Cigarette smoke can cause cellular oxidative stress that contributes to various adverse health effects associated with smoke exposure, partially due to reactive oxygen species (ROS) present in cigarette smoke. Reduction of abundant ROS in the cigarette mainstream smoke (MSS) is of importance for human health. In this work, a simple, rapid, and reliable fluorescence evaluation of scavenging efficiency of antioxidants as potential filter additives against ROS in cigarette smoke is reported. This method was based on the combination of model glass reactor and a fluorescence assay of ROS in cigarette smoke using dihydrorhodamine 6 G (DHR-6 G). The antioxidant was added into a glass reactor attached to cigarette filter, which simplified the preparation of combined filter containing additives. The ROS scavenging efficiency of antioxidants was then determined using spectrofluorimetry by the change in fluorescence intensity of whole smoke-bubbled solutions before and after addition of antioxidants into the glass reactor. The proposed method was successfully applied to the determination of ROS scavenging efficiency of several potential additives, such as tert-butylhydroquinone (TBHQ), vitamin C, β-carotene, grape seed extract, and Ginkgo biloba extract. Moreover, the relationship between MSS ROS scavenging efficiency and antioxidant activities (DPPH radicals scavenging efficiency and Fe2+ reducing power) of these compounds was also investigated.  相似文献

Cigarette smoke contains free radicals that are considered to be a major group of carcinogens. In the present study, we used plant antioxidants to treat cigarette filters for scavenging gas phase free radicals in cigarette smoke. The scavenging activities of four antioxidants against gas phase free radicals were analyzed using an improved spin trap protocol. The scavenging rates ranked as lycopene > grape seed extract > pycnogenol > tomato powder. A scavenging rate of 62% (p < 0.001) was obtained from the lycopene treatment. The use of plant antioxidants as potential scavengers for reducing free-radicals in cigarette smoke is discussed.  相似文献

A series of shikonin derivatives have been designed and their radical scavenging activity has been characterized by the B3LYP/6‐31+G(d) approach. The hydrogen bond properties of the studied structures were investigated using the atoms in molecules (AIM) theory. The calculated results reveal that the hydrogen bond is important for good scavenging activity. The introduction of electron‐drawing (electron‐donating) groups increases (decreases) the scavenging activities of radical and radical cations of shikonin derivatives. Shikonin derivatives appear to be good candidates for the single‐electron‐transfer mechanism, particularly for ? N(CH3)2 derivative. Taking this system as an example, we present an efficient method for the investigation of radical scavenging activity from theoretical point of view. With the current work, we hope to highlight the radical scavenging activity of hydroxynaphthoquinones derivatives and stimulate the interest for further studies and exploitation in pharmaceutical industry.  相似文献

Solar radiation and cigarette smoke are two environmental risk factors known to affect skin integrity. Although the toxic effects of these factors on skin have been widely studied separately, few studies have focused on their interaction. The objective of this study was to evaluate and understand the synergistic harmful effects of cigarette smoke and solar rays on human primary keratinocytes. The keratinocytes were exposed to cigarette smoke extract (CSE) and then irradiated with a solar simulator light (SSL). The viability, as determined by measuring metabolic activity of skin cells, and the levels of global reactive oxygen species (ROS) were evaluated after exposure to CSE and SSL. The combination of 3% CSE with 29 kJ m−2 UVA caused a decrease of 81% in cell viability, while with 10% to 20% CSE, the cell viability was null. This phototoxicity was accompanied by an increase in singlet oxygen but a decrease in type I ROS when CSE and SSL were combined in vitro. Surprisingly, an increase in the CSE's total antioxidant capacity was also observed. These results suggest a synergy between the two environmental factors in their effect on skin cells, and more precisely a phototoxicity causing a drastic decrease in cell viability.  相似文献

