利用分子模拟、荧光光谱、紫外吸收光谱等方法,研究了4-羟基-2,2’,3,4’-四溴二苯醚(4-OHBDE-42)与人血清白蛋白(HSA)的相互作用。三维荧光分析表明,4-OH-BDE-42的存在降低了HSA的荧光强度,且使HSA的微环境和构象发生变化。荧光光谱和紫外吸收光谱显示,4-OH-BDE-42与HSA结合后显著猝灭了HSA的内源性荧光,猝灭机制为静态猝灭与非辐射能量转移。结合常数Ka106L·mol-1,表明两者的结合作用较强,结合距离r为3.66nm。根据热力学参数分析,ΔH0,ΔS0,即4-OH-BDE-42与HSA之间结合的主要作用力为疏水作用,这与分子对接、结合自由能分析结论一致。结合自由能贡献分析表明,LYS199、GLU292、ARG257、ARG218、ALA291、HIS242为4-OH-BDE-42与HSA结合的关键氨基酸残基。  相似文献

用荧光光谱法研究了在生理pH条件下杀虫剂辛硫磷与牛血清白蛋白(BSA)的相互作用. 结果表明: 辛硫磷对BSA的荧光有较强的猝灭作用, 该猝灭属于静态猝灭. 根据猝灭结果求得了不同温度下辛硫磷与牛血清白蛋白结合作用的结合位点数、结合常数及反应热力学参数, 并据此确定它们之间主要的相互作用力为疏水作用力. 用同步荧光光谱法探讨了辛硫磷对BSA构象的影响.  相似文献

The interactions between daunorubicin and human serum albumin (HSA) were studied using the spectrofluorimetric method. The binding constants of daunorubicin with HSA were obtained at different temperatures. The effects of various metal ions on the binding constants of daunorubicin with HSA were also studied. The optimum conditions of fluorometric determination of daunorubicin were studied and the developed method was successfully applied to the determination of daunorubicin in serum samples.  相似文献

In this article the interaction between methyl violet (MV) and bovine serum albumin (BSA) was studied with spectroscopy. The results indicated that the fluorescence intensity of BSA was quenched strongly by MV through a static quenching procedure. The association constants, the number of binding sites and basic thermodynamic parameters were obtained based on fluorescence quenching data. The effect of MV on the conformation of BSA had been investigated with synchronous fluorescence spectroscopy and circular dichroism (CD) spectrum.  相似文献

The interaction between vinpocetine(VPC) and human serum albumin(HSA) in physiological buffer(pH 7.40) was investigated by fluorescence,FT-IR,UV-vis absorption and molecular modeling.VPC effectively quenched the intrinsic fluorescence of HSA via static quenching.The binding site number n and apparent binding constant K_a,corresponding thermodynamic parametersΔG,ΔH andΔS at different temperatures were calculated.The synchronous fluorescence and FT-IR spectra were used to investigate the structural change of HSA molecules with addition of VPC.Molecular modeling indicated that VPC could bind to the site I of HSA and hydrophobic interaction was the major acting force,which was in agreement with the binding mode study.  相似文献

以光谱技术和微量热技术相结合的方法研究水溶液中氟罗沙星分子及铁存在时与人血清白蛋白之间结合作用的机制,用荧光淬灭法测得两反应的结合常数分别为K=1.19×l05L·mol-1和1.0565×105L·mol-1,结合位点数0.97和0.835.依据Fōrster非辐射能量转移机制,得到氟罗沙星与人血清白蛋白间的结合距离(r=3.23nm).用同步荧光技术考察氟罗沙星对人血清白蛋白构象的影响.微量热法测得氟罗沙星与人血清白蛋白反应的ΔH≈0,ΔS>0,氟罗沙星分子在铁离子存在时与人血清白蛋白反应的ΔH<0,并且ΔS>0表明它们的作用都主要为静电力.由于焓熵互补的作用,两反应的自由能没有发生大的变化.  相似文献

The binding of nevadensin to human serum albumin (HSA) in aqueous solution was investigated for the first time by molecular spectroscopy and modeling at pH 7.4. Spectrophotometric observations are rationalized in terms of a static quenching process and binding constant (KaKb) and the number of binding sites (n ≈ 1) were evaluated by fluorescence quenching methods. Thermodynamic data showed that nevadensin was included in the hydrophobic cavity of HSA mainly via hydrophobic interactions. The value of 3.09 nm for the distance r between the donor (HSA) and acceptor (nevadensin) was derived from the fluorescence resonance energy transfer. Spectrophotometric techniques were also applied to investigate the structural information of HSA molecules on the binding of nevadensin and the results showed that the binding of nevadensin to HSA did not change significantly molecular conformation of HSA in our experimental conditions. Furthermore, the study of molecular modeling also indicated that nevadensin could strongly bind to the site I (subdomain IIA) of HSA mainly by a hydrophobic interaction and there are hydrogen bond interactions between nevadensin and the residues Arg-218, Arg-222, Lys-195, and Asp-451. As compared to the other flavonoids, the flavonoids containing methoxy groups which are in aromatic rings can bind to HSA with higher affinity.  相似文献

