A series of 8-(substituted)aryloxycaffeine were prepared from 8-bromocaffeine and (substituted)phenols by modified Ullmann reaction. In vitro antibacterial activity, inhibitory activity on topoisomerase II and pharmacological activities were evaluated for the synthesized 8-(substituted)aryloxycaffeine. Among the synthesized compounds, 8-(5-chloropyridin-3-yloxy)caffeine (3k) showed strong inhibitory activity (MIC = 15.6 μg/mL) against the tested gram negative (−) bacteria Salmonella enteritidis. 8-(quinolin-8-yloxy)caffeine (3g) showed the strongest inhibitory activity against topoisomerase II. And the compounds 8-(6-methylpyridin-2-yloxy)caffeine (3j) and 8-(3-chloro-6-(trifluoromethyl)pyridin-2-yloxy)caffeine (3m) showed analgesic effect without the central nervous system stimulation.  相似文献

An efficient catalytic system of CuI/8‐hydroxyquinalidine was developed for the coupling of aryl iodides and indole as well as some azoles. The reaction could be carried out at 90°C under the condition of relatively low catalyst loading, affording various N‐arylindoles and N‐aryl azoles in good yields. Over bimetallic Au/Cu catalyst supported on magnetic Fe3O4 nanoparticles, water-mediated bromamine acid could be selectively converted into 4,4'-diamino-1,1'-dianthraquinonyl-3,3'-disulfonic acid (DAS) with a yield of 88.67%. The magnetic catalyst could be readily separated and reused.  相似文献

四环杂萜类天然产物Liphagal含有新奇的[6-7-5-6]四环骨架, 是磷酸肌醇3-激酶(PI3K)的有效抑制剂之一, 可以选择性抑制细胞增殖和促进细胞死亡, 在治疗炎症、免疫紊乱、癌症以及心血管疾病中有潜在的医用价值. 从廉价的2,4,5-三甲氧基苯甲醛出发, 通过Wittig反应、Cu催化的环化反应、选择性氢化、分子内Friedel-Crafts反应, 以直线步骤最长7步, 8.72%的总收率实现了8-epi-Liphagal高级中间体10的合成.  相似文献

A simple and efficient methodology for the synthesis of triphenylamines has been demonstrated using copper catalyst with a ligand and tripotassium phosphate as the base. The effect of parameters such as catalyst precursors, ligands, bases and solvents were studied and a series of triphenylamines were obtained with moderate to excellent yields in DMF. An enantioselective total synthesis of the polycyclic diterpene (+)‐chatancin, a potent PAF antagonist, is reported. Proceeding in seven steps from dihydrofarnesal, this synthetic route was designed to circumvent macrocyclization‐based strategies to complex, cyclized cembranoids. The described synthesis requires only six chromatographic purifications, is high yielding, and avoids protecting‐group manipulations. Total synthesis of cytotoxic marine alkaloids, lamellarins D, L, and N, has been achieved by using Hinsberg-type pyrrole synthesis and palladium-catalyzed Suzuki-Miyaura coupling of the 3,4-dihydroxypyrrole bistriflate 6 as the key reactions.