Caffeic acid (CA) and its analogues such as rosmarinic acid are well known as antioxidative agents. Exposure to UVA is known to generate reactive oxygen species (ROS) such as singlet oxygen (1O2) and superoxide anion radical (*O2-) in the skin of animals, which in turn induces skin photodamage and photoaging. Because CA and its analogues quench 1O2, these compounds were topically applied to the abdominal skin of live hairless mice and were found to suppress ROS generation upon UVA exposure. Furthermore, the generation of UVA-induced ROS was also suppressed in the skin of mice that were orally given CA. In order to understand the mechanism by which CA blocks ROS production in UVA-exposed skin, the pharmacokinetics of CA upon oral administration to mice was followed and CA was found to efficiently distribute in the skin. These results suggest that skin damage by UVA-induced ROS generation is reduced by oral supplementation of CA, which has a scavenging and quenching activity against ROS.  相似文献

研究了一种准确测定主流卷烟烟气中固相自由基总量的新方法. 通常人们采用剑桥滤片来捕集主流卷烟烟气中的固相自由基, 但它并不适合用于准确测定主流卷烟烟气中的固相自由基的总量, 研究发现有超过20%的主流卷烟烟气中的固相自由基可以透过剑桥滤片. 找到了一种电子顺磁共振波谱(EPR)信号很小的、适合用来捕集主流卷烟烟气中的固相自由基总量的过滤片. 采用了以在相同条件下测得的1,1-二苯基-2-苦基肼基(1,1-Diphenyl-2-picryl-hydrazyl, 简称DPPH)的EPR信号强度与强煤的EPR信号强度相比得到的相对EPR信号强度与自由基的自旋数建立校准曲线的方法来计算烟气固相自由基的量, 降低了使用不同的电子自旋共振波谱仪造成的对同一种检测样品测定结果的差异. 通过不同实验室内和实验室间的验证实验, 证明了此方法有很好的重复性和重现性.  相似文献

An improved gas chromatography-mass spectrometry (GC-MS) method was described for the analysis of carbonyl compounds in cigarette mainstream smoke (CMS) after 2,4-dinitrophenylhydrazine (DNPH) derivatization. Besides formaldehyde, acetaldehyde, acetone, acrolein, propionaldehyde, methyl ethyl ketone, butyraldehyde, and crotonaldehyde that are routinely analyzed in cigarette smoke, this technique separates and allows the analysis of several C4, C5 and C6 isomeric carbonyl compounds. Differentiation could be made between the linear and branched carbon chain components. In cigarette smoke, the branched chain carbonyls are found at higher level than the linear chain carbonyls. Also, several trace carbonyl compounds such as methoxyacetaldehyde were found for the first time in cigarette smoke. For the analysis, cigarette smoke was collected using DNPH-treated pads, which is a simpler procedure compared to conventional impinger collection. Thermal decomposition of DNPH-carbonyl compounds was minimized by the optimization of the GC conditions. The linear range of the method was significantly improved by using a standard mixture of DNPH-carbonyl compounds instead of individual compounds for calibration. The minimum detectable quantity for the carbonyls ranged from 1.4 to 5.6 microg/cigarette.  相似文献

A sampling system has been set up to monitor a group of volatile smoke analytes (nitric oxide, acetaldehyde, acetone, benzene, toluene, 1,3 butadiene, isoprene and carbon dioxide) from mainstream cigarette smoke on a puff-resolved basis. The system was able to record gas evolution profiles during puffing and interpuff periods without interruption (e.g. taking clearing puffs). Gas phase smoke analytes were sampled as close to the mouth end of the cigarette filter as possible in order to minimise any dead volume effect. The results revealed that, for some volatile species, a significant fraction (e.g. up to 30% for benzene) in the cigarette mainstream smoke had been generated during the preceding smoulder period. These species were trapped or absorbed within the cigarette rod and then subsequently eluted during the puff. The identification of the two sources of the mainstream smoke, a smouldering source and a puffing source, has not been reported before. The observation contributes to the fundamental knowledge of the cigarette smoke formation and may have implications on wider smoke chemistry and associated effects.  相似文献

