Development of an electrochemical DNA biosensor for the direct detection and discrimination of double-stranded oligonucleotide (dsDNA) corresponding to hepatitis C virus genotype 3a, without its denaturation, using a gold electrode is described. The electrochemical DNA sensor relies on the modification of the gold electrode with 6-mercapto-1-hexanol and a self-assembled monolayer of 14-mer peptide nucleic acid probe, related to the hepatitis C virus genotype 3a core/E1 region. The increase of differential pulse voltammetric responses of methylene blue, upon hybridization of the self-assembled probe with the target ds-DNA to form a triplex is the principle behind the detection and discrimination. Some hybridization experiments with non-complementary oligonucleotides were carried out to assess whether the developed DNA sensor responds selectively to the ds-DNA target. Diagnostic performance of the biosensor is described and the detection limit was found to be 1.8 × 10−12 M in phosphate buffer solution, pH 7.0. The relative standard deviation of measurements of 100 pM of target ds-DNA performed with three independent probe-modified electrodes was 3.1%, indicating a remarkable reproducibility of the detection method.  相似文献

We have investigated the interaction between diclofenac and deoxyribonucleic acid (DNA) by the electrochemical method. On glassy carbon electrode, the voltammetric curve of diclofenac showed an oxidation peak, which was obviously influenced by the addition of DNA. Accordingly, a series of electrochemical experiments were carried out to elucidate the interaction between diclofenac and DNA. Based on the diclofenac–DNA interaction, a biosensor for diclofenac was developed. When a film of DNA was immobilized on the electrode surface, DNA showed an oxidation signal promoted by graphene oxide. Due to the diclofenac–DNA interaction, the peak current of DNA decreased and the peak potential shifted positively with increasing the concentration of diclofenac. The response of the biosensor was linear to the concentration of diclofenac in the range of 1 to 130?μM. Using such a biosensor, the photodegradation of diclofenac was successfully monitored.  相似文献

This paper describes a disposable indicator-free electrochemical DNA biosensor applied to the detection of apolipoprotein E (apoE) sequences in PCR samples. In the indicator-free assays, the duplex formation was detected by measuring the electrochemical signal of the guanine base of nucleic acids. The biosensor format involved the immobilisation of an inosine-modified (guanine-free) probe onto a screen-printed electrode (SPE) transducer and the detection of the duplex formation in connection with the square-wave voltammetric measurement of the oxidation peak of the guanine of the target sequence.The indicator-free scheme has been characterised using 23-mer oligonucleotides as model: parameters affecting the hybridisation assay such as probe immobilisation conditions, hybridisation time, use of hybridisation accelerators were examined and optimised.The analysis of PCR samples (244 bp DNA fragments, obtained by amplification of DNA extracted from human blood) required a further optimisation of the experimental procedure. In particular, a lower steric hyndrance of the probe modified surface was essential to allow an efficient hybridisation of the target DNA fragment. Negative controls have been performed using the PCR blank and amplicons unrelated to the immobilised probe. A 10 min hybridisation time allowed a full characterisation of each sample.  相似文献

The electrochemical behavior of ceftriaxone was investigated on a carbon-nanotube-modified glassy carbon (GC-CNT) electrode in a phosphate buffer solution, pH = 7.40, and the results were compared with those obtained using the unmodified one [glassy carbon (GC) electrode]. During oxidation of ceftriaxone, an irreversible anodic peak appeared, using both modified and unmodified electrodes. Cyclic voltammetric studies indicated that the oxidation process is irreversible and diffusion-controlled. The number of electrons exchanged in the electrooxidation process was obtained, and the data indicated that ceftriaxone is oxidized via a one-electron step. The results revealed that carbon nanotube promotes the rate of oxidation by increasing the peak current. In addition, ceftriaxone was oxidized at lower potentials, which thermodynamically is more favorable. These results were confirmed by impedance measurements. The electron-transfer coefficients and heterogeneous electron-transfer rate constants for ceftriaxone were reported using both the GC and GC-CNT electrodes. Furthermore, the diffusion coefficient of ceftriaxone was found to be 2.74 × 10−6 cm2 s−1. Binding of ceftriaxone to human serum albumin forms a kind of electroreactive species. The percentage of interaction of ceftriaxone with protein was also addressed. A sensitive, simple, and time-saving differential-pulse voltammetric procedure was developed for the analysis of ceftriaxone, using the GC-CNT electrode. Ceftriaxone can be determined with a detection limit of 4.03 × 10−6 M with the proposed method.  相似文献

