Silica Gel,an Effective Catalyst for the Reaction of Electron-Deficient Nitro-Olefins with Nitrogen Heterocycles

The reaction of electron-deficient olefins with nitrogen heterocycles such as pyrrole and indole was examined in the presence of silica gel at room temperature under stirring at solvent-free conditions. It was found that silica gel is an effective catalyst for this conjugate addition. This work resulted in the formation of monosubstituted pyrroles selectively as a major product except in a few cases where disubstituted pyrroles were also formed as side products. Thus, a simple, rapid, efficient, environmentally benign, and solvent-free method was established.     

Peptide-Catalyzed Stereoselective Conjugate Addition Reaction of Aldehydes to C-Substituted Maleimides

Catalytic stereoselective additions with maleimides are useful one-step reactions to yield chiral succinimides, molecules that are widespread among therapeutically active compounds but challenging to prepare when the maleimide is C-substituted. We present the tripeptide H-Pro-Pro-Asp-NHC₁₂H₂₅ as a catalyst for conjugate addition reactions between aldehydes and C-substituted maleimides to form succinimides with three contiguous stereogenic centers in high yields and stereoselectivities. The peptidic catalyst is so chemoselective that no protecting group is needed at the imide nitrogen of the maleimides. Derivatization of the succinimides was straightforward and provided access to chiral pyrrolidines, lactones, and lactams. Kinetic studies, including a Hammett plot, provided detailed insight into the reaction mechanism.

Thermal and Microwave‐Assisted Conjugate Additions of Indole on Electron‐Deficient Nitro‐olefins

Conjugate addition of indole on nitro‐olefins was carried out by three different methods: (i) thermal heating in a sealed tube, (ii) reaction with indolyl magnesium iodide, and (iii) microwave irradiation. Comparison of the results indicated that the microwave technique is most efficient.

以乙酰乙酸乙酯为原料,经缩合、氧化、还原和氯代等反应合成了3-甲基-5-碘甲基-1H-吡咯-2,4-二(甲酸乙酯)(5);5分别与乌洛托品,吡啶和三甲胺经取代反应合成了3个吡咯季铵盐6a~6c,其中5和6a~6c均为新化合物,其结构经1H NMR,IR和ESI-MS表征。     

Microwave-Assisted Synthesis,Biological Activity Evaluation,Molecular Docking,and ADMET Studies of Some Novel Pyrrolo [2,3-b] Pyrrole Derivatives

Novel pyrrolo [2,3-b] pyrrole derivatives were synthesized and their hypolipidemic activity was assessed in hyperlipidemic rats. The chemical structures of the new derivatives were confirmed through spectral analysis. Compounds 5 and 6 were revealed to be the most effective hypolipidemic agents, with considerable hypocholesterolemic and hypotriglyceridemic effects. They appear to be promising candidates for creating new powerful derivatives with anti-atherosclerotic and hypolipidemic properties. As for antimicrobial activity, some of the tested compounds showed moderate activity against Pseudomonas aeruginosa: compound 2 revealed an MIC value of 50 μg/mL, compared to 25 μg/mL for ciprofloxacin. Compound 3 showed good antimicrobial activity against Staphylococcus aureus, comparable to ciprofloxacin, and roughly half the activity of ampicillin, according to MIC values. Compound 2 has an MIC approximately 25% of that of clotrimazole against Candida albicans. Compound 2 also showed the highest antioxidant activity with 59% inhibition of radical scavenging activity. Additionally, the cytotoxic activity of these new derivatives 1–7 was investigated and most of them showed good anticancer activity against the three tested cell lines.     

The Oxidation of Pyrrole

The dearomatization of heterocycles has been a powerful means for producing functional molecules in synthesis. In the case of pyrroles, reductive methods (such as the Birch reduction) have been most widely exploited, while oxidative methods are generally dismissed as too difficult or unpredictable to be useful. However, since the early twentieth century considerable research has been carried out on the controlled oxidation of pyrroles to give highly functionalized products, using a variety of oxidants. This review presents a summary of all work up until the present day in the area of pyrrole oxidation, looking at the use of peroxide, singlet oxygen, hypervalent iodine reagents, a range or organic and inorganic oxidants, and electrochemical approaches. It also offers some perspective on the potential future role of pyrrole oxidation in synthesis.

以乙酰乙酸苄酯(4)为原料, 经Knorr缩合制备了2,4-二甲基-3-丙酸甲酯基-5-羧酸苄酯基-1-氢吡咯 (2) 和2,4-二甲基-3-羧酸甲酯基-5-羧酸苄酯基-1-氢吡咯(3)。乙醚介质中,溴水氧化条件下,吡咯 (2) 发生自身缩合生成二吡咯甲烷 (7),吡咯 (3)无反应发生。在Pb(OAC)₄氧化条件下,当其浓度为吡咯 (3)浓度的2倍时,在80 ℃,吡咯 (3) 完全转华。所得产物经HCl甲醇溶液回流,以82%的产率制备了新型2-甲氧基甲基-3-羧酸甲酯基-4-甲基-5-羧酸苄酯基-1氢吡咯 (1)。吡咯(1)、(2)、(3)及二吡咯(7)的结构用核磁、元素分析、质谱和红外等测试技术进行了表征。吡咯(1)的构建对进一步研究吡咯构效关系具有一定参考价值。     

Phosphorus-containing Derivatives of Indole and Pyrrole. (Review)

Data on the synthesis and chemical properties of the phosphorus-containing derivatives of indoles and pyrroles, published in 1976-1998, are reviewed. The introduction of substituents containing tri- and tetracoordinated phosphorus atom into the heterocycle and the side chain is discussed. Information on the biological activity is presented.     