Polyphenols and flavonoids possess a variety of biological activities including antioxidant and anti-tumor activities. Ixora parviflora is a member of the flavonoid-rich Rubiaceae family of flowering plants and used as folk medicine in India. The aim of this study was to investigate the antioxidant activity of Ixora parviflora extract (IPE) in a cell-free system and erythrocytes, and the ability of IPE to inhibit reactive oxygen species (ROS) generation in human fibroblasts (Hs68) after ultraviolet (UV) exposure. Various in vitro antioxidant assays were employed in this study. The extraction yield of IPE was 17.4 ± 3.9%, the total phenolic content of IPE was 26.2 μg gallic acid equivalent (GAE)/mg leaves dry weight and the total flavonoids content was 54.2 ± 4.4 μg quercetin equvalent (QE)/mg extract. The content of chlorogenic acid was 9.7 ± 1.2 mg/g extract. IPE at 1000 μg/mL exhibited a reducing capacity of 90.5 ± 0.6%, a 1,1-diphenyl-2-picrylhydrazy (DPPH) radical scavenging activity of 96.0 ± 0.4%, a ferrous chelating activity of 72.2 ± 3.5%, a hydroxyl radical scavenging activity of 96.8 ± 1.4%, and a hydrogen peroxide scavenging activity of 99.5 ± 3.3%. IPE at 500 μg/mL also possessed inhibitory activity against 2,2'-azobis (2-methylpropionamidine) dihydrochloride (AAPH)-induced hemolysis of erythrocytes (89.4 ± 1.8%) and resulted in a 52.9% reduction in ROS generation in UV-exposed fibroblasts. According to our findings, IPE is a potent antioxidant and a potential anti-photoaging agent.  相似文献

The electrochemical behavior of an indole library of compounds, including several tryptophan and tryptamine derivatives previously demonstrated to be active against several reactive oxygen species (ROS), was investigated. For this purpose, a voltammetry study was undertaken and the oxidation potential was correlated to the scavenging activity reported for the studied indoles. All the compounds showed an oxidation potential peak lower than that observed for indole (Epox = 1.035 V), but higher than that described for the antioxidant melatonin (Epox = 0.715 V). The electrochemical behavior showed a high correlation with the scavenging activity of peroxyl radical, for selected compounds.  相似文献

In this study, the concentrations of Cd in smoke condensate of Paraguayan and Brazilian full flavor normal size cigarettes were determined. A “fishtail” chimney was used to confine and direct cigarette smoke to an impaction trap followed by 3 MCE (mixed cellulose ester) filters for smoke collection. The smoke condensate collected at the first two collection stages (chimney and impactor) was prepared as Triton X-100 slurry samples and the 3 MCE filters (third collection stage) used to collect smoke were microwave digested. Samples were analyzed by Thermospray Flame Furnace Atomic Absorption Spectrometry (TS-FF-AAS) for Cd and the results obtained were compared with Graphite Furnace Atomic Absorption Spectrometry (GFAAS). No significant differences were found between the values obtained with TS-FF-AAS and GFAAS at a 95% confidence level. The total concentration of Cd in the smoke of Brazilian and Paraguayan cigarettes tested ranged from 187 to 281 ng/cigarette and from 176 to 271 ng/cigarette, respectively. The LODs for Cd using TS-FF-AAS and GFAAS, were 5.8 and 1.5 ng/cigarette, respectively, for the analysis of smoke condensate from 1 cigarette. The main advantages of using the simple TS-FF-AAS system are its low running costs and the analysis time when compared to GFAAS.  相似文献

以自旋标记荧光探针4-((9-acridinecarbonyl)amino)-2,2,6,6-tetramethylpiperidin-1-oxyl(TEMPO-9-AC)研究了卷烟主流烟气中的碳中心自由基。以吸烟机抽吸卷烟,以弱荧光的TEMPO-9-AC作为捕集剂捕集卷烟主流烟气中的碳中心自由基,生成稳定的强荧光的碳中心自由基捕集加合物,以高效液相色谱-三重四极杆串联质谱联用仪(HPLC-MS/MS)对其结构进行了确认,并建立了高效液相色谱-荧光检测(HPLC-FLD)的定量方法。结果表明,方法的检出限为0.318 nmol/cig,相对标准偏差(RSD)为3.5%~9.7%;利用TEMPO-9-AC捕集体系鉴别出了10种碳中心自由基;对5种代表性卷烟烟气中的碳中心自由基进行了定量计算,获得了它们在主流烟气中碳中心自由基的含量数据,并发现碳中心自由基总量与焦油释放量之间具有高度的相关性。本法检出限低,重复性好,适用于卷烟主流烟气中碳中心自由基的结构验证及释放量的检测分析。  相似文献

