The synthesis of the spiroacetal-containing anti-Helicobacter pylori agents (3S,2'S,5'S,7'S)- (ent-CJ-12,954) and (3S,2'S,5'R,7'S)- (ent-CJ-13,014) has been carried out based on the convergent union of a 1:1 mixture of heterocycle-activated spiroacetal sulfones and with (3S)-phthalide aldehyde . The synthesis of the (3R)-diastereomers (3R,2'S,5'S,7'S)- and (3R,2'S,5'R,7'S)- was also undertaken in a similar manner by union of (3R)-phthalide aldehyde with a 1:1 mixture of spiroacetal sulfones and . Comparison of the (1)H and (13)C NMR data, optical rotations and HPLC retention times of the synthetic compounds (3S,2'S,5'S,7'S)- and (3S,2'S,5'R,7'S)- and the (3R)-diastereomers (3R,2'S,5'S,7'S)- and (3R,2'S,5'R,7'S)-, with the naturally occurring compounds, established that the synthetic isomers and were in fact enantiomeric to the natural products CJ-12,954 and CJ-13,014. The (2S,8S)-stereochemistry in protected dihydroxyketone , the precursor to the mixture of spiroacetal sulfones and was established via union of readily available (S)-acetylene with aldehyde in which the (4S)-stereochemistry was established via asymmetric allylation. Deprotection and cyclization of protected dihydroxyketone afforded an inseparable 1:1 mixture of spiroacetal alcohols and that were converted into a 1:1 inseparable mixture of spiroacetal sulfones and . Phthalide-aldehyde was prepared via intramolecular acylation of bromocarbamate in which the (3S)-stereochemistry was established via asymmetric CBS reduction of ketone .  相似文献

From a MeOH extract of the aerial part of Piper futokadsura, the tetrahydrofuran lignans, futokadsurin A [(7S,8S,7'S,8'R)-3,4,3'-trimethoxy-4'-hydroxy-7,7'-epoxylignan], futokadsurin B [(7R,8R,7'R,8'S)-3,4-dimethoxy-3',4'-methylenedioxy-7,7'-epoxylignan], and futokadsurin C [(7R,8R,7'S,8'S)-3,4-methylenedioxy-3',4'-dimethoxy-7,7'-epoxylignan] were isolated, together with nine known neolignans. In addition, L-tryptophan, pellitorine, phytol, elemicin, and 1,2,4-trimethoxyphenyl-5-aldehyde were isolated. The structures of the new compounds were elucidated using spectroscopic methods. These lignans inhibited nitric oxide production by a murine macrophage-like cell line (RAW 264.7), which was activated by lipopolysaccharide and interferon-gamma.  相似文献

Various conflicting data on the rearrangement and absolute stereochemistry of hydroxylignano-9,7'-lactones are resolved using 18O labeled compounds, also confirmed by an X-ray analysis of a pure lignano-9,7'-lactone enantiomer, obtained for the first time. Under NaH/DMF rearrangement conditions a silyl protected hydroxylignano-9,9'-lactone underwent an unexpected silyl migration.  相似文献

One new phenylpropanoid, turformosin A (1), and one new triterpene, turformosinic acid (2), together with 16 known compounds, were isolated from the stems of Turpinia formosana Nakai. All structures were elucidated on the basis of spectroscopic analysis, including 1D- and 2D-NMR techniques and MS analysis. Selected isolated compounds were evaluated for in vitro cytotoxicity against four human cancer cell lines and antioxidant scavenging effects on DPPH. (-)-(7'S,8'S)-threo-carolignan X (3) exhibited cytotoxicity against Hep2, WiDr, Daoy, and MCF-7 cell lines with ED(50) values of 3.60, 4.45, 6.07, and 13.7 μg/mL, respectively. Turformosin A (1), (-)-(7'S,8'S)- threo-carolignan X (3), methoxyhydroquinone-4-β-D-glucopyranoside (5), and methoxy-hydroquinone-1-β-D-glucopyranoside (6), exhibited similar anti-oxidative activity. Hep2 cells treated with 10 μg/mL of 3 showed elevation of sub-G1 population (from 20% at 8 h to 60% at 48 h), and activation of caspase-9/caspase-3/PARP cascade. Compound 3 induced intrinsic apoptotic pathway in Hep2 cells with dose and time dependence (10 μg/mL for 8 h).  相似文献