An amperometric glucose biosensor was developed based on the immobilization of glucose oxidase in the organically modified silicate (ormosil)-polyvinyl acetate (PVA) matrix onto a Prussian Blue (PB)-modified glassy carbon electrode. A higher stability PB-modified electrode was prepared by the electrochemical deposition of FeCl3, K3[Fe(CN)6] and ethylenediamine tetraacetic acid (EDTA) under cyclic voltammetric (CV) conditions. The effects of the potential range of CV conditions, electrolyte cations, applied potential, pH, temperature and co-existing substances were investigated. The detection limit of the glucose biosensor was 8.1 μmol·L−1 (S/N = 3) with a linear range from 20 μmol·L−1 to 2 mmol·L−1 (R = 0.9965). The biosensor presented a fast response and good selectivity. Additionally, excellent reproducibility and stability of the biosensor were observed. A screen-printed carbon working electrode within a commercially available screen-printed three-electrode assembly was modified by using a composite of multiwalled carbon nanotubes (MWCNT) dispersed in polyethylenimine (PEI) followed by covering with the calf thymus dsDNA layer. Several electrochemical methods were used to characterize the biosensor and to evaluate damage to the surface-attached DNA: square wave voltammetry of the [Ru(bpy)3]2+ redox indicator and mediator of the guanine moiety oxidation, cyclic voltammetry and electrochemical impedance spectroscopy in the presence of the [Fe(CN)6]3−/4− indicator in solution. Due to high electroconductivity and large surface area of MWCNT and positive charge of PEI, the MWCNT–PEI composite is an advantageous platform for the DNA immobilization by the polyelectrolyte complexation and its voltammetric and impedimetric detection. In this respect, the MWCNT–PEI interface exhibited better properties than the MWCNT–chitosan one reported from our laboratory previously. A deep DNA layer damage at incubation of the biosensor in quinazoline solution was found, which depends on the quinazoline concentration and incubation time. New voltammetric DNA biosensor based on a microcrystalline natural graphite–polystyrene composite film in the role of a transducer was used for the investigation of the interaction between model carcinogenic substance (2-aminofluorene; one of the most extensively studied examples of the aromatic amine class of carcinogens) and calf thymus double-stranded DNA (dsDNA). The layer of dsDNA immobilized at the electrode surface was utilized as a biocomponent responsive interface. The biosensor was characterized regarding the detection of DNA damage (induced by direct interaction with 2-aminofluorene) using square wave voltammetric responses of the guanine and adenine moieties and cyclic voltammetric responses of the anionic redox indicator [Fe(CN)6]4–/3– present in the solution.  相似文献

An electrochemical biosensor for the detection of DNA based a peptide nucleic acid (PNA) capture probe (CP) modified indium tin oxide electrode (ITO) is described in this report. After hybridization, a threading intercalator, N,N′-bis[(3-propyl)-imidazole]-1,4,5,8-naphthalene diimide (PIND) imidazole complexed with Ru(bpy)2Cl (PIND-Ru, bpy = 2,2′-bipyridine), was introduced to the biosensor. PIND-Ru selectively intercalated to double-stranded DNA (ds-DNA) and became immobilized on the biosensor surface. Voltammetric tests showed highly stable and reversible electrochemical oxidation/reduction processes and the peak currents can directly be utilized for DNA quantification. When the tests were conducted in an amine-containing medium, Tris-HCl buffer for example, a remarkable improvement in the voltammetric response and noticeable enhancements of voltammetric and amperometric sensitivities were observed due to the electrocatalytic activity of the [Ru(bpy)2Cl] redox moieties. Electrocatalytic current was observed when as little as 3.0 attomoles of DNA was present in the sample solution.  相似文献

