SERS of C(60)/C(70) adsorbed on gold nanoparticles coated on filter paper or filter film was studied. As a new SERS substrate, dried gold-coated filter paper or filter film has a high SERS activity, whose enhancement factor can be up to about 10(5), because it avoided the influence of solvents in C(60)/C(70) solution and water in gold hydrosols. The influence of the gold thickness coated on filter paper or filter film to SERS of C(60)/C(70) adsorbed on gold nanoparticles was mainly discussed. It is indicated that the SERS effect of C(60)/C(70) was very sensitive to the distribution and aggregated characteristics of gold nanoparticles, and the SERS intensity of each mode increased at its own proportion, but it integrally tended to saturation when the thickness of colloidal gold coatings increased.  相似文献

This work is focused on studying the grafting of gold nanoparticles (Np) on a cystamine self-assembled monolayer on gold, in order to build sensitive immunosensors. The synthesis and deposition of gold nanoparticles, 13 and 55 nm sizes, were characterised by combining Polarisation Modulation Infrared Reflection-Absorption Spectroscopy (PM-IRRAS), X-ray Photoelectron Spectroscopy (XPS) Surface Enhanced Raman Scattering (SERS), and Atomic Force Microscopy (AFM) which all indicated the formation of a dispersed layer of nanoparticles. This observation is explained by the compromise between the high reactivity of amine-terminated layers towards gold, and interparticle repulsions. Nps were then functionalised with antibody probes, and the recognition by an anti-rIgG was assayed both on planar and Np gold surfaces.The important result is that nanoparticles of 55 nm are preferable for the following reasons: they enable to build a denser and well dispersed layer and they increase both the number of receptors (IgGs) and their accessibility. Beside these geometric improvements, a net enhancement of the Raman signal was observed on the 55 nm nanoparticle layer, making this new platform promising for optical detection based biosensors.  相似文献

Surface-enhanced Raman scattering (SERS) hybrid probes are characterized by the typical spectrum of a reporter molecule. In addition, they deliver information from their biological environment. Here, we report SERS hybrid probes generated by conjugating different reporter molecules to bovine serum albumin (BSA) and using gold nanoparticles as plasmonic core. Advantages of the BSA-conjugate hybrid nanoprobes over other SERS nanoprobes are a high biocompatibility, stabilization of the gold nanoparticles in the biological environment, stable reporter signals, and easy preparation. The coupling efficiencies of the BSA–reporter conjugates were determined by MALDI-TOF-MS. The conjugates’ characteristic SERS spectra differ from the spectra of unbound reporter molecules. This is a consequence of the covalent coupling, which leads to altered SERS enhancement and changes in the chemical structures of the reporter and of BSA. The application of the BSA–reporter conjugate hybrid probes in 3T3 cells, including duplex imaging, is demonstrated. Hierarchical cluster analysis and principal components analysis were applied for multivariate imaging using the SERS signatures of the incorporated SERS hybrid nanoprobes along with the spectral information from biomolecules in endosomal structures of cells. The results suggest more successful applications of the SERS hybrid probes in cellular imaging and other unordered high-density bioanalytical sensing.

Aggregation of gold nanoparticles of increasing size has been studied as a consequence of adsorption of 2-aminothiophenol (ATP) on gold nanoparticle surfaces. The capping property of ATP in the acidic pH range has been accounted from UV-vis absorption spectroscopy and surface-enhanced Raman scattering (SERS) studies. The effect of nanoparticle size (8-55 nm) on the nature of aggregation as well as the variation in the optical response due to variable degree of interparticle coupling effects among the gold particles have been critically examined. Various techniques such as transmission electron microscopy, X-ray diffraction, zeta-potential, and average particle size measurement were undertaken to characterize the nanoparticle aggregates. The aggregate size, interparticle distances, and absorption band wavelengths were found to be highly dependent on the pH of the medium and the concentration of the capping agent, ATP. The acquired SERS spectra of ATP relate the interparticle spacing. It has been observed that the SERS signal intensities are different for different sized gold nanoparticles.  相似文献

