To obtain a highly selective material for the antibiotic chloramphenicol, which has several harmful side effects in humans, different molecularly imprinted polymers (MIPs) were prepared. In order to avoid a major traditional drawback associated with MIPs of residual template bleeding, molecules that are structurally related to chloramphenicol were used as templates for polymer synthesis. Chromatographic evaluation indicated that the employed template imparted a significant influence on the recognition properties of the corresponding polymer. A strong retention of chloramphenicol under nonpolar elution conditions (k = 68.03, IF = 17.72) and under aqueous elution conditions (k = 92.44, IF = 1.35) was achieved. After chromatographic evaluation, the MIP was utilized as the recognition sorbent in a solid-phase extraction to determine chloramphenicol using either an organic or aqueous washing solvent. Recoveries of nearly 100% from the chloramphenicol standard solution and nearly 90% from honey samples spiked with chloramphenicol were attained. Furthermore, the applicability of the MIP for sample cleanup was demonstrated.  相似文献

A molecularly imprinted polymer designed for the selective extraction of donepezil from serum samples was synthesized using a noncovalent molecular imprinting approach. The molecularly imprinted polymer was evaluated chromatographically and then its affinity for donepezil was confirmed by solid‐phase extraction. The optimal conditions for solid‐phase extraction were provided by cartridge conditioning using acidified water purified from a Milli‐Q system, sample loading under basic aqueous conditions, clean‐up using acetonitrile, and elution with methanol/tetrahydrofuran. Desirable molecular recognition properties of the molecularly imprinted polymer led to good donepezil recoveries (90–102%). The data indicated that the imprinted polymer has a perfect selectivity and affinity for donepezil and could be used for selective extraction and analysis of donepezil in human serum.  相似文献

The molecularly imprinted polymer (MIP) was synthesized and used as dispersant of matrix solid‐phase dispersion (MSPD) for the extraction of chloramphenicol (CAP) in soil samples. The satisfactory recovery of CAP was obtained by the optimized extraction conditions: 1:2 as the ratio of sample to MIPs; 5 min as the dispersion time; 30% aqueous methanol as washing solvent and methanol as elution solvent. The CAP extracted from soil was determined by LC‐MS/MS. The slight ion suppression phenomenon was observed for the CAP when the sample was cleaned up by MSPD with MIP as dispersant, when compared with C18 as MSPD dispersant, which caused significant ion suppression. LOD of CAP is 4.1 ng/g. RSDs of intra‐ and inter‐day tests ranging from 3.1 to 6.2% and from 3.9 to 8.3% are obtained. At all three fortified levels (20, 100 and 500 ng/g), recoveries of CAP are in the range of 86.9–92.6%. The effect of ageing time of spiked soil sample on the CAP recovery was examined. The CAP recovery decreased from 91.0 to 36.9% when the ageing time changed from 1 day to 4 wk.  相似文献

A new molecularly imprinted polymer for extraction of crocin from saffron stigmas was prepared using gentiobiose (a glycoside moiety in crocin structure) as a template. Crocin binding to gentiobiose imprinted polymer (Gent‐MIP) was studied in comparison with a blank nonimprinted polymer in aqueous media. Affinity of the Gent‐MIP for the crocin was more than the nonimprinted polymer at all concentrations. In Scatchard analysis, the number of binding sites in each gram of polymer (maximum binding sites) and dissociation constant of crocin to binding sites were 18.4 μmol/g polymer and 11.2 μM, respectively. The Gent‐MIP was then used as the sorbent in an SPE method for isolation and purification of crocin from methanolic extract of saffron stigmas. The recovery of crocin, safranal and picrocrocin was determined in washing and elution steps. The Gent‐MIP had significantly higher affinity for crocin than other compounds and enabled selective extraction of crocin with a high recovery (84%) from a complex mixture. The results demonstrated the possibility of using a part of a big molecule in preparing a molecularly imprinted polymer with a good selectivity for the main structure.  相似文献

