Upon collision‐induced activation, gaseous sodium adducts generated by electrospray ionization of disodium salts of 1,2‐ 1,3‐, and 1,4‐benzene dicarboxylic acids (m/z 233) undergo an unprecedented expulsion of CO2 by a rearrangement process to produce an ion of m/z 189 in which all three sodium atoms are retained. When isolated in a collision cell of a tandem‐in‐space mass spectrometer, and subjected to collision‐induced dissociation (CID), only the m/z 189 ions derived from the meta and para isomers underwent a further CO2 loss to produce a peak at m/z 145 for a sodiated arene of formula (Na3C6H4)+. This previously unreported m/z 145 ion, which is useful to differentiate meta and para benzene dicarboxylates from their ortho isomer, is in fact the sodium adduct of phenelenedisodium. Moreover, the m/z 189 ion from all three isomers readily expelled a sodium radical to produce a peak at m/z 166 for a radical cation [(?C6H4CO2Na2)+], which then eliminated CO2 to produce a peak at m/z 122 for the distonic cation (?C6H4Na2)+. A simple and sensitive liquid chromatography tandem multiple‐stage mass spectrometry (HPLC/MS/MS) method suitable for bulk lisinopril analysis was developed, by which lisinopril and its RSS isomer were separated and differentiated. In the collision‐induced dissociation (CID) mass spectra of the [M + H]+ ions, the abundance of the fragment ion of m/z 246 for lisinopril was about two times higher than the ion of m/z 245; however, the former fragment ion was noted to be a little lower than the latter for RSS isomer at all collision energies. In the CID mass spectra of the [M + Li]+ ion, the abundance of the rearrangement ion of m/z 315 for the RSS isomer was about three times higher than that for lisinopril. Furthermore, the difference was supported by the results of energy‐resolved mass spectrometry (ERMS) in the test range of collision energies. Similar differences were also observed between the CID mass spectra of lisinopril and RSS isomer methylester, which indicated that the RSS isomer could be rapidly characterized by the CID mass spectra of both the protonated and lithium adduct ion. Elemental compositions of all the ions were confirmed by Fourier Transform ion cyclotron resonance ESI mass spectrometry (FT‐ICR‐ESI/MS). Incorporation of deuterium from deuterium oxide (2H2O) into biological components is a commonly used approach in metabolic studies. Determining the dilution of deuterium in the body water (BW) pool can be used to estimate body composition. We describe three sensitive GC/MS/MS methods to measure water enrichment in BW. Samples were reacted with NaOH and U‐13C3‐acetone in an autosampler vial to promote deuterium exchange with U‐13C3‐acetone hydrogens. Headspace injections were made of U‐13C3‐acetone‐saturated air onto a 30‐m DB‐1MS column in electron impact‐mode. Subjects ingested 30 ml 2H2O, and plasma samples were collected. BW was determined by standard equation. Dual‐energy X‐ray absorptiometry scans were performed to calculate body mass, body volume and bone mineral content. A four‐compartmental model was used to estimate body composition (fat and fat free mass). Full‐scan experiments generated an m/z 45 peak and to a lesser extent an m/z 61 peak. Product fragment ions further monitored included 45 and 46 using selected ion monitoring (Method1), the 61 > 45 and 62 > 46 transition using multiple reaction monitoring (MRM; Method2) and the neutral loss, 62 > 45, transition (Method3). MRM methods were optimized for collision energy (CE) and collision‐induced dissociation (CID) argon gas pressure with 6 eV CE and 1.5 mTorr CID gas being optimal. Method2 was used for final determination of 2H2O enrichment of subjects because of lower natural background. N‐Boc/Fmoc/Z‐N′‐formyl‐gem‐diaminoalkyl derivatives, intermediates particularly useful in the synthesis of partially modified retro‐inverso peptides, have been characterized by both positive and negative ion electrospray ionization (ESI) ion‐trap multi‐stage mass spectrometry (MSn). The MS2 collision induced dissociation (CID) spectra