3-Bromo-1,5-dimethyl-4-nitropyrazole does not react upon heating with aqueous ammonia, while 1,5-dimethyl-3,4-dinitropyrazole under the same conditions yields 3-amino-1,5-dimethyl-4-nitropyrazole, which is formed from 3-bromo-1,5-dimethyl-4-nitropyrazole in the presence of a copper catalyst. The amination of 1-methyl-3,4-dinitropyrazole-5-carboxylic acid is accompanied by decarboxylation, which is characteristic for 4-substituted 1-methylpyrazole-5-carboxylic acids upon heating in aqueous ammonia or water.  相似文献

The reactions of 6,6-dialkyl-5,7-dioxo-4,8-dioxaspiro[2.5]octane-1,1,2,2-tetracarbonitriles with primary aliphatic alcohols lead to the formation of alkyl 2,2,3,3-tetracyanocyclopropanecarboxylates; the reactions of the same compounds with ketone oximes give 2-amino-4,4-bis(alkylideneaminooxy)-6-(alkylidene-aminooxycarbonyl)-3-azabicyclo[3.1.0]hex-2-ene-1,5-dicarbonitriles, while with aldehyde oximes 2-amino-2-oxo-1,5-dicyano-3-azabicyclo[3.1.0]hex-2-ene-6-carboxylic acid is formed.  相似文献

Condensation of 5(3)-amino-3(5)-arylpyrazoles with 4-hydroxy-6-methylpyran-2-one leads to 5,7-dimethyl-2-arylpyrazolo[1,5-a]pyrimidines, 5-alkoxycarbonylmethyl-7-methyl-2-arylpyrazolo[1,5-a]pyrimidines and their isomeric 7-alkoxycarbonylmethyl-5-methyl-2-arylpyrazolo[1,5-a]pyrimidines. These compounds result from competitive reactions and from different cyclization pathways. Structure and mechanism of formation of these new products are reported.  相似文献

An efficient chemoenzymatic process has been developed for preparation of 7-amino-3-[Z-2-(4-methylthiazol-5-yl)vinyl]-3-cephem-4-carboxylic acid, featuring removal of para-methoxybenzyl by trichloroacetic acid and cleavage of phenylacetyl E-isomer by immobilized penicillin acylase enzyme. The E-isomer of 7-amino-3-[Z-2-(4-methylthiazol-5-yl)vinyl]-3-cephem-4-carboxylic acid could be easily decreased to less than 0.2 % by salt formation. Importantly, trichloroacetic acid and immobilized penicillin acylase enzyme could be recovered and reused. The enzyme reaction could be run in a flow reactor. Only two crystallizations are involved as the purification procedure in the six-step sequence.  相似文献

The stability of solutions of esters of 6′-carbamoylmethylthio-5′-cyano-2′-methyl-1′,4′-dihydro-4,4′-bipyridine-3′-carboxylic acids was investigated by HPLC. The corresponding esters of 6′-carbamoylmethylthio-5′-cyano-4,4′-bipyridine-3′-carboxylic acids,esters of 8-cyano-5-methyl-3-oxo-7-(4-pyridyl)-2,3-dihydro-7H-thiazolo-[3,2-a]pyridine-6-carboxylic acids, methyl 3-amino-2-carbamoyl-6-methyl-4-(4-pyridyl)-4,7-dihydrothieno[2,3-b]pyridine-5-carboxylate, and methyl 3-amino-2-carbamoyl-6-methyl-4-(4-pyridyl)-thieno[2,3-b]pyridine-5-carboxylate were synthesized as standard compounds (typical impurities). Analysis by HPLC was realized under the conditions of reverse-phase chromatography. It was established that solutions of the investigated compounds (with mixtures of acetonitrile with phosphate buffer, having pH values of not less than 3 and not more than 5, as solvents) are stable for one month when the solutions are stored in a place protected against light. It is also necessary to use chromatographic systems in which the aqueous components have pH 3–5 during determination of the purity of the esters of 6′-carbamoylmethylthio-5′-cyano-2′-methyl-1′,4′-dihydro-4, 4′-bipyridine-3′-carboxylic acid by HPLC in order to separate the analyzed sorbates and their typical impurities more completely. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 6, pp. 841–848, June, 2007.  相似文献

5-(3-Indolyl)isoxazole-3-carboxylic acid and its amide and hydrazide were obtained from ethyl 5-(3-indolyl)isoxazole-3-carboxylate. When 5-(3-indolyl)isoxazole-3-carboxylate is heated, it undergoes decarboxylation with isomerization to 3-(-cyanoacetyl)indole; when it is heated in alcohol with hydrazine and phenylhydrazine in the presence of copper, it undergoes isomerization to 5-(3-indolyl)pyrazole-3-carboxylic and 1-phenyl-5-(3-indolyl)-pyrazole-3-carboxylic acids. 5-(3-Indolyl)pyrazole-3-carboxylic acid hydrazide is formed when a solution of ethyl 5-(3-indolyl)isoxazole-3-carboxylate is refluxed with hydrazine in 96% alcohol.See [1] for communication CXIV.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, pp. 936–938, July, 1978.  相似文献

all-endo-3-amino-5-hydroxybicyclo[2.2.2]octane-2-carboxylic acid (13) and all-endo-5-amino-6-(hydroxymethyl)bicyclo[2.2.2]octan-2-ol (10) were prepared via dihydro-1,3-oxazine or g-lactone intermediates by the stereoselective functionalization of an N-protected derivative of endo-3-aminobicyclo[2.2.2]oct-5-ene-2-carboxylic acid (2). Ring closure of b-amino ester 4 resulted in tricyclic pyrimidinones 15 and 16. The structures, stereochemistry and relative configurations of the synthesized compounds were determined by IR and NMR.  相似文献