The performance of a molecularly imprinted polymer (MIP) as a selective solid-phase extraction sorbent for the clean-up and preconcentration of the antibiotic chloramphenicol is described. The MIP was prepared using chloramphenicol as the template, (diethylamino)ethyl methacrylate as the functional monomer, and ethylene glycol dimethacrylate as the cross-linking monomer, and using tetrahydrofuran as the solvent. Detection of chloramphenicol was carried out by square-wave voltammetry at electrochemically activated carbon fiber microelectrodes. Chloramphenicol was eluted from the MIP microcolumn with methanol. Different experimental variables (sample pH, eluent volume, analyte and eluent flow rates and sample volume) associated with the rebinding/elution process were optimized. For a 250 mL sample, a nominal enrichment factor of 500 was attained, and for a chloramphenicol concentration of 3.0x10(-8) mol L(-1) (9.7 microg L(-1)) a recovery of 96+/-4% was obtained. A range of linearity for chloramphenicol between 3.0x10(-8) and 1.0x10(-5) mol L(-1) was obtained by loading 17 mL of analyte solutions of different concentration, eluting with 0.5 mL methanol, evaporating under a stream of nitrogen and dissolving the residue in phosphate buffer of pH 7.8. The MIP selectivity was evaluated by checking several substances with similar molecular structures to that of chloramphenicol. The applicability of the MIP for both clean up and preconcentration was demonstrated by determining chloramphenicol in ophthalmic solutions and spiked milk at different concentration levels.  相似文献

This work reports the preparation of molecularly imprinted polymer (MIP) particles for selective extraction and determination of aluminum ion from aqueous media. Polymerization was achieved in a glass tube containing AlCl3, morin, 4-vinylpyridine, ethyleneglycoldimethacrylate, 2,2′ - azobisisobutyronitrile. The polymer block obtained was ground and sieved (55–75 μ m) and the Al (III)-morin complex was removed from polymer particles by leaching with 2 M of HCl, which leaves a cavity in the polymer particles. The polymer particles, both prior to and after leaching, have been characterized by IR and differential scanning calorimetry (DSC) studies. The effect of different parameters, such as pH, adsorption and desorption time, type and least amount of eluent for elution of complex from polymer were evaluated. Extraction efficiencies more than 99% were obtained by elution of the polymers with 15 mL of methanol-acetonitrile mixture (1:2) followed by ICP-OES. The limit of detection of the proposed method was 3.5 μ g L?1. A dynamic linear range (DLR) in the range of 20 to 200 μ g L?1 was obtained. The relative standard deviation (RSD) was less than 6.5%. The influence of various cationic and anionic interferences on percent recovery of complex was studied. The method was applied to the recovery and determination of aluminum in different real samples.  相似文献

Stir‐bar sorptive extraction is based on the partitioning of target analytes between the sample (mostly aqueous‐based liquid samples) and a stationary phase‐coated magnetic stir bar. Until now, only PDMS‐coated stir bars are commercially available, restricting the range of applications to the non‐selective extraction of hydrophobic compounds due to the apolar character of PDMS. In this work, a novel stir bar coated with molecularly imprinted polymer as selective extraction phase for sorptive extraction of thiabendazole (TBZ) was developed. Two different procedures, based on physical or chemical coating, were assessed for the preparation of molecularly imprinted stir bars. Under optimum conditions, recoveries achieved both in imprinted and non‐imprinted polymer stir bars obtained by physical coating were very low, whereas TBZ was favourably retained by imprinted over non‐imprinted polymer stir bars obtained by chemical coating and thus the latter approach was used in further studies. Different parameters affecting both stir‐bars preparation (i.e. cross‐linker, porogen, polymerization time) and the subsequent selective extraction of TBZ (i.e. washing, loading and elution solvents, extraction time) were properly optimized. The molecularly imprinted coated stir bars were applied to the extraction of TBZ from citrus samples (orange, lemon and citrus juices) allowing its final determination at concentrations levels according to current regulations.  相似文献

A molecularly imprinted polymer (MIP) was synthesized and evaluated to selectively extract ephedrine from human plasma. The MIP synthesis was performed in chloroform with methacrylic acid as a functional monomer and the target alkaloid as a template molecule. The resulting MIP was applied to the selective extraction of ephedrine from a pure aqueous medium. A recovery about 74% was obtained using the MIP with only 7% on the nonimprinted polymer (NIP). A very straightforward selective SPE procedure was then successfully applied to the direct extraction of ephedrine from spiked human plasma with a high extraction recovery (68%) on the MIP with no recovery on the NIP. Moreover, the MIP was used for the selective extraction of catecholamine neurotransmitters, i.e. adrenaline and noradrenaline.  相似文献