Efficient methodology for alkylation of vinylnitroso compounds with carbon nucleophiles

A diverse array of nitrosoalkenes derived from both acyclic and cyclic ketones, as well as aldehydes, via the Denmark protocol using α-chloro-O-TBS-oximes can be trapped efficiently in situ by a wide variety of potassium ester enolates to afford conjugate addition products in good yields.     

Microwave-Assisted Michael Addition of Diethyl Malonate with Ferrocenyl Substituted Chalcones Under Solvent-Free Conditions

The article describes an efficient, economical, and environmentally friendly approach for Michael addition of diethyl malonate to ferrocenyl substituted chalcones in the presence of microwave irradiation under solvent-free conditions, affording the corresponding Michael adducts in moderate to good yields of 53–94%.

Pyrrole thin films deposited on paper by pulsed RF plasma

Attempts were made to plasma deposit an oriented π-conjugated polymer of pyrrole (Py) on paper surfaces in order to produce electrically conductive layers. The N/C atomic ratio of 0.13–0.24 was observed for all treatment conditions. This implies the nature of the deposition formed on the paper surface via pulsed plasma is different from that of pyrrole monomer. An increase in conductivity of all pyrrole-plasma treated papers was observed. The 50 W RF-power with 5 min plasma exposed paper sample shows 8.15 × 10⁻⁹ S·cm⁻¹ conductivity. The conductivity measurements indicated a plasma-enhanced ring-opening reaction mechanism of pyrrole.      

Stereodivergent Construction of 1,3-Chiral Centers via Tandem Asymmetric Conjugate Addition and Allylic Substitution Reaction

Herein, we report a synthesis of cyclohexanones bearing multi-continuous stereocenters by combining copper-catalyzed asymmetric conjugate addition of dialkylzinc reagents to cyclic enones with iridium-catalyzed asymmetric allylic substitution reaction. Good to excellent yields, diastereoselectivity and enantioselectivity can be obtained. Unlike the stereodivergent construction of adjacent stereocenters (1,2-position) reported in the literature, the current reaction can achieve the stereodivergent construction of nonadjacent stereocenters (1,3-position) by a proper combination of two chiral catalysts with different enantiomers.     

Microwave-Assisted Synthesis of Disulfides

Abstract

A new microwave-assisted synthesis methodology for the preparation of substituted disulfide derivatives is presented. 4-Substituted sulfenimides were reacted with 4-substituted thiols under neat (to right doughy consistency) conditions in chloroform, with both microwave heating and conventional methods. The resulting 4-substituted disulfide derivatives were obtained at higher yields and in shorter reaction times with microwave heating. Their chemistry was confirmed by ¹H-NMR, ¹³C-NMR, infrared (IR), and elemental analysis.

A Facile Procedure for Conjugate Addition of Amines to Electron Deficient Alkenes with Metal Oxide as Catalyst

A novel efficient procedure has been developed for the conjugate addition of amines to electron deficient alkenes. A series of metal oxides was synthesized for catalyzing the conjugate addition of amines and alkenes. After optimizing the reaction conditions, SrO was chosen as the most efficient catalyst for the reactions. The results show that the catalyst is very efficient for the conjugate addition of amines to electron deficient alkenes with the excellent yields in several minutes. Operational simplicity...     

芳基吡咯衍生物的合成及其发光性质

本文通过Sonogashira偶联反应合成了一类含有苯甲酸甲酯基团的吡咯衍生物MB3PE2和MB3,其结构为典型的给体（D,吡咯）-受体（A,苯甲酸甲酯基）结构,该类结构表现为可调的荧光性质。通过系统研究它们在各个状态下的发光性质,阐明了结构与性能的关系。实验结果表明,共轭基团的加入对D-A共轭结构的化合物发光性质影响明显,且不同状态下的影响程度显著不同。MB3PE2具有π-T-型D-A结构而MB3是T-型D-A结构。它们在溶液状态下都具有明显的溶剂效应,但是其溶液态的荧光量子产率明显不同,π-T-型D-A结构的MB3PE2是T-型MB3的2.6倍。该结果对设计溶液态下高量子效率的D-A荧光化合物具有一定的指导意义。     

Plasma Copolymerization of Pyrrole with Si-Compounds

The plasma copolymerization of pyrrole with hexamethyldisiloxane, hexamethyldisilane and hexamethyldisilazane was investigated with regard to the polymer deposition characteristics and the properties of the product films. Deposition rates were evaluated by monitoring the weight increase of the deposits over time, and the effect of varying the experimental set up parameters was determined. The results of deposition rates and quantitative IR analysis suggested that the increasing amounts of Si-components were incorporated into the polymer matrix as the supply ratio of the Si-monomer was increased. The Si-component in the copolymer brought some dramatic effects on the physical properties of the films and changed the hydrophobicity of the surfaces, as reflected in the advancing and receding contact angle measurements. The experimental results obtained are discussed in terms of the contribution of the polar and dispersion components of surface energy.     

Nucleophilic Addition of Highly Hindered Amines to Electron-Deficient Acetylenes

Even the most highly hindered amines such as diisopropylamine, isopropyl-t-butylamine, 2,2,6,6-tetramethylpiperidine, 4-oxo-2,2,6,6-tetramethyl piperidine and hexamethyldisilazane react with electron-deficient alkynes to give the Michael addition products.
