The hitherto unreported oxazastibinanes 3 have been synthesized by the sodium borohydride reduction of 3‐phenyl‐1‐arylamino‐3‐oxopropane ( 1 ) and subsequent cyclization of the disodium salt of 3‐phenyl‐1‐arylamino‐3‐hydroxypropane ( 2 ) with R3SbBr2 (R = Ph, p‐tolyl, or mesityl). These compounds have been characterized by elemental analyses, molecular weight determination, and by IR, far IR, 1H, and 13C NMR spectral studies. © 2003 Wiley Periodicals, Inc. The interactions of a series of platinum(II) Schiff base complexes with c‐myc G‐quadruplex DNA were studied. Complex [PtL 1a ] ( 1 a ; H2L 1a =N,N′‐bis(salicylidene)‐4,5‐methoxy‐1,2‐phenylenediamine) can moderately inhibit c‐myc gene promoter activity in a cell‐free system through stabilizing the G‐quadruplex structure and can inhibit c‐myc oncogene expression in cultured cells. The interaction between 1 a and G‐quadruplex DNA has been examined by 1H NMR spectroscopy. By using computer‐aided structure‐based drug design for hit‐to‐lead optimization, an in silico G‐quadruplex DNA model has been constructed for docking‐based virtual screening to develop new platinum(II) Schiff base complexes with improved inhibitory activities. Complex [PtL 3 ] ( 3 ; H2L 3 = N,N′‐bis{4‐[1‐(2‐propylpiperidine)oxy]salicylidene}‐4,5‐methoxy‐1,2‐phenylenediamine) has been identified with a top score in the virtual screening. This complex was subsequently prepared and experimentally tested in vitro for its ability to stabilize or induce the formation of the c‐myc G‐quadruplex. The inhibitory activity of 3 (IC50=4.4 μM ) is tenfold more than that of 1 a . The interaction between 1 a or 3 with c‐myc G‐quadruplex DNA has been examined by absorption titration, emission titration, molecular modeling, and NMR titration experiments, thus revealing that both 1 a and 3 bind c‐myc G‐quadruplex DNA through an external end‐stacking mode at the 3’ terminal face of the G‐quadruplex. Such binding of G‐quadruplex DNA with 3 is accompanied by up to an eightfold increase in the intensity of photoluminescence at λmax=652 nm. Complex 3 also effectively down‐regulated the expression of c‐myc in human hepatocarcinoma cells.  相似文献

Synthesis and Structure of Highly Functionalized 2, 3‐Dihydro‐1H‐1, 3, 2‐diazaboroles A series of differently substituted 2, 3‐dihydro‐1H‐1, 3, 2‐diazaboroles has been prepared by various methods. 1, 3‐Di‐tert‐butyl‐2‐trimethylsilylmethyl‐1H‐1, 3, 2‐diazaborole ( 7 ), 2‐isobutyl‐1, 3‐bis(1‐cyclohexylethyl)‐1H‐1, 3, 2‐diazaborole ( 8 ), 1, 3‐bis‐(1‐cyclohexylethyl)‐2‐trimethylsilylmethyl‐1H‐1, 3, 2‐diazaborole ( 9 ) 1, 3‐bis(1‐methyl‐1‐phenyl‐propyl)‐2‐trimethylsilylmethyl‐1H‐1, 3, 2diazaborole ( 10 ) and 2‐bromo‐1, 3‐bis(1‐methyl‐1‐phenyl‐propyl)‐1H‐1, 3, 2‐diazaborole ( 11 ) were formed by reaction of the corresponding 1, 4‐diazabutadienes with the boranes Me3SiCH2BBr2, iBuBBr2 and BBr3 followed by reduction of the resulting borolium salts [R1 = tBu, Me(cHex)CH, [Me(Et)Ph]C; R2 = Me3SiCH2, iBu, Br] with sodium amalgam. Treatment of 11 and 12 with silver cyanide afforded the 2‐cyano‐1, 3, 2‐diazaboroles 13 and 14 . An alternative route to compound 8 is based on the alkylation of 2‐bromo‐1, 3, 2‐diazaborole 12 with isobutyllithium. Equimolar amounts of 13 and isobutyllithium give rise to the formation of 15 . The new compounds were characterized by 1H‐, 13C‐, 11B‐NMR, IR and mass spectra. The molecular structures of 7 and meso ‐10 were confirmed by x‐ray structural analysis.  相似文献

