Abstract

A simple, sensitive, reliable, and rapid HPTLC method has been developed for the determination of pitavastatin calcium in tablet dosage form. Identification and determination were performed on aluminum backed silica gel 60F254 washed with methanol. The mobile phase of ethyl acetate‐methanol‐ammonia‐1 drop formic acid (7:2:0.8) calibration plots were established showing the dependence of response (peak area) on the amount chromatographed. The spot were scanned at 245 nm. The method has a linear range of 50–250 ng/spot. The method was validated for selectivity, repeatability, and accuracy. The method was used for determination of the compound in commercial pharmaceutical dosage forms. It is a more effective option than other chromatographic techniques in routine quality control.  相似文献

A simple reverse-phase method for the selective quantification of pitavastatin calcium (PIT) and its related substances was developed. The method demonstrated an excellent separation between PIT and each of 15 impurities (including its isomers and degradants) within a short run time of 12 min by HPLC. A rapid resolution similar to that of UHPLC was achieved using high flow rate on superficially porous C18 stationary phase. A synergistic combination of quality by design approach and use of a superficially porous column delivered a HPLC method with ultra-high performance. Forced degradation studies proved the method to be highly specific (mass balance > 98 %) and the structures of major degradation products were proposed based on LC–MS analysis. The results of validation proved the method to be highly precise (%RSD < 4), accurate (recoveries in range of 100 ± 7 %), linear (r 2 > 0.999) and sensitive (LOQ ≤ 0.02 % and LOD ≤ 0.005 %) for all the impurities and drug. Use of multivariate analysis helped to incorporate high robustness in the method. The method is valuable for quantification of PIT and its related substances in both drug substance and oral solid dosage form.

An efficient and stereoselective syntheses of all the possible stereoisomers of coronafacic acid (CFA) has been developed. The stereochemistries of C3a and C7a were controlled in a diastereoselective Diels-Alder type cycloaddition using a chiral auxiliary. CFA and 6-epi-CFA were synthesized by hydrogenation of a common intermediate. During the synthesis of 6-epi-CFA, we established that its cis-fused configuration is important for the introduction of C4-C5 double bond by dehydration. This report is the first practical synthesis of both 6-epi-CFA, and its enantiomer.  相似文献

Abstract— The composition of retinal isomers in bacteriorhodopsin (bR) in purple membrane (PM) was determined by photoelectric response measurements using a sandwich-type electrochemical cell. The measured amplitude of the photocurrent obtained from a dark-adapted sample was 55% lower than that from a light-adapted sample. This ratio, 55:45, would correspond to the 13- cis /aU- trans isomer ratio of retinal in the dark if the 13- cis form of the pigment did not give a response. This amplitude change correlated with the visible spectral shift of bR. The isomer ratio in the dark depended only weakly on the temperature of the electrolyte, whereas the retinal isomerization rate strongly depended on the temperature and the pH of the electrolyte in the cell. Our results indicate that photoelectric response is elicited only by a species originating from bR containing all- trans retinal and that the behavior of the response in the dark is associated with the pKa of the proton release kinetics of Asp-85.  相似文献

For the first time, in this study electrochemical oxidation behavior of pesticide maneb is evaluated. Due to the structure electroanalytical quantification of maneb has not been exploited enough. Maneb electrochemical behavior was investigated using glassy carbon (GC), graphene modified glassy carbon (GR/GC) and boron doped diamond (BDD) electrodes. It is shown that only BDD shows satisfactory results toward maneb detection. Based on this, a simple, sensitive and selective electroanalytical method for determination of pesticide maneb using differential pulse voltammetry (DPV) is proposed, with a working linear range of 80–3000 nM and the limit of detection of 24 nM. The developed methodology has been applied for the determination of maneb in river water samples with satisfactory recovery. Additionally, this green method, being simple, fast, and free of chemical‐reduction reagents, offers several advantages over modified electrodes and expands the scope of BDD based electrochemical sensing devices, with promise for wider applications in environmental analysis.  相似文献

Strigolactones (SLs) are signaling molecules involved in plant development and governing interactions with soil microorganisms in the rhizosphere as well as the germination of parasitic weeds. Developing their use in Crop Protection is a promising approach to a sustainable agriculture by mitigating biotic and abiotic stresses. Recently, a new class of lactam analogues of SL has emerged, namely strigolactams, displaying outstanding potency to induce the germination of parasitic weed O. cumana as well as enhanced chemical and soil stability. Herein, we describe the stereoselective synthesis of GR24 and GR18 lactams harnessing the chemistry of chiral keteniminium (KI) salts, in particular the unprecedented reactivity of chloro-substituted KI, supported by DFT calculations. We disclose subsequently the biological activity on corn of the 32 stereopure strigolactams prepared, highlighting the crucial influence of stereochemistry and lactam substitution, rationalized by docking analyses. Finally, we performed stability studies in soil, which reveal that stereoisomers display very different half-lives, reflecting the significant impact of stereochemistry on degradation kinetics.  相似文献

Two trans stereoisomers of 3‐methylcyclopentadecanol (=muscol), (1R,3R)‐ 2 and (1S,3S)‐ 2 , were efficiently synthesized from (3RS)‐3‐methylcyclopentadecanone (=muscone; (3RS)‐ 1 ) by a highly stereoselective reduction (Scheme). L‐Selectride® (=lithium tri(sec‐butyl)borohydride) was used, followed by the enantiomer resolution by lipase QLG (Alcaligenes sp.). The cis stereoisomers of muscol, (1S,3R)‐ 2 and (1R,3S)‐ 2 , were obtained by the Mitsunobu inversion of (1R,3R)‐ 2 and (1S,3S)‐ 2 , respectively (Scheme). The absolute configuration of (1R,3R)‐ 2 was determined by X‐ray crystal‐structure analysis of its 3‐nitrophthalic acid monoester, 2‐[(1R,3R)‐3‐methylcyclopentadecyl hydrogen benzene‐1,2‐dicarboxylate ((1R,3R)‐ 3b ), and by oxidation of (1R,3R)‐ 2 to (3R)‐muscone.  相似文献

