Herein, we report the synthesis of biocompatible triplex Ag@SiO2@mTiO2 core–shell nanoparticles (NPs) for simultaneous fluorescence‐surface‐enhanced Raman scattering (F‐SERS) bimodal imaging and drug delivery. Stable Raman signals were created by typical SERS tags that were composed of Ag NPs for optical enhancement, a reporter molecule of 4‐mercaptopyridine (4‐Mpy) for a spectroscopic signature, and a silica shell for protection. A further coating of mesoporous titania (mTiO2) on the SERS tags offered high loading capacity for a fluorescence dye (flavin mononucleotide) and an anti‐cancer drug (doxorubicin (DOX)), thereby endowing the material with fluorescence‐imaging and therapeutic functions. The as‐prepared F‐SERS dots exhibited strong fluorescence when excited by light at 460 nm whilst a stable, characteristic 4‐Mpy SERS signal was detected when the excitation wavelength was changed to longer wavelength (632.8 nm), both in solution and after incorporation inside living cells. Their excellent biocompatibility was demonstrated by low cytotoxicity against MCF‐7 cells, even at a high concentration of 100 μg mL?1. In vitro cell cytotoxicity confirmed that DOX‐loaded F‐SERS dots had a comparable or even greater therapeutic effect compared with the free drug, owing to the increased cell‐uptake, which was attributed to the possible endocytosis mechanism of the NPs. To the best of our knowledge, this is the first proof‐of‐concept investigation on a multifunctional nanomedicine that possessed a combined capacity for fast and multiplexed F‐SERS labeling as well as drug‐loading for cancer therapy.  相似文献

The one‐dimensional (1D) transition‐metal oxide MoO3 belt is synthesized and characterized with X‐ray diffraction, scanning electron microscopy, and Raman spectroscopy. Charge‐transfer‐(CT) enhanced Raman scattering of 4‐mercaptobenzoic acid (4‐MBA) on a 1D MoO3 belt was investigated experimentally and theoretically. The chemical enhancement of surface‐enhanced Raman scattering (SERS) of 4‐MBA on the MoO3 belt by CT is in the order of 103. The SERS of 4‐MBA was investigated theoretically by using a quantum chemical method. The remote SERS of 4‐MBA along the 1D MoO3 belt (the light excitation to one side of the MoO3 belt, and the SERS spectrum is collected on the other side of the MoO3 belt) is also shown experimentally, which provides potential applications of SERS. The incident polarization dependence of remote SERS spectra has also been investigated experimentally.  相似文献

The availability of sensitive, reproducible, and stable substrates is critically important for surface‐enhanced Raman spectroscopy (SERS)‐based applications, but it presently remains a challenge. In this work, well‐aligned zinc gallate (ZnGa2O4) nanorod arrays grown on a Si substrate by chemical vapor deposition were used as templates to fabricate SERS substrates by deposition of Ag nanoparticles onto the ZnGa2O4 nanorod surfaces. The coverage of the Ag nanoparticles on the ZnGa2O4 nanorod surfaces was easily controlled by varying the amount of AgNO3. SERS measurements showed that the number density of Ag nanoparticles on the ZnGa2O4 nanorod surfaces had a great effect on SERS activity. The SERS signals collected by point‐to‐point and SERS mapping images showed that as‐prepared SERS substrates exhibited good spatial uniformity and reproducibility. Detection of melamine molecules at low concentrations (1.0×10?7 M ) was used as an example to show the possible application of such a substrate. In addition, the effect of benzoic acid on the detection of melamine was also investigated. It was found that the SERS signal intensity of melamine decreased greatly as the concentration of benzoic acid was increased.  相似文献

An ultrasensitive surface‐enhanced Raman spectroscopy (SERS) sensor based on rolling‐circle amplification (RCA)‐increased “hot‐spot” was developed for the detection of thrombin. The sensor contains a SERS gold nanoparticle@Raman label@SiO2 core‐shell nanoparticle probe in which the Raman reporter molecules are sandwiched between a gold nanoparticle core and a thin silica shell by a layer‐by‐layer method. Thrombin aptamer sequences were immobilized onto the magnetic beads (MBs) through hybridization with their complementary strand. In the presence of thrombin, the aptamer sequence was released; this allowed the remaining single‐stranded DNA (ssDNA) to act as primer and initiate in situ RCA reaction to produce long ssDNAs. Then, a large number of SERS probes were attached on the long ssDNA templates, causing thousands of SERS probes to be involved in each biomolecular recognition event. This SERS method achieved the detection of thrombin in the range from 1.0×10?12 to 1.0×10?8 M and a detection limit of 4.2×10?13 M , and showed good performance in real serum samples.  相似文献

