The chiral separation of amino acid derivatives by ligand-exchange electrophoresis in a microchannel chip was performed for the first time. A Cu(II) complex with L-prolinamide was used as a chiral selector. The migration behaviors of eleven NBD-DL-amino acids were investigated by ligand-exchange capillary electrophoresis (LE-CE). The enantiomer of five NBD-amino acids (Ser, Thr, Val, Phe and His) could be separated by LE-CE using a 20 mM ammonium acetate buffer (pH 9.0) containing 10 mM copper acetate, 20 mM L-prolinamide and 1 mM SDS. NBD-His was eluted in the order D-form and L-form, while the elution order of another enantiomers was L-form and D-form. Under this condition, the enantioseparation of these five NBD-amino acids by ligand-exchange microchip electrophoresis (LE-ME) was investigated using a glass microchip. The enantioseparation of NBD-Ser, -Thr and -His could be successfully accomplished by LE-ME. LE-ME was superior to LE-CE in terms of the short migration time and a good enantiomeric separation.  相似文献

A ligand-exchange capillary electrophoresis was explored, with L-ornithine as the ligand and copper(II) as the central ion. Its applicability was demonstrated with underivatized and dansyl amino acids, a dipeptide, and drugs with amino alcohol structure. The enantioselectivity was found to be strongly dependent on pH and copper(II)-L-Orn complex concentration. Due to the adsorption of the positively charged species onto the capillary inner walls, the chiral separation selectivity is very high while the efficiency is relatively low. Permanent 1,3-propanediamine-coated capillaries show an improved separation efficiency and theoretical plate numbers increasing from 10(4) to 10(5). Similar phenomena were observed when sodium dodecyl sulfate (SDS) micelles were added to the copper(II) complex solution. The poor separation efficiency of chiral compounds in uncoated capillaries may result from the low rate of the ligand-exchange reactions, and the high enantioselectivity may derive from the complexing process in the adsorbed phase.  相似文献

D-Penicillamine is demonstrated for the first time as a chiral ligand for the enantioseparation of dansyl amino acids based on ligand-exchange micellar electrokinetic chromatography (LE-MEKC). Copper(II) was used as the central ion in the ternary complex. The effect of surfactant on the resolution was significant. A concentration of 20 mM sodium dodecyl sulfate (SDS) was shown to be necessary for the separation. Other important parameters, such as the concentration ratio of D-penicillamine (D-PEN) to Cu2+, the kind of metal central ion, the type and pH value of buffer, were also investigated. N-Acetyl-D-penicillamine and L-valine (Val), with similar structure to D-penicillamine, were applied as their copper(II) complexes as chiral selector and the chiral recognition mechanism is briefly discussed. Under optimum experimental conditions, i.e., 20 mM NH4OAc, pH 6.5, a 2:1 concentration ratio of D-penicillamine to Cu(II), 4 mM CuSO4 and 8 mM D-penicillamine, the chiral separation of eight pairs of different dansyl amino acid enantiomers was accomplished with resolution ranging from 1.1 to 5.9. When L-PEN was used instead of D-PEN, reversal of the migration order was observed.  相似文献

Enantiomers of dansylated DL-amino acids were resolved by chiral copper (II)-N-n-dodecyl-L-proline (1) complexes incorporated in micelles of sodium dodecyl sulfate (SDS) in electrokinetic capillary chromatography (EKC). This resolution is caused by formation of diastereomeric ternary complexes consisting of chiral ligand 1, central copper (II) ion and enantiomeric amino acid derivatives in micellar phase. However, the resolution was not observed when SDS with an anionic polar head grop was replaced with dodecyl trimethylammonium brode (DTMAB) with a cationic polar head group. The ratio between copper (II) ion and 1 in the complex in either SDS or DTMAB was measured by UV-visible spectra, which respond to the d-d transition of copper (II). Mechanism of separation should be discussed in terms of effect of surfactant structures on constitution of copper (II) ion and 1 in the micellar phase and that of arene substituent structures linked to sulfonamide units in amino acid derivatives to be separated.  相似文献

