In this study, the simultaneous extraction of estrone (E1), 17β-estradiol (E2), estriol (E3), ethinylestradiol (EE2), and their glucuronated and sulfated metabolites in milk was optimized using solid-phase extraction (SPE). The aim of this research was to analyze estrogens and their conjugated metabolites by liquid chromatography with tandem mass spectrometry (LC–MS/MS) in a single run, without the need to perform enzymatic cleavage and derivatization. Two SPE cartridges in tandem were used, consisting of sorbents based on the hydrophilic–lipophilic balance and amine-functionalized packing materials. To monitor analyte loss at every step of the SPE procedure 14C-labeled E2 was spiked into the milk sample and the radioactivity was monitored at all stages of the SPE. In addition, non-radiolabeled standards of estrogens and metabolites were used to optimize solvent systems for the SPE and LC–MS/MS. The optimized method described in this paper can achieve recoveries ranging from 72% to 117% for the free estrogens (E1, E2, E3, and EE2), and 62% to 112% for seven conjugated metabolites. The three doubly conjugated, highly polar metabolites included in this study gave lower recoveries (≤43%) due to poor retention in SPE. Finally, commercial milk samples were analyzed for the presence of estrogens and their conjugated metabolites. Estrone (concentration range: 23–67 ng/L) was found to be the major free estrogen present in all milk samples. Estradiol was consistently observed in milk, but the concentrations were below the limit of detection (LOD of 10 ng/L), and no estriol and ethinylestradiol were detected. Several conjugated estrogen metabolites were identified, 17β-estradiol-3-glucuronide (71–289 ng/L), estrone-3-sulfate (60–240 ng/L), 17β-estradiol-3,17β-sulfate (<LOD to 30 ng/L), and estrone-3-glucuronide (<LOQ of 25 ng/L). An analytical method was developed for the determination in urine of 2 metabolites of diazinon: 6-methyl-2-(1-methylethyl)-4(1H)-pyrimidinone (G-27550) and 2-(1-hydroxy-1-methylethyl)-6-methyl-4(1H)-pyrimidinone (GS-31144). Two of the urine sample preparation procedures presented rely on gas chromatography/mass selective detection (GC/MSD) in the selected ion monitoring mode for determination of G-27550. For fast sample preparation and a limit of quantitation (LOQ) of 1.0 ppb, urine samples were purified by using ENV+ solid-phase extraction (SPE) columns. For analyte confirmation at an LOQ of 0.50 ppb, classical liquid/liquid partitioning was used before further purification in a silica SPE column. An SPE sample preparation procedure and liquid chromatography/electrospray ionization/mass spectrometry/mass spectrometry (LC/ESI/MS/MS) were used for both G-27550 and GS-31144. The limit of detection was 0.01 ng for G-27550 with GC/MSD, and 0.016 ng when LC/ESI/MS/MS was used for both G-27550 and GS-31144. The LOQ was 0.50 ppb for G-27550 when GC/MSD and the partitioning/SPE sample preparation procedure were used, and 1.0 ppb for the SPE only sample preparation procedure. The LOQ was 1.0 ppb for both analytes when LC/ESI/MS/MS was used.  相似文献

A solid-phase extraction (SPE)-gas chromatography (GC)-mass spectrometry (MS) analytical method for the simultaneous separation and determination of endocrine disrupting chemicals (EDCs) from water samples is described in detail. Important and contrasting EDCs including estrone, 17beta-estradiol, 17beta-ethynylestradiol, 16beta-hydroxyestrone, 4-nonylphenol, bisphenol A and 4-tert-octylphenol were selected as the target compounds. The SPE technique, followed by the derivatisation with bis (trimethylsilyl) trifluoroacetamide was used for the extraction recoveries of target compounds from water samples. A number of parameters that may affect the recovery of EDCs, such as the type of SPE cartridges, eluents, as well as water properties including pH value, and concentration of salts and humic substances were investigated. It is shown that the Oasis cartridges produced the best recoveries of target EDCs while ethyl acetate was efficient in eluting EDCs from SPE cartridges. The recovery of some EDCs was enhanced by the addition of salt, but reduced by the increase in pH value and humic acid concentration. The optimised method was further verified by performing spiking experiments in natural river water and seawater matrices, with good recovery and reproducibility for all the selected compounds. The established method was successfully applied to environmental water samples from East and West Sussex, UK, for the determination of the target EDCs.  相似文献

