Summary EU Directive 90/128/EEC prescribes a specific migration limit (SML) of 0.05 mg kg−1 for the aliphatic diaminem-xylylenediamine (XDA) into food or food simulants, but there is no generally accepted method of analysis available for testing compliance with this restriction.m-XDA has been determined in olive oil (EU food simulant for fatty foods) by high-performance liquid chromatography (HPLC) with fluorescence detection, after extraction into an aqueous phase and derivatization with fluorescamine. The method is appropriate for the quantitative determination ofm-XDA at a minimum level of 0.02 mg kg−1 in this food simulant. The method should also be applicable to other fatty-food simulants (sunflower oil and HB 307).  相似文献

Fumonisins B1 (FB1) and fumonisin B2 (FB2) are the main members of a family of mycotoxins produced by Fusarium verticillioides, Fusarium proliferatum, and other fungi species of the section Liseola. The present work shows the results of comparative studies using two different procedures for the analysis of fumonisins in maize and maize-based samples. The studied analytical methods involve extraction with methanol/water, dilution with PBS, and clean-up through immunoaffinity columns. Two reagents (o-phthaldialdehyde and naphthalene-2,3-dicarboxaldehyde) were studied for formation of fluorescent derivatives. The separation and identification were carried out by high-performance liquid chromatography with fluorescence detection. The optimized method for analysis of fumonisins in maize involved extraction with methanol/water (80:20), clean-up with an immunoaffinity column, and derivatization with naphthalene-2,3-dicarboxaldehyde (NDA). The limit of detection was 20 μg kg−1 for FB1 and 15 μg kg−1 for FB2. Recoveries of FB1 and FB2 ranged from 79% to 99.6% for maize fortified at 150 μg kg−1 and 200 μg kg−1, respectively, with within-day RSDs of 3.0 and 2.7%. The proposed method was applied to 31 samples, and the presence of fumonisins was found in 14 samples at concentrations ranging from 113 to 2,026 μg kg−1. The estimated daily intake of fumonisins was 0.14 μg kg−1 body weight per day.  相似文献

A new procedure for spectrofluorimetric determination of free and total glycerol in biodiesel samples is presented. It is based on the oxidation of glycerol by periodate, forming formaldehyde, which reacts with acetylacetone, producing the luminescent 3,5-diacetyl-1,4-dihydrolutidine. A flow system with solenoid micro-pumps is proposed for solution handling. Free glycerol was extracted off-line from biodiesel samples with water, and total glycerol was converted to free glycerol by saponification with sodium ethylate under sonication. For free glycerol, a linear response was observed from 5 to 70 mg L−1 with a detection limit of 0.5 mg L−1, which corresponds to 2 mg kg−1 in biodiesel. The coefficient of variation was 0.9% (20 mg L−1, n = 10). For total glycerol, samples were diluted on-line, and the linear response range was 25 to 300 mg L−1. The detection limit was 1.4 mg L−1 (2.8 mg kg−1 in biodiesel) with a coefficient of variation of 1.4% (200 mg L−1, n = 10). The sampling rate was ca. 35 samples h−1 and the procedure was applied to determination of free and total glycerol in biodiesel samples from soybean, cottonseed, and castor beans.  相似文献

To evaluate the pharmacokinetics of a novel analogue of ginkgolide B, 10-O-dimethylaminoethylginkgolide B (XQ-1) in rat plasma in pre-clinical studies, a sensitive and specific liquid chromatographic method with electrospray ionization mass spectrometry detection (LC–ESI–MS) was developed and validated. After a simple extraction with ethyl acetate, XQ-1 was analyzed on a Shim-pack C18 column with a mobile phase of a mixture of 1 μmol L−1 ammonium acetate containing 0.02% formic acid and methanol (55:45, v/v) at a flowrate of 0.3 mL min−1. Detection was performed in selected ion monitoring (SIM) mode using target ions at [M + H]+ m/z 496.05 for XQ-1 and m/z 432.10 for the internal standard (lafutidine). Linearity was established for the concentration range from 2 to 1,000 ng mL−1 . The extraction recoveries ranged from 86.0 to 89.9% in plasma at concentrations of 5, 50, and 500 ng mL−1. The lower limit of quantification was 2 ng mL−1 with 100 μL plasma. The validated method was successfully applied to a pharmacokinetic study after intragastic administration of XQ-1 mesylate in rats at a dose of 20 mg kg−1.  相似文献

