Interactions and uses of antisense peptides in affinity technology.   总被引:3,自引:0,他引:3  
Antisense peptides, amino acid sequences encoded in the antisense strand of DNA, can interact with significant affinity and selectivity with their corresponding sensepeptides. Experimentally, sense-antisense peptide recognition has been observed repeatedly. However, skepticism about the biological relevance of this phenomenon has persisted. This is due in part to the unexpected and somewhat couterintutive nature of the interaction as well as to its non-universality as an empirical observation. Nonetheless, antisense peptides in several cases investigated so far have been used as immobilized ligands for the successful affinity chromatographic separation of native (sense) peptides and proteins. For example, immobilized antisense peptides corresponding to Arg8-vasopressin (AVP) have been used to separate vasopressin from oxytocin chromatographically as well as to affinity capture AVP-receptor complex. These results, together with improved understanding of the general features of amino acid sequence which drive antisense-sense peptide interactions as well as new ideas for making antisense peptides chimeras, are beginning to suggest improved ways to make antisense-related peptides as affinity agents for separation as well as for other biotechnology applications.  相似文献

Enzyme-based biocatalysis is emerging as an advanced technique to develop green processes that help to maintain the sustainability of the environment. The bioremediation of toxic organic pollutants and waste to bioenergy production using enzymes as biocatalysts is rapidly growing due to its eco-friendly and sustainable nature. Additionally, a range of microbial species that typically grow on organic wastes can be used to produce these enzymes in an efficient manner. This is seen as a potential strategy for the development of cost-effective manufacturing for a number of biotechnological applications. The present study discusses biocatalysis as a promising and sustainable method toward the bioremediation of hazardous organic pollutants as well as for bioenergy production, based on the immense potential of enzymes as biocatalysts. Emphasis has been placed on evaluating the critical elements that can enhance the production of enzymes used as biocatalysts, as well as their functional effectiveness and stability.  相似文献

The series of new bis(naphthalic anhydrides) was prepared through Diels-Alder cycloaddition. The Diels-Alder cycloaddition was used as a synthetic route to new phenylated monomers as well as to polymers. All polymers synthesized revealed to be soluble in a wide range of organic solvents such as toluene, THF, chloroform, and displayed high thermostability. Therefore, they can be processed easily and are promising candidates for advanced coating systems as well as for electrooptical applications.  相似文献

The utility of diphosphorous tetraiodide as a new, mild, condensing agent for synthesis of oxadiazole is described. These data indicate the simple dehydration of oximes to 1,2,5-oxadiazole as well as the rearrangements of oximes to normal Beckmann product 1,2,4-oxadiazole. However, mono-oxime of benzil undergoes abnormal Beckman rearrangement to benzaldehyde as major product. The described method is simple and important for the synthesis of the oxadiazoles as well as for nitriles.  相似文献

A copper(II)-catalyzed, sequential Michael addition-aldol condensation reaction of N-carboxybenzyl-protected aminobenzaldehyde with various α,β-unsaturated N-acyl pyrroles is described. Substrate scope was found to include both aryl and aliphatic N-acyl pyrroles as the Michael acceptors, and isolated product yields as high as 93% were observed. The use of acetonitrile as the reaction solvent proved to be crucial for catalysis, both to function as a labile ligand for copper, as well as an agent to minimize hydrolytic catalyst poisoning.  相似文献

We have investigated the overall excitation energy relaxation dynamics in linear porphyrin arrays as well as the energy transport phenomena by attaching an energy acceptor to one end of a linear porphyrin array by using steady state and time-resolved spectroscopic measurements. We have revealed that the solvation dynamics as well as the conformational dynamics contributes significantly to the energy relaxation processes of linear porphyrin arrays. Consequently, long porphyrin arrays no longer serve as good energy transmission elements in donor-acceptor linked systems due to conformational heterogeneities which provide the non-radiative deactivation channels as energy quenchers.  相似文献