[reaction: see text] (-)-(6S,7S,8S,9R,10S,2'S)-Membrenone-A and (-)-(6S,7S,8S,9R,10S)-membrenone-B were prepared in 11 steps (3% and 2.4% overall yield, respectively). Key steps included a tin(II)-mediated aldol followed by a syn selective reduction, giving the C7-C9 stereocenters, a second chain extending aldol coupling, and a p-TsOH-promoted cyclization/dehydration giving the common gamma-dihydropyrone precursor. We have thus established that synthetic (-)-(6S,7S,8S,9R,10S,2'S)-membrenone-A, (-)-(6S,7S,8S,9R,10S)-membrenone-B, and (-)-(6S,7S,8S,9R,10S)-membrenone-C are the enantiomers of the natural products.  相似文献

The petrol extract from mace of Myristica argentea Warb. afforded six phenylpropenes, three lignans, three neolignans and a dilignan, bis erythro-argenteane (4) or rel-(8R,8'S,8"S,8"'R)-5',5"'-bis(7-(3,4-methylenedioxyphenyl)-7'-(4'-hydroxy-3'-methoxyphenyl)-8.8'-lignane]. Time-dependent density functional theory (TD-DFT) is applied to the UV-vis absorption and circular dichroism (CD) spectra of a series of transition metals (M=Ru, Zn, Fe) complexed with an enantiopure hemicage ligand, (-)-(5R,5'R,5' 'R,7R,7'R,7' 'R,8S,8'S,8' 'S)-8,8',8' '-[(2,4,6-trimethyl-1,3,5-benzenetriyl)tris(methylene)]tris[5,6,7,8-tetrahydro-6,6-dimethyl-3-(2-pyridinyl)-5,7-methanoisoquinoline (1). The electronic spectra of the Ru and Fe complexes contain two regions, one featuring low-energy 1MLCT transitions and the other higher energy 1LC transitions; the Zn analog possesses only the 1LC transitions due to its filled 3d shell. TD-DFT is able to identify correctly these transitions in the spectra, as well as to reproduce experimental spectra accurately, with regard to both the transition energies and the relative intensities of the different transitions. Additionally, it is possible to use TD-DFT to assign the absolute configuration at the metal center with high confidence by matching the experimental and calculated spectra.  相似文献

Two new stereoisomers of tetrahydrofuranoid lignans, 7S,8R,7'S,8'R- (1) and 7R,8S,7'S,8'R-3,4,3',4'-tetramethoxy-9,7'-dihydroxy-8.8',7.O.9'-lignan (2) along with nine known lignans including tetrahydrofuranoids (3, 4) and tetrahydrofurofuranoids (5-11) were isolated from a CHCl(3)-soluble fraction of the flower buds of Magnolia fargesii. Two tetrahydrofuranoids, magnostellin A (3) and lariciresinol dimethyl ether (4) were isolated from this species for the first time. The structures of these compounds (1-11) were identified by spectroscopic methods as well as by comparison with published values. Absolute configurations of new stereoisomers (1, 2) were determined by the Mosher's esterification method and Circular Dichroism (CD) studies. All the isolates (1-11) were evaluated for their antioxidant activities using modified superoxide radical-scavenging assay. Compounds 5-8 showed the potent superoxide radical-scavenging activities with the ED(50) values of 19.2, 19.2, 16.5, and 27.7 microM, respectively, as compared with standard antioxidants (BHA: 22.8 microM; Trolox: 940 microM).  相似文献