Soft single-photon ionisation (SPI)–time-of-flight mass spectrometry (TOFMS) has been used to investigate the effect of different cigarette-lighting devices on the chemical composition of the mainstream smoke from the first cigarette puff. Lighting devices examined were a Borgwaldt electric lighter, a propane/butane gas lighter, a match, a candle, and the burning zone of another cigarette. To eliminate the effects of the different masses of tobacco burnt by use of the different lighting methods a normalisation procedure was performed which enabled investigation of changes in the chemical patterns of the resulting smoke. When another cigarette was used as the lighting device, elevated levels of ammonia and other nitrogen-containing substances were observed. These are high in the sidestream smoke of the cigarette used for lighting and would be drawn into the mainstream smoke of the cigarette being lit. In contrast, smoke from the cigarette lit by the electric lighter contained slightly higher normalised amounts of isoprene. Lighting the cigarette by use of a candle resulted in larger amounts of substances, e.g. benzene, which most probably originated from thermal decomposition of wax. The composition of the first puff of smoke obtained by use of the three lighting methods with open flames (gas lighter, match, and candle) was usually similar whereas the composition of the smoke produced by use of the electric lighter and the cigarette as the lighter were more unique. The chemical patterns generated by the different lighting devices could, however, be separated by principal-component analyses. Two additional test series were also studied. In the first the cigarette was lit with an electric lighter, then extinguished, the ash was cut off, and the cigarette was re-lit. In the second the cigarette was heated in an oven to 80 °C for 5 min before being lit. These treatments did not result in changes in the chemical composition compared with cigarettes lit in the ordinary way. HPLC validated hexane bark extract of Onosma echioides L. root (OE) was evaluated for cure of human diabetic neuropathy in human neuroblastoma cell line. HPLC analysis was performed. Human neuroblastoma cells were grouped into control, normal glucose, high glucose (HG) and HG plus different concentrations of OE extract (10, 25 and 50 μg/mL). MTT, DCFH-DA staining and nuclear condensation assays were performed on neuroblastoma cells to evaluate antiproliferative activity, ROS activity level and apoptotic effect of OE. HPLC analysis revealed the existence of maximum yield of shikonin in n-hexane extract of OE. Exposure with different concentrations of OE effectively decreased ROS level and apoptosis of cells and as a result improved the viability of cells in a dose dependent manner in response to HG-induced oxidative stress. Thus, OE possesses the property to cure human diabetic neuropathy and further can be clinically tested for its use in diabetic neuropathy.  相似文献

The yield increase of secondary metabolite production was examined in plant cell cultures with the use of relatively low to high doses gamma irradiation. Suspension culture of Lithospermum erythrorhizon cells was irradiated to 2, 16, and 32 Gy. The gamma irradiation significantly stimulated the shikonin biosynthesis of the cells and increased the total shikonin yields (intracellular+extracellular shikonin yields) by 400% at 16 Gy and by only 240% and 180% at 2 and 32 Gy, respectively. One of the key enzymes for the shikonin biosynthesis of cells, p-hydroxylbenzoic acid (PHB) geranyltransferase, was found to be stimulated by the gamma-radiation treatments. The activity of PHB geranyltransferase was increased at 2 and 16 Gy with a negligible change at 32 Gy. In contrast, the activity of PHB glucosyltransferase was slightly changed at all doses of gamma radiation compared with the control cells. Therefore, the increase in PHB geranyltransferase activity leads to the accumulation of secondary metabolites such as a shikonin, which may contribute to plant defense against the stresses induced by gamma irradiation.  相似文献