Zinc complexes supported by tertiary 1,3,5‐triazapenta‐1,3‐dienate ligand (L1) and N ‐benzoyl‐N′ ‐arylbenzamidinate [aryl =2,6‐diisopropylphenyl (L2), phenyl (L3)] ligands have been synthesized and characterized. The reaction of L1H with ZnEt2 affords a mononuclear zinc complex [L1ZnEt] ( 1 ) in good yield. Tetra nuclear zinc complex [(L1)2Zn4O(OAc)4] ( 2 ) is prepared by treating L1H with one equivalent of Zn(OAc)2 in toluene. Further, dinuclear zinc complexes [L2ZnEt]2 ( 3 ) and [L3ZnEt]2 ( 4 ) are obtained in good yields from L2H and L3H with ZnEt2 in toluene respectively. The complexes 1–4 have been characterized by 1H/13C NMR spectroscopy and single crystal X‐ray diffraction studies. All of the complexes have been explored for their catalytic activity toward the ring‐opening polymerization (ROP) of ε ‐caprolactone. It has been found that complex 1 is an active catalyst for the polymerization of ε ‐caprolactone in presence of a cocatalyst benzyl alcohol (BnOH). While complex 2 is as active as 1 there is no need for a cocatalyst for the polymerization to proceed. Dinuclear zinc complexes 3 and 4 show very high activity for the ROP of ε ‐caprolactone (CL) and rac ‐lactide (LA) without requiring a cocatalyst. The resultant polymers are found to have very high molecular weight (M n = 296 X 103 g mol−1) and relatively narrow polydispersity index compared to 1 and 2 .  相似文献

The complex poly[[aqua(μ2‐phthalato‐κ2O1:O2){μ3‐2‐[3‐(pyridin‐2‐yl)‐1H‐pyrazol‐1‐yl]acetato‐κ4N2,N3:O:O′}{μ2‐2‐[3‐(pyridin‐2‐yl)‐1H‐pyrazol‐1‐yl]acetato‐κ3N2,N3:O}dizinc(II)] dihydrate], {[Zn2(C10H8N3O2)2(C8H4O4)(H2O)]·2H2O}n, has been prepared by solvothermal reaction of 2‐[3‐(pyridin‐2‐yl)‐1H‐pyrazol‐1‐yl]acetonitrile (PPAN) with zinc(II). Under hydrothermal conditions, PPAN is hydrolyzed to 2‐[3‐(pyridin‐2‐yl)‐1H‐pyrazol‐1‐yl]acetate (PPAA). The structure determination reveals that the complex is a one‐dimensional double chain containing cationic [Zn4(PPAA)4]4+ structural units, which are further extended by bridging phthalate ligands. The one‐dimensional chains are extended into a three‐dimensional supramolecular architecture via hydrogen‐bonding and π–π stacking interactions.  相似文献

