We report on novel diblock copolymers of poly(N‐vinylcaprolactam) (PVCL) and poly(N‐vinyl‐2‐pyrrolidone) (PVPON) (PVCL‐b‐PVPON) with well‐defined block lengths synthesized by the MADIX/reversible addition‐fragmentation chain transfer (RAFT) process. We show that the lower critical solution temperatures (LCST) of the block copolymers are controllable over the length of PVCL and PVPON segments. All of the diblock copolymers dissolve molecularly in aqueous solutions when the temperature is below the LCST and form spherical micellar or vesicular morphologies when temperature is raised above the LCST. The size of the self‐assembled structures is controlled by the molar ratio of PVCL and PVPON segments. The synthesized homopolymers and diblock copolymers are demonstrated to be nontoxic at 0.1–1 mg mL?1 concentrations when incubated with HeLa and HEK293 cancer cells for various incubation times and have potential as nanovehicles for drug delivery. © 2014 Wiley Periodicals, Inc. A series of novel temperature‐ and pH‐responsive graft copolymers, poly(L ‐glutamic acid)‐g‐poly(N‐isopropylacrylamide), were synthesized by coupling amino‐semitelechelic poly(N‐isopropylacrylamide) with N‐hydroxysuccinimide‐activated poly(L ‐glutamic acid). The graft copolymers and their precursors were characterized, by ESI‐FTICR Mass Spectrum, intrinsic viscosity measurements and proton nuclear magnetic resonance (1H NMR). The phase‐transition and aggregation behaviors of the graft copolymers in aqueous solutions were investigated by the turbidity measurements and dynamic laser scattering. The solution behavior of the copolymers showed dependence on both temperature and pH. The cloud point (CP) of the copolymer solution at pH 5.0–7.4 was slightly higher than that of the solution of the PNIPAM homopolymer because of the hydrophilic nature of the poly(glutamic acid) (PGA) backbone. The CP markedly decreased when the pH was lowered from 5 to 4.2, caused by the decrease in hydrophilicity of the PGA backbone. At a temperature above the lower critical solution temperature of the PNIPAM chain, the copolymers formed amphiphilic core‐shell aggregates at pH 4.5–7.4 and the particle size was reduced with decreasing pH. Novel thermoresponsive double‐hydrophilic fluorinated block copolymers were successfully synthesized by reversible addition‐fragmentation chain transfer (RAFT) polymerization. Poly[N‐(2,2‐difluoroethyl)acrylamide] (P2F) was synthesized via RAFT polymerization of N‐(2,2‐difluoroethyl)acrylamide (M2F) using 2‐dodecylsulfanylthiocarbonylsulfanyl‐2‐methylpropionic acid (DMP) as the chain transfer agent (CTA) and 2,2′‐azobisisobutyronitrile (AIBN) as the initiator. The resulting P2F macroCTA was further chain extended with N‐(2‐fluoroethyl)acrylamide (M1F) to yield poly{[N‐(2,2‐difluoroethyl)acrylamide]‐b‐[N‐(2‐fluoroethyl)acrylamide]} (P2F‐b‐P1F) block copolymers with different lengths of the P1F block. Molecular weight and molecular weight distribution were determined by gel permeation chromatography. The average molecular weight (Mn) of the resulting polymers ranged from 2.9 × 104 to 5.8 × 104 depending on the length of the P1F block. The molecular weight distribution was low (Mw/Mn = 1.11–1.19). Turbidimetry by UV‐Visble (UV‐Vis) spectroscopy, dynamic light scattering, and in situ temperature‐dependent 1H NMR measurements demonstrated that the P2F block underwent a thermal transition from hydrophilic to hydrophobic, which in turn induced self‐assembly from unimers to aggregates. Transmission electron microscopy studies demonstrated that polymeric aggregates formed from an aqueous solution of P2F‐b‐P1F at 60 °C were disrupted by cooling down to 20 °C and regenerated by heating to 60 °C. Self‐immolative polymers (SIPs) undergo depolymerization in response to the cleavage of stimuli‐responsive end‐caps from their termini. Some classes of SIPs, including polycarbamates, have depolymerization rates that depend on environmental factors such as solvent and pH. In previous work, hydrophobic SIPs have been incorporated into amphiphilic