A study on a rhodium(II )‐catalyzed reaction of N‐sulfonyl‐1,2,3‐triazoles with 2‐alkenols is reported. The reaction is initiated by insertion of an α‐imino carbene into the O–H linkage of alcohol, forming a 2‐alkenoxy enamide intermediate. A thermal [3,3]‐sigmatropic rearrangement follows to yield 2‐substituted 2‐amino ketone in a stereoselective manner. The successful application of this methodology to a formal synthesis of (–)‐α‐conhydrine is also described.  相似文献

In recent years, the microwave‐assisted ionic‐liquid method has been accepted as a promising methodology for the preparation of nanomaterials and cellulose‐based nanocomposites. Applications of this method in the preparation of cellulose‐based nanocomposites comply with the major principles of green chemistry, that is, they use an environmentally friendly method in environmentally preferable solvents to make use of renewable materials. This minireview focuses on the recent development of the synthesis of nanomaterials and cellulose‐based nanocomposites by means of the microwave‐assisted ionic‐liquid method. We first discuss the preparation of nanomaterials including noble metals, metal oxides, complex metal oxides, metal sulfides, and other nanomaterials by means of this method. Then we provide an overview of the synthesis of cellulose‐based nanocomposites by using this method. The emphasis is on the synthesis, microstructure, and properties of nanostructured materials obtained through this methodology. Our recent research on nanomaterials and cellulose‐based nanocomposites by this rapid method is summarized. In addition, the formation mechanisms involved in the microwave‐assisted ionic‐liquid synthesis of nanostructured materials are discussed briefly. Finally, the future perspectives of this methodology in the synthesis of nanostructured materials are proposed.  相似文献

Convenient and efficient one pot method for the synthesis of 2,5‐disubstituted‐1,3,4‐oxadiazoles based on the reaction of N,N‐dimethyl amides with acid hydrazides has been developed. The methodology is applied to a wide range of difference aryl hydrazide and difference N,N‐dimethyl amides to 2,5‐disubstituted‐1,3,4‐oxadiazoles yield the in good to excellent yields. A ligand‐enabled Pd‐catalyzed Suzuki–Miyaura coupling of haloterthienyls for the synthesis of various thiophene‐based π‐conjugated oligomers including quinquethiophenes is demonstrated. An indolyl phosphine ligand plays an important role in this transformation. Thiopheneboronic acids were well applied, which might open up a window for the application of thiopheneboronic acids in the synthesis of thiophene‐based π‐conjugated oligomers in materials chemistry.  相似文献

A novel and efficient one‐pot method has been developed for the synthesis of cyclopropane‐fused bicyclic amidines on the basis of a CuBr2‐mediated oxidative cyclization of carbanions. The usefulness of this unique multicomponent strategy has been demonstrated by the use of a wide variety of substrates to furnish novel cyclopropane‐containing amidines with a quaternary center in very good yields. This ketenimine‐based approach provides straightforward access to biologically active and pharmaceutically important 3‐azabicyclo[n .1.0]alkane frameworks under mild conditions. The synthetic power of this methodology is exemplified in the concise synthesis of the pharmaceutically important antidepressant drug candidate GSK1360707 and key intermediates for the synthesis of amitifadine, bicifadine, and narlaprevir.  相似文献

In the last twenty years DMC has been employed as an efficient methylating and methoxycarbonylating agent with several monodentate and bidentate nucleophiles, showing great selectivity and unexpected results. In this short review we report on yet another application of DMC chemistry i.e. the synthesis of 5‐membered N‐ and O‐heterocycles. In these reactions DMC acts as a sacrificial molecule since it is not present in the final products, but only in the reaction intermediates as an halogen‐free leaving group. This DMC‐based synthesis of heterocycles resulted of general application, as it is effective for aliphatic and aromatic 1,4‐diols, incorporating several functionalities (primary, secondary, tertiary, allylic, phenolic), as well as, for bifunctional compounds i.e. 4‐amino‐1‐butanol. This synthetic procedure was also employed for industrially relevant compounds such as (‐)‐norlabdane oxide and isosorbide showing to maintain the chiral integrity of the substrate. In one case intramolecular cyclisation of isosorbide was also observed to achieve a strained tricyclic derivative. Comparing this reaction methodology with a chlorine based procedure, the DMC‐mediated pathway is quantitative, occurs in one step, does not require any chlorine‐based chemical or strong acid and does not produce any chlorinated waste material.  相似文献