1,1-Dimethylhydrazine, also known as unsymdimethylhydrazine (UDMH) and vinyl acetate (VA), are both classified by the International Agency for Research on Cancer as 2B carcinogens (possibly carcinogenic to humans) and listed as cigarette smoke constituents; however, there is little or no quantitative data available on them. For UDMH in cigarette smoke, neither a yield nor a method has been published. For VA, the most recent information on yields dates back to 1965. To bridge this gap, we have developed new gas chromatographic-mass spectrometric methods for both compounds to determine their yields in cigarette smoke. UDMH is determined by derivatization with 2-nitrobenzaldehyde in methanol and is not found in cigarette smoke at levels above the detection limit of 19 ng/cig. In further experiments, when UDMH is added to the smoke stream or air stream of lit or unlit cigarettes, the derivative 2-nitrobenzaldehyde-2,2-dimethylhydrazone is found only in the air stream of the unlit cigarettes. From this, we conclude that UDMH is either not formed during smoking at all or, if it is, it reacts immediately and quantitatively with other smoke constituents (e.g., aldehydes) and is therefore not detectable in cigarette smoke. VA is determined by trapping in acetone at -78 degrees C and is found at a concentration of 270 ng/cig for a standard reference cigarette with a cellulose acetate filter (the reference cigarette 1 R4F). In the literature, VA is reported at concentrations of 1.6 microg/cig for a cigarette with a cellulose acetate/charcoal filter and 4 microg/cig for a cigarette with a cellulose acetate filter and for an unfiltered cigarette.  相似文献

Four phenanthrene and one aporphine alkaloids semi-synthesized from boldine were evaluated for their inhibitory effect on reactive oxygen species (ROS) generation. ROS generation by neutrophils stimulated with N-formyl-methionyl-leucyl-phenylalanine was inhibited in a concentration dependent manner. Alkaloids exerted similar inhibitory effect in the hypoxanthine-xanthine oxidase system than in stimulated neutrophils, which could be attributed to a direct ROS scavenging activity. None of the alkaloids assayed had any effect on xanthine oxidase activity. Therefore the synthesized alkaloids might constitute an alternative therapy in inflammation disorders in which ROS generation is involved.  相似文献

Based on the asymmetric reduction of alkannin ketone derivative 4 by diisopinocampheylchloro-borane(DIP-Cl), a series of shikonin and alkannin derivatives was designed, synthesized and their anticancer activities against various kinds of cell lines were evaluated. The in vitro biological experiments demonstrated that compound S-10, a 5,8-O-dimethyl-1,4-dioxime alkannin derivative, not only displayed excellent antiproliferative activity against cancer cells, but also exhibited low toxicity towards normal cells. It could induce HCT-116 cell apoptosis, but had no impact on the cell cycle. The underlying anticancer mechanism of S-10 is most likely different from other shikonin and alkannin derivatives.  相似文献

Although nitrogen dioxide (NO(2)) has been previously reported to be present in cigarette smoke, the concentration estimates were derived from kinetic calculations or from measurements of aged smoke, where NO(2) was formed some time after the puff was taken. The objective of this work was to use tunable infrared laser differential absorption spectroscopy (TILDAS) equipped with a quantum cascade (QC) laser to determine if NO(2) could be detected and quantified in a fresh puff of cigarette smoke. A temporal resolution of approximately 0.16s allowed measurements to be taken directly as the NO(2) was formed during the puff. Sidestream cigarette smoke was sampled to determine if NO(2) could be detected using TILDAS. Experiments were conducted using 2R4F Kentucky Reference cigarettes with and without a Cambridge filter pad. NO(2) was detected only in the lighting puff of whole mainstream smoke (without a Cambridge filter pad), with no NO(2) detected in the subsequent puffs. The measurement precision was approximately 1.0 ppbVHz(-1/2), which allows a detection limit of approximately 0.2 ng in a 35 ml puff volume. More NO(2) was generated in the lighting puff using a match or blue flame lighter (29+/-21 ng) than when using an electric lighter (9+/-3 ng). In the presence of a Cambridge filter pad, NO(2) was observed in the gas phase mainstream smoke for every puff (total of 200+/-30 ng/cigarette) and is most likely due to smoke chemistry taking place on the Cambridge filter pad during the smoke collection process. Nitrogen dioxide was observed continuously in the sidestream smoke starting with the lighting puff.  相似文献
