Summary A cerebroside fraction was obtained from the fruit bodies offomitopsis pinicola using column chromatography and then separated into six compounds by reversed-phase HPLC. The sugar component of all cerebrosides wasD-glucose. The major fatty acids were 2-hydroxyfatty acids (C14–C18), the long chain base was identified as 9-methyl-C18-4,8-sphingadienine which is widely distributed in fungi and reported to be essential for the fruit-inducing activity of fungi. Based on degradation studies, fast atom bombardment mass spectrometry, and different1H and13C NMR investigations, the structure of the main cerebroside (1) was determined to be (4E,8E,2S,3R,2R)-N-2-hydroxypalmityl-1-O--D-glucopyranosyl-9-methyl-4,8-sphingadienine.

Zusammenfassung Aus den Fruchtkörpern vonfomitopsis pinicola wurde ein Cerebrosidgemisch erhalten und durch Säulenchromatographie und HPLC in sechs Verbindungen aufgetrennt. Der Zuckerbaustein aller Cerebroside warD-Glucose. Die Fettsäurekomponenten waren 2-Hydroxyfettsäuren mit einer Kettenlänge zwische C14 und C18. Der Basenteil konnte als 9-Methyl-C18-4,8-sphingadienin identifiziert werden. Diese Verbindung ist in Pilzen weit verbreitet und für die Fruchtbildung verantwortlich. Aus Abbaustudien, FAB-MS und verschiedenen1H- und13C-NMR-Messungen wurde die Struktur des Hauptcerebrosids (1) als (4E,8E,2S,3R,2R)-N-2-hydroxypalmityl-1-O--D-glucopyranosyl-9-methyl-4,8-sphingadienin ermittelt.

Two new compounds were isolated from two Far-Eastern starfish species, Henricia sanguinolenta and H. leviuscula leviuscula, collected in the Sea of Okhotsk, viz., the glycoside sanguinoside C, (20R,22E,24R,25S)-3-O-(2,3,4-tri-O-methyl--xylopyranosyl)-24-methyl-5-cholest-22-ene-3,4,6,8,15,26-hexol, and a steroidal ketone, (20R,24S)-3,6,24-trihydroxy-5-cholestan-15-one. They exhibit moderate cytostatic activity with respect to the eggs of sea urchin Strongylocentrotus intermedius.  相似文献

Chemical investigation of the Red Sea soft coral Sarcophyton auritum led to the isolation and structure elucidation of two new diterpene cembranoids; 2-epi-sarcophine (2) and (1R,2E,4S,6E,8R,11R,12R)-2,6-cembradiene-4,8,11,12-tetrol (4), as well as two known diterpene cembranoids, reported for the first time from this species, namely sarcophine (1) and (+)-7α,8β-dihydroxydeepoxysarcophine (3). Structure elucidation was achieved using spectroscopic techniques, including 1D and 2D NMR and HRMS. The isolated cembranoids were found to display high cytotoxicity against HepG2 (liver cancer cell line) and MCF-7 (breast cancer cell line).  相似文献

A new polyhydroxylated macrolide, named mangiferaelactone (1) was isolated from a solid culture of the endophytic fungus Pestalotiopsis manguiferae, together with ten known compounds [(6S,1′S)-LL-P880α; (6S,1′S,2′R)-LL-P880β; (1′S,2′R)-LL-880γ; (1′R)-dehydropestalotin; (−)-5-carboxylmellein; (−)-5-methylmellein; (−)-5-hydroxylmethylmellein; arabenoic acid; 5,6-dihydro-4-methoxy-2H-pyran-2-one; and the (−)-2-hexylidene-3-methylsuccinic acid]. P. manguiferae was isolated from Hyptis dilatata, a small shrub common in the central region of Panama. The structure of compound 1 was elucidated by a combination of spectroscopic methods (IR, MS, optical rotation, 1D and 2D NMR spectroscopy). The absolute configuration of 1 was established as 4R,7R,8R,9S by application of vibrational circular dichroism (VCD). Compound 1 showed a minimum inhibitory concentration (MIC) of 1.6863 mg/mL against Listeria monocytogenes, and 0.5529 mg/mL against Bacillus cereus. No activity was observed for compound 1 against Plasmodium falciparum or Trypanosoma cruzi; likewise, no cytotoxic activity was observed against A2058 and H522-T1 cells.  相似文献

New cerebroside with the structure of 1-(-d-glucopyranosyloxy)-2-N-(2-hydroxyacyl)-erythro-2-amino-9-methyloctadeca-4E,8E-dien-3-ol (1) was isolated from the soft coral Cladiellasp. collected on the seaboard of the Andaman Islands (Indian Ocean). The acyl groups in compound 1 were identified as residues of d-2-hydroxy-C20:0, -C21:0, and -C22:0 acids.  相似文献