In continuation of our studies into the mass spectrometric detection of natural lignans and their identification in complex mixtures such as crude plant extracts, the electrospray ionization tandem mass spectrometric (ESI-MS/MS) fragmentation of Delta(7,8)-unsaturated dibenzylbutyrolactone-type lignans (lign-7-eno-9,9'-lactones) was studied in detail. It is demonstrated that the characteristic fragmentation allows unambiguous identification including distinction between constitutional isomers. These lignans containing an alpha,beta-unsaturated lactone structure exist as equilibrium mixtures of E- and Z-isomers indistinguishable by mass spectrometry, but it is shown that chromatographic retention time can be used to distinguish between the isomeric forms. Based on these observations, re-analysis of the dichloromethane extract obtained from flowering aerial parts of Linum usitatissimum L. by high-performance liquid chromatography (HPLC)/ESI-MS/MS led to the identification of eighteen lignans of these types (five lignano- and one lignenolactone previously reported along with five further lignano- as well as seven lignenolactones hitherto unreported for this plant). The simultaneous identification of eighteen different lignans in the complex matrix of a crude plant extract by a single analysis demonstrates the potential of this method, which will certainly lead to new insights into the lignan composition and metabolism of different Linum species and many other plants.  相似文献

An overview of the synthesis of the fungal metabolites (+)-dermolactone, (-)- semixanthomegnin, (+)- and (-)-mellein, (-)-ochratoxin alpha, (-)-(1R,3S)-thysanone, the enantiopure ventiloquinones L, E and G, and 8-desmethyleleutherin from a common chiral intermediate, is presented. The reaction of chiral (2R,1'S)- or (2S,1'S)-2-(1-aminoalkyl)epoxides 1 or 2 with CO2, generated from acidic treatment of an aqueous solution of NaHCO3 at room temperature, efficiently afforded enantiopure cyclic carbonates 3 or 4, respectively, with total selectivity. Compounds 3 and 4 were readily transformed into the corresponding diols 7 and 8 by reaction with LiAlH4 or by basic hydrolysis. When compounds 3 or 4 were allowed to react with methyllithium at -78 degrees C, O1-acetylalkane-1,2-diols 9 and 10 were obtained with total or high selectivity.  相似文献

A variety of substituted 2'-deoxyadenosin-5'-yl radicals 3 were generated under different reaction conditions. Radicals 3 underwent intramolecular cyclization onto the C8-N7 double bond of the adenine moiety leading to aminyl radicals (5'S,8R)-4 and (5'R,8R)-4 and, eventually, to the corresponding cyclonucleosides 5 and 6. The effect of the solvent, the nature of the substituents, and the generation method of radicals 3 on the stereoselectivity of the C5'-radical cyclization have been considered. The observed increase of the (5'S)/(5'R) ratio by increasing the bulkiness of the R1 group is explained in terms of steric repulsion between R1 and the purine moiety which favors the C5'-endo conformation, whereas the effect of the water solvent in promoting the (5'R)-stereoselective cyclization is ascribed to intermolecular hydrogen bonding stabilizing the C5'-exo conformation.  相似文献

Novel fluorene–isoindole-containing light-emitting mono- and oligomers were prepared. The synthesis of the monomers N-(4-bromo-phenyl)-2-(7-bromo-9,9-diethylfluoren-2-yl)-isoindole-1-carboxamide, N-(7-bromo-9,9-dibutylfluoren-2-yl)-2-(7-bromo-9,9-dibutylfluoren-2-yl)-isoindole-1-carboxamide, and N-[7-bromo-9,9-bis(2-ethylhexyl)-fluoren-2-yl]-2-[7-bromo-9,9-bis(2-ethylhexyl)-fluoren-2-yl]-isoindole -1-carboxamide was carried out by a three-component reaction of ortho-phthalaldehyde with the corresponding amine and isocyanide partners. The Ni(0) mediated polymerization reactions of the obtained monomers gave the corresponding mixture of oligomers from two up to twelve repeat units. The optical properties were also studied and it was found that the phenylene-containing oligomer emitted green light in dichloromethane solution, while both difluorene-containing oligomers, under the same conditions, proved to be blue light-emitters with good quantum efficiency.  相似文献