A novel and simple method for the selective cleanup and preconcentration of fluoroquinolone antibiotics in environmental water samples has been developed using molecularly imprinted polymer solid-phase extraction (MISPE). The molecularly imprinted polymer (MIP) has been prepared using enrofloxacin (ENR) as the template and a stoichiometric quantity of urea-based functional monomer to target the single oxyanionic moieties in the template molecule. The selectivity of the material for enrofloxacin, and structurally related and non-related compounds, has been evaluated using it as stationary phase in liquid chromatography. The novel polymer and the corresponding non-imprinted material (NIP) have been characterised using nitrogen adsorption-desorption isotherms and scanning electron microscopy. Various parameters affecting the extraction efficiency of the materials in the MISPE procedure were evaluated in order to achieve optimal preconcentration and to reduce non-specific interactions. The optimized MISPE/HPLC with fluorescence detection (FLD) method allows direct extraction of the antibiotics from the aqueous samples followed by a selective washing with acetonitrile/water (0.1M 2-[4-(2-hydroxyethyl)-1-piperazinyl]ethanesulfonic acid (HEPES) buffer, pH 7.5) (10/90, v/v) and elution with 2% trifluoracetic acid in methanol. Good recoveries and precision, ranging between 66 and 100% (RSD: 2-12%, n=3) for danofloxacin, enrofloxacin, oxolinic acid and flumequine, and moderate recoveries (15-40%, RSD 4-9%, n=3) for norfloxacin, ciprofloxacin, lomefloxacin and sarafloxacin, have been obtained for river water samples fortified with 0.50, 0.75 and 1.0microgL(-1) of all the antibiotics. The method detection limits ranged between 0.01 and 0.30microgL(-1) for all the antibiotics tested, when 100mL water samples were processed. The results demonstrate the applicability of the optimized method for the selective extraction of fluoroquinolones in environmental water samples at the ngL(-1) level.  相似文献

以红霉素为模板分子,采用溶胶-凝胶印迹技术,合成对红霉素具有特异选择识别性能的印迹聚合材料.采用红外光谱、扫描电子显微镜、热重分析以及比表面积测定技术对印迹聚合物的结构和表面性能进行详细探讨,结果表明所得印迹聚合物比表面积为266.2m2/g,粒径为600nm左右的颗粒材料.选择3种抗生素类药物进行选择吸附研究,结果表明,该聚合物对红霉素的分离因子最高达到2.20.优化固相萃取条件,结果表明用甲醇:乙酸(75:25,V/V)作为洗脱剂时对红霉素的洗脱效果最好.结合液相色谱技术,该印迹材料成功的分离和富集红霉素肠溶片中的红霉素,回收率达到104.5%。  相似文献

A simple method based on matrix solid-phase dispersion for selective extraction of anthraquinones from rhubarb samples was developed using a molecularly imprinted polymer as sorbent. The molecularly imprinted polymer was prepared using emodin as the template molecule, methacrylic acid as the functional monomer, and ethylene glycol dimethacrylate as the cross-linking agent. The polymer was characterized by scanning electron microscopy and Fourier-transform infrared spectrometry. Isothermal adsorption and dynamic adsorption experiments were performed. The best extraction conditions for anthraquinones were obtained at a ratio of molecularly imprinted polymer to sample of 1:1, a dispersion time of 5 minutes, with 5% aqueous methanol as the washing solvent, and an elution solvent of methanol-acetic acid (99:1, v/v). Once the matrix solid-phase dispersion process was optimized, the extract was reacted with 8% hydrochloric acid for hydrolysis. The anthraquinones extracted from rhubarb were determined by liquid chromatography. The detection limits of chrysophanol, emodin, physcion, and aloe-emodin were 0.23, 0.24, 0.28, and 0.27 µg mL?1, respectively. The proposed method was compared with the method in Chinese pharmacopoeia, and the results show that the extraction yield of anthraquinones obtained by molecularly imprinted polymer–matrix solid-phase dispersion method was higher. Moreover, the proposed method is faster and simpler and can achieve extraction and purification in the same system.  相似文献

An experimental investigation is presented for separation of pentachlorophenol (PCP) and methyl orange (MO) from an aqueous solution by use of cetyltrimethylammonium bromide (CTAB) as a regulator in the paper capillary permeation adsorption process (PCPA). The effects of pH, concentration of CTAB, and ionic strength on the separatability of the molecules were studied. The maximum separatability of nearly 100% PCP and MO was obtained in the optimum pH range 5–11 and 7–11, respectively. The ionic strength has an appreciable effect on separatability. The recovery of PCP and MO from paper by elution with acetone: isooctane solvent gradients was also studied. The selective separation of PCP and MO admixed in aqueous solution by a developed solvent elution technique was also reported. It was believed that PCP and MO were separated as their ion pairs with CTA cations by adsorption on the fiber surface.  相似文献