Complexes of copper(II), nickel(II), cobalt(II), and zinc(II) with 2-[2-(6-methylbenzothiazolyl)azo]-5-dimethylaminobenzoic acid have been prepared and characterized by elemental analysis, vibrational spectra, magnetic susceptibility measurements, conductance measurements and e.p.r. spectra. Stability constants have been evaluated potentiometrically. Electronic spectra, magnetic susceptibility measurements and molecular modeling studies support a distorted square planar geometry around the metal ions. Vibrational spectra indicate the coordination of the azo group, nitrogen of benzothiazole, the carboxylate anion and the acetate ion on complexation with the metal ion. All complexes are found to be monomers. The stability of the complexes follow the order: copper(II) > nickel(II) > cobalt(II) > zinc(II).  相似文献

Enantiomer separations of underivatised amino acids were carried out by using ligand exchange capillary electrophoresis (LECE). Chiral discrimination is based on the formation of ternary complexes between copper(II), a chiral selector (L-proline or trans-4-hydroxy-L-proline) and an amino acid. All amino acids containing aromatic moieties or not were detected at 214 nm because of their interactions with copper(II). In order to reduce copper(II) adsorption onto capillary walls, we used hexadimethrine bromide to reverse the electroosmotic flow. Using this original strategy, the studied enantiomers migrated in the opposite direction of the anodic electroosmosis. After optimising the analytical conditions taking into account the chiral resolution and the detection sensitivity, we performed very satisfactory enantioseparations not only of aromatic amino acids (tryptophan, tyrosine, phenylalanine and histidine) but also of aliphatic amino acids (threonine, serine, isoleucine and valine). These enantioseparations were performed in a short analysis time at 35 °C. In order to rationalise the obtained results, we evaluated the complexation constants corresponding to the formed ternary complexes by capillary electrophoresis and we used molecular mechanics modelling.  相似文献

Summary Complexes of furan and thiophene azo-oximes with iron(II), cobalt(III), nickel(II) and copper(II) have been prepared and characterised. Iron(II), cobalt(III) and copper(II) complexes are diamagnetic in the solid state. The diamagnetism of the copper(II) chelates is suggestive of antiferromagnetic interaction between two copper centres.1H n.m.r. spectral data suggest atrans-octahedral geometry for the tris-chelates of cobalt(III). Nickel(II) complexes are paramagnetic, in contrast to the diamagnetism of the analogous complexes of arylazooximes. The electronic spectra are suggestive of octahedral geometry for the iron(II), cobalt(III) and nickel(II) complexes, andD 4h -symmetry for copper(II). Infrared data indicate N-bonding of the oximino-group to the metal ions.  相似文献

Salicylidene-N-anilinoacetohydrazone (H(2)L(1)) and 2-hydroxy-1-naphthylidene-N-anilinoacetohydrazone (H(2)L(2)) and their iron(III), manganese(II), cobalt(II), nickel(II), copper(II) and zinc(II) complexes have been synthesized and characterized by IR, electronic spectra, molar conductivities, magnetic susceptibilities and ESR. Mononuclear complexes are formed with molar ratios of 1:1, 1:2 and 1:3 (M:L). The IR studies reveal various modes of chelation. The electronic absorption spectra and magnetic susceptibility measurements show that the iron(III), nickel(II) and cobalt(II) complexes of H(2)L(1) have octahedral geometry. While the cobalt(II) complexes of H(2)L(2) were separated as tetrahedral structure. The copper(II) complexes have square planar stereochemistry. The ESR parameters of the copper(II) complexes at room temperature were calculated. The g values for copper(II) complexes proved that the Cu-O and Cu-N bonds are of high covalency.  相似文献

