In 2002, a new class of thymidylate synthase (TS) involved in the de novo synthesis of dTMP named Flavin-Dependent Thymidylate Synthase (FDTS) encoded by the thyX gene was discovered; FDTS is present only in 30% of prokaryote pathogens and not in human pathogens, which makes it an attractive target for the development of new antibacterial agents, especially against multi-resistant pathogens. We report herein the synthesis and structure-activity relationship of a novel series of hitherto unknown pyrido[1,2-e]purine-2,4(1H,3H)-dione analogues. Several synthetics efforts were done to optimize regioselective N1-alkylation through organopalladium cross-coupling. Modelling of potential hits were performed to generate a model of interaction into the active pocket of FDTS to understand and guide further synthetic modification. All those compounds were evaluated on an in-house in vitro NADPH oxidase assays screening as well as against Mycobacterium tuberculosis ThyX. The highest inhibition was obtained for compound 23a with 84.3% at 200 µM without significant cytotoxicity (CC50 > 100 μM) on PBM cells. 相似文献

Dipolar cycloaddition of the N-substituted C-(diethoxyphosphonyl)nitrones with N3-allyl-N1-benzylquinazoline-2,4-diones produced mixtures of diastereoisomeric 3-(diethoxyphosphonyl)isoxazolidines with a N1-benzylquinazoline-2,4-dione unit at C5. The obtained compounds were assessed for antiviral and antibacterial activities. Several compounds showed moderate inhibitory activities against VZV with EC50 values in the range of 12.63–58.48 µM. A mixture of isoxazolidines cis-20c/trans-20c (6:94) was found to be the most active against B. cereus PCM 1948, showing an MIC value 0.625 mg/mL, and also was not mutagenic up to this concentration. 相似文献

The heterocyclizations from various methyl (2-nitrobenzoyl)carbamates to substituted quinazoline-2,4(1H,3H)-diones under hydrogenation conditions were investigated in this study. In the presence of p-toluenesulfonic acid monohydrate in methanol, various quinazoline-2,4(1H, 3H)-diones were obtained in good to excellent yields within 12?h. The reaction was proposed to proceed through the cascade reactions of nitro reduction and condensation. 相似文献

对羟苯基丙酮酸双氧化酶(EC 1.13.11.27, HPPD)是一个重要的除草剂作用靶标. 为了寻找具有高效除草活性的新型HPPD抑制剂, 在前期研究的基础上设计合成了24个含有喹唑啉二酮结构的三酮类化合物9a~9x. 所合成的化合物均经过1H NMR, 13C NMR和 HRMS的表征. 以来源于拟南芥的HPPD (AtHPPD)为测试对象进行酶抑制活性筛选, 结果表明所合成的大部分化合物对AtHPPD均表现出了较好的抑制效果, 其中化合物9i的Ki 值为0.005 μmol/L, 显著优于商品化对照药剂硝磺草酮(Ki=0.013 μmol/L). 进一步温室盆栽筛选结果表明, 多数化合物在150 g ai/ha的剂量下对六种供试杂草中的至少一种表现出80%以上的防效, 特别是化合物9g在37.5 g ai/ha的低剂量下对所测试的六种杂草中的四种仍表现出了大于85%的抑制效果, 同时9g 在150 g ai/ha的剂量下对水稻和小麦均表现出了很好的作物安全性, 可以作为先导化合物供进一步深入研究. 相似文献