The synthesis of the reactive PN(CH) ligand 2‐di(tert‐butylphosphanomethyl)‐6‐phenylpyridine ( 1H ) and its versatile coordination to a RhI center is described. Facile C?H activation occurs in the presence of a (internal) base, thus resulting in the new cyclometalated complex [RhI(CO)(κ3P,N,C‐ 1 )] ( 3 ), which has been structurally characterized. The resulting tridentate ligand framework was experimentally and computationally shown to display dual‐site proton‐responsive reactivity, including reversible cyclometalation. This feature was probed by selective H/D exchange with [D1]formic acid. The addition of HBF4 to 3 leads to rapid net protonolysis of the Rh?C bond to produce [RhI(CO)(κ3P,N,(C?H)‐ 1 )] ( 4 ). This species features a rare aryl C?H agostic interaction in the solid state, as shown by X‐ray diffraction studies. The nature of this interaction was also studied computationally. Reaction of 3 with methyl iodide results in rapid and selective ortho‐methylation of the phenyl ring, thus generating [RhI(CO)(κ2P,N‐ 1Me )] ( 5 ). Variable‐temperature NMR spectroscopy indicates the involvement of a RhIII intermediate through formal oxidative addition to give trans‐[RhIII(CH3)(CO)(I)(κ3P,N,C‐ 1 )] prior to C?C reductive elimination. The RhIIItrans‐diiodide complex [RhI(CO)(I)23P,N,C‐ 1 )] ( 6 ) has been structurally characterized as a model compound for this elusive intermediate.  相似文献

The synthesis, characterization and cytotoxic properties in vitro of tri‐n‐butyltin 1‐(4‐chlorobenzoyl)‐5‐methoxy‐2‐methyl‐1H‐indole‐3‐acetate ( 1 ), tri‐phenyltin 1‐(4‐chlorobenzoyl)‐5‐methoxy‐2‐methyl‐1H‐indole‐3‐acetate ( 2 ), tetra‐n‐butyltin[bis‐1‐(4‐chlorobenzoyl)‐5‐methoxy‐2‐methyl‐1H‐indole‐3‐acetato]distannoxane ( 3 ) and di‐n‐butyltin bis‐1‐(4‐chlorobenzoyl)‐5‐methoxy‐2‐methyl‐1H‐indole‐3‐acetate ( 4 ) are described. These compounds have been characterized by 1H, 13C and 119Sn NMR spectroscopy in solution and 119Sn NMR in the solid state, infrared spectroscopy, elemental analysis and X‐ray diffraction for compound 1 . The growth inhibition effects of compounds 1–4 against the lung adenocarcinoma cell line SK‐LU‐1 as well as the cervical cancer cell line HeLa were determined. Fluorescence properties of four based derivatives [An] (where n = 1–4) and their Cu(II) heterochelates of the type [Cu(An)(CQ)(OH)]?xH2O {where A1 = 3‐(2‐oxo‐2H‐chromen‐3‐yl)‐4H‐furo[3,2‐c]chromen‐4‐one, A2 = 8‐methyl‐3‐(2‐oxo‐2H‐chromen‐3‐yl)‐4H‐furo[3,2‐c]chromen‐4‐one, A3 = 6‐methyl‐3‐(2‐oxo‐2H‐chromen‐3‐yl)‐4H‐furo[3,2‐c]chromen‐4‐one, A4 = 8‐chloro‐3‐(2‐oxo‐2H‐chromen‐3‐yl)‐4H‐furo[3,2‐c]chromen‐4‐one and x = 3, 2, 4, 1} were studied at room temperature. The fluorescence spectra of heterochelates show red shift, which may be due to the chelation by the ligands to the metal ion. It enhances ligand ability to accept electrons and decreases the electron transition energy. The kinetic parameters such as order of reaction (n), energy of activation (Ea), entropy (ΔS#), pre‐exponential factor (A), enthalpy (ΔH#) and Gibbs free energy (ΔG#) have been reported. The antimicrobial activity of Clioquinol and Cu(II) heterochelates have been determined and described. All the heterochelates showed a more effective antimicrobial activity than the free ligand. Synthesis of a series of cyclic fused‐isoxazolines has been accomplished by regioselective and diastereoselective 1,3‐dipolar cycloaddition of 3‐methylindan‐1‐one enamines ( 1a , 1b , 1c ) and 3‐phenylindan‐1‐one enamines ( 2a , 2b , 2c ) to arylnitrile oxides ( 3d , 3e , 3f , 3g , 3h ). The structure of the cycloadducts was elucidated by 1H and 13C NMR spectroscopy. The proposed regio‐ and stereochemistry of fused‐compounds ( 4 ) and ( 5 ) has also been corroborated by two single‐crystal X‐ray diffraction studies carried out on 4‐methyl‐8b‐morpholinyl‐3‐(p‐tolyl)‐4H‐3a,8b‐dihydroindeno[2,3‐d]isoxazoline ( 4be ) and 3‐(p‐anisyl)‐4‐phenyl‐8b‐pyrrolidinyl‐4H‐3a,8b‐dihydroindeno[2,3‐d]isoxazoline ( 5af ) and by means of density functional theory calculations.  相似文献