For electrocatalytic determination of NADH, a graphite electrode modified with titania sol-gel/Meldola’s Blue/MWCNT/Nafion nanocomposite was proposed. The composition of the matrix film was optimised in terms of the content of carbon nanotubes and Nafion. Incorporation of a redox mediator, Meldola’s Blue, into the nanocomposite film enabled electrocatalytic determination of NADH at a low potential, −50 mV. For determination of ethanol, alcohol dehydrogenase (ADH) was immobilized into the matrix layer. Experimental conditions affecting the biosensor response were examined, including enzyme loading, temperature of measurement and pH of background electrolyte. Assessments of the analytical characteristics of the biosensor were performed with respect to sensitivity, limit of detection, operational stability, repeatability and reproducibility. The proposed biosensor showed electrocatalytic activity toward oxidation of ethanol with sensitivity of 2.24 μA L mmol−1, linear range from 0.05 to 1.1 mmol L−1, and limit of detection of 25 μmol L−1. The apparent Michaelis-Menten constant was 1.24 mmol L−1, indicating a high biological affinity of ADH/titania sol-gel/Meldola’s Blue/MWCNT/Nafion electrode for ethanol. The developed biosensor was tested in determinations of ethanol content in alcoholic beverages.  相似文献

In the present study, a comparison between a disposable electrochemical DNA biosensor and a Vibrio fischeri-based luminescent sensor for the detection of toxicants in water samples was made.In order to realize this study, a disposable electrochemical DNA biosensor has been reported. The DNA biosensor is assembled by immobilizing double stranded Calf Thymus DNA onto the surface of a disposable carbon screen-printed electrode. The oxidation signal of the guanine base, obtained by a square wave voltammetric scan, is used as analytical signal to detect the DNA damage; the presence of low molecular weight compounds with affinity for nucleic acids is measured by their effect on the guanine oxidation peak.Wastewater samples provided during First European Interlaboratory Exercise on water toxicity in the course of the project SWIFT-WFD were analyzed, and biosensor results were compared with a currently used toxicity test ToxAlert®100 based on the bioluminescence inhibition of Vibrio fischeri. This test have been used because is rapid, easy handling and cost effectively responses for the toxicity assessment in real water samples.The results showed a promising correlation between two tests used for the detection of toxic compounds in water samples.  相似文献

A novel voltammetric sensor, based on single-walled carbon nanotubes (SWNT) dispersed in Nafion and modified glassy carbon electrode (GCE), was fabricated and used to determine the trace amounts of dihydromyricetin (DMY). The electrochemical behavior of DMY at this sensor was investigated in 0.1 mol L−1 sulfuric acid solutions + 0.1 mol L−1 NaCl by cyclic voltammetry and squarewave voltammetry. Compared with bare GCE, the electrode presented an excellent response of DMY through an adsorption-controlled quasi-reversible process. Under the optimum conditions, the response peak currents were linear relationship with the DMY concentrations in the range of 1.0 × 10−7–1.0 × 10−5 mol L−1 with a detection limit of 9 × 10−8 mol L−1. Based on this voltammetric sensor, a simple and sensitive electroanalytical method for DMY was proposed and applied to quantitative determination of DMY in Ampelopsis grossedentata samples. In addition, the oxidation mechanism was proposed and discussed, which could be a reference for the pharmacological action of DMY in clinical study.  相似文献

Molecular wires containing copper(II) (CuMW), in the form of the coordination polymer (Cu(II)4(bpp)4(maa)8(H2O)2).2H2O (bpp=1,3-bis(4-pyridyl)propane, maa=2-methylacrylic acid), and multiwalled carbon nanotubes (CNT) have been combined to prepare a paste electrode (CuMW/CNT/PE). The voltammetric response of the CuMW/CNT/PE to metformin (MET) was significantly greater than that of electrodes prepared from other materials, because of both the surface effect of CuMW and CNT and coordination of MET with the Cu(II) ion in the CuMW. A novel voltammetric method for determination of MET is proposed. In pH 7.2 Britton–Robinson buffer, using single sweep voltammetry, the second-order derivative peak current for oxidation of MET at 0.97 V (relative to SCE) increased linearly with MET concentration in the range 9.0 × 10−7–5.0 × 10−5 mol L−1 and the detection limit was 6.5 × 10−7 mol L−1. A novel, sensitive and selective electrochemical hybridisation biosensor was developed for the detection of the hepatitis B virus (HBV) using a manganese(II) complex as electrochemical indicator and a DNA probe-modified carbon paste electrode as the biosensor (DNA/CPE). The results showed that this complex could be accumulated electrochemically the immobilised dsDNA layer rather than in the single-stranded DNA (ssDNA) layer. On the basis of this, the manganese complex was used as an electrochemical hybridisation indicator for the detection of oligonucleotides related to HBV. The hybridisation event was evaluated on the basis of the difference between the reduction signals of the manganese(II) complex with the probe DNA prior to and post hybridisation with a target sequence using a differential pulse mode. Several factors affecting the immobilisation and hybridisation of oligonucleotides as well as the indicator’s accumulation were investigated. Experiments with a non-complementary and mismatch sequences demonstrated the good selectivity of the biosensor. Using this approach, the HBV target oligonucleotide’s sequence could be quantified over arange from 0.22 ng L?1 to 5.40 ng L?1, with a linear correlation coefficient of 0.9994 and the limit of detection of 0.07 ng L?1.  相似文献