In this article a new analytical method for the confirmation and quantification of abamectin residues in avocados is described. The method allows a fast analysis of abamectin homologues using microwave assisted extraction (MAE), solid-phase extraction (SPE) and high-performance liquid chromatography (HPLC) with fluorescence (FL) detection using trifluoroacetic anhydride (TFAA) and N-methylimidazole (NMIM) as derivatizing agents. The mobile phase consisted of water, methanol and acetonitrile (5:47.5:47.5 v/v/v) and was pumped at a rate of 1.1 mL min−1 (isocratic elution). Homogenized avocado samples were extracted once with 20 mL acetonitrile:water 4:1 (v/v) in a microwave oven for 26 min at 700 W with a maximum temperature of 80 °C. MAE operational parameters were optimized by means of an experimental design. Extracts were cleaned using C18 SPE cartridges. Average recoveries of the method at four spiked levels (0.005, 0.01, 0.10 and 1.0 mg kg−1) were found to be in the range 90–100% with good precision (RSD < 12%). The limits of detection (LODs) and quantification (LOQs) of the whole method were 0.001 and 0.003 mg kg−1, respectively, which are lower than the maximum residue limit (MRL) established by the Spanish and the European legislation in avocados (0.01 mg kg−1). Several avocado samples previously treated with the pesticide were also analyzed.  相似文献

Ultra-performance liquid chromatography coupled to tandem mass spectrometry (UPLC–MS–MS) has been used for screening and quantification of 32 pesticides and metabolites in two fruit matrices. The compounds investigated belonged to different chemical families of insecticides, acaricides, fungicides, and herbicides; several metabolites were also included. Quantification was conducted using matrix-matched standards calibration; response was a linear function of concentration in the range tested (10–500 ng mL−1). The method was validated with blank samples of lemon and raisin spiked at 0.01 and 0.1 mg kg−1, and recoveries were satisfactory, between 70 and 110%, for most of the pesticides tested and relative standard deviations were below 15% (n = 5 at each spiking level). Excellent sensitivity resulted in limits of detection for all compounds well below 0.01 mg kg−1, with the limit of quantification being validated at 0.01 mg kg−1. The UPLC system generates narrow peaks (approx. 5 s), thus increasing peak height and improving sensitivity. This improved separation efficiency facilitates adequate resolution not only of the analytes but also of matrix interferences compared with conventional HPLC. The method developed could also resolve some geometric isomers. The main advantage of this approach is the high sample throughput achieved because of the short analysis time, which enables satisfactory separation of all the compounds in less than 5 min per sample.  相似文献

A rapid and accurate method for the quantification of cis-abienol in oriental tobacco leaves by normal phase liquid chromatography was developed. Freeze-dried tobacco samples were sonicated in methylene chloride for 10 min. The supernatant was purified using a silica gel solid phase extraction cartridge. Ten milliliter of the resulting methylene chloride eluate was collected, then separated on a 250 × 4.6 mm, 5 μm particle-size CN column with n-hexane: ethyl acetate, 100:2 (v/v) at a flow rate of 1 mL min−1. cis-Abienol was detected by UV absorption at 254 nm. The linear range was from 2.14 × 10−4 to 4.28 × 10−2 mg mL−1 and the correlation coefficient was 1.000. The average recovery was 98.7, 105.2 and 103.1% in five replicated sets of tobacco samples spiked with 0.2856, 0.7140 and 1.904 mg cis-abienol. The relative standard deviations (RSDs) were 1.04, 0.63 and 1.25%, respectively (n = 5). Limit of detection (S/N = 3) was 21.84 μg g−1 and limit of quantification (S/N = 10) was 72.80 μg g−1. The method was found to be suitable for determination of cis-abienol in oriental tobacco leaves. Furthermore, pure cis-abienol used for method validation was obtained by preparative reversed phase high-performance liquid chromatography. Identification was performed by UV detection, nuclear magnetic resonance and mass spectrometry.  相似文献

Summary A new gas chromatographic method using an automatic purge-and-trap system coupled to a GC with mass selective detection to analyze styrene at the parts-per-trillions (ng kg−1) level is described. The method shows a good sensitivity and the detection limit is 10 ng kg−1 with a relative standard deviation (RSD) of 4.7 % for 164 ng kg−1 styrene in olive oil. This analytical method has been successfully applied to the analysis of styrene in extra-virgin olive oil from the European market.  相似文献

