Gemini surfactants: new synthetic vectors for gene transfection   总被引:5,自引:0,他引:5  
The superior surfactant properties of cationic gemini surfactants are applied to the complex problem of introducing genes into cells. Of almost 250 new compounds tested, of some 20 different structural types, a majority showed very good transfection activity in vitro. The surfactant is shown to bind and compact DNA efficiently, and structural studies and calculations provide a working picture of the "lipoplex" formed. The lipoplex can penetrate the outer membranes of many cell types, to appear in the cytoplasm encapsulated within endosomes. Escape from the endosome--a key step for transfection--may be controlled by changes in the aggregation behavior of the lipoplex as the pH falls. The evidence suggests that DNA may be released from the lipoplex before entry into the nucleus, where the new gene can be expressed with high efficiency.  相似文献

The supramolecular structural nature of some cationic liposomes-DNA complexes, currently used as vehicles in non-viral gene delivery, has been elucidated by recent X-ray diffraction experiments. The relationship between the chemico-physical properties of these self-assembled structures and their transfection efficiency is extensively studied. Here we report a first comprehensive structural study by using energy dispersive X-ray diffraction, of the complex DC-Chol--DOPE/DNA (3beta[N-(N',N'-dimethylaminoethane)-carbamoyl]cholesterol dioleoylphosphatidylethanolamine/DNA), which has been classified as one of the most effective in in-vivo experiments. Our results show that DC-Chol--DOPE/DNA lipoplexes have a columnar inverted hexagonal structure, which is not influenced by the cationic liposome/DNA charge ratio. The transfection efficiency of C6 rat glioma cells by DC-Chol--DOPE/DNA lipoplexes and the toxicity of lipoplexes to cells are dramatically affected by cationic liposome/DNA weight ratio.It seems therefore that the lipoplex structures have not any influence on transfection efficiency and toxicity in our experimental system.  相似文献

We have characterized the phase behavior of mixtures of the cationic surfactant cetyltrimethylammonium bromide (CTAB) and the organic salt 3-sodium-2-hydroxy naphthoate (SHN) over a wide range of surfactant concentrations using polarizing optical microscopy and X-ray diffraction. A variety of liquid crystalline phases, such as hexagonal, lamellar with and without curvature defects, and nematic, are observed in these mixtures. At high temperatures the curvature defects in the lamellar phase are annealed gradually on decreasing the water content. However, at lower temperatures these two lamellar structures are separated by an intermediate phase, where the bilayer defects appear to order into a lattice. The ternary phase diagram shows a high degree of symmetry about the line corresponding to equimolar CTAB/SHN composition, as in the case of mixtures of cationic and anionic surfactants.  相似文献

We applied a molecular assembly formed in an aqueous surfactant mixture of cationic cetyltrimethylammonium bromide (CTAB) and anionic sodium octylsulfate (SOS) as templates of mesoporous silica materials. The hexagonal pore size can be controlled between 3.22 and 3.66 nm with the mixed surfactant system. In addition, we could observe the lamellar structure of the mixed surfactants with precursor molecules, which strongly shows the possibility of precise control of both the pore size and the structure of pores by changing the mixing ratio of surfactants. Moreover, use of the cationic surfactant having longer hydrophobic chain like stearyltrimethylammonium bromide (STAB) caused the increase in d(100) space and shifted the point of phase transition from hexagonal phase to lamellar phase to lower concentration of SOS.  相似文献

The phase behavior of a series of carbohydrate-based gemini surfactants with varying spacer lengths was studied using static and dynamic light scattering between pH 2 and 12. Cryo-electron microscopy pictures provide evidence for the different morphologies present in solution. The spacer length of the gemini surfactants was varied from two to 12 methylene units. At near neutral pH, spherical vesicles were obtained for gemini surfactants with a spacer shorter than 10 methylene units, whereas nonspherical vesicles were obtained for spacer lengths of 10 and 12. Upon decreasing the pH, the vesicles underwent transitions toward worm-like micelles and spherical micelles for a spacer length of six and larger, whereas for shorter spacers, these transitions are not observed. For the shortest spacer at low pH, perforated vesicles are observed, and vesicles built from the gemini surfactant with a spacer of four methylene units only underwent a transition toward worm-like micelles. Upon increasing the pH to slightly basic values, flocculation followed by redispersion upon charge reversal was observed up to a spacer length of eight methylene units. The redispersal is explained by hydroxide-ion binding to the uncharged vesicular surface. By contrast, vesicles formed from the gemini surfactants with 10 and 12 methylene units only undergo a transition toward inverted phases. The observations can be understood in terms of the packing parameter.  相似文献

