Abstract

Hydrophobic interaction chromatography (HIC) with a polar bonded phase (“Acetamide”) developped for size exclusion chromatography (SEC) is described. Retention of proteins depends on the surface area of the stationary phase, the pH and ionic strength of the eluent. For efficient separation the pore diameter should be 25 nm or more. The surface area should be large to achieve retention even at low ionic strength. Separation is only possible with a gradient from high to low ionic strength. Gradient volumes of 10 empty column volumes with column lengths above 15 cm are recommended. Selectivity can be optimized via pH adjustment. The advantage of this column packing is its applicability for two different separation modes: SEC and HIC.  相似文献

Abstract

This paper examines the use of 15–20 micron wide-pore silica-based ether bonded phases for the preparative hydrophobic interaction chromatography of proteins. In particular, silyl ethers are immobilized on large particle silica in an analogous manner to previously developed ether bonded 5 um analytical supports. The preparative supports are reproducibly prepared and exhibit constant chromatographic retention for at least five months of continual use. Preparative columns can be operated for protein chromatography with peak shapes and capacity as predicted by the Snyder gradient elution model. Moreover, similar retention times are obtained relative to those on the 5 um analytical columns, enabling the direct transition and scale-up of separation. Gradient optimization is seen to directly parallel that performed on 5 um bonded ether analytical columns. Acceptable chromatographic resolution was obtained with sample capacity of >15 mg protein/ml column volume using a repetitive injection technique. A column clean-up strategy is examined for rapid and safe removal of contaminants. An illustrative example of use of the bonded ether preparative columns is made by application to soybean trypsin inhibitor purification. Initial results are presented on a column-switching method for the analytical monitoring of preparative separation.  相似文献

Abstract

Various microparticulate siliceous bonded stationary phases having weakly hydrophobic ligates were developed for HPLC of proteins and t-RNA's by hydrophobic interaction chromatography (HIC). It was confirmed that optimal separation of different types of biopolymers can be obtained by using a set of stationary phases having appropriate hydrophobic properties. Thus, the separation of t-RNA's is best carried out on stationary phases which are more hydrophobic than those optimal for HIC of proteins. Plots of log k' of both proteins and t-RNA's against the salt molality in the eluent yielded straight lines at sufficiently high salt concentrations in the eluent. The limiting slopes represent the hydrophobic interaction parameter for the particular chromatographic system and can serve as measures of the hydrophobic character of either the biopolymer or the stationary phase. Stationary phases with covalently bound polyether chains at the surface were found to be most suitable for HIC of proteins and t-RNA's.  相似文献

Regularities were revealed for the retention of different classes of organic compounds on stationary phases based on C60 and carbon nanotubes and on phases of mixed composition (C60 and dibenzo-24-crown-8, C60 and dibenzo-30-crown-10, and C60 and -cyclodextrin) in gas chromatography. A synergistic effect was observed for the stationary phases of mixed composition. Quantitative estimation of the dispersion potential demonstrated that the C60-based stationary phase differs significantly from hydrocarbon adsorbents and more closely resembles nonpolar liquid stationary phases. Under the conditions of solid-phase microextraction, an adsorbing element based on fullerene C60 can be used for the preconcentration of aromatic hydrocarbons.  相似文献

以苯乙烯-二乙烯基苯微球为基质,建立了一种新型离子色谱固定相的制备方法。在基质微球表面合成一层聚缩水甘油甲基丙烯酸酯聚合物层(GMA),随后与甲胺及1,4-丁二醇二环氧甘油醚(BDDE)交替反应,与其表面接枝上带正电荷的季铵基团,可用于阴离子的分离。通过改变接枝反应的次数,控制交换树脂的交换容量。自制色谱固定相能够用于7种常规阴离子分离分析,并用于自来水中常规阴离子的检测。分离检测结果可与商用离子色谱柱相比,同时水负峰能与氟离子完全分离,不影响氟离子的定量检测。  相似文献