An improved HPLC procedure for the separation of phospholipids is described. The method described utilizes a solvent mixture of acetonitrile-methanol–water-trifluoroacetic acid (100:25:1.7:2.5, v/v) as the mobile phase, which is more compatible with the pump than mobile phases containing inorganic acids. Separation was by isocratic elution on a Hypersil silica column coupled to an evaporative light scattering detector. Complete separation of phosphatidylserine (PS), phosphatidylethanolamine (PE), phosphatidylcholine (PC) and sphingomyelin (SM) was achieved in less than 20 min. The detection limits for PS, PE, PC and SM were 50, 50, 80 and 150 ng (S/N = 3), respectively. Human, bovine and porcine erythrocyte ghost membranes and animal tissues have been successfully analyzed for their phospholipid contents.  相似文献

We described the simple, selective, and rapid method for determination of fucoidans using methylene blue staining of sulfated polysaccharides, immobilized into filter paper and consequent optic density (at A 663?nm) measurement of the eluted dye from filter paper. This solid-phase method allows selective determination of 1?C20???g fucoidan in presence of potentially interfering compounds (alginic acid, DNA, salts, proteins, and detergents). Further, we demonstrated the alternative way of using image processing software for fucoidan quantification without extraction of methylene blue dye from stained spots of fucoidan?Cdye complex.  相似文献

The title compounds 1(a,b) and 1(c,d) were synthesized by (i) non-chelation controlled sodium borohydride reduction of their corresponding α-N,N-dibenzylaminoketones and (ii) the Aldol-type condensation of optically pure N,N-dibenzylaminoaldehydes with the lithium derivative of isobutyronitrile, respectively. Attempted inversion of configurations of these secondary alcohols using the Moriarty method yielded 4-chloro-3-N,N-dibenzylaminonitrile via an aziridinium ion intermediate instead of the expected inverted alcohols.  相似文献

A novel, fast and sensitive enantioselective HPLC assay with a new core–shell isopropyl carbamate cyclofructan 6 (superficially porous particle, SPP) chiral column (LarihcShell-P, LSP) was developed and validated for the enantiomeric separation and quantification of verapamil (VER) in rat plasma. The polar organic mobile phase composed of acetonitrile/methanol/trifluoroacetic acid/triethylamine (98:2:0.05: 0.025, v/v/v/v) and a flow rate of 0.5 mL/min was applied. Fluorescence detection set at excitation/emission wavelengths 280/313 nm was used and the whole analysis process was within 3.5 min, which is 10-fold lower than the previous reported HPLC methods in the literature. Propranolol was selected as the internal standard. The S-(−)- and R-(+)-VER enantiomers with the IS were extracted from rat plasma by utilizing Waters Oasis HLB C18 solid phase extraction cartridges without interference from endogenous compounds. The developed assay was validated following the US-FDA guidelines over the concentration range of 1–450 ng/mL (r2 ≥ 0.997) for each enantiomer (plasma) and the lower limit of quantification was 1 ng/mL for both isomers. The intra- and inter-day precisions were not more than 11.6% and the recoveries of S-(−)- and R-(+)-VER at all quality control levels ranged from 92.3% to 98.2%. The developed approach was successfully applied to the stereoselective pharmacokinetic study of VER enantiomers after oral administration of 10 mg/kg racemic VER to Wistar rats. It was found that S-(−)-VER established higher Cmax and area under the concentration-time curve (AUC) values than the R-(+)-enantiomer. The newly developed approach is the first chiral HPLC for the enantiomeric separation and quantification of verapamil utilizing a core–shell isopropyl carbamate cyclofructan 6 chiral column in rat plasma within 3.5 min after solid phase extraction (SPE).  相似文献

The current research endeavours quality-by-design (QbD)-aided chromatographic techniques for the quantification of gliclazide (GLZ) in bulk and pharmaceutical dosage forms. Analytical QbD was initiated by assigning both an analytical target profile (ATP) and critical analytical attributes (CAAs). Furthermore, risk evaluation studies, along with factor screening studies, helped identify critical method parameters (CMPs). Optimisation was carried out using a 32 full factorial design by utilising the identified CMP, that is, flow rate (X1) and pH of buffer (X2) at three different levels along with evaluation of the selected CAA, that is, the retention time (Y1) and the peak area (Y2). In addition, the influence of sole and interactive CMPs on CAAs was checked using the data obtained statistically and with response surface plots. The confirmation of significance (P?<?0.05) of the method parameters was determined using analysis of variance (ANOVA). Chromatographic separation was achieved using a stainless-steel C8 column (25 cm?×?4 mm) in isocratic elution mode using phosphate buffer (pH 3.4) and HPLC-grade acetonitrile (50:50 v/v) as the eluent. The flow rate was adjusted to 1 mL min?1 and the eluent was detected at 230 nm. The validated method, alongside subsequent stress degradation studies conducted according to the ICH guidelines, further favours it as a highly efficient method for the analysis of regular drugs as well as their degraded products. The method proposed above provided a successful demonstration of the QbD-based approach in developing an extremely sensitive and dependable technique for estimating the GLZ for routine analysis and pre-clinical applications.
