Background  

Apoptosis plays a key role in cell death observed in neurodegenerative diseases marked by a progressive loss of neurons as seen in Alzheimer's disease. Although the exact cause of apoptosis is not known, a number of factors such as free radicals, insufficient levels of nerve growth factors and excessive levels of glutamate have been implicated. We and others, have previously reported that in a stable HT22 neuronal cell line, glutamate induces apoptosis as indicated by DNA fragmentation and up- and down-regulation of Bax (pro-apoptotic), and Bcl-2 (anti-apoptotic) genes respectively. Furthermore, these changes were reversed/inhibited by estrogens. Several lines of evidence also indicate that a family of cysteine proteases (caspases) appear to play a critical role in neuronal apoptosis. The purpose of the present study is to determine in primary cultures of cortical cells, if glutamate-induced neuronal apoptosis and its inhibition by estrogens involve changes in caspase-3 protease and whether this process is mediated by Fas receptor and/or mitochondrial signal transduction pathways involving release of cytochrome c.  相似文献

Background  

Although octopamine has long been known to have major roles as both transmitter and modulator in arthropods, it has only recently been shown to be functionally important in molluscs, playing a role as a neurotransmitter in the feeding network of the snail Lymnaea stagnalis. The synaptic potentials cannot explain all the effects of octopamine-containing neurons on the feeding network, and here we test the hypothesis that octopamine is also a neuromodulator.  相似文献

Background  

Debate has been ongoing on the relative merits of olfactory ensheathing cells (OECs) and Schwann cells as candidates for transplant-mediate repair of CNS lesions. Both glial cells exhibit similar molecular and cellular properties and to date there has been no antigenic marker identified that can clearly distinguish the two cell types. This inability to distinguish between the two cells types prevents confirmation of a controversial statement that cultures of OECs are contaminated with Schwann cells. Recently, proteomic analysis of foetal OECs and adult Schwann cells identified an actin-binding protein, calponin, as a specific marker for OECs. However, at the same time a recent report suggested that adult OECs do not express calponin. It was not clear if this discrepancy was due to methodology, as cells had to be treated with proteinase K to maximize calponin staining or developmental differences with only foetal/neonatal OECs expressing calponin. For this reason we have examined calponin expression in the peripheral olfactory system of embryonic and neonatal rats in vivo and from cells in vitro to assess if calponin is expressed in a developmental manner.  相似文献

Background  

Glutamate, a major excitatory amino acid neurotransmitter, causes apoptotic neuronal cell death at high concentrations. Our previous studies have shown that depending on the neuronal cell type, glutamate-induced apoptotic cell death was associated with regulation of genes such as Bcl-2, Bax, and/or caspase-3 and mitochondrial cytochrome c. To further delineate the intracellular mechanisms, we have investigated the role of calpain, an important calcium-dependent protease thought to be involved in apoptosis along with mitochondrial apoptosis inducing factor (AIF) and caspase-3 in primary cortical cells and a mouse hippocampal cell line HT22.  相似文献

Background  

Extracellular-signal regulated kinase (ERK1/2) is activated by nerve damage and its activation precedes survival and proliferation of Schwann cells. In contrast, activation of caspase 3, a cysteine protease, is considered as a marker for apoptosis in Schwann cells. In the present study, axonal outgrowth, activation of ERK1/2 by phosphorylation (p-ERK 1/2) and immunoreactivity of cleaved caspase 3 were examined after immediate, delayed, or no repair of transected rat sciatic nerves.  相似文献

With a Curie point at 370?K, the half-metal (La0.7Sr0.3)MnO3 (LSMO) is one of the most interesting candidates for electronic devices based on tunnel magnetoresistance. SrTiO3 (STO) is up to now the best substrate for the epitaxy of suitable thin films of LSMO. The pseudocubic unit cell of rhombohedral LSMO has a parameter a LSMO such that (a STO ? a LSMO)/a LSMO = +?0.83% (where a STO is the parameter of cubic STO) and an angle of 90.26°. As strained growth is tetragonal, relaxation implies recovery of both the pseudocubic parameter and of the original angle. In the LSMO layers that we prepare by pulsed-laser deposition, we show that these two processes are quite independent. The angular distortion is partially recovered by twinning in films 25?nm thick, while recovery of the parameter never occurs in the thickness range that we explored (up to 432?nm). A relaxation, however, takes place above a thickness of 100?nm, associated with a transition from two-dimensional to three-dimensional columnar growth. It is accompanied by chemical fluctuations. Our magnetic measurements exhibit Curie temperatures and magnetic moments very close to the bulk values in those layers where the crystal parameter is strained but the angle partially relaxed.  相似文献

