A comparison of rigorous and approximate models for the build-up of epoxy-amine networks with simultaneous etherification is presented. Contrary to a recent publication, it is shown that naïve statistical models perform very well in predicting statistical parameters when the different directions of network growth are distinguished in the statement of the recursive procedure. Slightly lower values of gel conversions are predicted by the approximate model, a fact that is consistent with the wider distribution of chain lengths resulting from the random recombination procedure. © 1993 John Wiley & Sons, Inc.  相似文献

Zidovudine (azidothymidine, AZT), a potent antiviral agent acting on acquired immunodeficiency syndrome virus, was examined with regard to permeation through rat and human skin. A steady state plasma concentration of AZT after transdermal application in rats estimated from both pharmacokinetics data after i.v. administration and penetration rate through excised rat skin from 10% oleic acid (OA) aqueous solution shows penetration about 85 times higher compared to that from 10% OA would be needed for therapeutic efficacy. A mixed-solvent system consisted of 5% Sefsol-318 (S-318), 10% OA, 10% N-methyl-2-pyrrolidone (MP), 20% propylene glycol (PG) and water showed promising characteristics as a vehicle in terms of permeability of AZT through excised rat skin. The maximum flux of 0.41 mumol/cm2/h was observed in excised human skin after application of a gel formulation including S-318, OA, MP and PG. The result suggests a possible use of the gel formulation to gain an effective plasma concentration in humans.  相似文献

A potential method for the discrimination and prediction of honey samples of various botanical origins was developed based on the non‐targeted volatile profiles obtained by solid‐phase microextraction with gas chromatography and mass spectrometry combined with chemometrics. The blind analysis of non‐targeted volatile profiles was carried out using solid‐phase microextraction with gas chromatography and mass spectrometry for 87 authentic honey samples from four botanical origins (acacia, linden, vitex, and rape). The number of variables was reduced from 2734 to 70 by using a series of filters. Based on the optimized 70 variables, 79.12% of the variance was explained by the first four principal components. Partial least squares discriminant analysis, naïve Bayes analysis, and back‐propagation artificial neural network were used to develop the classification and prediction models. The 100% accuracy revealed a perfect classification of the botanical origins. In addition, the reliability and practicability of the models were validated by an independent set of additional 20 authentic honey samples. All 20 samples were accurately classified. The confidence measures indicated that the performance of the naïve Bayes model was better than the other two models. Finally, the characteristic volatile compounds of linden honey were tentatively identified. The proposed method is reliable and accurate for the classification of honey of various botanical origins.  相似文献

The Science Wars have pitted defenders of science against those accused of attacking it with the weapons of constructivism and relativism. I argue that this defensive stance is in large part a consequence of two other -isms, organized skepticism and naïve methodism, that play a significant, if mostly unconscious, role in how scientists tend to think about science, and suggest that increased awareness of these -isms may help dissipate the perceptions of hostility.  相似文献

Summary: In this work, the use of a temperature-sensitive polymer gel, poly(N-isopropylacrylamide), for the concentration of whey proteins was studied. The studied variables were: gel mass/solution volume ratio and concentration temperature. The concentration percentage and the selectivity were determined. The gel 20 × 5 (20% w/w total monomer/solution and 5% w/w crosslinking agent/total monomer), contacted with whey proteins solutions, at 5 °C and at 20 °C, was capable of concentrating the solution, in protein, from 10 to 33%, depending on the gel mass/solution volume ratio. The separation efficiencies, for the different studied systems, varied from around 40 to 80%. The results were discussed in the context of gels thermodynamics and through correlations between synthesis parameters and structure of the obtained gels. The obtained results for the concentration of whey proteins solutions, by using temperature-sensitive polymer gel, poly(N-isopropylacrylamide), showed that the Gel Process can indeed be used as an advantageous alternative for such separation, either from an economic or from an environmental view point.  相似文献

