微波促进下2-氨基-3-氰基-4-芳基-4H- 吡喃化合物的一锅法合成

标题化合物可在微波辐射下以良好的产率经一锅法合成，为2-氨基-4H-吡哺类化合物的制备提供了一种快速便捷的有效方法。

Previously unknown 5-azolylpyrans were obtained by reactions ofN-acetonyl- andN-phenacylimidazoles, -triazoles, and -tetrazoles with arylmethylenemalononitriles. Regioselective Synthesis and Properties of 6-Amino-3-carbamoyl-5-cyano-3,4-dihydrospirocyclohexane-4-pyridine-2-thiol and 5-Cyano-3-thiocarbamoyl-4-spirocyclohexanepiperidine-2,6-dione

Treatment of monothiomalonodiamide with cyclohexylidenemalononitrile or cyclohexylidenecyanoacetic ester in the presence of sodium ethylate gave 6-amino-3-carbamoyl-5-cyano-3,4-dihydrospirocyclohexane-4-pyridine-2-thiol and 5-cyano-3-thiocarbamoyl-4-spirocyclohexanepiperidine-2,6-dione. Their alkylation and hydrolysis have been studied.

4-羟基-2-吡啶酮是天然生物碱结构母核,同时也是一类重要的医药中间体,该类衍生物具有抗真菌、抗菌和抗肿瘤等多种生物活性.因此,其合成得到了广泛的研究和关注.综述了近年来4-羟基-2-吡啶酮类化合物的合成方法及其研究进展.     

4-羟基-2-吡啶酮的合成

以氰乙酸乙酯和原乙酸三甲酯为原料,经缩合反应制得2-氰基-3-甲氧基丁烯-2-羧酸乙酯(1);1与二甲基甲酰胺缩二甲醇缩合得2-氰基-5-(二甲氨基)-3-甲氧基-2,4-戊二烯酸乙酯(2);2在80%醋酸中环合得4-甲氧基-2-吡啶酮-3-甲酸乙酯(3);3在HBr中脱甲基和脱羧合成了4-羟基-2-吡啶酮,总收率31%,其结构经1H NMR,IR和MS确证。     

Synthesis of 3-acetyl-2-amino-4-pyridone

A method hxsynthesizing 3-acetyl-2-amino-4-pyyridone has been suggested. The synthesis involves the condensation of dimethylformamide dimethylacetal at the NH₂ group of diphenylboron chelate diacetylketene aminal followed by its intramolecular cyclization under the action of NaOMe in MeOH.Translated fromIzvestiya Akademii Nauk. Three-component synthesis of 4-aryl-5-cyano-2-hetarylaminopyrimidines

Three-component condensation of benzooxazol-2-yl-, benzothiazol-2-yl-, and 5-ethoxycarbonyl-4-methyl-1,3-thiazol-2-ylguanidines with orthoesters and aroylacetonitriles afforded 4-aryl-5-cyano-2-hetarylaminopyrimidines. The Synthesis of 2-Alkylthio-3-alkyl-5-arylmethylidene4H-imidazol-4-ones

2-alkylthio-3-alkyl-5-arylmethylidene-4H-imidazol-4-ones were synthesized by the S-alkylation and N-alkylation of 2-thioxo-5-arylmethylidene-4-imidazolidinones, which were obtained via a tandem aza-Wittig reaction of vinyliminophosphoranes, carbon disulfide, and excess ammonium hydroxide (28% NH₃ in water).

Synthesis and Some Chemical Conversions of 2-([2,2]-5-Paracyclophanyl)pyrrole

2-([2,2]-5-Paracyclophanyl)pyrrole has been synthesized and nitration and formylation of it have been effected. The 1--cyanoethyl derivative and 2-formyl-5-paracyclophanyl-3H-pyrrolizine have been obtained from 5-formyl-2-([2,2]-5-paracyclophanyl)pyrrole by the action of acrylonitrile and acrolein respectively under Michael reaction conditions.     