By reaction of 2-[(1RS, 2RS)-2-hydroxy-1-methyl-propyl]-2-phenyl-1,3-dithiane (1 a) withcis-2-butene oxide, subsequent reduction and acetalizationc-4,t-5-dimethyl-r-2,c-6-diphenyl-1,3-dioxane (3 a) andt-4,c-5-dimethyl-r-2,c-6-diphenyl-1,3-dioxane (3 b) were synthesized as model compounds. For the same purpose by aldol reaction of cyclohexanone and reduction (1RS, 2SR)-2[(RS)-hydroxy-(4-methoxyphenyl)methyl]cyclohexanol (7 a), (1RS, 2RS)-2[(SR)-hydroxy-(4-methoxyphenyl)methyl]cyclohexanol (8 a), and (1RS, 2RS)-2[(RS)-hydroxy-(4-nitrophenyl)methyl]cyclohexanol (8 b) and by acetalization (2 , 4 , 4 a, 8 a)-2,4-bis(4-methoxyphenyl)hexahydro-4H-1,3-benzodioxin (9 a) and (2 , 4 , 4 a, 8 a)-2,4-bis(4-nitrophenyl)hexahydro-4H-1,3-benzodioxin (10 b) were obtained. FromPrins reactions, starting with 2-butene3 a,c-4,c-5-dimethyl-r-2,c-6-diphenyl-1,3-dioxane (3 c),r-4,t-5-dimethyl-c-6-phenyl-1,3,2-dioxathiane-2,2-dioxide (4), and (2Z, 4E)-1,5-diphenyl-4-methyl-2,4-pentadien-1-on (5), and starting with cyclohexene (E)-3-(4-methoxybenzylidene)cyclohexenyl-4-methoxyphenyl ketone (11) have been isolated in low yields.

The reaction of (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediol with glutaraldehyde has been studied. It has been established on the basis of AM1 and PM3 calculations and 1H NMR spectra recorded in the presence of the shift reagent Eu(fod)3 that (1S,3S,4S,7R,11R)-3-(4-nitrophenyl)-11-aza-2,6-dioxatricyclo[5,3,1,04,11]undecane is formed as the result of the reaction.  相似文献

Regioselectivity and diastereoselectivity of the addition of BrCCl3 to (R)-3-(E)-cinnamoyl-4-phenyloxazolidin-2-one (1) and (R)-3-(E)-acryloyl-4-phenyloxazolidin-2-one (2) catalyzed by Fe(CO)5 or initiated with benzoyl peroxide were investigated. Stereochemistry of the reaction of BrCCl3 with the a-complexes (4R,S,S)-2-(3-(E)-cinnamoyl-4-phenyl-oxazolidin-2-one)irontetracarbonyl (3a) and (4R,R)-2-(3-acryloyl-4-phenyloxazolidin-2-one)irontetracarbonyl (4b) was also studied. The results obtained allow the following conclusions to be drawn: (1) the thermal Kharasch reaction catalyzed by Fe(CO)5 proceeds by a redox catalysis mechanism; (2) iron (in any of its oxidation states) is not coordinated to olefins in the transition state of the reaction; (3) the transfer of the halogen atom on the radical adduct probably occurs inside a radical-iron cation pair.Yu. In the presence of lipase from the yeast Candida cylindracea, partial acetylation of (±)-2-[6-benzyloxy-2,5,7,8-tetramethylchroman-2-yl]ethanol with vinyl acetate gives S-(+)-acetate whose alkaline hydrolysis affords (S)-(–)-alcohol. Repeated enzymatic acetylation of the residual alcohol up to 39% conversion afforded the R-enantiomer. The enantiomeric alcohols were oxidized to (S)- or (R)-aldehydes having the same sign of []D as the original alcohols. These alcohols were converted into S-(+)- and R-(–)-enantiomers of the antioxidant MDL-73404, a hydrophilic analog of -tocopherol.  相似文献

A synthesis is reported for (4R,5R)-and (4S,5S)-4-hydroxymethyl-5-(4-nitrophenyl)oxazolidin-2-ones and (1′R,4R)-and (1′S,4S)-4-[hydroxy(4-nitrophenyl)methyl]oxazolidin-2-ones from (1R,2R)-and (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediols. The synthesis characterized by cationic olefin cyclizations accomplished using ketone enol esters and odor of novel (1R,6S)- and (1S,6R)-2,2,6-trimethylcyclohexyl methyl ketones (5) are described. The stereoselective syntheses of (E)-(1R,6S)- and (E)-(1S,6R)-1-(2,2,6-trimethylcyclohexyl)-2-buten-1-one (6) and (1R,6S)-ethyl 2,2,6-trimethylcyclohexylcarboxylate (7), useful raw materials for flavor and fragrance, starting from the (1R,6S)- and (1S,6R)-5 are also described.  相似文献