block copolymers and used to prepare nanoassemblies. However, stimuli‐responsive hydrophilic blocks have not previously been incorporated. In this work, we synthesized amphiphilic copolymers composed of a hydrophobic polycarbamate SIP block and a hydrophilic poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) block connected by a UV light‐responsive linker end‐cap. It was hypothesized that after assembly of the block copolymers into nanoparticles, chain collapse of the PDMAEMA above its lower critical solution temperature (LCST) might change the environment of the SIP block, thereby altering its depolymerization rate. Self‐assembly of the block copolymers was performed, and the depolymerization of the resulting assemblies was studied by fluorescence spectroscopy, dynamic light scattering, and NMR spectroscopy. At 20 °C, the system exhibited a selective response to the UV light. At 65 °C, above the LCST of PDMAEMA, the systems underwent more rapid depolymerization, suggesting that the increase in rate arising from the higher temperature dominated over environmental effects arising from chain collapse. © 2018 Wiley Periodicals, Inc. Polyisobutylene‐b‐poly(N,N‐diethylacrylamide) (PIB‐b‐PDEAAm) well‐defined amphiphilic diblock copolymers were synthesized by sequential living carbocationic polymerization and reversible addition‐fragmentation chain transfer (RAFT) polymerization. The hydrophobic polyisobutylene segment was first built by living carbocationic polymerization of isobutylene at ?70 ° C followed by multistep transformations to give a well‐defined (Mw/Mn = 1.22) macromolecular chain transfer agent, PIB‐CTA. The hydrophilic poly(N,N‐diethylacrylamide) block was constructed by PIB‐CTA mediated RAFT polymerization of N,N‐diethylacrylamide at 60 ° C to afford the desired well‐defined PIB‐b‐PDEAAm diblock copolymers with narrow molecular weight distributions (Mw/Mn ≤1.26). Fluorescence spectroscopy, transmission electron microscope, and dynamic light scattering (DLS) were employed to investigate the self‐assembly behavior of PIB‐b‐PDEAAm amphiphilic diblock copolymers in aqueous media. Poly(N‐isopropylacrylamide) (PNIPAAm) homopolymers synthesized by reversible addition–fragmentation chain transfer polymerization were used as macro‐chain‐transfer agents to synthesize smart amphiphilic block copolymers with a switchable hydrophilic–hydrophobic block of PNIPAAm and a hydrophilic block of poly(N‐dimethylacrylamide). All polymers were characterized by gel permeation chromatography, 1H NMR, and differential scanning calorimetry. The reversible micelles formed by the block copolymers of various compositions in aqueous solutions were characterized by 1H NMR, dynamic light scattering, and tensiometry. Micelles were observed in the aqueous solutions when the temperature was increased to 40 °C because of the collapse of the PNIPAAm structure, which led to a PNIPAAm hydrophobic block. The drug loading capacity was illustrated with the use of the solvatochromic Reichardt's dye and measured by ultraviolet–visible. © 2005 Wiley Periodicals, Inc. A series of poly(N‐isopropylacrylamide)‐co‐poly(Nε‐benzyloxycarbonyl‐L ‐lysine) graft copolymers (PNIPAm‐co‐PZLLys) with different side chains (degree of polymerization, DP = 5~40) and unit ratios (from 30 to 70 mol %) were prepared via free radical polymerization, followed by cleaving benzyloxycarbonyl groups (Z groups) to obtain the double hydrophilic graft copolymer, poly(N‐isopropylacrylamide)‐co‐poly(L ‐lysine) (PNIPAm‐co‐PLLys). The pH‐ and temperature‐response properties of the graft copolymers in aqueous solution were studied. The experimental results indicate L15‐N30 and L15N‐70, that is, the PNIPAm‐co‐PLLys having the poly(L ‐lysine) of DP = 15 as side chains as well as 30 and 70 mol %, respectively, of PNIPAm as backbone, have coil‐to‐helix transitions from pH 6 to pH 12 at room temperature and form uniform nanoscale micelle‐like dispersions in aqueous solution at pH 12. The graft copolymers also could form uniform and nanoscale micelle‐like structures at 50 °C in pH 6 buffer solution due to slightly polymer aggregation. With temperature and pH increased, both the deprotonated PLLys side chains and PNIPAm backbone become hydrophobic, leading to polymer precipitation. These results illustrate that a double tunable hydrophilic graft copolymer had been successfully synthesized via a simple radical polymerization, and could form micelles without serious polymer aggregation at a lower pH and a higher temperature. © 2011 Wiley Periodicals, Inc. A series of well‐defined amphiphilic graft copolymers bearing hydrophobic poly(tert‐butyl acrylate) backbone and hydrophilic poly[poly(ethylene glycol) methyl ether methacrylate)] (PPEGMEMA) side chains were synthesized by sequential reversible addition fragmentation chain transfer (RAFT) polymerization and single‐electron‐transfer living radical polymerization (SET‐LRP) without any polymeric functional group transformation. A new Br‐containing acrylate monomer, tert‐butyl 2‐((2‐bromoisobutanoyloxy)methyl)acrylate (tBBIBMA), was first prepared, which can be homopolymerized by RAFT to give a well‐defined PtBBIBMA homopolymer with a narrow molecular weight distribution (Mw/Mn = 1.15). This homopolymer with pendant Br initiation group in every repeating unit initiated SET‐LRP of PEGMEMA at 45 °C using CuBr/dHbpy as catalytic system to afford well‐defined PtBBIBMA‐g‐PPEGMEMA graft copolymers via the grafting‐from strategy. The self‐assembly behavior of the obtained graft copolymers in aqueous media was investigated by fluorescence spectroscopy and TEM. These copolymers were found to be stimuli‐responsive to both temperature and ions. Finally, poly(acrylic acid)‐g‐PPEGMEMA double hydrophilic graft copolymers were obtained by selective acidic hydrolysis of hydrophobic PtBA backbone while PPEGMEMA side chains kept inert. © 2011 Wiley Periodicals, Inc. In this study polyrotaxane (PR)‐based triblock copolymers were first synthesized via the atom transfer radical polymerization (ATRP) of N‐isopropylacrylamide initiated with the self‐assembly of a distal 2‐bromoisobutyryl end‐capped Pluronic 17R4 with a varying amount of α‐cyclodextrins (α‐CDs) in the presence of CuCl/PMDETA at 25°C in aqueous solution. The α‐CDs entrapped on the copolymer chain were then linked with hexamethylene diisocyanate to give rise to novel slightly cross‐linked polyrotaxanes (SCPRs) in DMF at 45°C. The structures of the PR‐based triblock copolymers and SCPRs were characterized by 1H NMR, 13C CP/MAS, GPC and TGA analyses. The number‐average molecular weight of the resulting SCPRs was nearly three and five times of their precursor after linking with a low polydispersity index range of 1.08–1.28. The thermo‐responsive transition of both PR‐based supramolecular polymers in aqueous solution was demonstrated by turbidity measurements and the self‐aggregated morphologies were also evidenced by TEM observations.  相似文献

Interaction between small molecules (iodine and 1-anilinonaphthalene-8-sulphonate) and poly-N-methylacetamide (PVMA), poly-N-vinylcaprolactam (PVCL), poly-N-vinylpyrrolidone (PVP) and copolymers of N-vinyleaprolactam with N-vinylpyrrolidone (CP VCL-VP) and with N-vinyl-N-methylacetamide (VMA) were studied. All the polymers formed complexes with the small molecules. The stability constant for the complexes with the probe increases in the series PVMA < PVP < copolymer < PVCL indicating the essential role of hydrophobic interactions in the complex formation. With increasing temperature of aqueous solution from 18 to 33°, the constant decreased for PVP, PVMA and copolymers with low VCL content and increased for PVCL. A folding of PVC coils was observed over the temperature range, demonstrating a relationship between the conformational changes of macromolecules and the polymer complex formation. Addition of PVCL, PVMA and PVP to aqueous I3? solution brings about a shift of λmax from 350 nm for free triiodide ions to 372, 367 and 364 nm respectively. It is explained by different dipole moments of the side substituents. The stability constant for the polymers and I3? was estimated as approx. 105 M. The ability of the polymers to form complexes depends on chainlength. The greatest loss of the ability was observed for PVCL with Mn changing from 6 × 103 to 2 × 103. Interaction of the polymers with I3? is accompanied by a conformational shrinking of coils. In the case of the probe, forces of electrostatic repulsion between negative charges of sulphonate groups result in an unfolding of polimer coils.  相似文献