Synthesis, Structure, and Some Properties of Substituted 3-Carbethoxy(methoxy)-5-cyano-1,2,3,4-tetrahydrospirocyclohexane-4-pyridine-2-thiones

The condensation of cyclohexylidenemalononitrile or cyclohexylidenecyanoacetic ester with thioamidoethyl(methyl)malonate in the presence of sodium ethylate gave 6-amino-3-carbethoxy-5-cyano-4-spirocyclohexane-1,2,3,4-tetrahydropyridine-2-thione and 3-carbethoxy(methoxy)-5-cyano-6-oxo-4-spirocyclohexanepiperidine-2-thiones which were used in the synthesis of the corresponding substituted 2-alkylthiotetrahydropyridines. 5-Carbethoxy-3-cyano-3-methyl-6-methylthio-4-spiro-cyclohexane-3,4-dihydropyridine-2(1H)-one was studied by X-ray crystallography.     

Efficient syntheses of ethyl 4-cyano-5-hydroxy-2-methyl-1H-pyrrole-3-carboxylate and ethyl (2Z)-(4-cyano-5-oxopyrrolidin-2-ylidene)ethanoate

Ethyl 2-chloroacetoacetate and its 4-chloro isomer react with cyanoacetamide in the presence of the mild, nonnucleophilic base, triethylamine under stoichiometric conditions to give high yields of ethyl 4-cyano-2-hydroxy-2-methyl-5-oxopyrrolidine-3-carboxylate and ethyl (4-cyano-2-hydroxy-5-oxopyrrolidin-2-yl)acetate, respectively. These, under acid-catalyzed dehydration conditions, afforded ethyl 4-cyano-5-hydroxy-2-methyl-1H-pyrrole-3-carboxylate and ethyl (2Z)-(4-cyano-5-oxopyrrolidin-2-ylidene)ethanoate, respectively. Similarly, the 4-chloro isomer reacted with ethyl cyanoacetate to give the novel product, diethyl 2-cyano-4-oxohexanedioate. The use of triethylamine enables access to a whole new library of pyrrole derivatives from easily accessible, commercially available starting materials. The reactions described in this Letter enable access to libraries of important pyrrole systems in any of the isotopically enriched forms.

Synthesis, properties, and structures of ammonium 4-aryl-5-cyano-2-oxo-1,2,3,4-tetrahydropyridine-6-thiolates

The reaction of arylcyanomethylenethiocetamides with Meldrum, acid gave Michael adducts as ammonium salts. When heated in alcohol, these salts undergo cyclization to ammonium 4-aryl-5-cyano-2-oxo-1,2,3,4-tetrahydropyridine-6-thiolates. The structure ofN-methylmorpholinium 4-(2′-chlorophenyl)-5-cyano-2-oxo-1,2,3,4-tetrahydropyridine-6-thiolate was established by X-ray structural analysis. Translated fromIzvestiya Akademii Nauk. 微波辅助合成5-亚甲基-2,3-二苯基-4-噻唑酮类衍生物

本文报道了一种通过微波辅助合成5-亚甲基-2,3-二苯基-4-噻唑酮类衍生物的新型方法。在以K2CO3 为碱、H2O 为溶剂、四丁基溴化铵（TBAB）为催化剂的条件下,2,3-二苯基-4-噻唑酮与取代醛在微波辅助下发生Knoevenagel 反应,生成5-亚甲基-2,3-二苯基-4-噻唑酮类衍生物,产率最高可达85%。     

One-step synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles. Molecular and crystal structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole

A one-step procedure was developed for the synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles involving the three-component reaction of 1-acetylindol-3(2H)-one with aromatic aldehydes and malononitrile in ethanol in the presence of triethylamine as the catalyst. The structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole was established by X-ray diffraction analysis.

Synthesis of 2-amino-4-aryl-3-cyano-5-hydroxyimino-4,5-dihydrothiophenes

Novel Stereoselective Synthesis and Molecular and Crystalline Structure of 3-Allyl-4-(4-bromophenyl)-3-cyano-6-oxopiperidine-2-thione

The stereoselective synthesis of 3-allyl-4-(4-bromophenyl)-3-cyano-6-oxopiperidine-2-thione has been carried out by refluxing N-methylmorpholinium 5-[1-(4-bromophenyl)-2-cyano-2-thiocarbamoylethyl]-2,2-dimethyl-1,3-dioxa-6-oxo-4-cyclohexen-4-olate with allyl bromide in ethanol. The structure of the product was confirmed by X-ray investigation.