Poly(ethylene terephthalate) (PET) was rapidly crystallized through thermoreversible gelation in a liquid ethylene glycol oligomer or in epoxy resin. The solutions formed gel rapidly on cooling. Polarized light microscopy and small-angle light scattering showed that these gels contain large, regular PET spherulites. The gels may be formed by two consecutive processes: the phase separation and crystallization, and gelation by formation of a three-dimensional PET network in the oligomer solvents, where the nodes of the network are PET spherulites. The crystallinity of PET recovered from polymer/oligomer gels is near 72% measured by wide-angle X-ray diffraction method, which is about 20% higher than PET samples crystallized by solution crystallization in small molecule solvent, high temperature annealing, and stretching techniques. It takes only a few minutes to form the highly crystalline phase PET in the PET/oligomer system, and the crystallinity of the dried gel is independent of the concentration of the original solution. Excimer-fluoresence and Raman spectroscopic studies indicated that PET recovered from the gels are in an ordered state with few chain entanglements. The entanglement density of the recovered PET recovered from a 20 wt % solution in ethylene glycol oligomer is as low as that of freeze-extracted PET from a 0.5 wt % solution in phenol. © 1998 John Wiley & Sons, Inc. A lot of population data of 30 deletion/insertion polymorphisms (DIPs) of the Investigator DIPplex kit in different continental populations have been reported. Here, we assessed genetic distributions of these 30 DIPs in different continental populations to pinpoint candidate ancestry informative DIPs. Besides, the effectiveness of machine learning methods for ancestry analysis was explored. Pairwise informativeness (In) values of 30 DIPs revealed that six loci displayed relatively high In values (>0.1) among different continental populations. Besides, more loci showed high population-specific divergence (PSD) values in African population. Based on the pairwise In and PSD values of 30 DIPs, 17 DIPs in the Investigator DIPplex kit were selected to ancestry analyses of African, European, and East Asian populations. Even though 30 DIPs provided better ancestry resolution of these continental populations based on the results of PCA and population genetic structure, we found that 17 DIPs could also distinguish these continental populations. More importantly, these 17 DIPs possessed more balanced cumulative PSD distributions in these populations. Six machine learning methods were used to perform ancestry analyses of these continental populations based on 17 DIPs. Obtained results revealed that naïve Bayes manifested the greatest performance; whereas, k nearest neighbor showed relatively low performance. To sum up, these machine learning methods, especially for naïve Bayes, could be used as the valuable tool for ancestry analysis.  相似文献

Polyacrylonitrile (PAN) is soluble in dimethyl sulfoxide (DMSO) and the resulting solution can gel by various mechanisms. The effects of temperature and water on the thermoreversible gelation of PAN have attracted much attention because of their importance in the fiber formation and film casting. Rheological tests were employed in this study to examine the gelation behavior and determine the gel point temperature of PAN-DMSO solution. Calorimetric studies of PAN-DMSO gels were conducted through Differential Scanning Calorimetry and no crystallization was discovered in these gels prepared at low temperature. X-ray diffraction of different PAN gels indicated that in the absence of water, gels resulted from PAN-DMSO solution by decreasing temperature were not crystallizable. In contrast, water-induced gelation led to crystallization of PAN gel. The water content in the formed gel is responsible for its crystallinity and average crystallite size.  相似文献

Cellulose gels were prepared from cellulose in lithium chloride/N,N-dimethylacetamide (LiCl/DMAc) solution. When the cellulose concentration in the solution is above the one at which cellulose molecules overlap, cellulose gels were formed. While the gel prepared by the addition of water was turbid, the one prepared by the ion exchange was colorless, transparent, and optically anisotropic. In order to explain this gelation behavior of cellulose, small-angle X-ray scattering (SAXS) measurements of the cellulose solutions and the gels were performed. The SAXS profiles of the cellulose solutions and the gels suggested that the large-scale fluctuation of the molecular chain density in the solution can be the origin of the molecular aggregates formed in the gel. Furthermore, the differences in the structure of the gels at the macroscopic and the molecular level were discussed in terms of the phase separation and the molecular association.  相似文献