This paper describes two isotachophoretic methods of metoprolol tartrate (MT) determination in pure and dosage forms. The first method was used for direct analysis where the following electrolyte system was applied: 10 mmol dm−3 3-morpholino-2-hydroxypropanesulfonic acid, 10 mmol dm−3 NaCl, 2 % hydroxyethylocelulose as leading (LE) and 10 mmol dm−3 glycyl-glycine as terminating (TE) electrolytes. The second method was used for indirect analysis of MT as tartrate ions. In this case, the leading electrolyte consisted of 10 mmol dm−3 HCl, β-alanine (BALA), pH 4-5, and the terminating one of 5 mmol dm−3 glutamic acid, 10 mmol dm−3 β-alanine. Calibration curves were calculated as follows: for system A: y = (0.52 ± 0.05)x − (0.9 ± 0.2) (LOD = 13.0 mg dm−3, LOQ = 31.7 mg dm−3); and for system B: y = (0.240 +- 0.001)x + (0.18 ± 0.06) (LOD = 1.8 mg dm−3, LOQ = 4.4 mg dm−3). The isotachophoretic method was compared with the pharmacopoeial one by statistical tests.  相似文献

A high-performance liquid chromatography–UV method for determining DCJW concentration in rat plasma was developed. The method described was applied to a pharmacokinetics study of intramuscular injection in rats. The plasma samples were deproteinized with acetonitrile in a one-step extraction. The HPLC assay was carried out using a VP-ODS column and the mobile phase consisting of acetonitrile–water (80:20, v/v) was used at a flow rate of 1.0 mL min−1 for the effective eluting DCJW. The detection of the analyte peak area was achieved by setting a UV detector at 314 nm with no interfering plasma peak. The method was fully validated with the following validation parameters: linearity range 0.06–10 μg mL−1 (r > 0.999); absolute recoveries of DCJW were 97.44–103.46% from rat plasma; limit of quantification, 0.06 μg mL−1 and limit of detection, 0.02 μg mL−1. The method was further used to determine the concentration–time profiles of DCJW in the rat plasma following intramuscular injection of DCJW solution at a dose of 1.2 mg kg−1. Maximum plasma concentration (C max) and area under the plasma concentration–time curve (AUC) for DCJW were 140.20 ng mL−1 and 2405.28 ng h mL−1.  相似文献

From vapor pressure osmometry data, the activity of water, osmotic coefficients and mean ionic activity coefficients of glycine (m=0.006−3.2 mol⋅kg−1), L-histidine (m=0.005−0.23 mol⋅kg−1), L-histidine monohydrochloride (m=0.008−0.63 mol⋅kg−1), glutamic acid (m=0.004−0.05 mol⋅kg−1), sodium L-glutamate (m=0.007−0.6 mol⋅kg−1), and calcium L-glutamate (m=0.008−0.6 mol⋅kg−1) have been obtained in aqueous solutions at 298.15 and 310.15 K. The Pitzer equations and the mean spherical approximation (MSA) are used for theoretical modeling. A simple and effective extraction method based on matrix solid-phase dispersion (MSPD) was developed to determine bifenthrin, buprofezin, tetradifon, and vinclozolin in propolis using gas chromatography–mass spectrometry in selected ion monitoring mode (GC–MS, SIM). Different method conditions were evaluated, for example type of solid phase (C18, alumina, silica, and Florisil), the amount of solid phase and eluent (n-hexane, dichloromethane, dichloromethane–n-hexane (8:2 and 1:1, v/v) and dichloromethane–ethyl acetate (9:1, 8:2 and 7:3, v/v)). The best results were obtained using 0.5 g propolis, 1.0 g silica as dispersant sorbent, 1.0 g Florisil as clean-up sorbent, and dichloromethane–ethyl acetate (9:1, v/v) as eluting solvent. The method was validated by analysis of propolis samples fortified at different concentration levels (0.25 to 1.0 mg kg−1). Average recoveries (four replicates) ranged from 67% to 175% with relative standard deviation between 5.6% and 12.1%. Detection and quantification limits ranged from 0.05 to 0.10 mg kg−1 and 0.15 to 0.25 mg kg−1 propolis, respectively.  相似文献