Water‐soluble cationic alkynylplatinum(II) 2,6‐bis(benzimidazol‐2′‐yl)pyridine (bzimpy) complexes have been demonstrated to undergo supramolecular assembly with anionic polyelectrolytes in aqueous buffer solution. Metal–metal‐to‐ligand charge transfer (MMLCT) absorptions and triplet MMLCT (3MMLCT) emissions have been found in UV/Vis absorption and emission spectra of the electrostatic assembly of the complexes with non‐conjugated polyelectrolytes, driven by Pt???Pt and π–π interactions among the complex molecules. Interestingly, the two‐component ensemble formed by [Pt(bzimpy‐Et){C?CC6H4(CH2NMe3‐4)}]Cl2 ( 1 ) with para‐linked conjugated polyelectrolyte (CPE), PPE‐SO3?, shows significantly different photophysical properties from that of the ensemble formed by 1 with meta‐linked CPE, mPPE‐Ala. The helical conformation of mPPE‐Ala allows the formation of strong mPPE‐Ala– 1 aggregates with Pt???Pt, electrostatic, and π–π interactions, as revealed by the large Stern–Volmer constant at low concentrations of 1 . Together with the reasonably large Förster radius, large HOMO–LUMO gap and high triplet state energy of mPPE‐Ala to minimize both photo‐induced charge transfer (PCT) and Dexter triplet energy back‐transfer (TEBT) quenching of the emission of 1 , efficient Förster resonance energy transfer (FRET) from mPPE‐Ala to aggregated 1 molecules and strong 3MMLCT emission have been found, while the less strong PPE‐SO3?– 1 aggregates and probably more efficient PCT and Dexter TEBT quenching would account for the lack of 3MMLCT emission in the PPE‐SO3?– 1 ensemble.  相似文献

1,3‐Dipolar cycloaddition reactions of N‐cyclohexyl maleimide ( 1 ) with azomethine N‐oxide ( 2 ) have afforded novel isoxazolidine ( 3 ) in excellent yield. Their structures have been characterized from their IR, 1H‐NMR, 13C‐NMR, 1H,1H‐COSY, MS(ESI), and elemental analysis techniques. In vitro antibacterial activity of the synthesized compounds were investigated against a representative panel of pathogenic strains specifically two Gram‐positive bacteria (Staphylococcus aureus and Streptococcus pyogenes ) and two Gram‐negative bacteria (Pseudomonas aeruginosa and Escherichia coli ) using agar‐well diffusion assay. Some of the compounds ( 3a , 3k , 3n , and 3o ) exhibited promising antibacterial activities. All the synthesized compounds have also been screened for their antioxidant activities and were found to be significantly active.  相似文献

A new cyclopeptide, clausenain I ( 1 ), has been isolated by a multi‐step chromatography procedure from Clausena anisum‐olens. Its structure was elucidated as cyclo (‐Gly1‐Ile2‐Ile3‐Val4‐Leu5‐Ile6‐Ile7‐Leu8‐Leu9‐) by extensive 2D‐NMR spectroscopic methods and chemical evidence. It is the first time that a natural cyclic peptide has been isolated from the genus Clausena.  相似文献

