4-Methylthiopyrido[1,2-α]pyrimidin-2-one and 2-hydroxypyrido[1,2-α]pyrimidine-4-thione derivatives were synthesized by the addition ofN-(4-R-pyrid-2-yl)acetoacetamides (R = H, Me) to CS2 under phase-transfer conditions followed by the alkylation of the reaction products with Mel. The molecular structure of 3-acetyl-4-methylthiopyrido[1,2-α]pyrimidin-2-one is established by X-ray analysis.  相似文献

A direct and efficient approach to synthesize imidazo[1,2-α]pyridines through three-component one-pot reaction of 2-aminopyridine,aldehyde and terminal alkyne catalyzed by Cu-nanoparticles under solvent-free conditions has been developed.This method provides a rapid access to substituted imidazo[1,2-α]pyridines with good yields(up to 85%).  相似文献

Polychlorinated imidazo[1,2-α]pyridines were synthesized as analogs of certain chlorinated benzimidazoles. The imidazo[1,2-α]pyridines were obtained by a condensation of ethyl bromoacetate and chlorinated 2-aminopyridines. These condensation products were treated with an ion exchange resin to effect an exchange of hydrobromide salts with hydrochloride salts. These compounds were subsequently treated with POCl3 to convert the imidazo[1,2-α]pyridin-2-ones into 2-chloroimidazo[1,2-a]pyridines.  相似文献

The reaction of 2-amino-1-(2-propynyl)pyridinium bromide with various iodobenzenes, catalyzed by Pd-Cu, in the presence of sodium lauryl sulfate as the surfactant and cesium carbonate as the base, in water, leads to the formation of 2-substituted imidazo[1,2-α]pyridines.  相似文献

Structure containing the CS bond exist widely in nature, drugs and chemical materials. Here, a novel sulfenylation protocol employing an aryl or alkyl halide and odorless and cheap S as reagents was developed, generating regioselective alkyl-S- and Ar-S-substituted imidazol[1,2-α]pyridine derivatives in good yields under relatively environmentally friendly and mild conditions. This protocol enriches current thioether-producing methods, making up for the shortcomings of previous sulfenylation methods which can only make MeS- and ArS-substituted imidazol[1,2-α]pyridine derivatives.  相似文献

The reaction of various anthranilic acid derivatives or their esters with 4-oxotetrahydrothiophene-3-carbonitrile 2, 2-oxocyclopentanecarbonitrile (9, n = 1) or 2-oxocyclohexanecarbonitrile (9, n = 2) in ethanol under reflux conditions giving rise the formation of single products isolated in each case after simple filtration. The products were characterized as pyrazolo[1,5-a]quinazolin-5-ones 4 instead of the expected pyrazol-3-amines 3. These cascade condensation–intramolecular acylation processes generated in one-step reactions from simple starting materials novel heterocyclic scaffolds ready for further functionalization. The present synthetic protocol provides acceptable yields of new tetracyclic products in high purity.  相似文献

An efficient, four-component reaction of isatin, 1,3-indanedione, ethyl acetoacetate, and ammonium acetate in ethanol/water (9:1) system furnished spiro[4H-indeno[1,2-b]pyridine-4,3′-[3H]indoles] at room temperature. Merits of the method are mild reaction conditions, simple workup procedure, and ambient temperature. The synthesized compounds exhibit excellent fluorescence properties.  相似文献

2-Aminocyclopenta[d]pyrimidines 3a–c were achieved via a one-pot, three-component reactions of cyclopentanone 1, aromatic aldehyde and guanidine hydrochloride(1:2:1 molar ratio). Also, cyclization of 2,5-bis-(arylmethylidene)cyclopentanones 2 with guanidine hydrochloride(1:1 molar ratio) in methanol in the presence of sodium methoxide afforded cyclopenta-[d]pyrimidines 3. Compound 3c has been shown to be a useful building block for the synthesis of some novel pyrimido[1,2-a]pyrimidines 5, 7and 12. The structures of the newly synthesized compounds were confirmed on the basis of analytical and spectral data.  相似文献

N-Substituted imidazo[4,5-e]benzo[1,2-c;3,4-c’]difuroxans were synthesized by phase-transfer catalyzed alkylation of imidazo[4,5-e]benzo[1,2-c;3,4-c’]difuroxan salts with various alkylating agents. Acid hydrolysis of N-(ethoxycarbonylmethyl)imidazo[4,5-e]benzo-[1,2-c;3,4-c’]difuroxans gives the corresponding carboxylic acids. Structures of the synthesized compounds were confirmed by NMR spectroscopy, high resolution mass spectrometry, IR spectroscopy, and elemental anslysis.

New series of pyrazolo[1,5-α]pyrimidine derivatives 7a-i,11a-c and Schiff bases 13a-c were synthesized and screened for their in vitro antitumor activity against three human carcinoma cell lines,namely colorectal carcinoma(HCT116),prostate adenocarcinoma(PC-3) and liver carcinoma(HepG-2) using MTT cytotoxicity assay at 100 μg/mL.Some of the tested compounds displayed good anticancer activities against HCT-116 and PC-3 cells.Whereas,compounds 7d and 11 a showed better antitumor activity than the rest of the compounds against both cell lines.A structure-activity relationship(SAR) has been discussed and structures of the newly synthesized compounds were confirmed by different spectral data(MS,IR,~1H NMR and ~(13)C NMR) and elemental analysis.  相似文献

The published route to a series of 6′-arylamino substituted photochromic spirooxazines has been investigated with improvements made to perform the synthesis in satisfactory yield. The route has been exemplified with several novel derivatives prepared (including hydroxyl functionalised). 开展了水相中硝基烯烃与氨基吡啶反应生成咪唑并[1,2-α]吡啶衍生物的微波辅助合成研究。结果表明:相同反应时间,在微波辅助条件下,生成咪唑并[1,2-α]吡啶衍生物的速率较常规加热条件下增大。一系列咪唑并[1,2-α]吡啶衍生物被高效的合成出来,最高产率达到85%。产物结构经~1H NMR,~(13)C NMR及MS得到确证。该方法具有产率高、操作简单、环境友好等优点。  相似文献

The reaction of 8-hydroxy-1,3-dimethyl-4H-cyclohepta[c]furan-4-one with the ethoxymethylene derivatives of malononitrile or ethyl cyanoacetate in the presence of KOH gave noncyclic cyanovinyl derivatives as their potassium salts rather than the expected α-pyrone derivatives. They are smoothly cyclized to the target α-pyrones by refluxing with acid. The corresponding 3-benzamido-2H-pyran-2-ones can be obtained in a single vessel from the 2-aryl-4-ethoxymethylene-4H-1,3-oxazol-5-ones using the same scheme. In our research on the ring enlargement of certain N-heterocycles, a general procedure for the ring enlargement with the insertion of an extra -CH2- was improved and shown in Scheme I1.Scheme IThe procedure had been carried out with such N-heterocycles as 2-methyl-1,2,3,4-tetrahydroisoquinoline2, tetrahydroberberine and strychnine3 with expected results.