of the sodium adduct of the formamides derived from the corresponding N‐Fmoc/Z‐amino acids, dipeptide and tripeptide acids show the [M + Na‐NH2CHO]+ ion, arising from the loss of formamide, as the base peak. Differently, the MS2 CID spectra of [M + Na]+ ion of all the N‐Boc derivatives yield the abundant [M + Na‐C4H8]+ and [M + Na‐Boc + H]+ ions because of the loss of isobutylene and CO2 from the Boc protecting function. Useful information on the type of amino acids and their sequence in the N‐protected dipeptidyl and tripeptidyl‐N′‐formamides is provided by MS2 and subsequent MSn experiments on the respective precursor ions. The negative ion ESI mass spectra of these oligomers show, in addition to [M‐H]?, [M + HCOO]? and [M + Cl]? ions, the presence of in‐source CID fragment ions deriving from the involvement of the N‐protecting group. Furthermore, MSn spectra of [M + Cl]? ion of N‐protected dipeptide and tripeptide derivatives show characteristic fragmentations that are useful for determining the nature of the C‐terminal gem‐diamino residue. The present paper represents an initial attempt to study the ESI‐MS behavior of these important intermediates and lays the groundwork for structural‐based studies on more complex partially modified retro‐inverso peptides. Electrospray‐generated precursor ions usually follow the ‘even‐electron rule’ and yield ‘closed shell’ fragment ions. We characterize an exception to the ‘even‐electron rule.’ In negative ion electrospray mass spectrometry (ES‐MS), 2‐(ethoxymethoxy)‐3‐hydroxyphenol (2‐hydroxyl protected pyrogallol) easily formed a deprotonated molecular ion (M‐H)? at m/z 183. Upon low‐energy collision induced decomposition (CID), the m/z 183 precursor yielded a radical ion at m/z 124 as the base peak. The radical anion at m/z 124 was still the major fragment at all tested collision energies between 0 and 50 eV (Elab). Supported by computational studies, the appearance of the radical anion at m/z 124 as the major product ion can be attributed to the combination of a low reverse activation barrier and resonance stabilization of the product ions. Under conditions of collision‐induced dissociation (CID), anions of α‐hydroxycarboxylic acids usually fragment to yield the distinctive hydroxycarbonyl anion (m/z 45) and/or the complementary product anion formed by neutral loss of formic acid (46 u). Further support for the known two‐step mechanism, involving an ion‐neutral complex for the formation of the hydroxycarbonyl anion from the carboxyl group, is herein provided by tandem mass spectrometric results and density functional theory computations on the glycolate, lactate and 3‐phenyllactate ions. A fourth, structurally related α‐hydroxycarboxylate ion, obtained by deprotonation of mandelic acid, showed only loss of carbon dioxide upon CID. Density functional theory computations on the mandelate ion indicated that similar energy inputs were required for a direct, phenyl‐assisted decarboxylation and a postulated novel rearrangement to a carbonate ester, which yielded the benzyl oxide ion upon loss of CO2. Rearrangement of the glycolate ion led to expulsion of carbon monoxide, whereas the 3‐phenyllactate ion showed the loss of water and formation of the benzyl anion and the benzyl radical as competing processes. The fragmentation pathways proposed for lactate and 3‐phenyllactate are supported by isotopic labeling. The relative computed energies of saddle points and product ions for all proposed fragmentation pathways are consistent with the energies supplied during CID experiments and the observed relative intensities of product ions. The diverse reaction pathways characterized for this set of four α‐hydroxycarboxylate ions demonstrate that it is crucial to understand the effects of structural variations when attempting to predict the gas‐phase reactivity and CID spectra of carboxylate ions. The gas‐phase reactions of Aryl―SF5·+ and Aryl―SO2F·+ have been studied with the electron ionization tandem mass spectrometry. Such reactions involve F‐atom migration from the S‐atom