A simple and straightforward methodology toward the synthesis of novel 1‐(4,5‐diphenylpyrimidin‐2‐yl)thiourea has been developed by a one‐step reaction of isoflavones with amidinothiourea. A series of 16 new compounds was synthesized. All compounds were characterized by FT‐IR, NMR, and elemental analysis. The structure of a typical compound was established by X‐ray diffraction. A variety of substrates can participate in the process with good yields and high purities, making this methodology suitable for library synthesis in drug discovery.  相似文献

Reported herein is an ortho‐oxygenative 1,2‐difunctionalization of diarylalkynes under merged gold/organophotoredox catalysis to access highly functionalized 2‐(2‐hydroxyaryl)‐2‐alkoxy‐1‐arylethan‐1‐ones. Detailed mechanistic studies suggested a relay process, initiating with gold‐catalyzed hydroalkoxylation of alkynes, to generate enol‐ether followed by a key formal [4+2]‐cycloaddition reaction. The successful application of the present methodology was also shown for the synthesis of benzofurans.  相似文献

cis‐2,6‐Tetrahydropyran is an important structural skeleton of bioactive natural products. A facile synthesis of cis‐2,6‐disubstituted‐3,6‐dihydropyrans as cis‐2,6‐tetrahydropyran precursors has been achieved in high regio‐ and stereoselectivity with high yields. This reaction involves a palladium‐catalyzed decarboxylative allylation of various 3,4‐dihydro‐2H‐pyran substrates. Extending this reaction to 1,2‐unsaturated carbohydrates allowed the achievement of challenging β‐C‐glycosylation. Based on this methodology, the total syntheses of (±)‐centrolobine and (+)‐decytospolides A and B were achieved in concise steps and overall high yields.  相似文献

A facile and benign synthetic strategy is proposed for the synthesis of 2‐arylpyrroloacridin‐1(2H )‐ones via a lactic acid‐catalyzed three‐component reaction of dimedone, various anilines, and isatins under solvent‐free conditions. Avoidance of hazardous organic solvents, the use of a one‐pot multicomponent procedure for the synthesis of 2‐arylpyrroloacridin‐1(2H )‐ones, operational simplicity, no need for column chromatography, lactic acid utilization as a bio‐based organic compound, reusability, homogeneity, and commercial availability of the catalyst, and superior synthetic performance are some important aspects of this methodology to access a series of pyrroloacridine motifs with potentially biological scaffolds.  相似文献

In contrast to the traditional multistep synthesis, herein an efficient and fewer‐steps new synthetic strategy is demonstrated for the facile preparation of organic‐electronically important D–π–A–π–D‐type oligoaryls through sequential direct C?H arylations. This methodology has shown that the synthesis of thieno[3,4‐c]pyrrole‐4,6‐dione (TPD)‐ or furano[3,4‐c]pyrrole‐4,6‐dione (FPD)‐centred target molecules could be accessed step‐economically either from the core structure (acceptor) or from the end structure (donor), which supplied a more flexible and succinct new synthetic alternative to the preparation of the π‐functional small‐molecule semiconducting materials. The limited availability of highly valuable arenesulfonyl fluorides seriously hinders their further application in many research fields including medicinal chemistry and chemical biological, organic synthesis, polymer preparation, etc. We report herein a mild and efficient copper‐free Sandmeyer‐type fluorosulfonylation reaction of various arenediazonium salts to prepare valuable arenesulfonyl fluorides using K2S2O5 as both a reductant and a practical sulfonyl source in combination with N‐fluorobenzenesulfonimide as an effective fluorine source. This methodology provides an attractive route to diverse important arenesulfonyl fluorides given the overall practicality and scope.  相似文献