Graphene‐enhanced Raman scattering (GERS) is emerging as an important method due to the need for highly reproducible, quantifiable, and biocompatible active substrates. As a result of its unique two‐dimensional carbon structure, graphene provides particularly large enhanced Raman signals for molecules adsorbed on its surface. In this work, the GERS signals of a test molecule, 4‐mercaptobenzoic acid (4‐MBA), with reproducible enhancement factors are discussed and compared with surface‐enhanced Raman scattering (SERS) signals from highly active substrates, covered with spherical silver nanoparticles. It is shown that chemical interactions between the molecule and graphene can result in a frequency shift in the graphene‐enhanced Raman signal of the molecule.  相似文献

Surface‐enhanced Raman scattering (SERS) is a process with origins, electromagnetic and chemical. The electromagnetic enhancement consists of the excitation of surface plasmons in the metallic support of the thin film. With only the electromagnetic enhancement mechanism, the surface spectra should not differ from volume Raman spectra. However, between SERS and volume Raman spectra, there are differences resulting from the chemical reactions taking place at the polymer/metal interface, intermediated by solvent molecules, that finally depend on the types of polymers and metallic supports. Polyaniline (PAN) is an excellent material to emphasize the chemical component of SERS. This is due to its particular structure with a repeating unit that contains two entities at different weights—a reduced state and an oxidized state–that, in turn, react differently with a metallic substrate. SERS spectra depend on the oxidizing properties of the metal surface, which involves an intermediate compound of the types Ag2O and Au2O3 when N‐methyl‐2‐pyrrolidinone is used as the solvent. This article presents new results concerning the surface chemical effects that produce variations of the PAN SERS spectra. The SERS spectra of the PAN emeraldine base (PAN‐EB) layered on Au support are characterized by a semiquinoid structure that we believe is induced on the intermediate compound Au2O3. In the presence of H2SO4, the SERS spectra change gradually as the degree of acid protonation doping increases. The SERS spectra of the fully protonated PAN‐EB are identical to those obtained on PAN emeraldine salt (PAN‐ES) synthesized by cyclic voltammetry in an acid medium and are invariable with the type of metallic support. The SERS spectra show that the emeraldine salt can be partially or totally deprotonated with water or NH4OH. The deprotonation is complete for the Ag support and partial for the Au support. The SERS spectra of the fully protonated PAN‐EB are characterized by a double band with maxima at about 1330 and 1370 cm−1. Although the generation process of positive charge on the macromolecular chain of PAN‐EB doped in the presence of (C4H9)4NBF4 is similar to that due to protonic acid doping, involving cation addition (C4H or H+ ions, respectively) in SERS spectra, the complex band situated at about 1330–1370 cm−1 no longer appears. The doping of PAN‐EB with FeCl3 produces two polymer forms: a salt type characterized by a protonated structure similar to that found for PAN‐ES and a base type similar to the leucoemeraldine form. © 2000 John Wiley & Sons, Inc. This letter reports the first observation of both surface enhanced Raman scattering (SERS) and surface enhanced resonance Raman scattering (SERRS) from the transition metal complex tris(2,2′-bipyridine)ruthenium (II), Ru(bpy)32+, adsorbed on a silver electrode from acetonitrile (ACN). The assignment of these spectra as valid examples of SERS and SERRS in a non-aqueous environment is based on the following criteria: (1) in situ demonstration of monolayer surface coverage of Ru(bpy)32+ using double potential step chronocoulometry (DPSCC); (2) the Raman signals are most intense after surface roughening by anodization; (3) the Raman spectra are potential dependent in the non-faradaic potential region; (4) the measured enhancement factors are greater ilian 106; (5) the surface spectra are frequency shifted relative to their bulk counterpart; and (6) several other molecules also exhibit non-aqueous SERS and SERRS behavior. These results are highly significant in that generality of surface enhanced Raman spectroscopy has been extended into the rich domain of nonaqueous electrochemistry.  相似文献