to the aryl group affording the product ion Aryl―F·+ by subsequent expulsion of SF4 or SO2, respectively. Especially, the 4‐pentafluorosulfanylphenyl cation 4‐SF5C6H4+ (m/z 203) from 4‐NO2C6H4SF5·+ by loss of ·NO2 could occur multiple F‐atom migration reactions to the product ion C6H4F3+ (m/z 133) by loss of SF2 in the MS/MS process. The gas‐phase reactions of 2,5‐xylylfluoroiodonium (pXyl―I+F, m/z 251) have also been studied using the electrospray tandem mass spectrometry, which involve a similar F‐atom migration process from the I‐atom to the aryl group giving the radical cation of 2‐fluoro‐p‐xylene (or its isomer 4‐fluoro‐m‐xylene, m/z 124) by reductive elimination of an iodine atom. All these gas‐phase F‐atom migration reactions from the heteroatom to the aryl group led to the aryl―F coupling product ions with a new formed CAryl―F bond. Upon collisional activation, gaseous metal adducts of lithium, sodium and potassium oxalate salts undergo an expulsion of CO2, followed by an ejection of CO to generate a product ion that retains all three metals atoms of the precursor. Spectra recorded even at very low collision energies (2 eV) showed peaks for a 44‐Da neutral fragment loss. Density functional theory calculations predicted that the ejection of CO2 requires less energy than an expulsion of a Na+ and that the [Na3CO2]+ product ion formed in this way bears a planar geometry. Furthermore, spectra of [Na3C2O4]+ and [39K3C2O4]+ recorded at higher collision energies showed additional peaks at m/z 90 and m/z 122 for the radical cations [Na2CO2]+? and [K2CO2]+?, respectively, which represented a loss of an M? from the precursor ions. Moreover, [Na3CO2]+, [39K3CO2]+ and [Li3CO2]+ ions also undergo a CO loss to form [M3O]+. Furthermore, product‐ion spectra for [Na3C2O4]+ and [39K3C2O4]+ recorded at low collision energies showed an unexpected peak at m/z 63 for [Na2OH]+ and m/z 95 for [39K2OH]+, respectively. An additional peak observed at m/z 65 for [Na218OH] + in the spectrum recorded for [Na3C2O4]+, after the addition of some H218O to the collision gas, confirmed that the [Na2OH] + ion is formed by an ion–molecule reaction with residual water in the collision cell. Low molecular weight polyisobutylenes (PIB) with chlorine, olefin and succinic acid end‐groups were studied using direct analysis in real time mass spectrometry (DART‐MS). To facilitate the adduct ion formation under DART conditions, NH4Cl as an auxiliary reagent was deposited onto the PIB surface. It was found that chlorinated adduct ions of olefin and chlorine telechelic PIBs, i.e. [M + Cl]? up to m/z 1100, and the deprotonated polyisobutylene succinic acid [M? H]? were formed as observed in the negative ion mode. In the positive ion mode formation of [M + NH4]+, adduct ions were detected. In the tandem mass (MS/MS) spectra of [M + Cl]?, product ions were absent, suggesting a simple dissociation of the precursor [M + Cl]? into a Cl? ion and a neutral M without fragmentation of the PIB backbones. However, structurally important product ions were produced from the corresponding [M + NH4]+ ions, allowing us to obtain valuable information on the arm‐length distributions of the PIBs containing aromatic initiator moiety. In addition, a model was developed to interpret the oligomer distributions and the number average molecular weights observed in DART‐MS for PIBs and other polymers of low molecular weight. Collision‐induced dissociation (CID) mass spectra of differently substituted glucosinolates were investigated under negative‐ion mode. Data obtained from several glucosinolates and their isotopologues (34S and 2H) revealed that many peaks observed are independent of the nature of the substituent group. For example, all investigated glucosinolate anions fragment to produce a product ion observed at m/z 195 for the thioglucose anion, which further dissociates via an ion/neutral complex to give two peaks at m/z 75 and 119. The other product ions observed at m/z 80, 96 and 97 are characteristic for the sulfate moiety. The peaks at m/z 259 and 275 have been