An optically active juvenile hormone analogue,S-(+)-methoprene (1), is synthesized in six steps from technical gradeS-(+)-3,7-dimethyl-1,6-octadiene ((+)-dihydromyrcene, e.e. –50%) by a novel procedure which begins with selective hydroalumination-oxidation to giveS-(-)-citronellol. This alcohol is oxidized to giveS-(-)-citronellal which on reaction with allylmagnesium chloride affords 6S, 10-dimethyl-1,9-undecadien-4 R/S-ol (5). Smidt-Moiseev oxygenation of 5 followed by dehydration leads to 6S, 10-dimethyl-3E,9-undecadien-2-one. The latter on treatment with isopropoxyethynylmagnesium bromide is transformed into isopropyl 3,7S,11-trimethyl-2E/Z,4,E,10-dodecatrienoate which upon Brown solvomercurationreduction in MeOH gives1 in 14% overall yield.Translated fromIzvestiya Akademii Nauk. The rotational barriers between the configurational isomers of two structurally related push–pull 4-oxothiazolidines, differing in the number of exocyclic CC bonds, have been determined by dynamic 1H NMR spectroscopy. The equilibrium mixture of (5-ethoxycarbonylmethyl-4-oxothiazolidin-2-ylidene)-1-phenylethanone (1a) in CDCl3 at room temperature to 333 K consists of the E- and Z-isomers which are separated by an energy barrier ΔG# 98.5 kJ/mol (at 298 K). The variable-temperature 1H NMR data for the isomerization of ethyl (5-ethoxycarbonylmethylidene-4-oxothiazolidin-2-ylidene)ethanoate (2b) in DMSO-d6, possessing the two exocyclic CC bonds at the C(2)- and C(5)-positions, indicate that the rotational barrier ΔG# separating the (2E,5Z)-2b and (2Z,5Z)-2b isomers is 100.2 kJ/mol (at 298 K). In a polar solvent-dependent equilibrium the major (2Z,5Z)-form (>90%) is stabilized by the intermolecular resonance-assisted hydrogen bonding and strong 1,5-type S · · · O interactions within the SCCCO entity. The 13C NMR ΔδC(2)C(2′) values, ranging from 58 to 69 ppm in 1ad and 49-58 ppm in 2ad, correlate with the degree of the push-pull character of the exocyclic C(2)C(2′) bond, which increases with the electron withdrawing ability of the substituents at the vinylic C(2′) position in the following order: COPh COEt > CONHPh > CONHCH2CH2Ph. The decrease of the ΔδC(2)C(2′) values in 2ad has been discussed for the first time in terms of an estimation of the electron donor capacity of the S fragment on the polarization of the CC bonds.  相似文献

The crystal structure of the paramagnetic bis(pyridine-2,6-dithiocarbomethylamide) nickel(II) nitrate (NiPDTA) is described: C18H22N6S4·(NO3)2·(H2O)1,5, monoclinic, C2/c,Z=4,a=14.705 (3) Å,b=23.254 (8) Å,c=8.383 (3) A, =98.18 (2)°,d x=1.55 gcm–3,d m=1.53 gcm–3. The structure was solved withPatterson and differenceFourier techniques and refined to a residual ofR=0.053. The nickel is surrounded by a square bipyramidal coordination of four thioamide sulfur atoms and two pyridine nitrogen atoms. Vibrational and electronic band positions for this compound are discussed.

C18H22N6S4Ni·(NO3)2·(H2O)1,5, monoklin, C2/c,Z=4,a=14,705 (3) Å,b=23,254 (8) Å,c=8,383 (3) A, =98,18 (2)°,d x=1,55gcm–3,d m=1,53gcm–3. Das Phasenproblem wurde mittelsPatterson-und Differenz-Fourier-Synthese bestimmt und die Struktur bis zu einem kristallographischenR-Faktor vonR=0.053 verfeinert. Das Nickel-Atom ist von vier Thioamid-Schwefelatomen und zwei Pyridin-Stickstoffatomen in quadratisch-bipyramidaler Anordnung umgeben. Schwingungsspektren und Anregungsspektren des Komplexes werden diskutiert.