Microfluidic LC systems present undeniable advantages over classical LC in terms of sensitivity. Hepcidin, a peptide marker of clinical disorders linked to iron metabolism, was used as model to demonstrate peptide quantification potentialities of LC-chip coupled to a nanoelectrospray source ion trap mass spectrometer in an aqueous sample. First, stable isotope labelled hepcidin was chosen as internal standard and gradient as well as sample compositions were optimised using design of experiments as development tool. The method was then prevalidated using accuracy profiles in order to select the most appropriate response function and to confirm the ability of the technique to quantify low hepcidin concentration. A reliable and very sensitive quantitation method was finally obtained using this integrated microfluidic technology. Indeed, good results with respect to accuracy, trueness and precision were achieved, as well as a very low limit of quantitation (0.07 ng/ml). Method suitability of nano-LC on chip tandem mass spectrometry for hepcidin quantitation was also demonstrated in complex media such as human plasma.  相似文献

The toxic bicyclic octapeptide α‐amanitin is mostly found in different species of the mushroom genus Amanita, with the death cap (Amanita phalloides) as one of the most prominent members. Due to its high selective inhibition of RNA polymerase II, which is directly linked to its high toxicity, particularly to hepatocytes, α‐amanitin received an increased attention as a toxin‐component of antibody‐drug conjugates (ADC) in cancer research. Furthermore, the isolation of α‐amanitin from mushrooms as the sole source severely restricts compound supply as well as further investigations, as structure–activity relationship (SAR) studies. Based on a straightforward access to the non‐proteinogenic amino acid dihydroxyisoleucine, we herein present a robust total synthesis of α‐amanitin providing options for production at larger scale as well as future structural diversifications.  相似文献

Aluminosilicates have traditionally been important materials for applications related to adsorbents, water softeners, catalysis and mechanical and thermal reinforcement due to their high surface area, excellent thermal/hydrothermal stability, high shape-selectivity and superior ion-exchange ability. Recently, their use as polymer fillers has allowed to increasingly extending their application range to innovative areas such as medical and biological fields as well as in sensors, filtration membranes, energy storage and novel catalysis routes. Further, the large versatility and tailoring possibilities of both filler and matrix indicates this area as one of the enabling key technologies of the near future.This work summarizes the main developments up to date in this increasingly interesting field, focuses on the main applications already developed as well as on the key challenges for the near future.  相似文献

Organic micropollutants such as estrogens occur in water in increasing quantities from predominantly anthropogenic sources. In water such micropollutants partition not only to surfaces such as membrane polymers but also to any other natural or treatment related surfaces. Such interactions are often observed as sorption in treatment processes and this phenomenon is exploited in activated carbon filtration, for example. Sorption is important for polymeric materials and this is used for the concentration of such micropollutants for analytical purposes in solid phase extraction. In membrane filtration the mechanism of micropollutant sorption is a relatively new discovery that was facilitated through new analytical techniques. This sorption plays an important role in micropollutant retention by membranes although mechanisms of interaction are to date not understood. This review is focused on sorption of estrogens on polymeric surfaces, specifically membrane polymers. Such sorption has been observed to a large extent with values of up to 1.2 ng/cm(2) measured. Sorption is dependent on the type of polymer, micropollutant characteristics, solution chemistry, membrane operating conditions as well as membrane morphology. Likely contributors to sorption are the surface roughness as well as the microporosity of such polymers. While retention-and/or reflection coefficient as well as solute to effective pore size ratio-controls the access of such micropollutants to the inner surface, pore size, porosity and thickness as well as morphology or shape of inner voids determines the available area for sorption. The interaction mechanisms are governed, most likely, by hydrophobic as well as solvation effects and interplay of molecular and supramolecular interactions such as hydrogen bonding, π-cation/anion interactions, π-π stacking, ion-dipole and dipole-dipole interactions, the extent of which is naturally dependent on micropollutant and polymer characteristics. Systematic investigations are required to identify and quantify both relative contributions and strength of such interactions and develop suitable surface characterisation tools. This is a difficult endeavour given the complexity of systems, the possibility of several interactions taking place simultaneously and the generally weaker forces involved.  相似文献

The essentials of the QCHB (quasi-chemical hydrogen-bonding) equation-of-state model are presented along with some applications for calculations of phase equilibria and interfacial properties of fluids and their mixtures. This is a model applicable to non-polar systems as well as to highly non-ideal systems with strong specific interactions, to systems of small molecules as well as to macromolecules, including polydisperse polymers, glasses, and gels, to liquids as well as to vapours including supercritical systems, to homogeneous as well as to inhomogeneous systems. A quasi-thermodynamic approach of inhomogeneous systems is used for modeling the fluid–fluid interface. Consistent expressions for the interfacial tension and interfacial profiles for various properties are presented. A satisfactory agreement is obtained between experimental and calculated surface tensions. Extension of the approach to mixtures is examined along with the associated problems for the numerical calculations of the interfacial profiles. A new equation is derived for the chemical potentials in the interfacial region, which facilitates very much the calculation of the composition profiles across the interface. The relation of the model with the COSMO-RS approach is also discussed.  相似文献