A novel molecularly imprinted polymer based on graphene oxide was prepared as a solid‐phase extraction adsorbent for the selective adsorption and extraction of cyromazine from seawater samples. The obtained graphene oxide molecularly imprinted polymer and non‐imprinted polymer were nanoparticles and characterized by scanning electron microscopy. The imprinted polymer showed higher adsorption capacity and better selectivity than non‐imprinted polymer, and the maximum adsorption capacity was 14.5 mg/g. The optimal washing and elution solvents for molecularly imprinted solid phase extraction procedure were 2 mL of acetonitrile/water (80:20, v/v) and methanol/acetic acid (70:30, v/v), respectively. The recoveries of cyromazine in the spiked seawater samples were in the range of 90.3–104.1%, and the relative standard deviation was <5% (n = 3) under the optimal procedure and detection conditions. The limit of detection of the proposed method was 0.7 μg/L, and the limit of quantitation was 2.3 μg/L. Moreover, the imprinted polymer could keep high adsorption capacity for cyromazine after being reused six times at least. Finally, the synthesized graphene oxide molecularly imprinted polymer was successfully used as a satisfied sorbent for high selectivity separation and detection of cyromazine from seawater coupled with high‐performance liquid chromatography.  相似文献

A bisphenol A (BPA) molecularly imprinted polymer, the composition of which was optimised using a chemometric approach, has been applied to the selective preconcentration of the template from aqueous samples. The selectivity of the polymer toward BPA and related compounds was evaluated chromatographically. The BPA-imprinted polymer was packed in a column and used for continuous on-column solid-phase extraction (MISPE) of aqueous samples followed by subsequent analysis by HPLC with fluorescence detection of the eluted fractions. The composition of the washing solvent applied in the MISPE procedure was optimised to favour the specific interactions of the MIP with BPA and to remove the non-selectively bound matrix components. The MISPE method has proven to be effective for selective preconcentration of BPA in aqueous samples (recoveries >84% obtained in the eluate for 10–100 mL sample volumes) enabling detection and quantification limits of 1.0 and 3.3 ng mL–1, respectively (based on 25 mL sample size). Analytical recoveries were between 92 and 101% for river water samples spiked with known amounts of BPA (30, 60, and 80 ng mL–1); relative standard deviations (RSD) were lower than 5.0%.  相似文献

Cotinine, the main metabolite of nicotine in human body, is widely used as a biomarker for assessment of direct or passive exposure to tobacco smoke. A method for molecularly imprinted solid-phase extraction (MISPE) of cotinine from human urine has been investigated. The molecularly imprinted polymer (MIP) with good selectivity and affinity for cotinine was synthesized using cotinine as the template molecule, methacrylic acid as the functional monomer, and ethylene glycol dimethacrylate as the cross-linker. The imprinted polymer was evaluated for use as a SPE sorbent, in tests with aqueous standards, by comparing recovery data obtained using the imprinted form of the polymer and a non-imprinted form (NIP). Extraction from the aqueous solutions resulted in more than 80% recovery. A range of linearity for cotinine between 0.05 and 5 μg mL−1 was obtained by loading 1 mL blank urine samples spiked with cotinine at different concentrations in acetate buffer of pH 9.0, and by using double basic washing and acidic elution. The intra-day coefficient of variation (CV) was below 7% and inter-day CV was below 10%. This investigation has provided a reliable MISPE–HPLC method for determination of cotinine in human urine from both active smokers and passive smokers. Figure  相似文献