attributed previously to glucose 1‐sulfate anion and 1‐thioglucose 2‐sulfate anion, respectively. However, based on our tandem mass spectrometric experiments, we propose that the peak at m/z 275 represents the glucose 1‐thiosulfate anion. In addition to the common peaks, the spectrum of phenyl glucosinolate (β‐D ‐Glucopyranose, 1‐thio‐, 1‐[N‐(sulfooxy)benzenecarboximidate] shows a substituent‐group‐specific peak at m/z 152 for C6H5‐C(?NOH)S?, the CID spectrum of which was indistinguishable from that of the anion of synthetic benzothiohydroxamic acid. Four pairs of positional isomers of ureidopeptides, FmocNH‐CH(R1)‐φ(NH‐CO‐NH)‐CH(R2)‐OY and FmocNH‐CH(R2)‐φ(NH‐CO‐NH)‐CH(R1)‐OY (Fmoc = [(9‐fluorenyl methyl)oxy]carbonyl; R1 = H, alkyl; R2 = alkyl, H and Y = CH3/H), have been characterized and differentiated by both positive and negative ion electrospray ionization (ESI) ion‐trap tandem mass spectrometry (MS/MS). The major fragmentation noticed in MS/MS of all these compounds is due to ? N? CH(R)? N? bond cleavage to form the characteristic N‐ and C‐terminus fragment ions. The protonated ureidopeptide acids derived from glycine at the N‐terminus form protonated (9H‐fluoren‐9‐yl)methyl carbamate ion at m/z 240 which is absent for the corresponding esters. Another interesting fragmentation noticed in ureidopeptides derived from glycine at the N‐terminus is an unusual loss of 61 units from an intermediate fragment ion FmocNH = CH2+ (m/z 252). A mechanism involving an ion‐neutral complex and a direct loss of NH3 and CO2 is proposed for this process. Whereas ureidopeptides derived from alanine, leucine and phenylalanine at the N‐terminus eliminate CO2 followed by corresponding imine to form (9H‐fluoren‐9‐yl)methyl cation (C14H11+) from FmocNH = CHR+. In addition, characteristic immonium ions are also observed. The deprotonated ureidopeptide acids dissociate differently from the protonated ureidopeptides. The [M ? H]? ions of ureidopeptide acids undergo a McLafferty‐type rearrangement followed by the loss of CO2 to form an abundant [M ? H ? Fmoc + H]? which is absent for protonated ureidopeptides. Earlier characterization of some hydrolysis products of AlCl3·6H2O was confirmed by electrospray ionization tandem mass spectrometry with increasing collision energy of projectile ions. At lower collision energies, the aqua ligands were stripped off. At higher energies, two hydroxo groups formed a bridging oxo group with loss of one water molecule. Aluminum complexes could also capture aqua ligands in the collision chamber so long as the parent ion did not fragment, and the fragment ion spectra broadened toward higher m/z values. For identification of ginsenoside enantiomers, electrospray ionization mass spectrometry (ESI‐MS) was used to generate silver complexes of the type [ginsenoside + Ag]+. Collision induced dissociation of the silver‐ginsenoside complexes produced fragment ions by dehydration, allowing differentiation of ginsenoside enantiomers by the intensity of [M + Ag ? H2O]+ ion. In the meanwhile, an approach based on the distinct profiles of enantiomer‐selective fragment ion intensity varied with collision energy was introduced to refine the identification and quantitation of ginsenoside enantiomers. Five pairs of enantiomeric ginsenosides were distinguished and quantified on the basis of the distribution of fragment ion [M + Ag ? H2O]+. This method was also extended to the identification of other type of ginsenoside isomers such as ginsenoside Rb2 and Rb3. For demonstrating the practicability of this novel approach, it was utilized to analyze the molar ratio of 20‐(S) and 20‐(R) type enantiomeric ginsenosides in enantiomer mixture in red ginseng extract. The generation of characteristic fragment ion [M + Ag ? H2O]+ likely results from the reduction of potential energy barrier of dehydration because of the catalysis of silver ion. A series of 4‐substituted 3‐hydroxyfurazans were subjected to electrospray ionization tandem mass spectrometry. At low collision energy, oxyisocyanate ([O=C=N–O]?, m/z 58) was formed as the predominant product ion from each deprotonated 3‐hydroxyfurazan, indicating cleavage of the heterocyclic ring. The facile energetics of this characteristic fragmentation process was confirmed by density functional computations.  相似文献