GalactosylatedN-3-guanidinopropylmethacrylamide-co-poly (ethylene glycol) methacrylate copolymers (galactosylated GPMA-co-PEGMA, GGP) were developed in order to promote transfection efficiency in the presence of serum in this report. First of all, the galactosylated PEGMA-co-GPMA copolymers were prepared via aqueous reversible addition – fragmentation chain transfer polymerization (RAFT) of poly (ethylene glycol) methacrylate (PEGMA) with long circulating chain segment and N-3-aminopropyl methacrylamide (APMA) followed by galactosylation and guanidinylation. After that, GGP/plasmid DNA complexes were examined by a dynamic light scattering and gel electrophoresis. It is showed that GGP copolymers have effective condensing ability. The cytotoxicity of GGP was measured by MTT assay. It was found that all the GGP/plasmid DNA complexes had less cytotoxic effects on HepG2 cells than HeLa cells, and the galactose groups reduced the cytotoxicity of complexes with high charge ratios to HepG2 cells. Finally, the transfection efficiency of the galactosylated PEGMA-co-GPMA copolymers was investigated by luciferase expression assay. The results revealed that the copolymers with galactose groups more than 5.83% could induce the asialoglycoprotein (ASGP) receptor mediated transfection, which improved the transfection efficiency in target cells. The GPMA-co-PEGMA copolymers with 54.57% hydrophilic chain segment PEG should prevent the aggregation of protein on the GGP/pDNA complexes, and GGP with 7.94% galactose graft exhibited the highest transfection in the presence of serum.  相似文献

The characteristics of protein detection and quantitation with SYPRO Ruby protein gel stain in one- and two-dimensional polyacrylamide gels were evaluated. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) analyses of three different purified recombinant proteins showed that the limits of detection were comparable to the limits of detection with ammoniacal silver staining and were protein-specific, ranging from 0.5 to 5 ng. The linearity of the relationship between protein level and SYPRO Ruby staining intensity also depended on the individual protein, with observed linear dynamic ranges of 200-, 500-, and, 1000-fold for proteins analyzed by SDS-PAGE. SYPRO Ruby protein gel stain was also evaluated in two-dimensional electrophoretic (2-DE) analysis of Escherichia coli proteins. The experiment involved analysis of replicates of the same sample as well as dilution of the sample from 0.5 to 50 nug total protein across gels. In addition to validating the 2-DE system itself, the experiment was used to evaluate three different image analysis programs: Z3 (Compugen), Progenesis (Nonlinear Dynamics), and PDQuest (Bio-Rad). In each program, we analyzed the 2-DE images with respect to sensitivity and reproducibility of overall protein spot detection, as well as linearity of response for 20 representative proteins of different molecular weights and pI. Across all three programs, coefficients of variation (CV) in total number of spots detected among replicate gels ranged from 4 to 11%. For the 20 representative proteins, spot quantitation was also comparable with CVs for gel-to-gel reproducibility ranging from 3 to 33%. Using Progenesis and PDQuest, a 1000-fold linear dynamic range of SYPRO Ruby was demonstrated with a single known protein. These two programs were more suitable than Z3 for examining individual protein spot quantity across a series of gels and gave comparable results.  相似文献

The performance of commercial and enzymatically modified size-exclusion (SE) gels in electrochromatography was compared for preparative protein separations. Dextran and agarose-based SE gels were subjected to enzymatic digestion under mild conditions. This treatment partially hydrolyzed the gel matrix modifying its pore size distribution. Enzymatic treatment of agarose-based SE gels was found to increase the resolution of the separation. Successful separation of preparative amounts of the A and B forms of beta-lactoglobulin (difference in electrophoretic mobility of 8.5%) was achieved with a high degree of purity using agarose-based SE gels. The four major whey proteins, beta-lactoglobulin, alpha-lactalbumin, BSA and immunoglobulins, were purified from an acid whey preparation. The degree of retention of a protein in electrochromatography followed their free-solution electrophoretic mobility (mu) when the protein was able to enter the gel pores and the ratio of diffusion/mu when the protein was excluded.  相似文献

A method is described for the preparation of rehydratable agarose gels, with specific application to the direct incorporation of 9 M urea and carrier ampholytes into rehydratable agarose gels for use in isoelectric focusing. After drying the agarose gel containing an uncharged linear polyacrylamide, one gel volume of a 9 M urea-carrier ampholyte solution is absorbed directly into the gel in 60 min, eliminating equilibration or dialysis of the gel in larger volumes of the 9 M urea-carrier ampholyte solution. Proteins with a molecular mass of 970,000 Da can be separated by isoelectric focusing in these rehydratable gels. The focused proteins can then be quantitatively transferred to nitrocellulose in less than 10 min, and any immunostaining procedure can be used to probe the blotted proteins. These agarose gels are easy to make, they rehydrate rapidly and they can be used in applications other than isoelectric focusing.  相似文献