An iron‐based cross‐dehydrogenative coupling (CDC) approach was applied for the diversity‐oriented synthesis of coumestrol‐based selective estrogen receptor modulators (SERMs), representing the first application of CDC chemistry in natural product synthesis. The first stage of the two‐step synthesis of coumestrol involved a modified aerobic oxidative cross‐coupling between ethyl 2‐(2,4‐dimethoxybenzoyl)acetate and 3‐methoxyphenol, with FeCl3 (10 mol %) as the catalyst. The benzofuran coupling product was then subjected to sequential deprotection and lactonization steps, affording the natural product in 59 % overall yield. Based on this new methodology other coumestrol analogues were prepared, and their effects on the proliferation of the estrogen receptor (ER)‐dependent MCF‐7 and of the ER‐independent MDA‐MB‐231 breast cancer cells were tested. As a result, new types of estrogen receptor ligands having an acetamide group instead of the 9‐hydroxyl group of coumestrol were discovered. Both 9‐acetamido‐coumestrol and 8‐acetamidocoumestrol were found more active than the natural product against estrogen‐dependent MCF‐7 breast cancer cells, with IC50 values of 30 and 9 nM , respectively.  相似文献

Mechanochemistry has recently emerged as an environmentally friendly solventless synthesis method enabling a variety of transformations including those impracticable in solution. However, its application in the synthesis of well‐defined nanomaterials remains very limited. Here, we report a new bottom‐up mechanochemical strategy to rapid mild‐conditions synthesis of organic ligand‐coated ZnO nanocrystals (NCs) and their further host–guest modification with β‐cyclodextrin (β‐CD) leading to water‐soluble amide‐β‐CD‐coated ZnO NCs. The transformations can be achieved by either one‐pot sequential or one‐step three‐component process. The developed bottom‐up methodology is based on employing oxo‐zinc benzamidate, [Zn44‐O)(NHOCPh)6], as a predesigned molecular precursor undergoing mild solid‐state transformation to ZnO NCs in the presence of water in a rapid, clean and sustainable process.  相似文献

A novel methodology is presented for the synthesis of 3‐substituted 2‐thioxo‐2,3‐dihydroquinazolin‐4(1H)‐one derivatives based on an efficient tandem multicomponent reaction using copper bromide as catalyst. This methodology is based on the multicomponent one‐pot reaction of methyl 2‐bromobenzoate, phenylisothiocyanate derivatives and sodium azide in the presence of copper bromide and l ‐proline under basic conditions. To show the generality of the method, various phenylisothiocyanates bearing electron‐donating or electron‐withdrawing functionalities were used and the desired products were obtained in high isolated yields.  相似文献

α‐amino nitrones are found to have a great significance in the synthesis of peptides. Simple reaction methodology, high yield, and atom efficiency in these reactions have made this protocol highly attractive. The synthetic route may be extended to the synthesis of dipeptides in enantiomerically pure form. A simple and straightforward methodology towards the synthesis of 2,4,5‐trisubstituted pyrimidines has been developed by one‐step reaction of isoflavones with thiourea or guanidine. Ten 2‐mercapto‐4,5‐diphenylpyrimidines and eight 2‐amino‐4,5‐diphenylpyrimidines were reported. All of them were characterized by FT‐IR, NMR, and elemental analysis. A variety of substrates can participate in the process with good yields and high purities, making this methodology suitable for library synthesis in drug discovery. J. We report an environmentally friendly, efficient and practical method for the synthesis of 2‐aminobenzothiazoles by a copper(II)‐catalyzed tandem reaction of 2‐haloanilines with isothiocyanates under ligand‐ and solvent‐free conditions in air. The developed methodology conforms to the principles of 'green chemistry' and addresses the shortage of such methods for the synthesis of 2‐aminobenzothiazoles. The rhodium‐catalyzed asymmetric N‐selective coupling of pyrazole derivatives with terminal allenes gives access to enantioenriched secondary and tertiary allylic pyrazoles, which can be employed for the synthesis of medicinally important targets. The reaction tolerates a large variety of functional groups and labelling experiments gave insights into the reaction mechanism. This new methodology was further applied in a highly efficient synthesis of JAK 1/2 inhibitor (R)‐ruxolitinib.  相似文献
