Abstract

The first completed chemical synthesis of Agrocinopine A (5) is presented. The key step in the synthesis was the pivaloyl chloride promoted condensation of a blocked arabinose 2-H-phosphonate derivative 3 with a protected sucrose derivative 1 having a free hydroxyl at 4′-OH. The resulting H-phosphonic acid diester was oxidised in situ with iodine to produce the corresponding phosphodiester 4, which was subsequently deblocked to give Agrocinopine A.  相似文献

An unsaturated 2-methoxy-substituted 1-O-alkylglycerol, (Z)-(2′R)-1-O-(2′-methoxynonadec-10′-enyl)-sn-glycerol, a new analog of bioactive ether lipids, was synthesized from oleic acid and 2,3-isopropylidene-sn-glycerol. The two key steps of this synthesis were the conversion of oleyl aldehyde to a monounsaturated epoxide using Matteson’s method followed by hydrolytic kinetic resolution and a nucleophilic epoxide opening by 2,3-isopropylidene-sn-glycerol in the presence of potassium tert-butoxide in anhydrous DMF, which appeared to be a good reagent for this purpose. Furthermore, the diol by-product of the HKR process was also easily converted back to the starting epoxide thus almost doubling the amount of target molecule.  相似文献

A synthesis of the enamine (−)-(1′S)-5-ethyl-1-(1′-phenylethyl)-1,2,3,4-tetrahydropyridine 4 and its application in a synthesis of (−)-(1′S,4aS,8aR)- and (+)-(1′S,4aR,8aS)-4a-ethyl-1-(1′-phenylethyl)-octahydroquinolin-7-ones 5 and 6 is described. A concise process of 1,3-diacetoxy-2-[2′-(2″,4″-difluorophenyl)prop-2′-en-1′-yl]propane has been developed. Diethyl malonate was C-alkylated with 2,3-dichloropropene and then the ester groups were reduced by LiAlH4, followed by acylation to provide 2-(2′-chloroprop-2′-en-1′-yl)-1,3-diacetoxypropane. The chloropropene was finally coupled with 2,4-difluorophenylmagnesium bromide and catalyzed by Fe(acac)3 to afford the title compound in good total yield.  相似文献

The title compound (C26H20ClN3O3) has been synthesized by 1,3-dipolar cycloaddition reaction from isatin, sarcosine and (Z)-4-(4-chlorobenzylidene)-2-phenyloxazol-5(4H)-one through a one-pot procedure, and its structure was confirmed by IR, 1H NMR, elemental analysis and single-crystal X-ray diffraction analysis. The crystal belongs to the triclinic system, space group P1, with a = 9.3903(19), b = 11.398(2), c = 12.603(3) , α = 83.495(3), β = 68.988(3), γ = 67.178(3)°, V = 1160.1(4) 3, Z = 2, Mr = 457.90, Dc = 1.311 g/cm3, μ = 0.198 mm-1, F(000) = 476, the final R = 0.0489 and wR = 0.1144 for 3109 observed reflections with I > 2σ(I).  相似文献

Three series of 2-(4′-alkoxybiphenyl-4-yl)-1H-benzimidazole derivatives (nM-x), which possessed 5-nitrobenzimidazole (nM-N series), benzimidazole (nM-H series) or 5-methylbenzimidazole (nM-M series) units at the end of the molecule, were synthesised and characterised by infrared, 1H- and 13C-nuclear magnetic resonance spectra, electrospray ionisation-mass spectrometry and elemental analysis. Their phase transition behaviour was investigated by differential scanning calorimetry, polarising optical microscopy and X-ray diffraction. All the compounds exhibited enantiotropic smectic mesophases with wide temperature domains for a carbon number in the alkoxy chain from 6 to 16, where the mesophase ranges were 14–91°C and 17–99°C during heating and cooling processes for the nM-N compounds, 7–25°C and 8–49°C for the nM-H compounds and 48–81°C and 52–85°C for the nM-M compounds, respectively. The effect of the length of alkoxy chain on mesomorphic properties was discussed. The nM-N and nM-M exhibited a much wider mesophase range whether during heating or cooling process than the corresponding nM-H series, especially for the longer terminal chain (n > 8), which indicated that the substituent in the benzimidazole moiety was helpful in increasing the mesophase stability.  相似文献

A dioxovanadium(V) complex, [VO2(moptpy)](ClO4) (1, moptpy = 4′-(3-methoxyphenyl)2,2′:6′2″-terpyridine), was synthesized and characterized by elemental analysis, ESI-MS, UV–vis, IR, and 1H, 13C, and 51V NMR. The cytotoxicity in vitro of 1 was evaluated against cancer cell lines HepG-2 (hepatocellular carcinoma), SGC-7901 (gastric carcinoma), SiHa (cervical cancer), BEL-7402 (hepatocellular), and rat PC-12 (pheochromocytoma) by the MTT method. The results demonstrated that 1 exhibits superior anticancer activity in vitro with much lower values of 50% inhibitive concentration (IC50) against the selected cell lines than cisplatin, and 1 shows the highest cytotoxic activity toward SGC-7901 cells (IC50 = 1.3 ± 0.1 μM) among the selected cell lines. A series of cellular morphological and staining experiments were carried out, and the results indicated that 1 can effectively induce apoptosis of SGC-7901 cells through a ROS-mediated mitochondrial dysfunction pathway. In addition, cellular incorporation and cell cycle analysis were also performed, and it was concluded from the experimental observations that 1 can efficiently enter into the cell nuclei, and the complex inhibits cell growth in SGC-7901 cell at G0/G1 phase.  相似文献

Abstract  

A series of 2-amino-4-aryl-6-(9,9′-spirobifluoren-2-yl)pyrimidines have been synthesized by the reaction of guanidine and chalcones under basic conditions. The physical and optical properties indicated that these compounds generally show good blue light emissions and excellent thermal stabilities. Intermolecular hydrogen bonds and interactions have strong influences on their properties.  相似文献

(E)-8, which can be stereospecifically prepared from (Z)-1-dimethylphenylsilyl-2-(2-thienyl)-1-trimethylstannyl-ethene, (Z)-6, or from (E)-2-(2-thienyl)ethenyldimethylphenylsilane, (E)-9, serves as an equivalent to the 2-ethenylthiophene d 1', d 5-synthon, 20, in an efficient two-step synthesis of 5-ethenyl-5′-(1-propynyl)-2,2′-bithiophene, 10, a naturally-occurring phototoxin. 2-Amino-4-(4′-hydroxyphenyl) thiazole 1a was prepared from reaction between p-hydroxyacetophenone, thiourea, and iodine; compound 1a was treated with several (aryl/hetro aryl) aldehydes to form 2-(substituted benzylideneamino)-4-(4′-hydroxyphenyl) thiazoles 2a–j, which were glucosylated by using acetobromoglucose as a glucosyl donor to afford 2-(substituted benzylideneamino)-4-(2, 3, 4, 6-tetera-o-acetyl-4′-o-β-d-glucosidoxyphenyl) thiazoles 3a–j, which further on during deacetylation produced 2-(substituted benzylideneamino)-4-(4′-o-β-d-glucosidoxyphenyl) thiazoles 4a–j. These compounds were evaluated for biological activity, and their structure was confirmed by IR, NMR, mass spectra, elemental, and chemical analysis.
