The tautomeric equilibria between 2-pyridone and 2-hydroxypyridine forms of methoxy, chloro, and nitro derivatives of 3-cyano-4-(2-, 3-, and 4-substituted phenyl)-6-phenyl-2(1H)-pyridones were evaluated from UV/Vis spectral data. Linear solvation energy relationships of Kamlet–Taft and Catalán-rationalized solvent have influence on tautomeric equilibria. Transmission of substituent effect was analyzed by the Hammett equation. Quantum chemical calculations were performed by density functional theory (B3LYP). The experimental data were interpreted with the aid of time-dependent density functional method. Electron density distribution was analyzed by Bader’s analysis. It was found that substituents of different electronic properties change the extent of conjugation, and affect intramolecular charge transfer character. Theoretical calculations and experimental results gave insight into the influence of the molecular conformation on the transmission of substituent effects, as well as on contribution of different solvent–solute interactions.  相似文献

The configurational isomers of 2,5-dimethoxy-1,4-bis-[2-(2,4-dimethoxyphenyl)ethenyl]benzene have been investigated by ab initio (4-21G level) and MOPAC-AM1 semiempirical methods. The calculations were guided by and compared with single crystal X-ray results of the trans,trans-isomer determined at 298 K and 103 K. The barriers of rotation of the latter isomer in the free and the solid state were enumerated. A comparison with related molecules, in particular 1,4-bis[2-(3,4,5-trimethoxyphenyl)ethenyl]benzene, is made. The positioning of substituents is shown to be of importance for ring rotational movements and NMR shifts, as well as for UV /Vis spectroscopy. © 1996 by John Wiley & Sons, Inc.  相似文献

A number of 1-methyl-4-[2-(1-naphthyl)ethenyl]pyridinium salts were synthesized by condensation of 1,4-dimethylpyridinium iodide with naphthalene-1-carbaldehydes containing dimethylamino groups in positions 4 and 5. The obtained salts are characterized by negative solvatochromism which is the most pronounced for those possessing 1,8-bis(dimethylamino)naphthalene (“proton sponge”) and 1,2,2,3-tetramethyl-2,3-dihydroperimidine residues.  相似文献

The structural characterisation of the molecule 1,4-bis[2-cyano-2-(o-pyridyl)ethenyl] benzene obtained through Knoevenagel condensation is reported. The single crystals, as light brown rods, were cultured from a chloroform solution using a slow evaporation method at ambient temperature. The compound crystallised in the monoclinic system belonging to the C2/c space group with a = 26.4556(9) Å, b = 3.73562(10) Å, c = 18.4230(6) Å, β = 109.841(4)° and the asymmetric unit comprising Z = 4. The structure is ordered and the molecules of the title compound exhibited a lattice with water molecules located at sites of inversion and two-fold axial symmetries. Thus, only halves of the molecules are symmetrically independent. The lattice is reported and contrasted with X-ray single-crystal diffraction and theoretical calculations of 1,4-bis(1-cyano-2-phenylethenyl)benzene. By using density functional theory (DFT) and second order Moller-Plesset (MP2) theoretical calculations, the ground state geometry in the whole molecule at the B3LYP/6-31+G(d,p), and MP2/6-31+G(d,p) theory levels, respectively, were optimised. The DFT calculations showed a quasi-planar structure of the molecule, whereas the wave function-based MP2 method afforded a non-planar optimised structure with significant torsion angles between the pyridine and phenyl rings.  相似文献

Abstract

The present study describes the synthesis of novel chalcone based 1-{3-[3-(substituted phenyl) prop-2-enoyl] phenyl} thioureas (4a-c) using Claisen Schmidt condensation and investigates their protective role in diabetic conditions and associated oxidative stress. Spectral properties for the synthesized compounds were studied. Novel compounds were screened for antihyperglycemic effect in streptozotocin (STZ)-induced diabetic rats in a 6?week study and compound 4b exhibited significant (p?≤?.05) results similar to the standard drug glipizide. Treatment of diabetic animals with compound 4b (10 and 20?mg/kg, body weight) for 12?weeks, reduced the increased blood glucose level significantly (p?≤?.01) and restored attenuated serum biochemical parameters to normal levels. Altered antioxidant enzyme activity was also considerably (p?≤?.01) restored to the standard normal range. β-apoptotic TUNEL assay indicated that compound 4b (AI: 1.2?±?0.05) could prevent further β-cell death in the pancreas of diabetic animals in a dose-dependent manner, which highlights its potentiality as an effective antihyperglycemic agent.  相似文献

通过E-[4-[2-(4-羟基苯基)乙烯基]吡啶与1,n-二溴烷烃亲核取代反应合成了4,4’-[1,n-亚烷基双[(E)-2-(4-氧苯基)乙烯基]]双吡啶[n=2(Ia),3(Ib),4(Ic),6(Id)].用元素分析、红外、紫外和质子核磁共振谱鉴定了Ia-Id的结构.将Ia-Id的稀溶液用中压汞灯和低压汞灯交替照射,发现其分子内光环加成反应的存在,并且随着亚烷基碳链的延长,反应速度加快.研究还发现锌离子可以与毗陡环上氮原子发生螯合作用使分子内光环加成反应加快.本文化合物荧光很弱,在较高浓度下有较强分子间激基缔合物荧光.  相似文献

In the present study o-phenylenediamine and naphtene-1-acetic acid/2-naphthoxyacetic acid were used as a starting material through a series of steps and 2-(naphthalen-1-ylmethyl/Naphthalen-2-yloxymethyl)-1H-benzimidazol-1-yl]acetohydrazide 5a, 5b were obtained. In the first series 1,3,4-oxadiazole derivatives have been synthesized from Schiff base of the corresponding hydrazide i.e. 2-[2-(naphthalen-1-ylmethyl)-1H-benzimidazol-1-yl]acetohydrazide 5a by using Chloramin-T. In the second series 1,3,4-oxadiazole has been synthesized from 2-{2-[(naphthalen-2-yloxy)methyl]-1Hbenzimidazol-1-yl}acetohydrazide 5b by using phosphorous oxychloride and aromatic acid. These compounds were evaluated by IR, NMR, Mass spectrometry, elemental analysis and finally in vitro anticancer evaluation was carried out by NCI 60 Cell screen at a single high dose (10–5 M) on various panel/cell lines. One compound 7c was found to be the most active on breast cancer cell line and compounds 4b and 7d were moderately active.  相似文献

The metabolites of (E) [corrected]-4-[1-[4-[2-dimethylamino)ethoxy]phenyl]- 2-(4-isopropylphenyl)-1-butenyl]phenyl monophosphate, TAT-59, (1), a potent antitumor agent for hormone-dependent tumors, and derivatives of TAT-59 were synthesized to confirm its proposed structure. The structure and the Z-configuration of the metabolites (2a-8a) were confirmed by comparison with synthesized authentic compounds. All of the metabolites and the derivatives of TAT-59 were tested for a binding affinity toward estrogenic receptors in vitro and antiuterotrophic activity in vivo. Most of the metabolites possessed remarkable binding affinity toward estrogenic receptors as well as fairly good antiuterotrophic activity.  相似文献