The development of effective chiral stationary phases (CSPs) and separation strategies for the liquid chromatographic (LC) resolution of enantiomers has been beneficial to many scientific disciplines. Over the last decade the number and type of CSPs has expanded tremendously. Not only have new classes of chiral selectors been introduced, but also many of the first CSPs have been changed and/or improved. The second or third generation of a CSP often can be different from the original. This can be confusing and intimidating to someone just entering the area of LC enantiomeric separations. Fortunately, all CSPs can be categorized in one or another of a few classes. There are generally one or two columns that can accomplish the majority of separations in each class. The most correct analysis of the compositions of diverse analytes mixtures is significant for analytical studies in different fields; however, many prevalent analytes cannot be identified employing traditional partition gas chromatographic methods. Thus, the increasing requirements on analytes of isomeric compounds and the problems encountered in their separation demand a study of more diverse analytical systems which are characterised by higher selectivity. Therefore, the selectivity and polarities of various liquid crystals (rod-like, banana-shape, biforked, oxygen, sulphur, nitrogen, and metal containing molecules, Schiff-base, and polymeric dendrimers) employed as liquid crystalline stationary phases (LCSPs) have been discussed from both points of views, namely, their analytical applications and thermodynamic characteristics of infinitely diluted probes with different acceptor–donor properties. Extreme particular effort has been paid to the different interdependencies between the bound up chemical structures of liquid crystal molecules with their different acceptor–donor properties and the connected resolution capabilities in the interpretation of the probe—LCSP systems, on the basis of the \( { \ln }V_{g\left( T \right)} = f\left( {\frac{1}{T}} \right) \) and \( { \ln }\left( {\frac{{a_{1} }}{{w_{1} }}} \right)^{\infty } = f\left( {\frac{1}{T}} \right) \) dependencies, with regard to the LCSP compositions, which have been controlled by the counterbalancing of the enthalpy and entropy factors. The properties of binary systems composed of liquid crystalline poly(propyleneimine) dendrimers—rod-like molecules of liquid crystals and effects of the dendrimer structure, the chemical nature, and molecular size of the non-mesogens on the ability to dissolve in the liquid crystalline phases, have been interpreted. Practical applications of metallomesogenes and chiral stationary phases for analytical separation of different organic substances have also been taken into consideration.

高效液相色谱(HPLC)不仅是一种有效的分析分离手段,也是一种重要的高效制备分离技术。色谱柱是HPLC系统的核心,不同性能的填料是HPLC广泛应用的基础。硅胶是开发最早、研究最为深入、应用最为广泛的HPLC固定相基质,其制备方法主要有喷雾干燥法、溶胶-凝胶法、聚合诱导胶体凝聚法及模板法等。近年来,亚2μm小粒径硅胶、核-壳型硅胶、双孔径硅胶、介孔性硅胶、有机杂化硅胶等新型硅胶应用于HPLC并取得了色谱分离技术的飞速发展,例如基于亚2μm填料的超高压液相色谱技术、基于核-壳型填料的快速分离技术、基于杂化硅胶填料的高温液相色谱技术等。硅胶经表面化学键合、聚合物包覆等有机改性可制得先进的大分子限进填料、温敏性填料、手性填料等,大大扩展了HPLC的应用范围。本文对液相色谱用硅胶的制备方法、改性与修饰方法以及硅胶基质固定相的评价方法加以系统综述,概述了新型硅胶在HPLC中的应用进展,并对硅胶基质填料的发展方向与应用前景进行了展望。  相似文献

以自行合成的8.0μm大孔单分散亲水性交联聚甲基丙烯酸环氧丙酯微球为基质,采用化学改性方法合成了邻二醇配基新型高效疏水作用色谱填料。详细评价了该填料对蛋白质的分离性能、动力学吸附容量、质量回收率、硫酸铵起始浓度和流动相pH值对蛋白保留的影响。结果表明,这种高效疏水作用色谱填料的分离性能优良,分离了5种标准蛋白,对溶菌酶的活性回收率为96%±5%,吸附容量高达36.5 g/L。  相似文献

Abstract

TSKgel Phenyl-5PW preparative column of 200 × 55 mm I.D. was evaluated with respect to resolution, sample loading capacity and applications to the purification of enzymes. The preparative column provided similar separations as analytical column (75 × 7.5 mm I.D.) and 150 × 21.5 mm I.D. preparative column. The sample loading capacity was 200 – 1000 mg depending on the sample. If the slight decrease in resolution is acceptable, much more samples could be applied. Lipoxidase, phosphoglucose isomerase and lactate dehydrogenase could be purified to a great extent with high recovery of activity (more than 80%).  相似文献

The development of new high-performance anion-exchange stationary phases is one of the priority tasks of modern ion chromatography (IC). Most of the research in this field is aimed at improving the selectivity of anion exchangers and increasing the sensitivity, efficiency, and rapidity of the analysis. The selectivity of the stationary phases is largely determined by the chemistry of the adsorbent surface; therefore, it is particularly important to understand the relationship between the structure of the functional layer of the anion exchanger and its chromatographic properties. This review is devoted to methods for modifying polymer matrices in order to obtain highly efficient anion exchangers for the separation of inorganic and organic anions.  相似文献