Quinoxaline-2,3(1H,4H)-dithione can be synthesized efficiently through the thionation of quinoxaline-2,3(1H,4H)-dione with phosphorous pentasulfide. It can also be obtained by reaction of 2,3-dichloroquinoxaline with thiourea or sodium hydrogen sulfide. The most common reactions of quinoxaline-2,3(1H,4H)-dithione involve deprotonation and electrophilic attack at the sulfur atoms to give various substituted derivatives and poly-fused heterocyclic ring systems. On the basis of the interesting structures and biological activities exhibited by several heterocyclic systems possessing the pyridone nucleus such as mappcine and camptothecin, we have planed to design the synthesis, crystal studies and antibacterial activity of the new 1-((2- chloroquinolin-3yl)-methyl)-pyridine-2(1H)-one building block. An X-ray analysis has provided valuable insight into the effect of steric factors on the three-dimensional shape of this compound which serves as a useful advanced intermediate in the synthesis of these biologically active molecules. 为了进一步研究3-取代苄基-6-三氟甲基嘧啶-2,4(1H,3H)-二酮衍生物的除草活性, 以期发现更高活性化合物, 合成了16个未见文献报道的3-取代苄基-6-三氟甲基嘧啶-2,4(1H,3H)-二酮衍生物, 其结构均经过1H NMR, IR和元素分析确证. 生测结果表明, 嘧啶环1-位取代基的变化, 不仅影响化合物的抑制活性与选择性, 可能还改变了化合物的作用方式. 定量的结构与活性关系研究表明, 当作用对象为油菜时, 化合物的活性可能主要与取代基R的摩尔分子折射常数有关; 当作用对象为稗草时, 化合物的活性可能主要与取代基R的电性参数有关. 1-位为氢时, 有利于对油菜生长的抑制; 1-位为甲基时, 有利于对稗草生长的抑制. 相似文献

Currently, studies are being conducted on the possible role of the cytoprotective effect of biologically active substances in conditions of cerebral hypoxia or cardiomyopathies. At the same time, oxidative stress is considered one of the important mechanisms of cellular cytotoxicity and a target for the action of cytoprotectors. The aim of this study is to search for derivatives of 3-(arylmethylamino)-6-methyl-4-phenylpyridin-2(1H)-ones. The probability of cytoprotective action was assessed by measuring cell viability using two tests (with neutral red dye and MTT test). It was found that some derivatives of 3-(arylmethylamino)-6-methyl-4-phenylpyridin-2(1H)-ones under the conditions of our experiment had a pronounced cytoprotective activity, providing better cell survival in vitro, including the MTT test and conditions of blood hyperviscosity. To correlate the obtained results in vitro, molecular docking of the synthesized derivatives was also carried out. The standard drug omeprazole (co-crystallized with the enzyme) was used as a standard. It was shown that all synthesized derivatives of 3-(arylmethylamino)-6-methyl-4-phenylpyridin-2(1H)-ones had higher affinity for the selected protein than the standard gastro-cytoprotector omeprazole. The studied derivatives of 3-(arylmethylamino)-6-methyl-4-phenylpyridin-2(1H)-ones also fully satisfy Lipinski’s rule of five (RO5), which increases their chances for possible use as orally active drugs with good absorption ability and moderate lipophilicity. Thus, the results obtained make it possible to evaluate derivatives of 3-(arylmethylamino)-6-methyl-4-phenylpyridin-2(1H)-ones as having a relatively high cytoprotective potential. 相似文献

A library of novel 1H-pyrimido[4,5-c]pyridazine-5,7-dione analogs was prepared in a parallel fashion via the cyclization of 6-(1-ethylhydrazinyl)-3-methylpyrimidine-2,4(1H,3H)-dione (4) with α-bromoketones and was then isolated by a two-step purification method with >90% purity. Both the library synthesis and purification method proved suitable for analogs with a high diversity of substitution at the 3-position including various alkyl, aryl, and heteroaryl groups. 相似文献

A novel series of pyrido[ 1,2-e]purin-4(3H)-one derivatives containing polar substituents on 5'-position were designed and prepared as potential PDE5 inhibitors. This paper reports the synthetic routes, 1H-NMR data, and the PDE5 inhibitory activities of the target compounds. A series of novel pyrimidin-4(3H)-one hydroxamate derivatives was designed, synthesized and studied for their activities against histone deacetylases(HDACs). The results indicate that all the compounds show HDACs inhibitiory activity. The antiproliferative activities of the compounds against HeLa and A549 cells were also investigated.