Absolute bond dissociation energies of water to sodium glycine cations and glycine to hydrated sodium cations are determined experimentally by competitive collision-induced dissociation (CID) of Na+Gly(H2O)x, x = 1–4, with xenon in a guided ion beam tandem mass spectrometer. The cross sections for CID are analyzed to account for unimolecular decay rates, internal energy of reactant ions, multiple ion–molecule collisions, and competition between reaction channels. Experimental results show that the binding energies of water and glycine to the complexes decrease monotonically with increasing number of water molecules. Ab initio calculations at four different levels show good agreement with the experimental bond energies of water to Na+Gly(H2O)x, x = 0–3, and glycine to Na+(H2O), whereas the bond energies of glycine to Na+(H2O)x, x = 2–4, are systematically higher than the experimental values. These discrepancies may provide some evidence that these Na+Gly(H2O)x complexes are trapped in excited state conformers. Both experimental and theoretical results indicate that the sodiated glycine complexes are in their nonzwitterionic forms when solvated by up to four water molecules. The primary binding site for Na+ changes from chelation at the amino nitrogen and carbonyl oxygen of glycine for x = 0 and 1 to binding at the C terminus of glycine for x = 2–4. The present characterization of the structures upon sequential hydration indicates that the stability of the zwitterionic form of amino acids in solution is a consequence of being able to solvate all charge centers.  相似文献

Dopamine [DA]+ (m/z 154), DA dimer [2DA‐H]+ (m/z 307) and DA quinone [DAQ]+ (m/z 152) are detected in positive ion mode electrospray ionization mass spectrometry (ESI MS) of dopamine in 50/1/49 (vol%) water/acetic acid/methanol. H/D exchange experiments support a covalent structure of DA dimer. Thus, ESI of DA may involve 1e?, 1H+ oxidation processes followed by rapid radical dimerization. The DA quinone signal is low in ESI MS, which indicates a low efficiency of the 2e?, 2H+ oxidation reaction. On‐line electrochemistry ESI MS (EC/ESI MS) with low electrochemical cell voltage floated on high ES voltage increases electrospray current and improves sensitivity for DA. The DA quinone signal increases and DA dimer signal decreases. A new configuration of the ESI MS instrument with a cone‐shaped capillary inlet significantly enhanced sensitivity of ESI and EC/ESI MS measurements. A DA quinone‐cysteine adduct [DAQ+Cys]+ was detected in solutions of DA with cysteine (Cys). ESI MS and EC/ESI MS indicate formation of the DA quinone‐cysteine adduct by 1e? pathway. Oxidation pathways in ESI MS are relevant to biological reactivity of DA and Cys.  相似文献

A new tandem time‐of‐flight mass spectrometer with an electrospray ionization ion source ‘ESI‐TOF/quadTOF’ was designed and constructed to achieve the desired aim of structural elucidation via high‐energy collision‐induced dissociation (CID), and the simultaneous detection of all fragment ions. The instrument consists of an orthogonal acceleration‐type ESI ion source, a linear TOF mass spectrometer, a collision cell, a quadratic‐field ion mirror and a microchannel plate detector. High‐energy CID spectra of doubly protonated angiotensin II and bradykinin were obtained. Several fragment ions such as a‐, d‐, v‐ and w‐type ions, characteristic of high‐energy CID, were clearly observed in these spectra. These high‐energy CID fragment ions enabled confirmation of the complete sequence, including leucine–isoleucine determinations. It was demonstrated that high‐energy CID of multiply protonated peptides could be achieved in the ESI‐TOF/quadTOF. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

