A highly selective synthesis of 2‐substituted benzimidazole derivatives from the reaction of o‐phenylendiamine derivatives and aromatic aldehydes in the presence of an organic salt, NH4OAc, in absolute ethanol is presented. The products were obtained by evaporating the solvent followed by a simple recrystallization with excellent yields.  相似文献

Abstract

The synthesis of β-cyclodextrin derivatives bearing one phosphate group on the primary rim is reported. These compounds were prepared in good to excellent yields, by reacting β-cyclodextrin with dialkyl chlorophosphates in the presence of 4-dimethyl amino pyridine (DMAP) catalyst and dimethylformamide (DMF) as solvent. The methodology described is highly selective and the purification of the title compounds is simple, because difficulties due to phosphate regioisomers mixture are avoided.  相似文献

Different novel phthalazino[2,3‐b]phthalazine‐5,7,12,14‐tetraones were synthesized in a simple and environmentally benign method from the reaction of phthalic anhydrides with semicarbazide or thiosemicarbazide using montmorillonite K‐10 clay as solid heterogeneous acidic catalyst and microwaves under solvent‐free conditions in good yields and short reaction times. Products were characterized by the elemental analysis, IR, NMR, and mass spectrometry.  相似文献

A facile synthetic route for the synthesis of bisphosphonates in phenols is described. Preparations of some of bisphosphonates, which are presently in clinical use such as risedronic acid and alendronate sodium, are synthesized following this new, simple method. This procedure can be useful for the synthesis of this class of bone‐resorptive inhibitors in bulk quantities.  相似文献

The synthesis of the aliphatic subunit 9 of the macrolide LL‐Z1640‐2 (I), starting from a 4‐deoxy‐D‐mannose derivative 2a, is described. The procedure includes the first successful application of a Vasella ring opening reaction for a 4‐deoxypyranoside. Nucleophilic addition of an alkynyllithium reagent to the aldehyde 4 led to the propargylic alcohol 7, which was converted to the advanced building block 9 in two further steps.  相似文献

A synthesis of N‐protected β‐aminomalonates starting from α‐amidosulfones under very mild and simple reaction conditions is described. Treatment of the sulfones with malonate esters in the presence of 2.5 equivalents of potassium carbonate affords the desired products in good yield. A variety of N‐Z and N‐Boc protected aliphatic and aromatic α‐aminosulfones and malonate esters have been successfully used as starting materials. Hydrolysis with concomitant decarboxylation of the N‐protected β‐aminomalonates provides a convenient access to racemic β‐amino acids.  相似文献

A convenient and efficient method for the synthesis of 1,5‐benzodiazepines by the reaction of o‐phenylenediamines (OPDA) and ketones with hydrogens at the α‐position has been developed. A new method for synthesis of 3-azido-3-deoxyxylofuranoside was described. D-ribose was methylated, then treated with methanesulfonyl chloride to give Ⅱ.Ⅱ was selectively replaced by benzoate at the 5-position to give Ⅲ. All these processes gave good yields. Ⅲ was treated with sodium azide to afford Ⅳ, whose structure was determined by ^1H NMR and ^13C NMR.  相似文献

A novel synthesis of 2,3,4‐tri‐O‐methyl‐D‐xylopyranose, 4, and its oxidation product 2,3,4‐tri‐O‐methyl‐D‐xylono‐1,5‐lactone, 5, are reported. The new synthesis applies a regioselective Wittig‐like reaction of tetra-O-acetyl-D-xylopyranase, 1, with triphenylphosphine and carbon tetrachloride to yield an O‐dichlorovinyl xyloside protected at C‐1, 2. The protecting group facilitates the permethylation of xylose and is removed under the methylation conditions, to yield tetra-O-acetyl-D-xylopyranase, 3. The anomeric methyl group was removed under mildly acidic conditions to give 2,3,4‐tri‐O‐methyl‐D‐xylopyranose, 4, in good yield. Compound 4 was oxidized using pyridinium chlorochromate to give the title compound, 5, in 95% yield.  相似文献

2‐Aza‐1,3‐butadienes have been synthesized from carbonyl compounds and 1,1,1,3,3,3‐hexamethyl‐disilazane in the presence of cobalt‐containing catalysts. The best yields (up to 95%) were achieved in the case of aldehydes branched in the α-position and 2-methylcyclohexanone. In the case of two α,β‐unsaturated ketones, pyridine derivatives were found as the main products.  相似文献

The ketoesters 3 and 4, obtained by the condensation of 2‐cyclohexanone carboxylate and 1‐chloro‐3‐pentanone, were heated with 2,3‐dichloro‐5,6‐dicyanobenzoquinone (DDQ) to yield the dienones 5 and 6, which on hydrolysis with potassium t‐butoxide and dimethyl sulfoxide afforded tetralin 8. These were converted to tetralone 10 by methylation and oxidation respectively. Further methylation of 10 yielded tetralone 11.  相似文献

Abstract

Isopropyl 6‐O‐acetyl‐3‐deoxy‐4‐S‐ethyl‐4‐thio‐α‐Dthreo‐hexopyranosid‐2‐ulose (3) was converted to the corresponding 3‐[bis(methylthio)methylene] derivative 4 with a push–pull activated C–C double bond. Treatment of 4 with hydrazine and methylhydrazine afforded the pyrano[3,4‐c]pyrazol‐5‐ylmethyl acetates 5a and 5b, respectively. Desulfurization of compound 4 with sodium boron hydride yielded the 3‐[(methylthio)methylene]hexopyranosid‐2‐ulose 7. Compound 7 was reacted with amines to furnish 3‐aminomethylene‐hexopyranosid‐2‐uloses 8, 9. Reaction of 7 with hydrazine hydrate, hydrazines, hydroxylamine, and benzamidine afforded the pyrazolo, isoxazalo, and pyrimido anellated pyranosides (1013).  相似文献

Treatment of 2‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)ethanal (3) with malononitrile in the presence of aluminium oxide provided 2‐cyano‐4‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)crotononitrile (4). Starting from 4, cyclization with sulphur and triethylamine yielded 2‐amino‐5‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)thiophene‐3‐carbonitrile (5). Further cyclization could be achieved with triethyl orthoformate/ammonia to furnish 4‐amino‐6‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)thieno[2.3‐d]pyrimidine (8).  相似文献

A simple, efficient, and general method has been developed for the synthesis of coumarin derivatives through a one‐pot reaction of aromatic aldehydes, 2,2‐dimethyl‐1,3‐dioxane‐4,6‐dione and 5,5‐dimethyl‐1,3‐cyclohexanedione, in the presence of TEBA under solvent‐free conditions using grinding methods. The mild reaction conditions, simple protocol, high yields, and cleaner reaction make this protocol practical and economically attractive.  相似文献

An efficient hydroxylation at the 5‐position of the C ring of camptothecins was accomplished with the complex of CuI and organic amines as catalyst in the presence of oxygen at room temperature in dimethyl formate (DMF). To be successfully hydroxylated, the insoluble camptothecin analogue was transformed to the corresponding 20 carbonates.  相似文献
