In situ vascular tissue engineering has been proposed as a promising approach to fulfill the need for small‐diameter blood vessel substitutes. The approach comprises the use of a cell‐free instructive scaffold to guide and control cell recruitment, differentiation, and tissue formation at the locus of implantation. Here we review the design parameters for such scaffolds, with special emphasis on differentiation of recruited ECFCs into the different lineages that constitute the vessel wall. Next to defining the target properties of the vessel, we concentrate on the target cell source, the ECFCs, and on the environmental control of the fate of these cells within the scaffold. The prospects of the approach are discussed in the light of current technical and biological hurdles.

Severe peripheral nervous system injuries currently hold limited therapeutic solutions. Existing clinical techniques such as autografts, allografts, and newer nerve guidance conduits have shown variable outcomes in functional recovery, adverse immune responses, and in some cases low or minimal availability. This can be attributed in part to the lack of chemical, physical, and electrical cues directing both nerve guidance and regeneration. To address this pressing clinical issue, electrospun nanofibers and microfibers composed of piezoelectric polyvinylidene flouride‐triflouroethylene (PVDF‐TrFE) have been introduced as an alternative template for tissue engineered biomaterials, specifically as it pertains to their relevance in soft tissue and nerve repair. Here, biocompatible scaffolds of PVDF‐TrFE are fabricated and their ability to generate an electrical response to mechanical deformations and produce a suitable regenerative microenvironment is examined. It is determined that 20% (w/v) PVDF‐TrFE in (6:4) dimethyl formamide (DMF):acetone solvent maintains a desirable piezoelectric coefficient and the proper physical and electrical characteristics for tissue regeneration. Further, it is concluded that scaffolds of varying thickness promoted the adhesion and alignment of Schwann cells and fibroblasts. This work offers a prelude to further advancements in nanofibrous technology and a promising outlook for alternative, autologous remedies to peripheral nerve damage. 相似文献

We report the production of chitosan‐based fibers and chitosan fiber‐mesh structures by melt processing (solvent‐free) to be used as tissue‐engineering scaffolds. The melt‐based approach used to produce the scaffolds does not change their main characteristics, including the surface roughness and microporosity. The porosity, pore size, interconnectivity and mechanical performance of the scaffolds are all within the range required for various tissue‐engineering applications. Biological assessments are performed in direct‐contact assays. Cells are able to colonize the scaffold, including the inner porous structure. The cells show high indices of viability in all of the scaffold types.

We have developed three‐dimensional electrospun microfibrous meshes of a novel star branched three‐arm poly(ε‐caprolactone) (*PCL) as potential scaffolds for tissue engineering applications. The processing conditions required to obtain uniform fibers were optimized by studying their influence on fiber morphology and size. Polymer molecular weight and solution feed rate influenced both the mesh microstructure and the tensile properties of the developed mats. Electrospun samples were also tested for their mechanical properties in wet conditions, showing higher yield strength and strain in comparison to that observed in dry conditions. Cell culture experiments employing MC3T3‐E1 osteoblast like cells showed good cell viability adhesion and collagen production on the *PCL scaffolds.

Tissue engineering scaffolds should provide a suitable porous structure and proper mechanical strength, which is beneficial for the delivery of growth factor and regulation of cells. In this study, the open‐porous polycaprolactone (PCL)/poly (lactic acid) (PLA) tissue engineering scaffolds with suitable porous scale were fabricated using different ratios of PCL/PLA blends. At the same time, the relationship of foaming process, morphology, and mechanical behavior in the optimized batch microcellular foaming process were studied based on the single‐factor experiment method. The porous structures and mechanical strength of the scaffolds were optimized by adjusting foaming parameters, including the temperature, pressure, and CO2 dissolution time. The results indicated that the foaming parameters influence the cell morphology, further determine the mechanical behavior of PCL/PLA blends. When the PCL content is high, with the increase of temperature and time, the cell diameter and the elastic modulus increased, and the tensile strength and elastic modulus increased with the increase of the average cell size, and decreased as the increase of the cell density. While when the PLA content was high, the cell diameter showed the same trend, and the tensile strength and elastic modulus were higher, and the elongation at break was lower, and tensile strength and elastic modulus decreased with the increase of the average cell size and increased with the increase of cell density. This work successfully fabricated optimized porous PCL/PLA scaffolds with excellent suitable mechanical properties, pore sizes, and high interconnectivity, indicating the effectiveness of modulating the batch foaming process parameters. 相似文献