We report the use of two different sizes of dogbone shaped gold nanoparticles as colloidal substrates for surface enhanced Raman spectroscopy (SERS) based detection of ultra-low levels of thiram, a dithiocarbamate fungicide. We demonstrate the ability to use a solution based, direct readout SERS method as a quantitative tool for the detection of ultra-low levels of thiram. The two different sizes of dogbone shaped gold nanoparticles are synthesized by using the seed-mediated growth method and characterized by using UV-visible spectroscopy and transmission electron microscopy (TEM). The smaller dogbone shaped nanoparticles have an average size of 43 ± 13 nm. The larger dogbone shaped gold nanoparticles have an average size of 65 ± 15 nm. The nanoparticle concentration is 1.25 × 10(11) nanoparticles per mL for the smaller dogbone shaped gold nanoparticles and is 1.13 × 10(11) nanoparticles per mL for the larger dogbone shaped gold nanoparticles. Different concentrations of thiram are allowed to bind to the two different sizes of dogbone shaped gold nanoparticles and the SERS spectra are obtained. From the calibration curve, the limit of detection for thiram is 43.9 ± 6.2 nM when the smaller dogbone shaped gold nanoparticles are used as colloidal SERS substrates In the case of the larger dogbone shaped gold nanoparticles, the limit of detection for thiram is 11.8 ± 3.2 nM. The lower limit of detection obtained by using the larger dogbone shaped gold nanoparticles as colloidal substrates is due to the lightning rod effect, higher contributions from the electromagnetic enhancement effect, and larger number of surface sites for thiram to bind.  相似文献

A rapid and sensitive method was developed here for separation and detection of multiple pathogens in food matrix by magnetic surface-enhanced Raman scattering (SERS) nanoprobes. Silica-coated magnetic probes (MNPs@SiO2) of ∼100 nm in diameter were first prepared via the reverse microemulsion method using cetyltrimethylammonium bromide as a surfactant and tetraethyl orthosilicate as the silica precursor. The as-prepared MNPs@SiO2 were functionalized with specific pathogen antibodies to first capture threat agents directly from a food matrix followed by detection using an optical approach enabled by SERS. In this scheme, pathogens were first immuno-magnetically captured with MNPs@SiO2, and pathogen-specific SERS probes (gold nanoparticles integrated with a Raman reporter) were functionalized with corresponding antibodies to allow the formation of a sandwich assay to complete the sensor module for the detection of multiple pathogens in selected food matrices, just changing the kinds of Raman reporters on SERS probes. Here, up to two key pathogens, Salmonella enterica serovar Typhimurium and Staphylococcus aureus, were selected as a model to illustrate the probability of this scheme for multiple pathogens detection. The lowest cell concentration detected in spinach solution was 103 CFU/mL. A blind test conducted in peanut butter validated the limit of detection as 103 CFU/mL with high specificity, demonstrating the potential of this approach in complex matrices.  相似文献

This report describes the synthesis of magnetic NiFe@Au (i.e., NiFe core with Au shell) nanoparticles as functional spectroscopic probes. Both of the magnetic NiFe nanoparticles and its composite NiFe@Au particles were synthesized in aqueous solution. It is more analogous with the biological organism environment. The composite nanoparticles were dispersible in aqueous solution and could be directed by a magnetic field. Such NiFe@Au nanoparticles have been shown to function as magnetic and spectroscopic nanoprobes for surface enhanced Raman scattering (SERS) detection of molecules attached to the surface of the nanoparticles. It shows more potential functional SERS nanoprobes for biomolecular separation and detection.  相似文献