The corresponding 5(4)-mercaptoimidazoles, from which various 5(4)-mercaptoimidazole-4(5)-carboxylic acid derivatives were synthesized, were obtained from the amide and ethyl ester of 5-diazoimidazole-4-carboxylic acid by substitution of the diazo group. 5-Diazo-imidazole-4-hydroxamic acid does not undergo substitution with sodium disulfide but does undergo cyclization to 3-N-hydroxyimidazo[4,5-d]-1,2,3-triazin-4-one under these conditions. 以2-氨基-6-甲基苯甲酸为起始原料,通过合环反应生成两种4-氧代-3(4H)-喹唑啉-5-羧酸衍生物(1a, 1b), 1a, 1b分别经高锰酸钾氧化合成了4-氧代-3(4H)-喹唑啉-5-羧酸(2a, 2b),其结构经1H NMR, 13C NMR和MS表征。研究了投料比{r[n(高锰酸钾) :n(5-甲基-3(4H)-喹唑啉-4-酮)]}和反应温度等对收率的影响。结果表明:在最佳反应条件(r=7:1,中性高锰酸钾氧化,于90 ℃反应)下合成2总收率可达66%。  相似文献

A new procedure for the synthesis of 5(3)-nitroamino-1,2,4-triazole-3(5)-carboxylic acid and 5,5′ -bi(3-nitroamino-1,2,4-triazole) potassium salt has been developed. It includes cyclization of [2-(N2-nitro-carbamimidoyl) hydrazino]oxoacetic acid and 2,2′-bis(N 2-itrocarbamimidoyl)oxalohydrazide, respectively, which are prepared by reaction of 1-amino-2-nitroguanidine with oxalic acid. The reaction of 5(4)-aminoimidazole-4(5)-carboxhydrazide with nitrous acid was investigated. A mixture of four compounds, viz., 5-diazoimidazole-4-carboxazide, 5-diazoimidazole-4-carboxylic acid, 5(4)-aminoimidazole-4(5)-carboxazide, and 2-azahypoxanthine, is formed under all of the investigated conditions, 5(4)-Azidoimidazole-4(5)-carboxamide derivatives were obtained in the reaction of diazoimidazole-carboxazide with various amines in protic and aprotic solvents. 5-N-(Piperidyl)-azoimidazole-4-carboxazide was isolated only in the reaction with piperidine in an aqueous medium.See [1] for Communication 8.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 11, pp. 1536–1540, November, 1980.  相似文献

Isomeric 4-acetyl-5-amino-3-methyl- and 4-acetyl-3-amino-5-methylpyrazoles (2, 3) were formed in the reaction of hydrazines with 3-[amino(methylthio)methylene]pentan-2,4-dione (1) (diacetylketeneN,S-acetal). Pyrazolo[3,4-d]pyrimidines (5a,b) were synthesized by condensation of 4-acetyl-5-amino-1,3-dimethylpyrazole (2a) with amide dimethylacetals followed by treatment with ammonium acetate. 1-Oxo-3-thioxo-5-hydroxy-2-acyl-2,3-dihydro-1H-1,3-thiazino[6,5-c]quinolines were obtained for the first time by condensation of 2-hydroxy-3-mercaptoquinoline-4-carboxylic acid with acyl isothiocyanates. An efficient synthesis of (R)-7-(3-benzothienylamido)deacetoxy-cephalosporanic acid (1) is described beginning from (R)-alpha-aminobenzo[b]thiophene-3- acetic acid (2) and 7-amino-3-methyl-3-cephem-4-carboxylic acid (7-ADCA). 5-Aryl-substituted 4,7(6,7)-dihydro-1,2,4-triazolo[1,5-a]pyrimidines were obtained by condensation of 3-amino-1,2,4-triazole with -dimethylaminopropiophenone hydrochlorides or crotophenone. The optimum route for the synthesis of methyl esters of N-[(4-substituted amino)-5-cyano-2-methylthiopyrimidin-6-yl]amino acids (which are starting materials for preparing the methyl esters of the corresponding 5-amino-4-(substituted amino)pyrrolo[2,3-d]pyrimidine-6-carboxylic acids) is via subsequent reactions of 4,6-dichloro-2-methylthiopyrimidine-5-carbonitrile with amines and methyl glycinate. In some examples, the reaction of methyl N-(4-chloro-2-methylthio-6-pyrimidinyl)aminoacetate with amines occurs to give the corresponding acid amides.