Various solid-phase extraction (SPE) procedures for clean-up, two perfluoroacylation reagents (pentafluoropropionic anhydride (PFPA) and heptafluorobutyric anhydride (HFBA)) and two chromatographic columns (HP-1701 and HP-5) have been assessed comparatively to achieve the determination of type B trichothecenes (deoxynivalenol (DON), nivalenol (NIV), 3- and 15-acetyldeoxynivalenol (3- and 15-ADON)) in wheat grain by gas chromatography (GC)-electron-capture detection (ECD). Spiked wheat samples were extracted with acetonitrile-water (84:16, v/v). Tested SPE procedures were MycoSep 225 column, Florisil and different cartridges prepared in the laboratory with mixtures of various sorbents like alumina, Celite 545, C18, silica and charcoal. We propose MycoSep 225 column, and cartridges made with alumina-charcoal-silica and alumina-charcoal-C18 silica mixtures as clean-up procedures on the basis of recovery values (89.6, 87.3 and 86.1% for deoxynivalenol, respectively, at 1.0 mg/kg spiking level). The two last procedures are less expensive. Pentafluoropropionic anhydride was more stable against moisture and less expensive, while recoveries were similar to those obtained with heptafluorobutyric anhydride. HP-1701 column can separate 3- and 15-acetyldeoxynivalenol derivatives while HP-5 cannot, although this last column provided lower bleed and better sensitivity.  相似文献

Several sample preparation methods have been assayed to analyze residues of fipronil in honey. After diluting the honey, liquid-liquid extraction (LLE) with organic solvents, and solid-phase extraction (SPE) on commercial cartridges or Florisil-packed column have been tested at three concentration levels: 0.01, 0.1 and 1 mg/kg. LLE with n-hexane or SPE on a Florisil column resulted to be the most suitable procedures to analyze fipronil at trace concentrations. Fipronil was quantified by gas chromatography (GC) with electron-capture detection (ECD) or mass spectrometric (MS) detection after conventional or matrix-matched calibration. The detection limit of fipronil in honey samples was below 1 microg/kg. The degradation of fipronil in honey has also been studied, being identified three minor degradation products in the extracts.  相似文献

The aim of this work is to establish a sensitive and reliable method for the analysis of the 16 priority Environmental Protection Agency-defined polycyclic aromatic hydrocarbons (PAHs) found in water samples. Gas chromatography (GC)-mass spectrometry (MS) and high-performance liquid chromatography (HPLC)-fluorescence detection (FLD)-UV techniques are optimized to obtain an adequate resolution of all compounds. Validation of the methods is carried out, and a good performance is observed for both techniques. The HPLC-FLD-UV technique is somewhat more sensitive than the GC-MS technique for the determination of PAHs; thus, the HPLC-FLD-UV method is used to follow up both the solid-phase extraction (SPE) analysis using cartridges and discs and the liquid-liquid extraction (LLE), which are also evaluated for the extraction of the PAHs. Low recoveries between 43% and 79% are obtained using SPE cartridges, and higher values are obtained using SPE discs (56-96%) and LLE (60-105%). Better results are obtained using the LLE technique, and, thus, analysis of real water samples is carried out using this technique. LODs between 0.6 and 21 ng/L and relative standard deviations less than 15% are obtained using a spiked water sample analyzed using the full LLE HPLC-FLD-UV method.  相似文献