A facile chiral ligand-exchange capillary electrophoretic method has been explored for the enantioseparation and UV detection of dansyl-amino acids with Zn(II) L-arginine complex as a chiral selecting system. Successful enantioseparation of 17 pairs of amino acid enantiomers has been achieved with a buffer of 100 mM boric acid, 5 mM ammonium acetate, 3 mM ZnSO4 and 6 mM L-Arg at pH 8.0, of which 10 pairs were fully resolved with resolution in between 1.59 and 4.21. This new method was shown to be applicable to the separation of some mixed pairs of amino acids and to the quantitative analysis of some real samples such as rice vinegars, with a linear range between 0.8 and 150 microg/mL, correlation coefficient above 0.99 and recovery in between 90.1 and 112.4%. It was found that amino acids with low resistance side chain(s), low tendency to form intramolecular hydrogen bond or high tendency to form intermolecular hydrogen bonds are more easily enantioseparated than those with extra carboxyl and/or phenyl groups. By the use of the suggested buffer, the running pH should be selected at 7.4-9.0 to compromise the resolution and elution speed.  相似文献

High-performance liquid chromatographic (HPLC) separation of stereoisomeric cyclic beta-substituted alpha-quaternary alpha-amino acids was performed by ligand-exchange on a copper(II)-D-penicillamine chiral stationary phase. The investigated amino acids are the 1-amino-2-methylcyclohexanecarboxylic acids, the 1-amino-2-hydroxycyclohexanecarboxylic acids, the 1-amino-2-methylcyclopentanecarboxylic acids and the trans-configured 1,2-diaminocyclohexanecarboxylic acids. The effects of the mobile phase composition (copper(II) concentration, type and content of organic modifier, pH) and the temperature on the enantio- and diastereoselectivity were studied and the conditions were optimised to resolve the four stereoisomers of each of the said amino acids in single chromatographic runs. A reversal of the elution order occurred for enantiomers of some of the amino acids in dependence on the acetonitrile content of the eluent. This phenomenon is explained by at least two different copper(II) complexes of the tridentate ligand penicillamine.  相似文献

A chiral ligand exchange capillary electrophoresis (CLE-CE) method using Zn(II) as the central ion and l-4-hydroxyproline as the chiral ligand coordinating with γ-cyclodextrin (γ-CD) was developed for the enantioseparation of amino acids (AAs) and dipeptides. The effects of various separation parameters, including the pH of the running buffer, the ratio of Zn(II) to l-4-hydroxyproline, the concentration of complexes and cyclodextrins (CDs) were systematically investigated. After optimization, it has been found that eight pairs of labeled AAs and six pairs of labeled dipeptides could be baseline-separated with a running electrolyte of 100.0 mM boric acid, 5.0 mM ammonium acetate, 3.0 mM Zn(II), 6.0 mM l-hydroxyproline and 4.0 mM γ-CD at pH 8.2. The quantitation of AAs and dipeptides was conducted and good linearity (r2 ≥ 0.997) and favorable repeatability (RSD ≤ 3.6%) were obtained. Furthermore, the proposed method was applied in determining the enantiomeric purity of AAs and dipeptides. Meanwhile, the possible enantiorecognition mechanism based on the synergistic effect of chiral metal complexes and γ-CD was explored and discussed briefly.  相似文献

Hyperbranched polyester polycarboxylic acids of the second and third generations for use as high-efficiency complexing agents are synthesized. On the basis of these compounds, new metal-polymer complexes of cobalt(II) and copper(II) are prepared for the first time. As evidenced by IR and ERR studies, the central atom in these metal polycarboxylates occurs in the axially symmetric system MO6. The thermal stability of polymer copper complexes is improved with increases in the content of metal ions, the degree of functionalization, and the generation number of the polyacid platform.  相似文献

Chiral resolution of native DL-malic acid was achieved by ligand-exchange capillary electrophoresis using copper(II)-L-tartrate as a chiral selector. Factors affecting chiral resolution, migration time, and peak area of malic acid were studied. The running conditions for optimum separation of malic acid were found to be 1 mM copper(II) sulfate-1 mM L-tartrate (pH 5.1) with an effective voltage of -20 kV at 30 degrees C, using direct detection at 280 nm, and resolution (Rs) of racemic malic acid was approximately 4. With this system, D- and L-malic acids in apple juice were analyzed successfully.  相似文献