We report a new simple method to fabricate a highly active SERS substrate consisting of poly‐m‐phenylenediamine/polyacrylonitrile (PmPD/PAN) decorated with Ag nanoplates. The formation mechanism of Ag nanoplates is investigated. The synthetic process of the Ag nanoplate‐decorated PmPD/PAN (Ag nanoplates@PmPD/PAN) nanofiber mats consists of the assembly of Ag nanoparticles on the surface of PmPD/PAN nanofibers as crystal nuclei followed by in situ growth of Ag nanoparticles exclusively into nanoplates. Both the reducibility of the polymer and the concentration of AgNO3 are found to play important roles in the formation and the density of Ag nanoplates. The optimized Ag nanoplates@PmPD/PAN nanofiber mats exhibit excellent activity and reproducibility in surface‐enhanced Raman scattering (SERS) detection of 4‐mercaptobenzoic acid (4‐MBA) with a detection limit of 10?10 m , making the Ag nanoplates@PmPD/PAN nanofiber mats a promising substrate for SERS detection of chemical molecules. In addition, this work also provides a design and fabrication process for a 3D SERS substrate made of a reducible polymer with noble metals.  相似文献

Anisotropic noble‐metal structures are attracting increasing attention because of interesting size‐ and shape‐dependent properties and have emerging applications in the fields of optics and catalysis. However, it remains a significant challenge to overcome chemical contributions and acquire molecular insight into the relationship between Raman enhancement and photocatalytic activity. This study gives visualized experimental evidence of the anisotropic spatial distribution of Raman signals and photocatalytic activity at the level of single nanometer‐thin Au microtriangles and microhexagons. Theoretical simulations indicate an anisotropic spatial distribution and sharpness‐dependent strength of the electric‐field enhancement. Analysis by using statistical surface‐enhanced Raman scattering (SERS) supports this view, that is, Raman enhancement is on the order of corner>edge>face for a single microplate, but SERS measurements at different depths of focus also imply a concentration‐dependent feature of SERS signals, especially at the corners and edges. Similarly, the SERS signals of product molecules in plasmonic photocatalysis also exhibit asymmetrical strengths at different corners of the same microplate. However, by examining the variations in the relative intensities of the SERS peaks, the difference in the photocatalytic activities at the corners, edges, and faces has been successfully calculated and is highly consistent with electric‐field simulations, thus indicating that an increased number of molecules adsorbed at specific sites does not necessarily lead to a higher conversion ratio in noble‐metal photocatalysis. Our strategy weakens the assumed impact of plasmonic local heating and, to a certain extent, excludes the influence of concentration effects and chemical contributions in noble‐metal photocatalysis, thus clearly profiling plasmon‐related characteristics. This study also promises a new research direction to understand the enhancement mechanism of SERS‐active structures.  相似文献

The chemisorption of 1,3,4‐oxadiazole‐2‐thiol (ODT) on gold nanorods has been investigated by using surface‐enhanced Raman spectroscopy (SERS) and density functional theory (DFT). Although most of the SERS spectra have remarkable similarity to the normal Raman spectra of the pure analyte, the adsorption of ODT on a gold surface leads to a drastic change in its Raman spectrum and distinct vibrational features are obtained with gold nanorods and spherical nanoparticles. Simulated Raman spectra for hybrid systems that consist of an oxadiazole moiety coordinated to a Au20 gold cluster provided valuable information about the coordination mode and enabled us to assign vibration modes.  相似文献

Graphene oxide‐wrapped gold nanorods (GO@AuNRs) offer efficient drug delivery as well as NIR laser photothermal therapy (PTT) in vitro and in vivo. However, no real‐time observation of drug release has been reported to better understand the synergy of chemotherapy and PTT. Herein, surface‐enhance Raman spectroscopy (SERS) is employed to guide chemo‐photothermal cancer therapy by a two‐step mechanism. In the presence of GO as an internal standard, SERS signals of DOX (doxorubicin) loaded onto GO@AuNRs are found to be pH‐responsive. Both DOX and GO show strong SERS signals before the DOX@GO@AuNRs are endocytic. However, when the DOX@GO@AuNRs enter acidic microenvironments such as endosomes and/or lysosomes, the DOX signals start decreasing while the GO signals remain the same. This plasmonic antenna could be used to identify the appropriate time to apply the PTT laser during chemo‐photothermal therapy.  相似文献