Abstract

A method for the determination of toxaphene in whole blood using gas chromatography (GC)-electron capture negative ion ECNI/MS and pure polychlorobornane congerners has been evaluated. Blood samples were extracted with hexane/acetone (9:1), extracts defatted with sulfuric acid, analytes fractionated on Carbopack-C/ Florisil (or silica gel) SPE columns then concentrated to final volumes for GC-ECNI/MS analysis. Scientific evidence has shown an association between organochlorine compounds (OCC) exposure and human health hazards. Concerning this, OCC detection in human adipose samples has to be considered a public health priority. This study evaluated the efficacy of various solid‐phase extraction (SPE) and cleanup methods for OCC determination in human adipose tissue. Octadecylsilyl endcapped (C18‐E), benzenesulfonic acid modified silica cation exchanger (SA), poly(styrene‐divinylbenzene (EN) and EN/RP18 SPE sorbents were evaluated. The relative sample cleanup provided by these SPE columns was evaluated using gas chromatography with electron capture detection (GC–ECD). The C18‐E columns with strong homogenization were found to provide the most effective cleanup, removing the greatest amount of interfering substance, and simultaneously ensuring good analyte recoveries higher than 70%. Recoveries > 70% with standard deviations (SD) < 15% were obtained for all compounds under the selected conditions. Method detection limits were in the 0.003–0.009 mg/kg range. The positive samples were confirmed by gas chromatography coupled with tandem mass spectrometry (GC‐MS/MS). The highest percentage found of the OCC in real samples corresponded to HCB, o,p′‐DDT and methoxychlor, which were detected in 80 and 95% of samples analyzed respectively. The aim of the present study was to set‐up a Fast gas chromatography/mass spectrometry method for the analysis of cholesterol oxidation products (COPs). A silylated mixture of seven oxysterol standards was injected into a Fast GC/MS system. A capillary GC column (10 m×0.1 mm internal diameter×0.1 μm film thickness) coated with 95% dimethyl‐ and 5% diphenyl‐polysiloxane, was used. The method gave a fast (total analysis time=3.5 min) and satisfactory resolution (R>1.2) of the COPs standards, with a good repeatability and sensitivity, similar to those of conventional GC/MS; recoveries were tested on mice liver. Fast GC/MS method suitability for COPs analysis in food was also tested on an oxidized sardine fillet, which had been previously saponified and purified by NH2 solid‐phase extraction (SPE); a good repeatability and sensitivity was also obtained. The analytical performance of the Fast GC/MS method for the determination of COPs, together with the consequent significant reduction of the analysis time and consumables, demonstrates that Fast GC/MS represents a valid alternative to conventional GC/MS and evinces the great potential of such an analytical technique, which could be applied for both food and biological samples.  相似文献

A liquid chromatography-mass spectrometry (LC-MS/MS) method using hydrophilic interaction liquid chromatography (HILIC) was developed for the analysis of neurotoxins β-N-methylamino-L-alanine (BMAA) and 2,4-diaminobutyric acid (DAB), using multiple reaction monitoring (MRM) scan mode. Oasis-MCX and Strata-X-C polymeric cation-exchange cartridges were used to clean extracts of cyanobacterial cultures, including two strains of Microcystis aeruginosa and one strain of Nostoc sp. The performance of the solid-phase extraction (SPE) cartridges for BMAA and DAB were evaluated using mixed standards and spiked cyanobacterial extracts, which demonstrated recoveries of BMAA and DAB ranging from 66% to 91%. Matrix effects in LC-MS/MS were evaluated, and while there was no effect on BMAA quantitation, suppression of DAB was found. Full scan (Q1) and enhanced product ion (EPI) monitoring showed that the DAB suppression may be due to closely eluting compounds, including lysine, histidine, arginine and three other compounds with [M + H](+) m/z of 88, 164 and 191. The procedures developed allow the sensitive and effective analysis of trace BMAA and DAB levels in cyanobacteria. While DAB was confirmed to be present, no BMAA was found in the cyanobacterial samples tested in the present study.  相似文献