设计合成了3种新颖的螺二芴键联富勒烯(C60/C70)吡咯烷衍生物, 其结构通过IR, 1H NMR, 13C NMR和MALDI-TOF进行确证, 其电化学性质用循环伏安法进行研究. 结果表明, C70衍生物6的还原电位较C60衍生物7分别向负电势移动0.1, 0.12和0.01 V. 同时, 使用纳秒和飞秒激光分别研究了化合物6, 7和8的光限幅性能, 其光限幅阈值分别为15.3, 23.3和13.7 J/cm2, 表明材料具有优异的光限幅性能.  相似文献

3'-Deoxy-4-O-methylepisappanol, a new 3-benzylchroman derivative, was isolated from Sappan Lignum, together with thirteen known chemical compounds identified as protosappanin A, sappanchalcone, sappanone B, palmitic acid, (+)-(8S,8'S)-bisdihydrosiringenin, brazilein, 3-deoxysappanchalcone, (+)-lyoniresinol, 3-deoxysappanone B, protosappanin B, isoprotosappanin B, 3'-O-methylbrazilin and brazilin, respectively. Among these known compounds, this is the first time that (+)-(8S,8'S)-bisdihydrosiringenin was obtained from the family Caesalpiniaceae.  相似文献

Three new lignan glycosides (1-3) were isolated from the stems of Akebia trifoliata. Their structures were elucidated as (7R,8R,7'R,8'R)3,3',5,5'tetramethoxy-4,4'dihydroxy-7,9':7',9-diepoxylignan-4-O-beta-D-glucopyranoside (1), (7S,8S,8'R)-4,4',9-trihydroxy-3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (2), (7R,8R,8'S)-4,4',9-trihydroxy3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7'-one 9-O-beta-D-glucopyranoside (3) by spectral analyses, primarily NMR, MS and CD. The NMR assignments for the compounds were carried out using 1H, 13C, DEPT, COSY, HSQC, HMBC and ROESY NMR experiments.  相似文献

Three new carotenoids: 7',8',9',10'-tetrahydro-β-cryptoxanthin, 7',8'-dihydrodiatoxanthin, and (3S,6S,6'S)-ε-cryptoxanthin were isolated from the skin, fins, and gonads of the Japanese common catfish, Silurus asotus, as minor carotenoids. Their structures were determined based on chemical and spectroscopic data. Furthermore, 9Z and/or 9'Z geometrical isomers of parasiloxanthin, 7',8'-dihydroparasiloxanthin, and 7',8'-dihydro-β-cryptoxanthin were characterized by (1)H-NMR.  相似文献

The reactivity of various 2-oxyallyl cations toward 2,2'-methylenedifuran (1b), 2,2'-(hydroxymethyl)difuran (1c), 2,2'-(trimethylsilylmethylene)difuran (1d), and di(2-furyl)methanone (1e) has been explored. Difuryl derivatives 1c, 1d, and 1e refused to undergo formal double [4+3]-cycloadditions. Conditions have been found to convert 1b into meso-1,1'-methylenedi[(1R,1'S,5S,5'R)- (3) and (+/-)-1,1'-methylenedi[(1RS,1'SR,5SR,5'RS)-8-oxabicyclo[3.2.1]oct-6-en-3-one] (4) that do not require CF(3)CH(OH)CF(3) as solvent. High yields of meso-1,1'-methylenedi[(1R,1'S,2S,2'R,4R,4'S,5S,5'R)- (5) and (+/-)-1,1'-methylenedi[(1RS,1'RS,2SR,2'SR,4RS,4'RS,5SR,5'SR)-2,4-dimethyl-8-oxabicyclo[3.2.1]oct-6-en-3-one] (6) have been obtained when 1b was reacted with 2,4-dibromopentan-3-one (7h) and NaI/Cu.  相似文献