This study reports an energy‐resolved mass spectrometric (ERMS) strategy for the characterization of position isomers derived from the reaction of hydroxyl radicals (OH) with diphenhydramine (DPH) that are usually hard to differentiate by other methods. The isomer analogues formed by OH attack on the side chain of DPH are identified with the help of a specific fragment ion peak (m/z 88) in the collision‐induced dissociation (CID) spectrum of the protonated molecule. In the negative ion mode, the breakdown curves of the deprotonated molecules show an order of stability (supported by density functional theory (DFT) calculations) ortho > meta > para of the positional isomers formed by the hydroxylation of the aromatic ring. The gas phase stability of the deprotonated molecules [M ? H]? towards the benzylic cleavage depends mainly on the formation of intramolecular hydrogen bonds and of the mesomeric effect of the phenol hydroxyl. The [M ? H]? molecules of ortho and meta isomers result a peak at m/z 183 with notably different intensities because of the presence/absence of an intramolecular hydrogen bonding between the OH group and C9 protons. The ERMS approach discussed in this report might be an effective replacement for the conventional methods that requires very costly and time‐consuming separation/purification methods along with the use of multi‐spectroscopic methods. Long‐chain ferulic acid esters, such as eicosyl ferulate ( 1 ), show a complex and analytically valuable fragmentation behavior under negative ion electrospay collision‐induced dissociation ((?)‐ESI‐CID) mass spectrometry, as studied by use of a high‐resolution (Orbitrap) mass spectrometer. In a strong contrast to the very simple fragmentation of the [M + H]+ ion, which is discussed briefly, the deprotonated molecule, [M – H]?, exhibits a rich secondary fragmentation chemistry. It first loses a methyl radical (MS2) and the ortho‐quinoid [M – H – Me]‐? radical anion thus formed then dissociates by loss of an extended series of neutral radicals, CnH2n + 1? (n = 0–16) from the long alkyl chain, in competition with the expulsion of CO and CO2 (MS3). The further fragmentation (MS4) of the [M – H – Me – C3H7]? ion, discussed as an example, and the highly specific losses of alkyl radicals from the [M – H – Me – CO]‐? and [M – H – Me – CO2]‐? ions provide some mechanistic and structural insights.  相似文献

Two new series of Boc‐N‐α,δ‐/δ,α‐ and β,δ‐/δ,β‐hybrid peptides containing repeats of L ‐Ala‐δ5‐Caa/δ5‐Caa‐L ‐Ala and β3‐Caa‐δ5‐Caa/δ5‐Caa‐β3‐Caa (L ‐Ala = L ‐alanine, Caa = C‐linked carbo amino acid derived from D ‐xylose) have been differentiated by both positive and negative ion electrospray ionization (ESI) ion trap tandem mass spectrometry (MS/MS). MSn spectra of protonated isomeric peptides produce characteristic fragmentation involving the peptide backbone, the Boc‐group, and the side chain. The dipeptide positional isomers are differentiated by the collision‐induced dissociation (CID) of the protonated peptides. The loss of 2‐methylprop‐1‐ene is more pronounced for Boc‐NH‐L ‐Ala‐δ‐Caa‐OCH3 (1), whereas it is totally absent for its positional isomer Boc‐NH‐δ‐Caa‐L ‐Ala‐OCH3 (7), instead it shows significant loss of t‐butanol. On the other hand, second isomeric pair shows significant loss of t‐butanol and loss of acetone for Boc‐NH‐δ‐Caa‐β‐Caa‐OCH3 (18), whereas these are insignificant for its positional isomer Boc‐NH‐β‐Caa‐δ‐Caa‐OCH3 (13). The tetra‐ and hexapeptide positional isomers also show significant differences in MS2 and MS3 CID spectra. It is observed that ‘b’ ions are abundant when oxazolone structures are formed through five‐membered cyclic transition state and cyclization process for larger ‘b’ ions led to its insignificant abundance. However, b1+ ion is formed in case of δ,α‐dipeptide that may have a six‐membered substituted piperidone ion structure. Furthermore, ESI negative ion MS/MS has also been found to be useful for differentiating these isomeric peptide acids. Thus, the results of MS/MS of pairs of di‐, tetra‐, and hexapeptide positional isomers provide peptide sequencing information and distinguish the positional isomers.