A molecularly imprinted polymer (MIP) was prepared using (?)‐norephedrine as the template, methacrylic acid as the functional monomer, ethylene glycol dimethacrylate as the cross‐linker and chloroform as the porogen. The MIP was used as a selective sorbent in the molecularly imprinted solid‐phase extraction (MIP‐SPE) of the psychoactive phenylpropylamino alkaloids, norephedrine and its analogs, cathinone and cathine, from Khat (Catha edulis Vahl. Endl.) leaf extracts prior to HPLC‐DAD analysis. The MIP was able to selectively extract the alkaloids from the aqueous extracts of Khat. Loading, washing and elution of the alkaloids bound to the MIP were evaluated under different conditions. The clean baseline of the Khat extract obtained after MIP‐SPE confirmed that a selective and efficient sample clean‐up was achieved. Good recoveries (90.0–107%) and precision (RSDs 2.3–3.2%) were obtained in the validation of the MIP‐SPE‐HPLC procedure. The content of the three alkaloids in Khat samples determined after treatment with MIP‐SPE and a commercial Isolute C18 (EC) SPE cartridge were in good agreement. These findings indicate that MIP‐SPE is a reliable method that can be used for sample pre‐treatment for the determination of Khat alkaloids in plant extracts or similar matrices and could be applicable in pharmaceutical, forensic and biomedical laboratories. A molecularly imprinted polymer (MIP) designed to enable the selective extraction of carbamazepine (CBZ) from effluent wastewater and urine samples has been synthesised using a non-covalent molecular imprinting approach. The MIP was evaluated chromatographically in the first instance and its affinity for CBZ also confirmed by solid-phase extraction (SPE). The optimal conditions for SPE consisted of conditioning of the cartridge using acidified water purified from a Milli-Q system, loading of the sample under basic aqueous conditions, clean-up using acetonitrile and elution with methanol. The attractive molecular recognition properties of the MIP gave rise to good CBZ recoveries (80%) when 100 mL of effluent water spiked with 1 μg L−1 was percolated through the polymer. For urine samples, 2 mL samples spiked with 2.5 μg L−1 CBZ were extracted with a recovery of 65%. For urine, the linear range was 0.05-24 mg L−1, the limit of detection was 25 μg L−1 and precision, expressed as relative standard deviation at 0.5 mg L−1 (n = 3), was 3.1% and 12.6% for repeatability and reproducibility between days, respectively.  相似文献

Molecular imprinting as a promising and facile separation technique has received much attention because of their high selectivity for target molecules. In this study, the superparamagnetic lysozyme surface-imprinted polymer was prepared by a novel fabricating protocol, the grafting of the imprinted polymer on magnetic particles in aqueous media was done by atom transfer radical polymerization (ATRP), and the properties of the imprinted polymer were characterized in detail. Its high selective adsorption and recognition to lysozyme demonstrated the separation ability of the magnetic imprinted material to template molecule, and it has been used for quick and direct separation of lysozyme from the mixture of standard proteins and real egg white samples under an external magnetic field. Furthermore, the elution of lysozyme from the imprinted material was achieved by PEG/sulphate aqueous two-phase system, which caused lysozyme not only desorption from the imprinted materials but also redistribution in the top and bottom phase of aqueous two-phase system. The aqueous two-phase system exhibited some of the extraction and enrichment effect to desorbed lysozyme. Our results showed that ATRP is a promising method for the protein molecularly imprinted polymer preparation.  相似文献

Solid-phase extraction (SPE) with a molecularly imprinted polymer (MIP) as sorbent has been investigated for the clean-up of the broad-spectrum bacteriostatic antibiotic chloramphenicol (CAP) in honey samples. The MIP was prepared by using methacrylic acid (MAA) as functional monomer, ethylene glycol dimethacrylate (EDMA) as cross-linker, chloroform as porogen and CAP as template molecule. The binding behaviour of the template CAP on the MIP was evaluated by high-performance liquid chromatography, and then the MIP was applied as a sorbent in SPE to selectively extract CAP from honey. It was shown that recoveries of nearly 100% of a CAP standard solution and up to 94% from spiked honey samples could be obtained after SPE.  相似文献

The design, synthesis, and evaluation of a molecularly imprinted polymer transaminase mimic is described. Methacrylic acid-ethylene glycol dimethacrylate copolymers were synthesized using, as a template, a transition state analogue (TSA) for the reaction of phenylpyruvic acid and pyridoxamine to yield phenylalanine and pyridoxal. Polymer suitability was established on the basis of (1)H NMR studies of template-functional monomer interactions. Polymer recognition characteristics were examined in a series of HPLC studies using the polymers as chromatographic stationary phases. Selectivity for the TSA, relative to substrates and products, was observed in both aqueous and nonpolar media. In the latter case (chloroform/AcOH, 96:4), an enantioseparation factor (alpha) of 2.1 was obtained, and frontal chromatographic studies revealed the presence of 11.9 +/- 0.2 micromol g(-1) (dry weight) of enantioselective sites. Polymers imprinted with the l-form of the oxazine-based TSA induced a 15-fold enhancement of the apparent reaction rate (app. V(max) 2.5 x 10(-7) mol s(-1); app. K(m) 8.2 x 10(-3) M) and enantioselective production of phenylalanine (32 +/- 4% ee) for reactions conducted in an aqueous buffer system. Substrate selectivity was evident, and a turnover number (k(cat)) of 0.1 s(-)(1) was determined. This is the first example of the catalysis of sigmatropic shifts in aqueous media by molecularly imprinted polymers.  相似文献