Two new glycosides, 7S,7'S,8R,8'R-icariol A(2)-9-O-beta-D-glucopyranoside (1) and 4-allyl-2-hydroxyphenyl 1-O-beta-D-apiosyl-(1-->6)-beta-D-glucopyranoside (2), were isolated from the dried whole plants of Glechoma hederacea L. (Labiatae) together with four known compounds, cistanoside E (3), dihydrodehydrodiconiferyl alcohol 4-O-beta-D-glucopyranoside (4), apigenin 7-O-beta-D-glucuronopyranoside (5) and luteolin 7-O-beta-D-glucopyranoside (6). The structures of the new compounds were elucidated on the basis of chemical and spectral analysis.  相似文献

In the search for complexes modeling the [Fe(CN)(2)(CO)(cysteinate)(2)] cores of the active centers of [NiFe] hydrogenases, the complex (NEt(4))(2)[Fe(CN)(2)(CO)('S(3)')] (4) was found ('S(3)'(2-)=bis(2-mercaptophenyl)sulfide(2-)). Starting complex for the synthesis of 4 was [Fe(CO)(2)('S(3)')](2) (1). Complex 1 formed from [Fe(CO)(3)(PhCH=CHCOMe)] and neutral 'S(3)'-H(2). Reactions of 1 with PCy(3) or DPPE (1,2-bis(diphenylphosphino)ethane) yielded diastereoselectively [Fe(CO)(2)(PCy(3))('S(3)')] (2) and [Fe(CO)(dppe)('S(3)')] (3). The diastereoselective formation of 2 and 3 is rationalized by the trans influence of the 'S(3)'(2-) thiolate and thioether S atoms which act as pi donors and pi acceptors, respectively. The trans influence of the 'S(3)'(2-) sulfur donors also rationalizes the diastereoselective formation of the C(1) symmetrical anion of 4, when 1 is treated with four equivalents of NEt(4)CN. The molecular structures of 1, 3 x 0.5 C(7)H(8), and (AsPh(4))(2)[Fe(CN)(2)(CO)('S(3)')] x acetone (4 a x C(3)H(6)O) were determined by X-ray structure analyses. Complex 4 is the first complex that models the unusual 2:1 cyano/carbonyl and dithiolate coordination of the [NiFe] hydrogenase iron site. Complex 4 can be reversibly oxidized electrochemically; chemical oxidation of 4 by [Fe(Cp)(2)PF(6)], however, led to loss of the CO ligand and yielded only products, which could not be characterized. When dissolved in solvents of increasing proton activity (from CH(3)CN to buffered H(2)O), complex 4 exhibits drastic nu(CO) blue shifts of up to 44 cm(-1), and relatively small nu(CN) red shifts of approximately 10 cm(-1). The nu(CO) frequency of 4 in H(2)O (1973 cm(-1)) is higher than that of any hydrogenase state (1952 cm(-1)). In addition, the nu(CO) frequency shift of 4 in various solvents is larger than that of [NiFe] hydrogenase in its most reduced or oxidized state. These results demonstrate that complexes modeling properly the nu(CO) frequencies of [NiFe] hydrogenase probably need a [Ni(thiolate)(2)] unit. The results also demonstrate that the nu(CO) frequency of [Fe(CN)(2)(CO)(thiolate)(2)] complexes is more significantly shifted by changing the solvent than the nu(CO) frequency of [NiFe] hydrogenases by coupled-proton and electron-transfer reactions. The "iron-wheel" complex [Fe(6)[Fe('S(3)')(2)](6)] (6) resulting as a minor by-product from the recrystallization of 2 in boiling toluene could be characterized by X-ray structure analysis.  相似文献