We report on the fabrication of a class of surface-enhanced Raman scattering(SERS)active thermometers,which consists of60 nm gold nanoparticles,encoded with Raman-active dyes,and a layer of thermoresponsive poly(N-isopropylacrylamide)(PNIPAM)brush with different chain lengths.These SERS-active nanoparticles can be optimized to maintain spectrally silent when staying as single particles in dispersion.Increasing temperature in a wide range from 25 to 55°C can reversibly induce the interparticle self-aggregation and turn on the SERS fingerprint signals with up to 58-fold of enhancement by taking advantage of the interparticle plasmonic coupling generated in the process of thermo-induced nanoparticles self-aggregation.Moreover,the most significative point is that these SERS probes could maintain their response to temperature and present all fingerprint signals in the presence of a colored complex.However,the UV-Vis spectra can distinguish the differences faintly and the solution color shows little change in such complex mixture.This proof-of-concept and Raman technique applied here allow for dynamic SERS platform for onsite temperature detection in a wide temperature range and offer unique advantages over other detection schemes.  相似文献

采用水合肼还原的方法制备了金银比例为1∶1的金银合金纳米粒子, 紫外可见吸收光谱显示合成的溶胶只有一个介于金和银之间的吸收峰, 证明了合金结构的形成. 通过氨基耦联方法将合金纳米粒子组装到硅片表面, 利用氯金酸与合金中银的反应对基底上合金纳米粒子表面进行了改性处理. 以吡啶为探针分子, 研究了表面处理前后基底的SERS效应的差别, 结果表明随着浸泡时间延长, 信号强度先逐渐增强后降低至不变, 这与合金纳米粒子表面结构的变化有关, 氯金酸与表面银的反应经历了两个过程, 即粒子表面形成小的孔洞(去合金过程)和AgCl(s)在粒子表面的沉积, 前者有利于SERS效应的提高, 而后者导致SERS效应快速衰减.  相似文献

Adsorption of 1,4-phenylenediisocyanide (PDI) has been studied for different-sized gold nanoparticles with mean diameters of 6, 14, 23, 40, 57, and 97 nm using UV-vis absorption spectroscopy and surface-enhanced Raman scattering (SERS). The SERS enhancement was found to be relatively weak for 6-nm particles due to less aggregation between PDI and gold particles. Concentration-dependent SERS spectra show that PDI was assumed to bridge two different gold particles at low concentrations of PDI, but as the concentration was increased, the bridge appeared to be broken, and PDI bonded to the gold particle only via one of its two isocyanide groups. For the 57- and 97-nm particles, however, the nu(NC)(free) stretching band in the SERS spectrum almost disappeared, even at a high bulk concentration of PDI, differently from the case of the smaller sizes (14, 23, and 40 nm). The 57- and 97-nm particles appeared to cross-link through the pendent isocyanide group even at a high bulk concentration. UV-vis absorption spectra indicated that PDI appeared to aggregate more extensively with increasing size in agreement with Raman data. Our result shows an example that the adsorption scheme of an aromatic diisocyanide may be varied depending on particle size as well as the bulk concentration.  相似文献

A simple and cost-effective chemical method was introduced to assemble gold (Au) nanoparticles on smooth silver (Ag) spheres for realizing surface-enhanced Raman scattering (SERS) enhancement by the replacement reaction between chloroauric acid and Ag spheres. In addition, the Ag-Au core-shell spheres were fabricated when a certain amount of chloroauric acid was used in the reaction solution. We found that the Ag particles decorated with small Au nanoparticles demonstrated the strongest SERS enhancement, while Ag-Au core-shell spheres showed the weakest enhancement.  相似文献

The paper proposes a simple and portable approach for the surface enhanced Raman scattering (SERS) spectroscopy in situ determination of carboxylated single walled carbon nanotubes (SWNTs) in river water samples. The method is based on the subsequent microfiltration of a bare gold nanoparticles solution and the water sample containing soluble carbon nanotubes by using a home-made filtration device with a small filtration diameter. An acetate cellulose membrane with a pore size of 0.2 μm first traps gold nanoparticles to form the SERS-active substrate and then concentrates the carbon nanotubes. The measured SERS intensity data were closely fit with a Langmuir isotherm. A portable Raman spectrometer was employed to measure SERS spectra, which enables in situ determination of SWNTs in river waters. The limit of detection was 10 μg L−1. The precision, for a 10 mg L−1 concentration of carbon nanotubes, is 1.19% intra-membrane and 10.5% inter-membrane.  相似文献

