propargyl- and 2 - benzyl - 2 - mehtyl - cyclopentane - 1,3 - dione have been developed. The alkylation of 2 - methyl - cyclopentane - 1,3 - dione with methyl bromoacetate was also intensively investigated.  相似文献