Summary Comparative results of pre-treatment methods for the extracts of polycyclic aromatic hydrocarbons (PAHs) from airborne particulate matter for quantitative determination by gas chromatography (GC) are presented. The first method included liquid-liquid extraction and column liquid adsorption chromatography. In the second procedure the extract was fractionated by liquid-liquid chromatography and liquid adsorption chromatography. In the last procedure two different solid phase extraction (SPE) cartridges examined makes it possible to separate PAHs and remove paraffinic compounds which is very important for the quantitative GC analysis of the PAHs. Recoveries of PAH standards and some their derivatives were determined and the contents of 15 of the most important PAHs, were compared. Summary The re-usability of C18 solid-phase extraction (SPE) cartidges was assessed utilizing two different analytical procedures developed for the analysis of erythromycin and josamycin in human serum and urine. A statistical procedure using confidence intervals was employed in order to determine a 10% change in drug recovery on re-use with a 95% degree of certainty. The results obtained indicated that the SPE cartridges could be successfully re-used up to three times for serum and urine samples containing physiological concentrations of erythromycin base and propionate. However, in the case of josamycin, results were inconsistent after the second re-use of the extraction cartridges for serum samples. Reproducible results, however, were still obtained for urine samples using the same SPE cartridges up to four times. The results indicate that although succesful re-use of SPE cartridges is possible, each drug and associated extraction conditions need to be carefully assessed prior to implementing such re-use.  相似文献

建立了基于通过型固相萃取小柱净化的超高效液相色谱-三重四极杆质谱联用(UPLC-MS/MS)同时快速准确测定牛蛙中9种雌激素(雌三醇(E3)、17β-雌二醇(β-E)、17α-雌二醇(α-E)、17α-炔二雌醇(EE2)、雌酮(EI)、己烯雌酚(DES)、己二烯雌酚(DE)、己烷雌酚(HEX)、醋酸双烯雌酚(DD))残留的检测方法。样品经乙腈提取,经PRiME HLB固相萃取柱净化,Waters Acquity UPLC BEH C_(18)柱(100 mm×2.1 mm,1.7μm)分离,以0.5 mmol/L氟化铵水溶液-乙腈体系为流动相梯度洗脱,流速为0.3 mL/min,采用电喷雾正负离子切换模式(ESI^(+)/ESI^(-))和多反应监测(MRM)扫描方式检测,基质匹配外标法定量分析。该研究优化了液相色谱条件,相比于乙酸铵水溶液-乙腈体系和氨水溶液-乙腈体系,0.5 mmol/L氟化铵水溶液-乙腈体系作为流动相时9种雌激素普遍具有更佳的灵敏度。相比于甲醇和乙酸乙酯,乙腈作为提取溶剂时9种雌激素的提取率提高15%~40%。考察了HLB、C_(18)、Silica、PRiME HLB共4种不同类型的固相萃取小柱的基质净化效应,结果表明,PRiME HLB柱具有更好的基质净化能力。经PRiME HLB净化后,所有化合物的回收率均在70%~125%之间。DD的回收率从47%提高到74%,DES的回收率从180%降低到123%,有效减弱了基质效应。在最佳的实验条件下,E3、β-E、α-E、EI、DE、HEX、DD的线性范围为0.5~100.0μg/L,EE2和DES的线性范围为1.0~100.0μg/L,9种雌激素在各自的线性范围内均有良好的线性关系,相关系数为0.9953~0.9994,方法检出限为0.17~0.33μg/kg,方法定量限为0.5~1.0μg/kg,在2.0、10.0、80.0μg/kg 3个加标水平下,9种雌激素的加标回收率为65.1%~128.2%,相对标准偏差为1.9%~17.6%。该方法操作简便、快速、灵敏,重复性好,可用于大批量样品的同时快速准确检测。  相似文献

An integrated gas chromatography/Fourier transform infrared spectrometry (GC/FT-IR) system developed for the analysis of environmental pollutants is described. The versatility of the system allows the utilization of many different techniques of sample introduction and manipulation during analysis. The sample can be introduced by direct injection or thermal desorption from an adsorbent cartridge, and can then be separated on one of two capillary columns and detected by FT-IR or an FID. Cold traps and collection cartridges incorporated in the system permit recovery and additional fractionation of samples. Recovered sample and sample fractions can then be re-analyzed by GC/FT-IR or subsequently analyzed by GC/MS or other methods.  相似文献