The Pb(Mg1/3Nb2/3)O3 (PMN) relaxor system is used as an example to analyze the temperature dependences of the low-frequency dielectric permitivity (?′(T)) measured during zero-field heating (ZFH) from T = 10 K to T = 300 K after using different field cooling (FC) conditions. No changes in the temperature dependences of the permittivity have been detected during the transition from a nonergodic relaxor state (NERS) into an ergodic relaxor state (ERS) (at T f ≈ 216 K). However, the difference Δ?′(T) between the curves corresponding to different field cooling conditions in the same electric field has different shapes and different values below and above T → (T f + 9 K)? (for E dc = 1.52 kV/cm). The reduced permittivities ?′r(T, f) recorded under different cooling conditions are shown to change their behavior when passing through T = T f + 9 K. In NERS, these curves diverge: the stronger the field (0 ≤ E dc ≤ 3 kV/cm), the larger the divergence. In ERS, however, the ?′r(T, f) curves coincide under different cooling conditions irrespective of the field. The character of the changes in Δ?′(T) and ?′r (T, f) during the NERS-ERS transition is frequency-independent. The difference in the behavior of the dielectric response during ZFH after cooling in different (ZFC, FC) modes (even in a weak field), for both transition through T f and cooling down to T = 10 K, indicates different NERSs forming under these conditions. The contribution to ?′(T) from slowly relaxing regions (ω ~ 0.1 mHz), whose polarization is reoriented after the field is turned off, is responsible for the fact that, during the NERS-ERS transition, the ?′r(T, f) curves coincide at a temperature that is higher than T = T f.  相似文献

A systematic study of the magnetic and transport properties of the system (La1−xGdx)0.67Ca0.33MnO3 (0⩽x⩽1) is presented in this article. Through this series, a great crystallographic distortion is achieved but keeping constant the electronic density. At low Gd3+ content, these ions behaves in a paramagnetic state following a Brillouin function. Low-temperature ferromagnetism remains unchanged. However, for the samples with large Gd3+ content a complex magnetic behavior arises from several competing interaction as super-exchange, double-exchange and also the interaction between Gd and Mn ions. Resistivity and thermopower follow a progressive increase as Gd3+ content does, but the polaron energy in the paramagnetic range remains independent of all these changes. Thermopower infinite value is also determined and analyzed.  相似文献

We report an electron paramagnetic resonance (EPR) investigation of the spin dynamics in the paramagnetic regime of the colossal magnetoresistive manganites La2/3Ca1/3Mn1?x Me x O3 (Me=Al, In;x≤0.05). The temperature dependences of the EPR linewidth and integral intensity have been analyzed in terms of the bottleneck spin relaxation and small-polaron hopping models. The exchange coupling integral between Mn3+ and Mn4+ ions and the polaron activation energy decrease with increasing doping level. A discussion is given concerning the factors which could explain the observed changes.  相似文献

The formation mechanism of CH3O by the adsorption and decomposition of CH3OH on clean and oxygen-precovered Cu2O(1 1 1) surface has been investigated with density functional theory method together with the periodic slab models. Two possible formation pathways of CH3O by CH3OH decomposition on oxygen-precovered (Opre) Cu2O(1 1 1) surface were proposed and discussed. One is the O-H bond-cleavage of CH3OH with H migration to Opre to form CH3O; the other is the C-O bond-scission of CH3OH with CH3 migration to Opre leading to CH3Opre. The calculated results show that the O-H bond-breaking path has the lowest activation barrier 26.8 kJ mol−1, the presence of oxygen-precovered on Cu2O(1 1 1) surface exhibits a high surface reactivity toward the formation of CH3O by the O-H bond-cleavage of CH3OH, and reduce the activation barrier of O-H bond-cleavage. The C-O bond-breaking path was inhibited by dynamics, suggesting that the O atom of CH3O is not from the oxygen-precovered, but comes from the O of CH3OH. Meanwhile, the calculated results give a clear illustration about the formation mechanism of CH3O in the presence of oxygen and the role of oxygen at the microscopic level.  相似文献

