A new synthesis of pipecolic acid and homopipecolic acid derivative was accomplished from 1-chloroalkyl p-tolyl sulfoxides having a 2-aminophenyl group at the ω-position by treatment with i-PrMgCl via the intramolecular reaction of magnesium carbenoid with N-magnesio arylamine. In a similar way, proline and pipecolic acid derivatives were synthesized from 1-chloroalkyl p-tolyl sulfoxides having an arylamino group at the ω-position. 相似文献

From heteroarylaminomethyleneoxazolones 4 , obtained from N-heteroarylformamidines 2 and 2-phenyl-5-oxo-4,5-dihydro-1,3-oxazole ( 3 ), the following β-heteroarylamino-α,β-dehydro-α-amino acid derivatives were prepared: methyl 8 and ethyl esters 9 , amides 10 and 11 , hydrazides 12 , and azides 15 . By catalytic hydrogenation the compounds 4 were converted into β-heteroarylamino substituted amides 18 and β-heteroarylamino-α-amino acids 20 . 相似文献

2-Alkoxy-4-heteroarylaminomethylene-5(4H)-thiazolones 4 were converted with various nucleophiles into β-heteroarylamino-α,β-dehydro-α-amino acid derivatives 11, 14, 15, 16, 17, 18 , and 19 . Reduction of 4 with sodium borohydride in ethanol saturated with gaseous ammonia afforded the corresponding β-heteroaryl-amino substituted alanyl amides 20 . N-Sulfinylimines derived from aromatic and aliphatic aldehydes react with nitroethane and NaOH, yielding mainly two diastereoisomeric β-nitroamines as the result of a highly diastereoselective reaction and further epimerization of the carbon linked to the nitro group. N-(Hydroxyalkyl) β-alanine ester which was obtained from amino alcohol and acrylate yielded polyamide at room temperature in the presence of a basic catalyst. Alkali and alkali earth metal alkoxides had a strong catalytic effect on the room-temperature polycondensation of N-(hydroxyethyl)-β-alanine esters. The catalytic activity of metal alkoxides decreased in the order: Li > Na > K > Cs and Ca > Zn > Mg. Aluminum and titanium alkoxide had a weak catalytic effect, while boron (III), tin (IV), antimony (V), and tellurium (VI) alkoxides did not show any catalytic activity for the polycondensation. It was also found that solvent had an effect on the course of the polycondensation of N-(hydroxyethyl)-β-alanine esters, and the highest molecular weight polymer was formed only in methanol solution. The solid-phase polycondensation of the low molecular weight prepolymer resulted in a high molecular weight polymer with an inherent viscosity of 1.0 in the presence of a catalytic amount of phosphoric acid. The polymer obtained is hydrophilic and its moisture absorption is more than twice that of nylon 6. 相似文献

An improved practical synthetic method was developed for the preparation of selenocarboxylates of amino acids through the reaction of the corresponding activated esters with sodium hydrogen selenide in alcoholic or aqueous medium. The protected α-amino selenocarboxylates reacted readily with sulfonyl azide to form N-(α-aminoacyl)sulfonamides in high yields. The commonly used protecting groups in amino acid and peptide chemistries are well tolerated under these reaction conditions. No protecting groups are needed for the side chains of Arg, Met, Ser, Tyr, and Trp. 相似文献

A protocol for stereoselective C-radical addition to a chiral glyoxylate-derived N-sulfinyl imine was developed through visible light-promoted photoredox catalysis, providing a convenient method for the synthesis of unnatural α-amino acids. The developed protocol allows the use of ubiquitous carboxylic acids as radical precursors without prior derivatization. The protocol utilizes near-stoichiometric amounts of the imine and the acid radical precursor in combination with a catalytic amount of an organic acridinium-based photocatalyst. Graft copolymerization of N-carboxy anhydride of β-benzyl-L -aspartate onto copoly(L -lysine γ-methyl-L -glutamate) was carried out in N,N-dimethylformaide which contained 3 v/v% of dimethyl sulfoxide to obtain multi-Nε-poly(β-benzyl-L -aspartyl)copoly(L -lysine γ-methyl-L gluta mate). The degree of polymerization of the branch chain attained was much influenced by the interval of the grafting sites of the copoly(L -lysine γ-methyl-L -glutamate). The solvent-induced two-step conformational transition of the multichain copoly(α-amino acid) was observed in the chloroform-dichloroacetic acid system at 25°C by the ORD technique. The stability of the α-helical conformation of the backbone polymer chain was decreased by the presence of poly(β-benzyl-L -aspartyl) branch chains that could form unstable α-helical conformations of opposite spirals. 相似文献