SERS active surfaces were prepared by depositing silver films using Tollen's reaction on to barium titanate beads. The SERS activity of the resulting surfaces was probed using two thiols (benzene thiol and 1,2-benzene dithiol) and rhodamine 6G. The intensity of the SERS signal for the three analytes was investigated as a function of silver deposition time. The results indicate that the SERS intensity increased with increasing thickness of the silver film until a maximum signal intensity was achieved; additional silver deposition resulted in a decrease in the SERS intensity for all of the studied molecules. SEM measurement of the Ag coated barium titanate beads, as a function of silver deposition time, indicate that maximum SERS intensity corresponded with the formation of atomic scale islands of silver nanoparticles. Complete silver coverage of the beads resulted in a decreased SERS signal and the most intense SERS signals were observed at deposition times of 30 min for the thiols and 20 min for rhodamine 6G.  相似文献

We report on silver–gold core-shell nanostructures that contain Methylene Blue (MB) at the gold–silver interface. They can be used as reporter molecules in surface-enhanced Raman scattering (SERS) labels. The labels are stable and have strong SERS activity. TEM imaging revealed that these nanoparticles display bright and dark stripe structures. In addition, these labels can act as probes that can be detected and imaged through the specific Raman signatures of the reporters. We show that such SERS probes can identify cellular structures due to enhanced Raman spectra of intrinsic cellular molecules measured in the local optical fields of the core-shell nanostructures. They also provide structural information on the cellular environment as demonstrated for these nanoparticles as new SERS-active and biocompatible substrates for imaging of live cells.

以对巯基苯甲酸为拉曼标记和自组装修饰分子, 在光亮金基底上修饰后作为检测基底, 在金纳米粒子表面修饰后获得具有表面增强拉曼光谱信号的标记金溶胶. 修饰的基底及纳米离子通过重金属离子与羧基端的配位而发生相互作用, 最终形成“金属基底-对巯基苯甲酸/重金属离子/对巯基苯甲酸-金属纳米颗粒”的三明治结构. 采用扫描电镜表征纳米粒子的组装及以表面增强拉曼光谱检测表面标记分子的信号, 以此实现重金属离子的检测. 以强螯合剂EDTA溶液淋洗三明治结构, 使重金属离子与金属基底以及纳米颗粒上的羧基的配位作用断裂, 获得可再次利用的修饰金基底.  相似文献

Saxitoxin is one of the most harmful paralytic shellfish toxins due to its high toxicity and adverse effects on the environment and human health. Aptasensors provide simple detection procedures because they have the advantages of chemical stability, easy synthesis and modification, and high convenience in signal transformation. Surface-enhanced Raman scattering (SERS) is an analytical technique that amplifies the analytical signals of molecules at extremely low concentrations, or even at the single molecule level, when the analyte is very close to rough metal surfaces or nanostructures. In this study, an SERS aptasensor is reported for the determination of saxitoxin for the first time. The optimized saxitoxin aptamer (M-30f) was modified on gold nanoparticles and served as the recognition element. Crystal violet was used as the Raman reporter without chemical bounding. The analytical principles of the aptasensor are that saxitoxin destabilized the conformations of the aptamer at high temperature conditions and altered the binding of crystal violet on the gold nanoparticles. In the presence of saxitoxin, the conformation of aptamer containing the G-quadruplex that selectively bound crystal violet unfolded to a large extent and hence the crystal violet molecules were released from gold nanoparticles with a reduced SERS signal. The effects of the gold nanoparticle size, the amount of DNA, aptamer density, sodium chloride concentration, and operation temperature upon the SERS determination were optimized. The resulting simple SERS aptasensor was developed with a satisfactory limit of detection (11.7?nM) and selectivity. The application for the analysis of real shellfish samples with simple procedures demonstrates that this SERS aptasensor is promising for on-site applications.  相似文献