Fosamprenavir is a pro-drug of the antiretroviral protease inhibitor amprenavir and is oxidizable at solid electrodes. The anodic oxidation behavior of fosamprenavir was investigated using cyclic and linear sweep voltammetry at boron-doped diamond and glassy carbon electrodes. In cyclic voltammetry, depending on pH values, fosamprenavir showed one sharp irreversible oxidation peak or wave depending on the working electrode. The mechanism of the oxidation process was discussed. The voltammetric study of some model compounds allowed elucidation of the possible oxidation mechanism of fosamprenavir. The aim of this study was to determine fosamprenavir levels in pharmaceutical formulations and biological samples by means of electrochemical methods. Using the sharp oxidation response, two voltammetric methods were described for the determination of fosamprenavir by differential pulse and square-wave voltammetry at the boron-doped diamond and glassy carbon electrodes. These two voltammetric techniques are 0.1 M H2SO4 and phosphate buffer at pH 2.0 which allow quantitation over a 4 × 10−6 to 8 × 10−5 M range using boron-doped diamond and a 1 × 10−5 to 1 × 10−4 M range using glassy carbon electrodes, respectively, in supporting electrolyte. All necessary validation parameters were investigated and calculated. These methods were successfully applied for the analysis of fosamprenavir pharmaceutical dosage forms, human serum and urine samples. The standard addition method was used in biological media using boron-doped diamond electrode. No electroactive interferences from the tablet excipients or endogenous substances from biological material were found. The results were statistically compared with those obtained through an established HPLC-UV technique; no significant differences were found between the voltammetric and HPLC methods.  相似文献

The application of a disposable electrochemical DNA biosensor to wastewater samples is reported. The DNA biosensor is assembled by immobilising double-stranded calf thymus DNA on the surface of a disposable, carbon screen-printed electrode (SPE). The oxidation signal of the guanine base, obtained by a square wave voltammetric scan, is used as analytical signal. The presence of compounds with affinity for DNA is measured by their effect on the guanine oxidation. The comparison of the results with a toxicity test based on bioluminescent bacteria has confirmed the applicability of the method to real samples.  相似文献

A simple and highly selective electrochemical method has been developed for the simultaneous determination of hydroquinone (HQ) and catechol (CC) at a glassy carbon electrode covalently modified with penicillamine (Pen). The electrode is used for the simultaneous electrochemical determination of HQ and CC and shows an excellent electrocatalytical effect on the oxidation of HQ and CC upon cyclic voltammetry in acetate buffer solution of pH 5.0. In differential pulse voltammetric measurements, the modified electrode was able to separate the oxidation peak potentials of HQ and CC present in binary mixtures by about 103 mV although the bare electrode gave a single broad response. The determination limit of HQ in the presence of 0.1 mmol L−1 CC was 1.0 × 10−6 mol L−1, and the determination limit of CC in the presence of 0.1 mmol L−1 HQ was 6.0 × 10−7 mol L−1. The method was applied to the simultaneous determination of HQ and CC in a water sample. It is simple and highly selective.  相似文献

Electrochemical oxidation of methylthiomethyleneisoquinolinium chloride (MTMIQ), the first alkylthiomethyl substituted ammonium salt, which is fully miscible with water has been investigated by voltammetric (SWV) method using glassy carbon electrode. On the electrode, MTMIQ undergoes oxidation at the potential near Ep = 0.07V (vs. Ag/AgCl/3 M KCl). The influence of the pH of buffers, amplitude, frequency, step potential on the received signal was studied. The best results were obtained with a citrate buffer at a pH of 5. The oxidation peak current used for MTMIQ voltammetric determination was in the range of 2–8×10−5 mol L−1, LOD = 3.7×10−6, LOQ = 1.2×10−5. The product of the oxidation was accumulated at the working electrode and was investigated by spectroscopic method. Mechanistic pathways of the oxidation have been proposed.   相似文献