Pr concentration dependence of the superconducting transition temperature Tc in the Ho1−xPrxBa2Cu3O7−δ system is determined from measurements of DC electrical resistance. This dependence coincides with that for the parallely studied Y1−xPrxBa2Cu3O7−δ reference system. Both systems have the same value of the critical concentration xc=0.58, in accordance with nearly equal ionic radii of Ho3+ and Y3+ ions. It has been shown that the Tc(x) curve can be described with a single mechanism based on a decreasing number of sheet holes trapped by PrIV-ions, if one takes also into account that the number of these ions changes with x.  相似文献

Strong phase competitions between the ferromagnetic metal and the charge-ordered-insulator have been induced in a wide temperature range of 10-256 K for the shear-strained La0.67Ca0.33MnO3/NdGaO3(0 0 1) films. Based on various magnetotransport measurements, the mobility of phase boundaries was inferred to change dramatically with temperature. In the high temperature range where the phase boundaries are movable, strong relaxation in resistivity was observed, while at the frozen temperatures lower than 40 K it is weakened. The resistivities tend to relax in accordance with the phase transitions driven by the temperature or magnetic field in the phase separation (PS) background. Moreover, to our surprise, while the melting fields of the insulating phase varied with film thicknesses, for a given film however, they stay unchanged when started with different phase fractions produced by the field or thermal cycling. The results show a crucial role of the inherent strain state in determining PS and phase competitions in these epitaxial thin films.  相似文献

The radiative lifetime of the 4s 5s (3S1) level in Ca was measured using two-step excitation of atoms in a beam which traverses a cold-cathode Penning discharge, where, by electron impact the atoms are excited to the metastable 4s 5p (3P2, 1, 0) states. Immediately thereafter, the beam is crossed with a pulse-modulated cw dye laser beam to populate the final state selectively. Using a pulse-modulation frequency of 0.5 MHz, a preliminary value of 12.4 ± 0.5 ns was obtained with a statistical error of 0.4%. The wide limits of systematic error reflect an assumed uncertainty in the excitation function and possible distortion in the fluorescence decay requiring further study.  相似文献

Results of an experiment to measure the 2P3/2?2S1/2 energy difference δ1 in the muonic (μ?4H e)+ ion are presented; we found δ1exp = 1.5274 ± 0.0009 eV. Comparison with QED predictions shows that there is agreement between theory and experiment within 0.0087 eV; this limitation is mainly due to the uncertainty with which the r.m.s. charge radius of the 4He nucleus is known.  相似文献

The electron paramagnetic resonance (EPR) spectra of mixed crystals (BaF2)1 − x (LaF3) x (x = 0, 0.001, 0.002, 0.005, 0.010, 0.020) doped with Ce3+ ions (0.1%) are investigated at a frequency v ≈ 9.5 GHz in magnetic fields up to 1.45 T at temperatures T = 10 and 15 K. The EPR spectrum of “pure” barium fluoride BaF2 (x = 0) is characterized by a single Ce3+-F center with tetragonal symmetry (i.e., the O center with g = 2.601 and g = 1.555). For a lanthanum trifluoride concentration x ≠ 0, the spectrum exhibits new lines due to the presence of the clusters containing Ce3+ and La3+ ions. The intensity of EPR signals from the O centers decreases rapidly as the lanthanum trifluoride concentration x increases. The lines attributed to a paramagnetic center with tetragonal symmetry and strongly anisotropic g factors (i.e., the K center with g = 0.725 and g = 2.52) are separated in the complex EPR spectrum with the use of the angular dependence of the EPR signal intensity measured for the samples with x ≥ 0.002.