In the present work, zinc oxide nanoparticles were treated with aminopropyl trimethoxy silane‐coupling agent and used as a new kind of reinforcement for a typical high performance bisphenol‐A‐based phthalonitrile resin. The resulted nanocomposites were characterized for their mechanical, thermal, and optical properties. Results from the tensile test indicated that the tensile strength and modulus as well as the toughness state of the matrix were all enhanced with the increasing of the nanoparticles amount. Thermogravimetric analysis showed that the starting decomposition temperatures and the residual weight at 800°C were highly improved upon adding the nanofillers. At 6 wt% nanoloading, the glass transition temperature and the storage modulus were considerably enhanced reaching about 359°C and 3.7 GPa, respectively. The optical tests revealed that the neat resin possesses excellent UV‐shielding properties, which were further enhanced by adding the nanofillers. Furthermore, the fractured surfaces of the nanocomposites analyzed by scanning electron microscope exhibited homogeneous and rougher surfaces compared with that of the pristine resin. Finally, the good dispersion of the reinforcing phase into the matrix was confirmed by a high resolution transmission electron microscope. Copyright © 2015 John Wiley & Sons, Ltd. 相似文献

A new approach to prepare and characterize double network elastomeric systems was investigated. A styrene‐ethylene‐co‐butylene‐styrene (SEBS) triblock copolymer system containing physical crosslinks was used to achieve a double network by additional crosslinking using ultra‐violet (UV) light. An ethylene–propylene–diene monomer (EPDM) terpolymer system containing chemical crosslinks was used to achieve a conventional double network using UV crosslinking. Properties from conventional monotonic tensile tests, dynamic mechanical analysis, and thermomechanical properties were investigated. These double network elastomers show a transition between competitive and collaborative behavior in their mechanical properties and lower coefficients of thermal expansion arising from a competition of the networks. © 2010 Wiley Periodicals, Inc. There is a clear need for novel in vitro models, especially for neuronal applications. Development of in vitro models is a multiparameter task consisting of cell‐, biomaterial‐, and environment‐related parameters. Here, three different human origin neuronal cell sources are studied and cultured in various hydrogel 3D scaffolds. For the efficient evaluation of complex results, an indexing method for data is developed and used in principal component analysis (PCA). It is found that no single hydrogel is superior to other hydrogels, and collagen I (Col1) and hyaluronan–poly(vinyl alcohol) (HA1‐PVA) gels are combined into an interpenetrating network (IPN) hydrogel. The IPN gel combines cell supportiveness of the collagen gel and stability of the HA1‐PVA gel. Moreover, cell adhesion is studied in particular and it is found that adhesion of neurons differs from that observed for fibroblasts. In conclusion, the HA1‐PVA‐col1 hydrogel is a suitable scaffold for neuronal cells and supports adhesion formation in 3D. 相似文献

Electrospun micro- and nanofiber scaffolds have gained interest in biomedical applications, especially in tissue engineering, because they can be used to reproduce the structure of the extracellular matrix (ECM) of natural tissue. The selection of the solvent is an important factor which affects the diameter, the surface morphology and the crystallinity of the electrospun fibers, and, accordingly, their mechanical properties as well as their degradation kinetics. Furthermore, the surface morphology of the electrospun fibres can be controlled by solvent vapour pressure to produce porous structures which might be helpful for cell adhesion and proliferation. In the present work, poly (L-lactic acid) (PLLA) has been electrospun using solvents with different vapour pressures to investigate the influences of the solvent vapour pressure on morphology, diameter, crystallinity and mechanical properties of the electrospun fiber scaffolds. The results show that the vapour pressure of the solvents (or solvent mixtures) play an important role in the fiber diameter and crystallinity. Furthermore, the crystallinity of the fibers is increased by lowering the vapour pressure of the used solvent. In addition, the mechanical properties (e.g., tensile strength and Young's modulus) are strongly dependent on morphological features such average fibers diameter. Tissue expansion is used by plastic/reconstructive surgeons to grow additional skin/tissue for replacing or repairing lost or damaged soft tissues. Recently, hydrogels have been widely used for tissue expansion applications. Herein, a self‐inflating tissue expander blend composition from three different molecular weights (2, 6, and 10 kDa) of poly (ethylene glycol) diacrylate (PEGDA) hydrogel with tunable mechanical and swelling properties is presented. The in vitro results demonstrate that, of the eight studied compositions, P6 (PEGDA 6 kDa:10 kDa (50:50)) and P8 (PEGDA 6 kDa:10 kDa (35:65)) formulations provide a balance of mechanical property and swelling capability suitable for tissue expansion. Furthermore, these expanders can be compressed up to 60% of their original height and can be loaded and unloaded cyclically at least ten times with no permanent deformation. The in vivo results indicate that these two engineered blend compositions are capable to generate a swelling pressure sufficient to dilate the surrounding tissue while retaining their original shape. The histological analyses reveal the formation of fibrous capsule at the interface between the implant and the subcutaneous tissue with no signs of inflammation. Ultimately, controlling the PEGDA chain length shows potential for the development of self‐inflating tissue expanders with tunable mechanical and swelling properties.