A new Schiff base has been synthesized from 4-aminoantipyrine and 3-formylsalicylic acid. The ligand has a dianionic tetradentate compartmental OONO donor system. The cobalt(II), nickel(II), copper(II) and dioxouranium(VI) complexes exist in phenolato-bridged dinuclear species, while palladium(II) gives a mononuclear complex with free –COOH groups. The complexes have been characterized by elemental analyses, i.r., u.v.-vis, thermal and magnetic measurements.  相似文献

Summary The doubly bidentate ON-NO donor Schiff base, prepared from salicylaldehyde and 4,4-diaminodiphenylmethane forms complexes with 11 metal: ligand stoichiometric ratios. The cobalt(II), copper(II) and nickel(II) complexes exhibit subnormal magnetic moments. All the six complexes possess high melting points and are sparingly soluble in common organic solvents. A dinuclear octahedral structure is proposed for the cobalt(II), copper(II), nickel(II) and zinc(II) complexes and a dinuclear tetrahedral configuration is suggested for the cadmium(II) and mercury(II) complexes on the basis of analytical, conductance, magnetic susceptibility, molecular weight, i.r. and electronic spectral data.  相似文献

The complexing reagent 2-thiophenaldehyde-4-phenyl-3-thiosemicarbazone (TAPT) was examined for high performance liquid chromatographic (HPLC) separations of cobalt(II), copper(II) and iron(II) or cobalt(II), nickel(II), iron(II), copper(II) and mercury(II) as metal chelates on a Microsorb C-18, 5-mum column (150x4.6 mm i.d.) (Rainin Instruments Woburn, MA, USA). The complexes were eluted isocratically with methanol:acetonitrile:water containing sodium acetate and tetrabutyl ammonium bromide (TBA). UV detection was at 254 nm. The solvent extraction procedure was developed for simultaneous determination of the metals, with detection limits within 0.5-2.5 mug ml(-1) in the final solution. The method was applied for the determination of copper, cobalt and iron in pharmaceutical preparation.  相似文献

An optimized flow-injection manifold for the chemiluminescence determination of cobalt(II), copper(II), iron(II) and chromium(III) by their catalytic effect on the luminol reaction is described. Detection limits are 0.0006, 0.08, 0.3 and 0.1 ng ml?1, respectively. The suppression effect of several carboxylic acids on the emission intensity is discussed. A procedure for the separation of cobalt(II), copper(II) and iron(II) on a low-capacity, silica-based cation-exchange column, using 5 mM oxalic acid at pH 4.2 as the mobile phase and post-column detection via the luminol reaction, is also described. Detection limits for cobalt(II) and copper(II) are 0.01 and 5 ng ml?1, respectively.  相似文献

The interaction of diglycollic acid ligand with iron(III), cobalt(II), nickel(II) and copper(II) salts was investigated potentiometrically and spectrophotometrically. Only 1:1 complexes were formed in solution and solid. The pK's of the ligand and its complexes were computed. The electronic absorption spectra of the complexes depict the octahedral geometry. The dependence of the chiral recognition ability and enantiomer migration order on the structure, substitution pattern and chirality of chiral selectors used in ligand-exchange capillary electrophoresis is investigated. As chiral selectors different N-alkyl derivatives of proline and hydroxyproline as their copper(II) complexes are used. The influence of the position and conformation of the hydroxy group in the hydroxyproline derivatives and of the structure and chirality of the side chain on enantioselectivity is investigated. Furthermore, the effect of surfactants such as sodium dodecyl sulfate and cetyl trimethyl ammonium bromide on resolution and enantiomer migration order is studied. The investigations were carried out with three aromatic amino acids as model compounds.  相似文献