Chiral carbonaceous nanotubes (CNT) were successfully used in plasmon‐free surface‐enhanced Raman scattering (SERS) for the first time. Further modification of TiO2 nanocrystals on the chiral CNTs successfully realized the recycling of SERS substrate as chiral CNT/TiO2 hybrids. The high SERS sensitivity of methylene blue (MB) over the chiral CNT/TiO2 hybrids is ascribed to the laser‐driven birefringence induced by the helical structure, which provides much more opportunities for the occurrence of Raman scattering. The TiO2 nanocrystals highly dispersed on the surface and inside the hollow cavity of chiral CNTs can completely degrade the MB under the solar light irradiation, leading to the self‐cleaning of SERS substrate. The present research opens a new way for the application of chiral inorganic materials in plasmon‐free SERS detection.  相似文献

To detect biomarkers from human exhalation, air flow dynamics on the nanoparticle surface were explored by a surface‐enhanced Raman scattering (SERS) sensor. A hollow Co‐Ni layered double hydroxide (LDH) nanocage on Ag nanowires (Ag@LDH) was prepared. Ag nanowires provided amplified Raman signals for trace determination; hollow LDH nanocages served as the gaseous confinement cavity to improve capture and adsorption of gaseous analytes. The Raman intensity and logarithmic analyte concentration exhibit an approximately linear relationship; the detection limit of SERS sensors for aldehyde is 1.9×10?9 v/v (1.9 ppb). Various aldehydes in mixed mimetic gas are distinguished by Raman spectra statistical analysis assisted by multivariate methods, including principal component analysis and hierarchical cluster analysis. The information was recorded in a barcode, which can be used for the design and development of a desktop SERS sensor analysis system for large‐scale lung cancer detection.  相似文献

Hollow gold nanospheres (HGNs) with nanopinholes were prepared by chemical reduction of HAuCl4 with aqueous KBH4 in the presence of the Bu4PBr template. The HGNs could be served as substrates for surface‐enhanced Raman scattering (SERS), which exhibited much higher surface enhancement than the solid gold nanoparticles in case of Rhodamine 6G as a molecule probe. Moreover, the HGNs could also be used in the modified electrode to detect DNA and superior to the solid counterpart of the similar size. Based on the detailed characterizations by transmission electron micrograph (TEM), field emission scanning electron micrograph (FESEM) and UV‐visible absorption spectroscopy, a plausible mechanism for forming HGNs was proposed. Meanwhile, the promoting effect of the hollow gold nanospheres on the SERS was discussed briefly.  相似文献

Surface enhanced Raman spectroscopy (SERS) is a powerful optical sensing technique that can detect analytes of extremely low concentrations. However, the presence of enough SERS probes in the detection area and a close contact between analytes and SERS probes are critical for efficient acquisition of a SERS signal. Presented here is a light‐powered micro/nanomotor (MNM) that can serve as an active SERS probe. The matchlike AgNW@SiO2 core–shell structure of the nanomotors work as SERS probes based on the shell‐isolated enhanced Raman mechanism. The AgCl tail serves as photocatalytic nanoengine, providing a self‐propulsion force by light‐induced self‐diffusiophoresis. The phototactic behavior was utilized to achieve enrichment of the nanomotor‐based SERS probes for on‐demand biochemical sensing. The results demonstrate the possibility of using photocatalytic nanomotors as active SERS probes for remote, light‐controlled, and smart biochemical sensing on the micro/nanoscale.  相似文献

Hydrogen sulfide (H2S) has emerged as an important gasotransmitter in diverse physiological processes, although many aspects of its roles remain unclear, partly owing to a lack of robust analytical methods. Herein we report a novel surface‐enhanced Raman scattering (SERS) nanosensor, 4‐acetamidobenzenesulfonyl azide‐functionalized gold nanoparticles (AuNPs/4‐AA), for detecting the endogenous H2S in living cells. The detection is accomplished with SERS spectrum changes of AuNPs/4‐AA resulting from the reaction of H2S with 4‐AA on AuNPs. The SERS nanosensor exhibits high selectivity toward H2S. Furthermore, AuNPs/4‐AA responds to H2S within 1 min with a 0.1 μM level of sensitivity. In particular, our SERS method can be utilized to monitor the endogenous H2S generated in living glioma cells, demonstrating its great promise in studies of pathophysiological pathways involving H2S.  相似文献