The interaction between biliverdin and bovine serum albumin (BSA) has been studied by steady fluorescence spectroscopy, synchronous fluorescence and resonance light scanning spectra. The binding of biliverdin to BSA quenches the tryptophan residue fluorescence and the results show that both static and dynamic quenching occur together with complex formation. The binding constant and binding sites of biliverdin to BSA at pH 7.1 are calculated to be 3.33 × 108 L/mol and 1.54, respectively, according to the double logarithm regression curve. In addition, the distance between the biliverdin and BSA is estimated to be 1.25 nm using Föster's equation on the basis of the fluorescence energy transfer. Furthermore the synchronous fluorescence spectra show that the microenvironment of the tryptophan residues has not obvious changes, which obeys the phase distribution model. Finally, the thermodynamic data show that biliverdin molecules enter the hydrophobic cavity of BSA via hydrophobic interaction.  相似文献

This study was designed to examine the interaction of 8-bromoadenosine with human serum albumin (HSA) by fluorescence spectroscopy in combination with molecular modeling under simulative physiological conditions. The results of fluorescence measurements indicate that 8-bromoadenosine has a strong ability to quench the intrinsic fluorescence of HSA through static quenching procedure. The binding constants (K) at different temperatures and thermodynamic parameters, enthalpy changes (ΔH) and entropy changes (ΔS) were calculated according to the fluorescence data. The results showed that the hydrophobic force played the major role in the binding of 8-bromoadenosine to HSA. The fluorescence experimental results were in agreement with the results obtained by molecular modeling study. The effects of some normal positive and negative ions on the binding constants were also discussed. Moreover, the synchronous fluorescence technique was used to characterize the interaction of 8-bromoadenosine to HSA and successfully applied to determine the total proteins in human serum, urine and saliva samples at room temperature under the optimum conditions with a wide linear range and satisfactory results.  相似文献

在模拟生理条件下(pH=7.40),应用荧光光谱、吸收光谱和同步荧光光谱法探讨了不同温度下尼泊金乙酯钠(Ethylparaben Sodium,EPS)与牛血清白蛋白(Bovine Serum Albumin,BSA)相互作用的机制.荧光光谱和吸收光谱分别表明:尼泊金乙酯钠对BSA内源荧光有明显猝灭作用,属于静态猝灭,二者的猝灭作用是由于尼泊金乙酯钠与BSA形成基态复合物导致的.运用双对数方程求得291、310 K时的结合常数分别为3.23×106、3.63×105L·mol-1,结合位点数分别为0.99和0.84.运用热力学方程得出热力学参数(ΔH,ΔS),通过ΔH0,ΔS0可知二者之间的作用力主要是氢键或范德华力.同步荧光光谱表明尼泊金乙酯钠对BSA结构的构象会产生影响.本研究为EPS的毒理和生物学效应提供了重要的参考信息.  相似文献

用荧光光谱法研究了生理酸度条件下,头孢噻肟对牛血清白蛋白,Cu(Ⅱ)对牛血清白蛋白以及Cu(Ⅱ)对头孢噻肟和牛血清白蛋白荧光光谱特性的影响。结果表明:Cu(Ⅱ)和头孢噻肟均可使牛血清白蛋白的荧光强度发生静态猝灭,并且在Cu(Ⅱ)存在下,头孢噻肟对牛血清白蛋白的荧光猝灭作用显著增强。根据荧光猝灭双倒数图计算头孢噻肟和牛血清白蛋白的结合常数为3.11×104L/mol,结合位点数为1.03;二元配合物Cu(Ⅱ)与牛血清白蛋白之间的结合常数为1.13×103L/mol,结合位点数为0.74。  相似文献

采用荧光光谱、紫外光谱、CD光谱法研究了K2Cr2O7与牛血清白蛋白(BSA)的相互作用。实验结果表明, 铬(Ⅵ)使BSA的紫外吸收降低,峰位红移,表明铬(Ⅵ)与BSA发生较强的相互作用;铬(Ⅵ)酸根离子与BSA形成基态复合物导致BSA内源荧光猝灭,猝灭机理主要为静态猝灭。测定了不同温度下该反应的热力学参数,ΔGθ<0,ΔHθ和ΔSθ分别为–12.60 kJ/mol 和 56.60 J/(mol·k), 表明上述作用过程是一个熵增加、自由能降低的自发分子间作用过程,铬(Ⅵ)酸根离子与BSA之间以静电作用力为主;非辐射能量转移机理确定了铬(Ⅵ)与牛血清白蛋白中色氨酸残基之间的距离 r=2.85 nm;同步荧光和CD光谱研究表明,铬(Ⅵ)使BSA的二级结构发生改变,α–螺旋含量降低,色氨酸残基所处微环境的极性减小。  相似文献