Cationic gemini surfactants, N,N-bis(dimethylalkyl)-alpha,omega-alkanediammonium dibromide [C(m)H(2m+1)(CH(3))(2)N(+)(CH(2))(s)N(+)(CH(3))(2)C(m)H(2m+1) x 2 Br(-), or m-s-m], have proven to be effective synthetic vectors for gene delivery (transfection). Complexes (lipoplexes) of gemini compounds, where m = 12, s = 3, 12 and m = 18 : 1(oleyl), s = 2, 3, 6, with DNA have been investigated using isothermal titration calorimetry (ITC), dynamic light scattering (DLS), zeta potential, atomic force microscopy (AFM) and circular dichroism (CD) techniques. The results show that lipoplex properties depend on the structural properties of the gemini surfactants, the presence of the helper lipid dioleoylphosphatidylethanolamine (DOPE), and the titration sequence. ITC data show that the interaction between DNA and gemini surfactants is endothermic and the observed enthalpy vs. charge ratio profile depends upon the titration sequence. Isoelectric points (IP) of lipoplex formation were estimated from the zeta potential measurements and show good agreement with the reaction endpoints (RP) obtained from ITC. DLS data indicate that DNA is condensed in the lipoplex. AFM images suggest that the lipoplex morphology changes from isolated globular-like aggregated particles to larger-size aggregates with great diversity in morphology. This change is further accentuated by the presence of DOPE in the lipoplexes. The results are interpreted in terms of some current models of lipoplex formation.  相似文献

The sodium salt of N,N'-hexane-bis (1-dodecen-1-ylsuccinamic acid) is an anionic dimeric (gemini) surfactant. A flooding penetration scan of this surfactant in water demonstrates a sequence of lyotropic phases at room temperature (20 degrees C). Preparation of surfactant-water mixtures has resulted in a phase diagram which shows that the same sequence of phases exists up to 100 degrees C. These phases are tentatively assigned to the sequence: micellar to normal hexagonal (H1) to cubic (V1) to lamellar (Lalpha). The interfacial tension at the n-heptane/water interface has been determined in the presence of this surfactant. The surfactant head group area at the interface is large (2.8+/-0.3 nm2 at 298 K) and the interfacial tension above the critical micelle concentration is low (7 mN m(-1)), but considerably higher than the ultra-low values that have been reported for cationic dimeric surfactants at various hydrocarbon-water interfaces.  相似文献

We present the exact solutions of the linear Poisson-Boltzmann equation for several problems relevant to electrostatics of DNA complexes with cationic lipids. We calculate the electrostatic potential and electrostatic energy for lamellar and inverted hexagonal phases, concentrating on the effects of dielectric boundaries. We compare our results for the complex energy with the known results of numerical solution of the nonlinear Poisson-Boltzmann equation. Using the solution for the lamellar phase, we calculate the compressibility modulus and compare our findings with the experimental data available. Also, we treat charge-charge interactions across, along, and between two low-dielectric membranes. We obtain an estimate for the strength of electrostatic interactions of one-dimensional DNA smectic layers across the lipid membrane. We discuss in the end some aspects of two-dimensional DNA condensation and DNA-DNA attraction in the DNA-lipid lamellar phase in the presence of di- and trivalent cations. We analyze the equilibrium DNA-DNA separations in lamellar complexes using the recently developed theory of electrostatic interactions of DNA helical charge motifs.  相似文献

Two phases coexist in an aqueous system that contains the two surfactants cationic gemini 12‐3‐12,2Br? and anionic SDS. An aqueous two‐phase system (ATPS) is formed in a narrow region of the ternary phase diagram different from that of traditional aqueous cationic‐anionic surfactant systems. In that region, the molar ratio of gemini to SDS varies with the total concentration of surfactants. ATPS not only has higher stability but also has longer phase separation time for the new systems than that of the traditional system. Furthermore, the optical properties of ATPS are different at different total concentrations. All of these experimental observations can be attributed to the unique properties of gemini surfactant and the synergy between the cationic gemini surfactant and the anionic surfactant SDS.  相似文献