Crosslinked networks were synthesized by copolymerization of mono‐functional tert‐butyl acrylate (tBA) with diethyleneglycol dimethacrylate (DEGDMA) or polyethylene glycol dimethacrylates (PEGDMA). By varying the chain length and concentration of the difunctional PEGDMA, we obtained tBA‐PEGDMA copolymer networks while by varying the concentration of difunctional DEGDMA, we obtained tBA‐DEGDMA crosslinked networks. The various materials were submitted to large deformations through uniaxial tension tests. For moderate weight percent of crosslinking agent, up to 20%, the networks showed standard S‐shape stress–strain curves, characteristic of rubber‐like elasticity. Two macromolecular models, the 8‐chain model and the full‐network model, were applied to fit the uniaxial tensile response of the materials. Both models provide good representations of the overall uniaxial stress–strain response of each material. After fitting to stress–strain data, the network models were employed to predict the shear modulus and the elongation at break. Neither the 8‐chain nor the full network model were capable of predicting the failure strain or shear modulus, indicating these models are best used to describe stress–strain relations rather than predict mechanical properties for the network polymers considered here. © 2008 Wiley Periodicals, Inc. A combination of hard sphere and high internal phase emulsion templating gives a platform for synthesizing hierarchically porous polymers with a unique topology exhibiting interconnected spherical features on multiple levels. Polymeric spheres are fused by thermal sintering to create a 3D monolithic structure while an emulsion with a high proportion of internal phase and monomers in the continuous phase is added to the voids of the previously constructed monolith. Following polymerization of the emulsion and dissolution of the templating structure, a down‐replicating topology is created with a primary level of pores as a result of fused spheres of the 3D monolithic structure, a secondary level of pores resulting from the emulsion's internal phase, and a tertiary level of interconnecting channels. Thiol‐ene chemistry with divinyladipate and pentaerythritol tetrakis(3‐mercaptopropionate) is used to demonstrate the preparation of a crosslinked polyester with overall porosity close to 90%. Due to multilevel porosity, such materials are interesting for applications in bone tissue engineering, possibly simulating the native sponge like bone structure. Their potential to promote ossteointegration is tested using human bone derived osteoblasts. Material–cell interactions are evaluated and they reveal growth and proliferation of osteoblasts both on surface and in the bulk of the scaffold. 相似文献

Accurate prediction and validation of the assembly of bioinspired peptide sequences into fibers with defined mechanical characteristics would aid significantly in designing and creating materials with desired properties. This process may also be utilized to provide insight into how the molecular architecture of many natural protein fibers is assembled. In this work, computational modeling and experimentation are used in tandem to determine how peptide terminal modification affects a fiber‐forming core domain. Modeling shows that increased terminal molecular weight and hydrophilicity improve peptide chain alignment under shearing conditions and promote consolidation of semicrystalline domains. Mechanical analysis shows acute improvements to strength and elasticity, but significantly reduced extensibility and overall toughness. These results highlight an important entropic function that terminal domains of fiber‐forming peptides exhibit as chain alignment promoters, which ultimately has notable consequences on the mechanical behavior of the final fiber products. 相似文献

Inverse opal scaffolds have recently emerged as a novel class of scaffolds with uniform and controllable pore sizes for tissue engineering to provide better nutrient transport, a uniform cell distribution, and an adjustable microenvironment for cell differentiation. However, when the pore size of the scaffold is much larger than the dimension of a cell, the cell actually encounters a local 2D environment and the void space associated with the pore can not be efficiently utilized. Here, we demonstrate that a truly 3D microenvironment can be created inside a pore by further functionalizing the as‐prepared inverse opal scaffold with a second polymer by freeze‐drying. The resultant inverse opal scaffold with hierarchically structured pores can enhance both cell proliferation and tissue infiltration. 相似文献

Polyelectrolyte multilayers (PEMs) with different polycation/polyanion pairs are fabricated by the layer‐by‐layer technique employing synthetic, natural, and both types of polyelectrolytes. The impact of the chemical composition of PEMs on cell adhesion is assessed by studying cell shape, spreading area, focal contacts, and cell proliferation for the A549 cell line. Cells exhibit good adhesion on PEMs containing natural polycations and poly(sodium 4‐styrenesulfonate) (PSS) as polyanion, but limited adhesion is observed on PEMs fabricated from both natural polyelectrolytes. PEMs are then assembled, depositing a block of natural polyelectrolytes on top of a stiffer block with PSS as polyanion. Cell adhesion is enhanced on top of the diblock PEMs compared to purely natural PEMs. This fact could be explained by the interdigitation between polyelectrolytes from the two blocks. Diblock PEM assembly provides a simple means to tune cell adhesion on biocompatible PEMs.