A series of new pyrazolone and pyrazole derivatives with expected antifungicidal activity have been prepared through the reactions 3‐phenyl‐1‐H‐pyrazol‐5(4H)‐one ( 3 ) and 4‐(dimethylaminomethylene)‐3‐phenyl‐1H‐pyrazol‐5(4H)‐one ( 5 ) with a variety of electrophilic reagents and nucleophilic reagents. The reactive 1 : 1 adducts in the reaction between Ph3P and dialkyl acetylenedicarboxylates have been trapped with ‘tosylmethyl isocyanide’ (TsMIC ; 1 ) to yield dialkyl 2‐[(4‐methylphenyl)sulfonyl]‐1H‐pyrrole‐3,4‐dicarboxylates 3 (Scheme 1). The structures of the highly functionalized compounds 3 were corroborated spectroscopically (IR, 1H‐ and 13C‐NMR, and EI‐MS) and by elemental analyses. A plausible mechanism for this type of cyclization is proposed (Scheme 2).  相似文献

A series of 3‐fluoropyrrolidines have been studied to investigate the influence of the stereoelectronic fluorine gauche effect on ring conformations in the solid state by single‐crystal X‐ray analysis and in solution phase by NMR spectroscopy. As part of these studies 1D 19F–1H heteronuclear NOE (HOESY) experiments have been optimised for applications to small molecules and are described in detail. These have been employed to estimate 19F–1H internuclear distances and were combined with vicinal 3J(F,H) and 3J(H,H) scalar coupling constants to analyse the ring conformations. Where possible the derived solution‐phase structural data have been compared with those of the crystalline state. The results demonstrate the influence of the gauche effect in stabilising Cγexo conformations of the fluorinated pyrrolidines. It was further shown that when steric interactions were also present, this conformational bias was diminished and the contribution of the alternative Cγendo conformation was seen to increase in solution at lower sample temperatures.  相似文献

1‐Phosphabicyclo[2.2.1]heptanes Exo‐endo‐ and exo‐exo‐2.6‐dimethyl‐1‐phosphabicyclo [2.2.1]heptane have been obtained by cyclization of 2‐methyl‐4‐(2‐propenyl)phospholane in the presence of the complex base, sodium salt of diethylenglycolmonoethylether ‐ sodium amide in THF (NAMEDEG). The bicyclic phosphanes are characterized by reac‐tions with selenium, sulfur, (CH3)2SeO, CH3I and HSO3F, respectively, elemental analysis, X‐ray crystal structure analysis as well as 1H, 13C, 31P NMR spectral measurements. The steric demand of these phosphanes as complex ligands has been estimated from the P, H coupling constants of the phosphonium fluorosulphates according to the Tolman model. The phosphane selenides were found to display the lowest values for the 1J(Se, P) coupling constant, found up to now for alicyclic and cyclic aliphatic tertiary phosphane selenides. The nJ(P, H)‐ and nJ(H, H)n=2, 3 coupling constants have been extracted from the proton spectra at 600 MHz by computerized analysis.  相似文献

Several derivatives of the new pyrimido[4′,5′:3,4]pyrazolo[1,2‐b]phthalazine‐4,7,12‐trione ring system have been prepared by the reaction of 3‐amino‐1‐aryl‐5,10‐dioxo‐5,10‐dihydro‐1H‐pyrazolo[1,2‐b]phthalazine‐2‐carbonitriles with aliphatic carboxylic acids in the presence of phosphoryl chloride (POCl3). The title compounds, 7‐aryl‐5,6‐dihydro‐14‐aza[1]benzopyrano[3,4‐b]phenanthren‐8H‐ones 3a , 3b , 3c , 3d , 3e , 3f , 3g , 3h , 3i , 3j , 3k , 3l have been synthesized by reacting various 4‐hydroxy coumarins 1a , 1b , 1c with 2‐arylidene‐1‐tetralones 2a , 2b , 2c , 2d in the presence of ammonium acetate and acetic acid under Krohnke's reaction condition. The structures of all the synthesized compounds were supported by analytical, IR, 1H‐NMR, and 13C‐NMR data. All the synthesized compounds 3a , 3b , 3c , 3d , 3e , 3f , 3g , 3h , 3i , 3j , 3k , 3l have been screened for their antibacterial activities against Escherichia coli (Gram ?ve bacteria), Bacillus subtilis (Gram +ve bacteria), and antifungal activity against Candida albicans (Fungi).