Aervalanata possesses various useful medicinal and pharmaceutical activities. Phytochemical investigation of the plant has now led to the isolation of a new 2α,3α,15,16,19-pentahydroxy pimar-8(14)-ene diterpenoid (1) together with 12 other known compounds identified as β-sitosterol (2), β-sitosterol-3-O-β-D-glucoside (3), canthin-6-one (4), 10-hydroxycanthin-6-one (aervine, 5), 10-methoxycanthin-6-one (methylaervine, 6), β-carboline-1-propionic acid (7), 1-O-β-D-glucopyranosyl-(2S,3R,8E)-2-[(2′R)-2-hydroxylpalmitoylamino]-8-octadecene-1,3-diol (8), 1-O-(β-D-glucopyranosyl)-(2S,3S,4R,8Z)-2-[(2′R)-2′-hydroxytetracosanoylamino]-8(Z)-octadene-1,3,4-triol (9), (2S,3S,4R,10E)-2-[(2′R)-2′-hydroxytetracosanoylamino]-10-octadecene-1,3,4-triol (10), 6′-O-(4″-hydroxy-trans-cinnamoyl)-kaempferol-3-O-β-D-glucopyranoside (tribuloside, 11), 3-cinnamoyltribuloside (12) and sulfonoquinovosyldiacylglyceride (13). Among these, six compounds (813) are reported for the first time from this plant. Cytotoxicity evaluation of the compounds against five cancer cell lines (CHO, HepG2, HeLa, A-431 and MCF-7) shows promising IC50 values for compounds 4, 6 and 12.  相似文献

Four new cerebrosides, gynuramides I?IV ( 1 ‐ 4 ), together with 37 known compounds were isolated from the rhizome of Gynura japonica. The structures of cerebrosides 1 ‐ 4 were determined by chemical and spectroscopic examination to be: (2S,3S,4R,8E)‐2‐[(R)‐2‐hydroxypentacosanoylamino]‐8‐en‐1,3,4‐octadecanetriol, (2S,3S,4R,8E)‐2‐[(R)‐2‐hydroxytetracosanoylamino]‐8‐en‐1,3,4‐octadecanetriol, (2S,3S,4R,8E)‐2‐[(R)‐2‐hydroxytricosanoylamino]‐8‐en‐1,3,4‐octadecanetriol, and (2S,3S,4R,8E)‐2‐[(R)‐2‐hydroxydocosanoylamino]‐8‐en‐1,3,4‐octadecanetriol.  相似文献

Six polyhydroxylated steroids and their derivatives were isolated from the starfish Lysastrosoma anthosticta collected in the Posyet Bay, Sea of Japan. These include a new glycoside of the steroid polyol, lysastroside A (1), which was identified as (25S)-26-O--d-xylopyranosyl-5-cholestane-3,6,8,15,16,26-hexaol, and the previously known pycnopodioside C monoglycoside (2), marthasterone sulfate (3), (25S)-5-cholestane-3,6,8,15,16,26-hexaol (4), (25S)-5-cholestane-3,6,7,8,15,16,26-heptaol (5), and (25S)-5-cholestane-3,6,7,8,15,16,26-heptaol (6). The compounds were tested for the haemolytic activity and the action on the embryogenesis of the sea urchin Strongylocentrotus intermedius.  相似文献

Starting from the esters (2E,4S)- 6 and (2E,4R)- 6 , bromo aldehydes (S)- 9 and (R)- 9 as well as bromo alcohols (S)- 10 and (R)- 10 , respectively, were prepared. Bromo alcohol (R)- 8 was converted to the diol (2E,4R)- 16 . Ozonolysis of the latter led to aldehyde (R)- 17 , which was transformed, by a Wittig reaction, to (2R,4E,6R)- 18 , corresponding to the C(7)-to-C(14) segment of phomenoic acid ( 1 ). Attempts to improve the yields by applying a Julia coupling of (R)- 23 , which was prepared from (2E,4R)- 7 , with (R)- 24 were unsuccessful. Finally, the coupling of the iodo derivative (2E,4S)- 28 with the lithiated derivative of 1,3-dithiane 30 by the Corey-Seebach ‘Umpolung’ led to (3S,4E)- 32 which is a derivative of the C(7)-to-C(14) segment of 1 , suitable for further transformations.  相似文献

Two new 24-hydroxylated asterosaponins,sodium(20R,24S)-6α-O-(4-O-sodiumsulfato-β-D-quinovopyranosyl)-5α-cholest-9(11)-en-3β,24-diol 3-sulfate(1) and sodium(20R,24S)-6α-O-[3-O-methyl-β-D-quinovopyranosyl-(1→2)-β-D-xylopyranosyl-(1→3)-β-D-glucopyranosyl]-5α-cholest-9(11)-en-3β,24-diol 3-sulfate(2),were isolated from the starfish Culcita novaeguineae.Their structures were elucidated by extensive spectral analysis and chemical evidences.  相似文献