Mixed surfactant systems have the potential to impart controlled combinations of functionality and pore structure to mesoporous metal oxides. Here, we combine a functional glucopyranoside surfactant with a cationic surfactant that readily forms liquid crystalline mesophases. The phase diagram for the ternary system CTAB/H(2)O/n-octyl-beta-D-glucopyranoside (C(8)G(1)) at 50 degrees C is measured using polarized optical microscopy. At this temperature, the binary C(8)G(1)/H(2)O system forms disordered micellar solutions up to 72 wt% C(8)G(1), and there is no hexagonal phase. With the addition of CTAB, we identify a large area of hexagonal phase, as well as cubic, lamellar and solid surfactant phases. The ternary phase diagram is used to predict the synthesis of thick mesoporous silica films via a direct liquid crystal templating technique. By changing the relative concentration of mixed surfactants as well as inorganic precursor species, surfactant/silica mesostructured thick films can be synthesized with variable glucopyranoside content, and with 2D hexagonal, cubic and lamellar structures. The domains over which different mesophases are prepared correspond well with those of the ternary phase diagram if the hydrophilic inorganic species is assumed to act as an equivalent volume of water.  相似文献

A detailed physicochemical study is presented on a new class of cationic amphiphiles, Sunfish amphiphiles, recently designed, synthesized, and tested for gene delivery. These materials have two hydrophobic tails, connected to the cationic pyridinium headgroup at the 1- and 4-positions. Two extreme morphologies can be visualized, i.e. one by back-folding involving association of both tails at one side of the pyridinium ring and one by independent unfolding of the tails, the two molecular geometries leading to considerable differences in the aggregate morphology. The behavior of six members of the Sunfish family in mixtures with DOPE, applying different conditions relevant for transfection, has been studied by a combination of techniques (DLS, DSC, NMR, SAXS, Cryo-TEM, fluorescence, etc.). The effects of structural parameters such as the presence of unsaturation in the tails and length of the alkyl chains on the properties of the aggregates have been assessed. A correlation of these structural data with cellular transfection efficiencies reveals that the highest transfection efficiency is obtained with those amphiphiles that are easily hydrated, form fluid aggregates, and undergo a transition to the inverted hexagonal phase in the presence of plasmid DNA (p-DNA) at physiological ionic strength.  相似文献

Cationic gemini surfactant dimethylene-1,2-bis(dodecyldiethylammonium bromide), referred to as C12C2C12(Et), was synthesized. The effect of sodium salicylate (NaSal) on the assembly formation and transition of this cationic gemini surfactant solution was studied. Addition of NaSal induced rich aggregate morphologies in the C12C2C12(Et) system. The microstructures and rheological responses resulting from the addition of NaSal were studied systematically to explore the interaction between gemini surfactants and hydrotropic salts. The rich aggregation behavior can be attributed to the special molecular structure of the gemini surfactant and the appropriate interaction between the surfactant and NaSal. The study of gemini surfactant and hydrotropic salt interaction brings promise for applications in materials synthesis as soft templates.  相似文献

The self-assembly behavior of a cationic surfactant (dodecyltrimethylammonium, DTA) with DNA as counterion in mixtures of water and n-alcohols (decanol, octanol, hexanol, butanol, and ethanol) was investigated. The phase diagrams were established and the different regions of the phase diagram characterized with respect to microstructure by (2)H NMR, small-angle X-ray scattering (SAXS), and other techniques. The DNA-DTA surfactant is soluble in all of the studied alcohols, showing increased solubility from decanol down to ethanol. All of the phase diagrams are analogous with respect to the occurrence of liquid crystalline (LC) regions, but the area of the LC region increases as one goes from decanol to ethanol. In all phase diagrams, hexagonal phases (of the reversed type) for the alcohol-rich side and lamellar phases for the other side were detected. For balanced proportions of the components, there is a coexistence of the lamellar and the hexagonal phase, here detected with a double quadrupole splitting in the (2)H NMR spectra. The correctness of the phase diagrams is confirmed by the fact that along the tie-lines the splitting magnitude remains nearly constant. All of the alcohols except for ethanol act as cosurfactants penetrating the DNA-DTA film. Adding salt to the ternary mixtures causes an increase in the unit cell dimension of the lamellar and the hexagonal phases. The phase diagram becomes more complicated when butanol is used for the alcohol phase. Here, there is the occurrence of a new isotropic phase with some properties analogous to those of the disordered sponge (L3) phase obtained for simple surfactant systems.  相似文献