The regio- and stereoselective addition of chlorosulfonyl isocyanate to (+)-3-carene 1 resulted in β-lactam 2, which was converted to N-Boc-β-amino acid 4, β-amino ester 7, and carboxamide derivatives 18 and 20 via N-Boc activation and mild ring opening. The corresponding β-amino ester 7 was transformed to 2-thioxopyrimidin-4-one 11 and 2,4-pyrimidinedione 13. LAH reduction of 5 and 7 resulted in amino alcohols 6 and 8. A protocol for the chemically divergent synthesis of β-lactams and α-amino acid derivatives with isothiourea (ITU) catalysis by switching solvents was developed. The stereospecific Mannich reaction occurring between imine and C(1)-ammonium enolate generated zwitterionic intermediates, which underwent intramolecular lactamization and afforded β-lactam derivatives when DCM and CH3CN were used as solvents. However, when EtOH was used as the solvent, the intermediates underwent an intermolecular esterification reaction, and α-amino acid derivatives were produced. Detailed mechanistic experiments were conducted to prove that these two kinds of products came from the same intermediates. This work reports a new method for the synthesis of quaternary α-alkenyl substituted amino acids by the enantio- and diastereoselective addition of imidazolidinone derivatives to alkynes and allenes. Further hydrolysis of the imidazolidinone products under acidic conditions afforded biologically relevant amino acid derivatives. This method is geometry-selective (E-isomer), enantio- and diastereoselective, and products were obtained in good to excellent yields. The utility of this new methodology is proved by its operational simplicity and the successful accomplishment of gram-scale reactions. Polymerization of the N-carboxy anhydride of N?-carbobenzyloxy-L -lysine in the presence of multifunctional polymeric initiator, copoly(L -lysine γ-methyl-L -glutamate) was studied in N,N-dimethylformamide containing 3% (v/v) of dimethyl sulfoxide. Multichain copoly(α-amino acid), i.e., multi-N?-poly(N?-carbobenzyloxy-L -lysine)copoly(L -lysine γ-methyl-L -glutamate), was obtained with linear poly(N?-carbobenzyloxy-L -lysine) as by-product that could be removed by reprecipitation as was evidenced by gel-permeation chromatography. The degree of polymerization of the branch polymer chains estimated by the osmometric molecular weight determination and amino acid analysis was between 20 and 60, which decreased with increasing lysine content of the polymeric initiator. The stability of α-helical conformation of the multichain copoly(α-amino acid) was studied in the chloroform–dichloroacetic acid system at 25°C by the ORD technique. The α-helical conformation of poly(N?-carbobenzyloxy-L -lysine) branches was less stable than those of linear poly(N?-carbobenzyloxy-L -lysine) and the core molecular chains of the multichain copoly(α-amino acid). 相似文献

The organocatalytic asymmetric Mannich reaction of ethyl 4-chloro-3-oxobutanoate with N-Boc-imines has been studied and Cinchona alkaloids and chiral thiourea-tertiary amines were evaluated as catalysts. A pyrrolidine-based thiourea-tertiary amine was identified as the best catalyst for the transformation. The Mannich adducts readily underwent a one-pot intramolecular cyclization in the presence of triethylamine. A number of O-ethyl tetronic acid derivatives were obtained in good yields and enantioselectivities (up to 91% ee). The products could be further converted to the heteroatomic mimics of prostaglandins via reaction with primary amines. 相似文献

A copper-catalysed regio- and diastereoselective borylamination of α,β-unsaturated esters with B2pin2 and hydroxylamines has been developed to deliver acyclic β-boryl-α-amino acid derivatives with high anti-diastereoselectivity (up to >99 : 1), which is difficult to obtain by the established methods. A chiral phosphoramidite ligand also successfully induces the enantioselectivity, giving the optically active β-borylated α-amino acids. The products can be stereospecifically transformed into β-functionalised α-amino acids, which are of potent interest in medicinal chemistry.A Cu-catalysed regio-, diastereo-, and enantioselective borylamination of α,β-unsaturated esters with B2pin2 and hydroxylamines has been developed.