Hydrolytically degradable, multilayered films ranging from 10 to 100 nm have been constructed by the layer-by-layer deposition of degradable polycations and oppositely charged polyanions. Polycations play dual roles in these systems, serving as structural components of the film as well as transient elements designed to trigger release; polyanions serve as structural components and as entities to be released or delivered. The films erode in a controlled manner under physiological conditions and are suitable for the incorporation and subsequent controlled release of functional polyanions such as DNA.  相似文献

A review of the chemical education research literature suggests that the term constructivism is used in two ways: experience-based constructivism and discipline-based constructivism. These two perspectives are examined as an epistemology in relation to the teaching and learning of the concept of idealization in chemistry. It is claimed that experience-based constructivism is powerless to inform the origin of such concepts in chemistry and while discipline-based constructivism can admit such theoretical concepts as idealization it does not offer any unique perspectives that cannot be obtained from other models. Chemical education researchers do not consistently appeal to constructivism as an epistemology or as a teaching/learning perspective and it is shown that, while it draws attention to worthwhile teaching/learning strategies, it cannot be considered as foundational to chemical education research and tends to be used more as an educational label than as an undergirding theory.  相似文献

Two new bridged nitraminotriazoles with bridging oxapropylene and nitrazapropylene moieties were synthesized, and converted into several salts, as well as from the hydrazonemethylene bridged nitraminotriazole. All compounds were fully characterized by NMR and IR spectroscopy, elemental analysis as well as differential thermal analysis. The sensitivity towards friction and impact were determined according to BAM standard technics and the energetic properties were calculated by using the EXPLO5 computer code. The neutral compounds as well as the various salts were examined in terms of their physicochemical properties and detonation performance to each other and compared to the commonly used secondary explosive RDX.  相似文献

The photocatalyzed [2 + 2 + 2]-cycloaddition of nitriles with 2 equiv of acetylene to 2-pyridines can be carried out under mild conditions and represents a valuable extension to common synthetical methods. For the ideal wavelength range (350-500 nm), lamps as well as sunlight can be used. Working at room temperature and in organic solvents such as toluene or hexane as well as in water gives satisfying results in many cases. However, it is also possible to vary the solvent and the reaction temperature of the photocatalyzed synthesis and to choose, with respect to the specific substrate, specific requirements for this particular reaction and general requirements of the method. This simple and selective method derives its potential mainly from the large variety of applicable nitriles. Suitable substrates include (functionalized) aliphatic and aromatic nitriles as well as cyanamides derived from secondary amines.  相似文献

The reduction of benzophenone by lithium and chloromagnesium alkoxides has been studied as well as the transformation of certain lithium alkoxides to the corresponding ketones by electron transfer. Fluorenone was reduced by lithium sec-butoxide to the corresponding lithium ketyl to the extent of 65%. Lithium 9-fluoroenolate underwent in tetrahydrofuran a spontaneous transformation to lithium fluorenone ketyl. This process was interpreted as involving 1,2-hydrogen shift in an oxygen-centred radical. A mechanism for the Meerwein-Pondorf-Verley-type reductions is proposed, invoking single electron as well as 1,2-hydrogen shift steps.  相似文献

Systematic studies on art objects using instrumental neutron activation analysis and neutron autoradiography have been carried out in the Institute of Nuclear Chemistry and Technology in collaboration with the Faculty of Art Conservation and Restoration of the Academy of Fine Arts in Cracow, as well as with other Academies of Fine Arts and museums in Poland. It was possible to accumulate a number of essential data on the concentration of trace elements particularly in chalk grounds and pigments (such as lead white, lead-tin yellow, smalt), Chinese porcelain, Thai ceramics, as well as in the clay fillings of sarcophagi of Egyptian mummies. The above mentioned examination of art objects prior to their conservation helps to determine precisely the materials used in the process of creating art objects, as well as to identify the approximate place of origin of particular materials.  相似文献