The phase and miscibility behavior of a triple-chain phosphatidylcholine (TPHPC) and a single-chain surfactant (CTAB) were investigated in aqueous dispersions and in monolayers at the air/water interface. CTAB can be incorporated in the TPHPC monolayer because of its complementary molecule shape and reduces the tilt angle of TPHPC. The type of phases and the phase sequence (L2 - LS) are the same in the pure TPHPC monolayer and in the TPHPC/CTAB (80:20 mol:mol) mixture. No indication of any ordering of adsorbed DNA was observed. In the aqueous dispersions, TPHPC exhibits an inverted hexagonal phase above the chain melting. The addition of 30 mol% CTAB leads to the appearance of a lamellar Lalpha phase. The binding of DNA to the mixture is obvious but this is accompanied by a separation of the two lipids what is supported by monolayer experiments. The system has no long-term stability. The main reason seems to be not only the stronger interaction of DNA with CTAB, but also especially the unexpected weak interaction between CTAB and TPHPC. The transfection efficiency is lower compared with lipofectamine. The main disadvantage of this system is the cytotoxicity of CTAB, which could not be lowered by incorporation of CTAB in the TPHPC bilayer.  相似文献

Interactions between DNA and the cationic gemini surfactant trimethylene-1,3-bis(dodecyldimethylammonium bromide) (12-3-12) in aqueous solution have been investigated by UV-vis transmittance, zeta potential, and fluorescence emission spectrum. Complexes of DNA and gemini surfactant are observed in which the negative charges of DNA are neutralized by cationic surfactants effectively. The DNA-induced micelle-like structure of the surfactant due to the electrostatic and hydrophobic interactions is determined by the fluorescence spectrum of pyrene. It is found that the critical aggregation concentration (CAC) for DNA/12-3-12 complexes depends little on the addition of sodium bromide (NaBr) because of the counterbalance salt effect. However, at high surfactant concentration, NaBr facilitates the formation of larger DNA/surfactant aggregates. Displacement of ethidium bromide (EB) by surfactant evidently illustrates the strong cooperative binding between surfactant and DNA. In contrast to that in the absence of surfactant, the added NaBr at high surfactant concentration influences not only the binding of surfactant with DNA, but also the stability of DNA/EB complex.  相似文献

The pH-dependent phase behavior and hydroxide-ion adsorption ability of a series of (reduced) carbohydrate-based gemini surfactants were studied between pH 2 and 12. Static and dynamic light scattering were employed to address transitions in the aggregate morphologies and cryo-electron microscopy was used to provide further evidence for the morphologies present in solution. Changes in aggregate structure as a result of a change in solution pH and an accompanying change in protonation state or a change in molecular structure can be rationalized in terms of the variations in the packing parameter. In this paper we have focused our attention on the size of the carbohydrate moiety, the carbohydrate stereochemistry and the nature of the spacer (hydrophobic vs hydrophilic). At near neutral pH, most of the gemini surfactants form vesicles. Upon lowering of the pH, the vesicles undergo a transition toward wormlike micelles followed by a transition to spherical micelles. Upon increasing the solution pH, flocculation occurs due to charge neutralization followed at still higher pH by redispersion and charge reversal of the vesicles through the specific adsorption of hydroxide ions to the vesicular surface. Upon decreasing head group size at constant, but low, degrees of protonation, the packing parameter has a tendency to become larger than one resulting in the formation of inverted phases. Upon further decrease in the head group size, oil droplets are observed. In case of a hydrophobic spacer, the carbohydrate stereochemistry affects the pH of the transitions, but not the type of the transitions. By contrast, for a hydrophilic spacer, the pH of the transitions remains unaffected. Adsorption of hydroxide ions at basic pH follows similar trends, but was only found for vesicles and oil droplets. The large range of structural variations that we have examined allows a better understanding of the requirements for the phase transitions for carbohydrate-based gemini surfactants as well as for the physisorption of hydroxide ions to interfaces in general.  相似文献