A sensitive and reliable method has been developed and validated for the determination of chloramphenicol in poultry and swine liver using SPE and ultra-performance liquid chromatography (UPLC)/MS/MS. The liver samples were extracted with ethyl acetate, defatted with n-hexane, and further cleaned up using SPE cartridges with polymeric sorbent. An Acquity BEH C18 column was used for gradient UPLC separation, with water and acetonitrile as the mobile phase. The multiple reaction monitoring mode was used for two precursor-product ion transitions for chloramphenicol and one for the internal standard. The method was validated at 0.1, 0.3, and 1.0 microg/kg. Mean recoveries from fortified samples ranged from 95.5 to 106.7% with an RSD of 12.2%. The method LOD was < 0.02 microg/kg.  相似文献

Environmental analytical chemistry has recently changed focus from analysis of non-polar, persistent contaminants (e.g. polychlorinated biphenyls and dioxins) to more polar and labile compounds that interfere with biological processes. For example, natural and synthetic estrogens and their metabolites have been detected in sewage treatment plant effluents at nanogram/liter concentrations that are similar to those at which both total sex reversal and intersex (containing both testes and ova) is induced in fish exposed to these compounds in laboratory experiments. The development of techniques for the analysis of natural and synthetic estrogens in biological fluids (i.e. serum and urine) has been a priority in the biomedical field. However, the recent recognition that estrogen hormones are contaminants in the environment that may contribute to endocrine disruption has focused attention on the need for highly sensitive and specific techniques that are applicable for trace analysis in complex environmental matrices. Three optimized mass spectrometric protocols have been developed for the determination and quantitation of steroid hormones in environmental matrices using gas chromatography/tandem mass spectrometry (GC/MS/MS), liquid chromatography/mass spectrometry selected ion monitoring, (LC/MS - SIM) and liquid chromatography/tandem mass spectrometry (LC/MS/MS). A facile and sensitive method utilizing capillary gas chromatography with nitrogen phosphorus detection (GC–NPD) has been developed and validated for simultaneous analysis of hexaconazole, myclobutanil, and tebuconazole, three broad-spectrum systemic fungicides, in apples and soil. Two samples were fortified with the three pesticides and subjected to ultrasonic extraction, followed by solid-phase extraction (SPE) to remove coextractives, before analysis by GC–NPD. SPE procedures were performed on PSA cartridges (500 mg, 3 mL), the analytes being eluted with n-hexane–acetone (9:1 v/v, 2 mL). Recovery of three pesticides from the fortified apple and soil samples ranged from 94.5 to 107.3% with relative standard deviations less than 9.7% at the three spike levels (0.01, 0.1, and 0.5 mg kg?1). Limits of quantification of the method for apple and soil were 0.01 mg kg?1, sufficiently below the maximum residue limits. Direct confirmation of the analytes in samples was achieved by gas chromatography–mass spectrometry (GC–MS).  相似文献

In this work, we have compared the selectivity of two commercial molecularly imprinted polymers (AFFINIMIP®SPE Estrogens and AFFINIMIP®SPE Zearalenone) for the extraction of 12 estrogenic compounds of interest (i.e. 17α‐estradiol, 17β‐estradiol, estrone, hexestrol, 17α‐ethynylestradiol, diethylstibestrol, dienestrol, zearalenone, α‐zearalanol, β‐zearalanol, α‐zearalenol and β‐zearalenol) from different water samples. High‐performance liquid chromatography coupled with ion trap mass spectrometry with electrospray ionization was used for their determination. Results showed that although both molecularly imprinted polymeric cartridges were specifically designed for different groups of analytes (natural estrogens like estradiol in the first case and zearalenone derivatives in the second) they nearly have the same extraction performance (with recovery values in the range 65–101%) for the same analytes in Milli‐Q water because of the cross‐reactivity of the polymer. However, when more complex water samples were analyzed, it was clear that the behavior was different and that the AFFINIMIP®SPE Estrogens showed less cross‐reactivity than the other cartridge. Validation of the proposed methodology with both cartridges revealed that the extraction was reproducible and that the final limits of detection of the proposed method were in the low ng/L range.  相似文献