We report on the microstructure and mechanical properties (elastic modulus) of concentrated depletion and thermal gels of octadecyl-coated silica particles for different values of the strength of interaction--polymer concentration for depletion gels and temperature for thermal gels. The depletion gels are composed of dense clusters and voids, while the thermal gels are devoid of clusters. Shear breaks up clusters in depletion gels while it induces clustering in the thermal gels. In both of these gels, the microstructure recovers to the presheared state upon cessation of shear. The recovery of the elastic modulus mimics the microstructure in the sense that the elastic modulus recovers to the presheared sheared state after shearing is stopped. Calculations of the gel boundary by modeling the interactions with an effective one-component square-well model reveals that suspensions with similar ranges of attraction gel at the same volume fraction at a fixed strength of attraction. Calculations of the elastic modulus using the na?ve mode coupling theory for depletion gels are in good agreement with experimental measurements provided clustering is taken into account and have the same magnitude as the elastic moduli of thermal gels with similar strengths of attraction. These calculations, in addition to the experimental observations reinforce the point that the microscopic parameter determining the elastic modulus of dense gels and its recovery is the localization length which is only a fraction of the particle diameter and not the structure on the length scale of the particle diameter and larger.  相似文献

以丙烯酸(AA)、甲基丙烯酸十八酯(OMA)、十二烷基硫酸钠(SDS)为原料,采用胶束共聚的方法合成了疏水缔合(HA)凝胶.在HA凝胶内部,表面活性剂SDS与疏水单体OMA组成的增溶胶束起到物理交联作用,将亲水的聚合物链交联起来.通过单向拉伸试验证实了该凝胶具有较高的机械性能.此外,也测试了HA凝胶在不同pH值溶液中的溶胀行为.结果显示,HA凝胶具有特殊的溶胀行为,其溶胀过程可以分为凝胶溶蚀、溶胀平衡和凝胶瓦解3个阶段.在强酸性条件下,凝胶的溶胀被抑制,没有出现凝胶瓦解阶段.在强碱性条件下,凝胶的溶胀被促进,溶胀平衡阶段被越过.盐的存在也会抑制HA凝胶的溶胀,但在SDS溶液中,溶液中的SDS会促使凝胶中的疏水改性聚合物溶解到溶液中去,组成新的缔合结构,而使溶液增稠。  相似文献

In 1996, we first reported self‐oscillating polymer gels exhibiting autonomous swelling‐deswelling oscillations driven by the Belousov‐Zhabotinsky reaction. In contrast to conventional stimuli‐responsive gels, the self‐oscillating gel can autonomously and periodically change its volume in a closed solution without any external stimuli. Since the first report, the novel concept of self‐oscillating gels has been expanded into various polymer and gel systems. One persistent problem with using therapeutic concentrations of glycyrrhizin (GZ) is that, at these high concentrations, it forms a gel in an aqueous solution. We previously solved this problem by dissolving GZ in a highly concentrated phosphate buffer. Unfortunately, the resulting GZ solution has a hyperosmotic pressure that renders it unsuitable for use in patients. The aim of this study was to prepare a highly concentrated GZ solution having an osmotic pressure ratio of 1 and a pH of 7.4. By adding small amounts of oil and using a 100 mM phosphate buffer, we achieved an emulsified GZ solution that is stable at room temperature and has a physiological osmotic pressure and pH. When clove oil was used as an emulsifier, the gel formation temperature of GZ solution decreased appreciably compared to that of GZ solution without clove oil. Using scanning electron microscopy (SEM), we examined the detailed characteristics of GZ gels prepared from solutions with or without clove oil. SEM of cross sections of GZ gels revealed an irregular structure in gels prepared with clove oil, indicating that clove oil prevented the formation of the intermolecular GZ networks typically characterized by gels derived from pure GZ solutions.  相似文献