The phase behavior as well as the microstructures of the cationic gemini surfactant and anionic conventional surfactant aqueous two-phase system (ASTP) have been studied. The ASTP formation can be attributed to the coexistence of different kinds of aggregates in the upper and lower phases. The effects of temperature, shearing, surfactant concentration and mixing molar ratio on the phase separation of the ASTP-forming systems are systematically investigated. The ASTP can be destroyed by applying shear and increasing temperature. In this process, the lamellar structures (flat bilayers) in the ASTP are transformed into vesicles. Variation of surfactant structure also affects the phase behavior and the aggregates transformation. Appropriate molecular packing is crucial for the formation of ASTP.  相似文献

An aqueous phase behavior was investigated for ionic liquid-related cationic gemini surfactant, [C12im-4-C12im][Br2], in which two dodecylimidazolium parts are linked by a spacer composed of four methylene units. Differential scanning calorimetry was used to detect phase boundaries among various phases, from which a temperature−composition (T−X) phase diagram was constructed. Lyotropic liquid-crystalline phases appearing in this mixture were characterized by means of polarized optical microscopy. It was revealed that (1) two types of liquid-crystalline phases, lamellar (La {{\hbox{L}}_\alpha } ) and hexagonal (H1), are formed; (2) H1 phase coexists with micellar solution (L1) over a wide concentration range from a few wt.% to ca. 80 wt.% of the surfactant; and (3) the hydrated solid with the composition of [C12im-4-C12im][Br2]·2H2O exhibits incongruent melting point at which the hydrated solid decomposes into H1 phase and unhydrated surfactant solid.  相似文献

应用紫外光谱、荧光探针、zeta 电位、动态光散射和凝胶电泳等方法探讨了阳离子gemini 表面活性剂C12H25N+(CH3)2―(CH2)6―(CH3)2N+C12H25·2Br-(12-6-12)与DNA之间的相互作用. 研究结果表明, 与传统表面活性剂相比, 偶联表面活性剂特殊的分子结构使其与DNA的作用更强烈. DNA引导表面活性剂在其链周围形成类胶束结构, 开始形成类胶束时对应的表面活性剂临界聚集浓度(CAC)比纯表面活性剂临界胶束浓度(CMC)低两个数量级. CAC与DNA的浓度无关, 而与表面活性剂之间的疏水作用以及表面活性剂与DNA之间的静电吸引作用密切相关. Zeta 电位和凝胶电泳结果显示了DNA链所带负电荷逐渐被阳离子表面活性剂中和的过程. 借助原子力显微镜(AFM)成功观察到了松散的线团状DNA, 球状体随机地分散在DNA链上形成类似于串珠的结构、尺寸较大的球形复合物以及其由于吸附多余的表面活性剂重新带正电而被溶解得到的较小DNA/12-6-12聚集体. 圆二色(CD)光谱结果显示, 12-6-12可以诱导DNA的构象发生改变.  相似文献

Aqueous lyotropic liquid crystal (LLC) assemblies with bicontinuous cubic morphologies (Q-phases) have shown promise in applications ranging from selective chemical separations to ion transporting media, yet universal design criteria for amphiphiles that adopt these unique structures remain elusive. Recent reports have demonstrated that cationic gemini surfactants exhibit a tendency to form bicontinuous cubic LLCs as compared to single-tail amphiphiles; however, the universality of this surfactant design motif in stabilizing Q-phases remains untested. Herein, we report the modular synthesis of a new class of anionic gemini surfactants derived from aliphatic carboxylic acids and demonstrate their unexpectedly strong propensity to form gyroid LLC phases with unprecedented stability between 25 and 100 °C over amphiphile concentration windows up to 20 wt % wide. By systematically varying the alkyl spacer length and surfactant counterions (Na(+), K(+), and (CH(3))(4)N(+)), we identify molecular motifs that favor formation of technologically useful bicontinuous cubic LLC morphologies.  相似文献