Dynamic phase transition and self‐assembly mechanism of thermosensitive poly(ethylene oxide)‐b‐poly(N‐vinylcaprolactam) (PEO‐b‐PVCL) copolymer are explored deeply. A gradual dehydration process with predominated hydrophobic interactions among copolymer chains in the phase transition process distinguishes the copolymer from homopolymer. PVCL in the inner zone is restricted and counter‐balanced by the PEO segments based on the sequence order of representative groups during the heating‐cooling cycles. Remarkably, PEO shell experiences unusual hydration process, which is first discovered. This hydrophilic shell plays as water absorption sponge layer and captures expelled water from PVCL core, accompanied by gradient distribution of water existed in the assembly structures. Peculiarly, pseudo‐linear changes of the integral area of free C?O are presented compared with inflection point in the hydrated C?O integral area, which propose that a part of hydrated C?O forms incomplete dehydrated states. Terpolymers composed of Nn‐propylacrylamide (NPAAm), butyl methacrylate (BMA), and N,N‐diethylaminoethyl methacrylate (DEAEMA) were prepared in an attempt to investigate the temperature‐induced phase transition and its mechanism. Poly(NPAAm) showed the lower critical solution temperature (LCST) around 24°C in water. With the incorporation of DEAEMA with NPAAm, the LCST change was characterized by an initial increase. However, the LCST was shifted to the lower temperature at the later stage. This might be explained in terms of hydrophilic/hydrophobic contribution of DEAEMA to the LCST. The swelling behavior of copolymer gel in the various solvents and spin‐lattice relaxation time (T1) study by NMR strongly suggested the hydrophilic/hydrophobic contribution of DEAEMA to the LCST depending on the local environment. © 1999 John Wiley & Sons, Inc. A novel amphiphilic thermosensitive poly(ethylene glycol)45b‐poly(methyl methacrylate46co‐3‐(trimethoxysilyl)propyl methacrylate)2b‐poly(N‐isopropylacrylamide)429 (PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429) triblock copolymer was synthesized via consecutive atom transfer radical polymerization techniques. The thermoinduced association behavior of the resulting triblock copolymers in aqueous medium was further investigated in detail by 1H NMR, transmission electron microscopy, and dynamic light scattering. The results showed that at the temperature (25 °C) below the LCST, PEG45b‐P(MMA46co‐MPMA2)‐b‐PNIPAAm429 triblock copolymers self‐assembled into the core crosslinked micelles with the hydrophobic P(MMA‐co‐MPMA) block constructing a dense core, protected by the mixed soluble PEG and PNIPAAm chains acting as a hydrophilic shell simultaneously. With an increase in temperature, the resulting core‐shell micelles converted into a new type of micelles with the hydrophilic PEG chains stretching out from the hydrophobic core through the collapsed PNIPAAm shell. On the other hand, at the temperature (40 °C) above the LCST, such triblock copolymers formed the crosslinked vesicles with the hydrophobic PNIPAAm and P(MMA‐co‐MPMA) blocks constructing a membrane core and the soluble PEG chains building the hydrophilic lumen and the shell. On further decreasing the temperature, the resulting vesicles underwent transformation from the shrunken to the expanded status, leading to the formation of swollen vesicles with enlarged size. This study is believed to present the first formation of two types of hybrid crosslinked self‐assemblies by thermoinduced regulation. © 2011 Wiley Periodicals, Inc. A series of random copolymers of