A surface-enhanced Raman spectroscopy (SERS) study of imidazolium ionic liquid stabilized gold(0) nanoparticles (GNPs) furnished previously unknown knowledge about the coordination and stabilization mode of the imidazolium cation. GNPs were prepared by hydrazine reduction of a chloroauric acid solution in 1-triethylene glycol monomethyl ether-3-methylimidazolium methanesulfonate 2 as ether-functionalized room-temperature ionic liquid (RTIL). UV-vis spectroscopy showed the presence of GNP aggregates as absorptions extended to the NIR region. A parallel coordination mode for the imidazolium cation of RTIL 2 on the GNP surface was observed by SERS, which occurred without the simultaneous coordination of the 1-triethylene glycol monomethyl ether-functionality. Instead of this, the ether-functionality was directed away from the GNP surface and acted as steric barrier between the GNPs/GNP aggregates, thus preventing further aggregation. These new insights suggest that the imidazolium cation is responsible for electrosteric stabilization.  相似文献

In the investigation of chemical pollutants, such as PAHs (Polycyclic Aromatic Hydrocarbons) at low concentration in aqueous medium, Surface-Enhanced Raman Scattering (SERS) stands for an alternative to the inherent low cross-section of normal Raman scattering. Indeed, SERS is a very sensitive spectroscopic technique due to the excitation of the surface plasmon modes of the nanostructured metallic film. The surface of quartz substrates was coated with a hydrophobic film obtained by silanization and subsequently reacted with polystyrene (PS) beads coated with gold nanoparticles. The hydrophobic surface of the SERS substrates pre-concentrates non-polar molecules such as naphthalene. Under laser excitation, the SERS-active substrates allow the detection and the identification of the target molecules localized close to the gold nanoparticles. The morphology of the SERS substrates based on polystyrene beads surrounded by gold nanoparticles was characterized by scanning electron microscopy (SEM). Furthermore, the Raman fingerprint of the polystyrene stands for an internal spectral reference. To this extent, an innovative method to detect and to quantify organic molecules, as naphthalene in the range of 1 to 20 ppm, in aqueous media was carried out. Such SERS-active substrates tend towards an application as quantitative SERS sensors for the environmental analysis of naphthalene.  相似文献

We have developed a new class of surface-enhanced Raman scattering beacons (SERS beacons) that can be turned on and off by long-range plasmonic coupling, induced by biomolecular recognition and binding events. The beacons are based on colloidal gold nanocrystals in two sizes (40 and 60 nm) and are prepared by spectral encoding with a Raman reporter molecule, functionalized with thiolated DNA probes, and stabilized and protected by low molecular weight poly(ethylene glycol)s (PEGs). The results show the SERS signal intensities increase by 40-200-fold when the nanoparticle beacons are activated by plasmonic coupling, much higher than the bright-to-dark intensity ratios reported for traditional molecular beacons. Multivalent gold nanoparticles also have exquisite specificity and are able to recognize single-base mismatches or mutations. This class of SERS nanoparticle beacons has novel mechanisms for molecular detection and signal amplification, and its long-range coupling nature raises new opportunities in developing plasmonic probes to detect proteins, cells, and intact viruses.  相似文献

表面增强拉曼散射(SERS)被用于检测细菌芽孢中的一种重要的标志物吡啶2,6-二羧酸(DPA)。以聚乙烯吡咯烷酮(PVP)为粘合剂,将60 nm的金粒子组装到表面打磨光滑的金电极上,制备稳定、灵敏的SERS基底。通过不同pH值下吸附在金基底上的DPA的SERS特征,考察DPA分子吸附构型发生的变化,并分析酸根离子对其吸附的影响。结果表明:在强酸条件下,DPA在Au NPs/PVP/Au基底上的SERS信号能达到最大增强;当pH值大于DPA二级解离常数时,DPA的SERS特征逐渐减弱。在DPA中引入不同酸根盐时,后者会取代纳米金表面的柠檬酸根所占的部分位点,改变Au NPs-Au基底的SERS增强性能。3种酸根吸附性能不同,所以获得的光谱强度存在差异。  相似文献