The mechanism of surface‐enhanced Raman spectroscopy (SERS) is not very clear in view of the magnitude of the contribution of electromagnetic factor as well as the chemical mechanism. This report presents the extent of adsorption at different temperatures in terms of signal enhancements in SERS employing silver nanoparticles (AgNPs) of various shapes as substrate and dye molecules, crystal violet or Rhodamine 6G, as model Raman probes. Initially, the SERS signal increases with increasing temperature until a maximum intensity is reached, before it gradually decreases with increasing temperature. This trend is independent of the shape of the Raman substrates and probes. However, the temperature at which maximum intensity is obtained may depend upon the nature of the Raman probe. The energetics involved in the chemisorption process between dye molecules and AgNPs were determined through isothermal titration calorimetry and their implications for the observed SERS signals were assessed. The maximum heat change occurred at the temperature at which the maximum signal enhancement in SERS was obtained and the enhanced interaction at optimum temperature was confirmed by absorption spectroscopy.  相似文献

Sensitive detection of molecules by using the surface‐enhanced Raman scattering (SERS) technique depends on the nanostructured metallic substrate and many efforts have been devoted to the preparation of SERS substrates with high sensitivity, stability, and reproducibility. Herein, we report on the fabrication of stable monolithic nanoporous silver (NPS) by chemical dealloying of Ag–Al precursor alloys with an emphasis on the effect of structural evolution on SERS signals. It was found that the dealloying conditions had great influence on the morphology (the ligament/pore size) and the crystallization status, which determined the SERS signal of rhodamine 6G on the NPS. NPS with small pores, low residual Al, and perfect crystallization gave high SERS signals. A high enhancement factor of 7.5×105 was observed on bare NPS obtained by dealloying Ag30Al70 in 2.5 wt % HCl at room temperature followed by 15 min aging at around 85 °C. After coating Ag nanoparticles on the NPS surface, the enhancement factor increased to 1.6×108 owing to strong near‐field coupling between the ligaments and nanoparticles.  相似文献

An ultrasensitive surface enhanced Raman scattering (SERS) method has been designed to selectively and sensitively detect lysozyme. The gold chip as the detection substrate, the aptamer‐based target‐triggering cascade multiple cycle amplification, and gold nanoparticles (AuNPs) bio‐barcode Raman probe enhancement on the gold substrate are employed to enhance the SERS signals. The cascade amplification process consists of the nicking enzyme signaling amplification (NESA), the strand displacement amplification (SDA), and the circular‐hairpin‐assisted exponential amplification reaction (HA‐EXPAR). With the involvement of an aptamer‐based probe, two amplification reaction templates, and a Raman probe, the whole circle amplification process is triggered by the target recognition of lysozyme. The products of the upstream cycle (NESA) could act as the “DNA trigger” of the downstream cycle (SDA and circular HA‐EXPAR) to generate further signal amplification, resulting in the immobility of abundant AuNPs Raman probes on the gold substrate. “Hot spots” are produced between the Raman probe and the gold film, leading to significant SERS enhancement. This detection method exhibits excellent specificity and sensitivity towards lysozyme with a detection limit of 1.0×10?15 M . Moreover, the practical determination of lysozyme in human serum demonstrates the feasibility of this SERS approach in the analysis of a variety of biological specimens.  相似文献

A surface‐enhanced Raman scattering (SERS) technique shows extraordinary features for a range of biological and biomedical applications. Herein, a series of novel bioorthogonal SERS nanoprobes were constructed with Gold nanoflower (AuNF) and Raman reporters, the signals of which were located in a Raman‐silent region of biological samples. AS1411 aptamer was also co‐conjugated with AuNF through a self‐assembled monolayer coverage strategy. Multiplex SERS imaging using these nanoprobes with three different bioorthogonal small‐molecule Raman reporters is successfully achieved with high multiplexing capacity in a biologically Raman‐silent region. These Raman nanoprobes co‐conjugated with AS1411 showed high affinity for tumor cells with overexpressed nucleolin and can be used for selective tumor cell screening and tissue imaging.  相似文献