N‐isopropylacrylamide (NIPAM) and sodium 2‐acrylamido‐2‐methyl‐1‐propanesulphonate (AMPS) was synthesized by free‐radical copolymerization. The content of AMPS in the copolymers ranged from 1.1 to 9.6 mol %. The lower critical‐solution temperature (LCST) of copolymers in water increased strongly with an increasing content of AMPS. The influence of polymer concentration on the LCST of the copolymers was studied. For the copolymers with a higher AMPS content, the LCST decreased faster with an increasing concentration than for copolymers with a low content of AMPS. For a copolymer containing 1.1 mol % of AMPS the LCST dropped by about 3 °C when the concentration increased from 1 to 10 g/L, whereas for a copolymer containing 9.6 mol % of AMPS the LCST dropped by about 10 °C in the concentration range from 2 to 10 g/L. It was observed that the ionic strength of the aqueous polymer solution very strongly influences the LCST. This effect was most visible for the copolymer with the highest content of AMPS (9.6 mol %) for which an increase in the ionic strength from 0.2 to 2.0 resulted in a decrease in the LCST by about 27 °C (from 55 to 28 °C), whereas for the copolymer containing 1.1 mol % of AMPS the LCST decreased only by about 6 °C (from 37 to 31 °C) when the ionic strength increased from 0.005 to 0.3. The reactivity ratios for the AMPS and NIPAM monomer pairs were determined using different methods. Poly(N‐vinylcaprolactam) (PVCL) and poly(oligo(ethylene glycol) methyl ether methacrylate) (POEGMA) are well known for their thermoresponsive behavior in aqueous solutions. Indeed, they display lower critical solution temperatures (LCST) in the physiological range, which makes them interesting for biomedical devices and use in drug delivery systems. Homopolymers of N‐vinylcaprolactam and di(ethylene glycol) methyl ether methacrylate as well as copolymers thereof were synthesized by solution and direct miniemulsion polymerizations. The cloud points of the copolymers in aqueous solution were investigated as a function of temperature, comonomer ratio, and in the presence of model pharmaceutical ingredients. By variation of the comonomer ratio, it was possible to control the cloud point temperature between 26 and 35 °C, which was found to be beneficial to attenuate the effect of the drugs that also altered the cloud points. © 2013 Wiley Periodicals, Inc. Stimuli‐responsive poly[(N‐isopropylacrylamide‐co‐maleic anhydride)‐g‐poly(ethylene oxide)]/poly(ethylene imine) macrobranched macrocomplexes were synthesized by (1) the radical copolymerization of N‐isopropylacrylamide and maleic anhydride with α,α′‐azobisisobutyronitrile as an initiator in 1,4‐dioxane at 65 °C under a nitrogen atmosphere, (2) the polyesterification (grafting) of prepared poly(N‐isopropylacrylamide‐co‐maleic anhydride) containing less than 20 mol % anhydride units with α‐hydroxy‐ω‐methoxy‐poly(ethylene oxide)s having different number‐average molecular weights (Mn = 4000, 10,000, or 20,000), and (3) the incorporation of macrobranched copolymers with poly(ethylene imine) (Mn = 60,000). The composition and structure of the synthesized copolymer systems were determined by Fourier transform infrared, 1H and 13C NMR spectroscopy, and chemical and elemental analyses. The important properties of the copolymer systems (e.g., the viscosity, thermal and pH sensitivities, and lower critical solution temperature behavior) changed with increases in the molecular weight, composition, and length of the macrobranched hydrophobic domains. These copolymers with reactive anhydride and carboxylic groups were used for the stabilization of penicillin G acylase (PGA). The conjugation of the enzyme with the copolymers significantly increased the thermal stability of PGA (three times at 45 °C and two times at 65 °C). © 2003 Wiley Periodicals, Inc. Well‐defined amphiphilic A8B4 miktoarm star copolymers with eight poly(ethylene glycol) chains and four poly(ε‐caprolactone) arms (R‐8PEG‐4PCL) were prepared using “click” reaction strategy and controlled ring‐opening polymerization (CROP). First, multi‐functional precursor (R‐8N3‐4OH) with eight azides and four hydroxyls was synthesized based on the derivatization of resorcinarene. Then eight‐PEG‐arm star polymer (R‐8PEG‐4OH) was prepared through “click” reaction of R‐8N3‐4OH with pre‐synthesized alkyne‐terminated monomethyl PEG (mPEG‐A) in the presence of CuBr/N,N,N′,N″,N″′‐ pentamethyldiethylenetriamine (PMDETA) in DMF. Finally, R‐8PEG‐4OH was used as tetrafunctional macroinitiator to prepare resorcinarene‐centered A8B4 miktoarm star copolymers via CROP of ε‐caprolactone utilizing Sn(Oct)2 as catalyst at 100 °C. These miktoarm star copolymers could self‐assemble into spherical micelles in aqueous solution with resorcinarene moieties on the hydrophobic/hydrophilic interface, and the particle sizes could be controlled by the ratio of PCL to PEG. © 2013 Wiley Periodicals, Inc. Amphiphilic, biocompatible poly(N‐vinylpyrrolidone)‐b‐poly(l ‐lactide) (PVP‐b‐PLLA) block polymers were synthesized at 60 °C using a hydroxyl‐functionalized N,N‐diphenyldithiocarbamate reversible addition–fragmentation chain transfer (RAFT) agent, 2‐hydroxyethyl 2‐(N,N‐diphenylcarbamothioylthio)propanoate (HDPCP), as a dual initiator for RAFT polymerization and ring‐opening polymerization (ROP) in a one‐step procedure. 4‐Dimethylamino pyridine was used as the ROP catalyst for l ‐lactide. The two polymerization reactions proceeded in a controlled manner, but their polymerization rates were affected by the other polymerization process. This one‐step procedure is believed to be the most convenient method for synthesizing PVP‐b‐PLLA block copolymers. A series of well‐defined double‐hydrophilic graft copolymers, consisting of poly(N‐isopropylacrylamide)‐b‐poly(ethyl acrylate) (PNIPAM‐b‐PEA) backbone and poly(2‐(dimethylamino)ethyl acrylate) (PDMAEA) side chains, were synthesized by the combination of single‐electron‐transfer living radical polymerization (SET‐LRP) and atom‐transfer radical polymerization (ATRP). PNIPAM‐b‐PEA backbone was first prepared by sequential SET‐LRP of N‐isopropylacrylamide and 2‐hydroxyethyl acrylate at 25 °C using CuCl/tris(2‐(dimethylamino)ethyl)amine as catalytic system followed by the transformation into the macroinitiator by treating the pendant hydroxyls with 2‐chloropropionyl chloride. The final graft copolymers with narrow molecular weight distributions were synthesized by ATRP of 2‐(dimethylamino)ethyl acrylate initiated by the macroinitiator at 40 °C using CuCl/tris(2‐(dimethylamino)ethyl)amine as catalytic system via the grafting‐from strategy. These copolymers were employed to prepare stable colloidal gold nanoparticles with controlled size in aqueous solution without any external reducing agent. The morphology and size of the nanoparticles were affected by the length of PDMAEA side chains, pH value, and the feed ratio of the graft copolymer to HAuCl4. © 2009 Wiley Periodicals, Inc. A series of well‐defined amphiphilic diblock copolymers consisting of hydrophobic polyisobutylene (PIB) and hydrophilic poly(2‐(diethylamino)ethyl methacrylate) (PDEAEMA) segments was synthesized via the combination of living carbocationic polymerization and reversible addition fragmentation chain transfer (RAFT) polymerization. Living carbocationic polymerization of isobutylene followed by end‐capping with 1,3‐butadiene was first performed at ?70 °C to give a well‐defined allyl‐Cl‐terminated PIB with a low polydispersity (Mw/Mn =1.29). This end‐functionalized PIB was further converted to a macromolecular chain transfer agent for mediating RAFT block copolymerization of 2‐(diethylamino)ethyl methacrylate at 60 °C in tetrahydrofuran to afford the target well‐defined PIB‐b‐PDEAEMA diblock copolymers with narrow molecular weight distributions (Mw/Mn ≤1.22). The self‐assembly behavior of these amphiphilic diblock copolymers in aqueous media was investigated by fluorescence spectroscopy and transmission electron microscope, and furthermore, their pH‐responsive behavior was studied by UV‐vis and dynamic light scattering. © 2014 Wiley Periodicals, Inc.