A very stable electroactive film of catechin was electrochemically deposited on the surface of activated glassy carbon electrode. The electrochemical behavior of catechin modified glassy carbon electrode (CMGCE) was extensively studied using cyclic voltammetry. The properties of the electrodeposited films, during preparation under different conditions, and the stability of the deposited film were examined. The charge transfer coefficient (α) and charge transfer rate constant (k s) for catechin deposited film were calculated. It was found that the modified electrode exhibited excellent electrocatalytic activity toward hydrazine oxidation and it also showed a very large decrease in the overpotential for the oxidation of hydrazine. The CMGCE was employed to study electrocatalytic oxidation of hydrazine using cyclic voltammetry, rotating disk voltammetry, chronoamperometry, amperometry and square-wave voltammetry as diagnostic techniques. The catalytic rate constant of the modified electrode for the oxidation of hydrazine was determined by cyclic voltammetry, chronoamperometry and rotating disk voltammetry and was found to be around 10−3 cm s−1 . In the used different voltammetric methods, the plot of the electrocatalytic current versus hydrazine concentration is constituted of two linear segments with different ranges of hydrazine concentration. Furthermore, amperometry in stirred solution exhibits a detection limit of 0.165 μM and the precision of 4.7% for replicate measurements of 40.0 μM solution of hydrazine.  相似文献

A ferrocenedicarboxylic acid modified carbon paste electrode was constructed and used as a fast and sensitive tool for the determination of captopril at trace level. It has been shown by direct current cyclic voltammetry and double step chronoamperometry that ferrocenedicarboxylic acid can catalyze the oxidation of captopril in aqueous buffer solution and produces a sharp oxidation peak current at about +0.49 vs. Ag/AgCl reference electrode. The square wave voltammetric peak currents of the electrode increased linearly with the corresponding captopril concentration in the range of 3.0 × 10−7–1.4 × 10−4M with a detection limit of 9.1 × 10−8 M. The influence of pH and potential interfering substances on the determination of captopril were studied. Electrochemical impedance spectroscopy was used to study the charge transfer properties at the electrode–solution interface. Finally, the sensor was examined as a selective, simple, and precise new electrochemical sensor for the determination of captopril in real samples, such as drug and urine, with satisfactory results.  相似文献

A nano-composite of DNA/poly(p-aminobenzensulfonic acid) bi-layer modified glassy carbon electrode as a biosensor was fabricated by electro-deposition method. The DNA layer was electrochemically deposited on the top of electropolymerized layer of poly(p-aminobenzensulfonic acid) (Pp-ABSA). Scanning electron microscopy, X-ray photoelectron spectroscopy and electrochemical impedance spectrum were used for characterization. It demonstrated that the deposited Pp-ABSA formed a 2-D fractal patterned nano-structure on the electrode surface, and which was further covered by a uniform thin DNA layer. Cyclic voltammetry and electrochemical impedance spectrum were used to characterize the deposition, and demonstrated the conductivity of the Pp-ABSA layer. The biosensor was applied to the detection of dopamine (DA) and uric acid (UA) in the presence of ascorbic acid (AA). In comparison with DNA and Pp-ABSA single layer modified electrodes, the composite bi-layer modification provided superior electrocatalytic actively towards the oxidation of DA, UA and AA, and separated the originally overlapped differential pulse voltammetric signals of UA, DA and AA oxidation at the bare electrode into three well-defined peaks at pH 7 solution. The peak separation between AA and DA, AA and UA was 176 mV and 312 mV, respectively. In the presence of 1.0 mM AA, the anodic peak current was a linear function of the concentration of DA in the range 0.19-13 microM. The detection limit was 88 nM DA (s/n=3). The anodic peak current of UA was also a linear function of concentration in the range 0.4-23 microM with a detection limit of 0.19 microM in the presence of 0.5 mM AA. The superior sensing ability was attributed to the composite nano-structure. An interaction mechanism was proposed.  相似文献