The values of the second dissociation constant, pK 2, and related thermodynamic quantities of N-[tris(hydroxymethyl)methyl-3-amino]propanesulfonic acid (TAPS) have already been reported at 12 temperatures over the temperature range 5–55 C, including 37 C. This paper reports the results for the pH of five equimolal buffer solutions with compositions: (a) TAPS (0.03 mol⋅kg−1) + NaTAPS (0.03 mol⋅kg−1); (b) TAPS (0.04 mol⋅ kg−1) + NaTAPS (0.04 mol⋅kg−1); (c) TAPS (0.05 mol⋅kg−1) + NaTAPS (0.05 mol⋅kg−1); (d) TAPS (0.06 mol⋅kg−1) + NaTAPS (0.06 mol⋅kg−1); and (d) TAPS (0.08 mol⋅kg−1) + NaTAPS (0.08 mol⋅kg−1). The remaining eight buffer solutions consist of saline media of the ionic strength I = 0.16 mol⋅kg−1, matching closely to that of the physiological sample. The compositions are: (f) TAPS (0.04 mol-kg−1) + NaTAPS (0.02 mol-kg−1) + NaCl (0.14 mol⋅kg−1); (g) TAPS (0.05 mol⋅kg−1) + NaTAPS (0.04 mol⋅kg−1) + NaCl (0.12 mol⋅kg−1); (h) TAPS (0.6 mol⋅kg−1) + NaTAPS (0.04 mol⋅kg−1) + NaCl (0.12 mol⋅kg−1); (i) TAPS (0.08 mol⋅kg−1) + NaTAPS (0.06 mol⋅kg−1) + NaCl (0.10 mol⋅kg−1); (j) TAPS (0.04 mol⋅ kg−1) + NaTAPS (0.04 mol⋅kg−1) + NaCl (0.12 mol⋅kg−1); (k) TAPS (0.05 mol⋅kg−1) + NaTAPS (0.05 mol⋅kg−1) + NaCl (0.11 mol⋅kg−1); (l) TAPS (0.06 mol⋅kg−1) + NaTAPS (0.06 mol⋅kg−1) + NaCl (0.10 mol⋅kg−1); and (m) TAPS (0.08 mol⋅kg−1) + NaTAPS (0.08 mol⋅kg−1) + NaCl (0.08 mol⋅kg−1). These buffers are recommended as a pH standard for clinical measurements in the range of physiological application. Conventional pH values, designated as pH(s), for all 13 buffer solutions from 5 to 55 C have been calculated. The operational pH values with liquid junction corrections, at 25 and 37 C for buffer solutions, designated above as (b), (c), (d), (e), (j), (l), and (m); have been determined based on the difference in the values of the liquid junction potentials between the accepted phosphate standard and the buffer solutions under investigation.  相似文献

A multiresidue method has been developed for the simultaneous determination of sulfadiazine, sulfathiazole, sulfapyridine, sulfamerazine, sulfamethoxydiazine, sulfamethylthiazole, sulfamethazine, sulfamonomethoxine, sulfamethoxypyridazine, sulfisoxazole, sulfamethoxazole, sulfadimethoxine and sulfaquinoxaline in natural animal casings by HPLC after solid-phase extraction. The sulfonamides were extracted with acetonitrile and the extract cleaned up with an Oasis MCX SPE cartridge prior to analysis. Separation was on a ZOBAX Eclipse XDB-C8 column using gradient elution with acetonitrile/methanol/0.1% acetic acid. The effect of separation conditions on chromatographic behavior and recovery has been studied. Calibration graphs were linear with very good correlation coefficients (r = 0.9983−0.9996) in the concentration range from 0.02 to 1 μg mL−1. The limits of quantitation (LOQ) for the 13 sulfonamides were in the range of 1.5–2.2 μg kg−1. Decision limits (CCα) and detection capabilities (CCβ) were in the range of 105.2–111.0 and 113.0–120.2 μg kg−1, respectively. The recovery for casings spiked with 1.5–100 μg kg−1 ranged from 65.2 to 85.9%. The relative standard deviations (RSDs) of the sulfonamides for six measurements at 100 μg kg−1 were from 2.2 to 7.7%. The applicability of the method to the analysis of salted swine casings, salted sheep casings and dry casing samples was demonstrated.  相似文献

This paper reports the results for the pa H of eight isotonic saline buffer solutions with an I=0.16 mol⋅kg−1 including compositions: (a) HEPES (0.01 mol⋅kg−1) + NaHEPES (0.01 mol⋅kg−1) + NaCl (0.15 mol⋅kg−1); (b) HEPES (0.02 mol⋅kg−1) + NaHEPES (0.02 mol⋅kg−1) + NaCl (0.14 mol⋅kg−1); (c) HEPES (0.03 mol⋅kg−1) + NaHEPES (0.03 mol⋅kg−1) + NaCl (0.13 mol⋅kg−1); (d) HEPES (0.04 mol⋅kg−1) + NaHEPES (0.04 mol⋅kg−1) + NaCl (0.12 mol⋅kg−1); (e) HEPES (0.05 mol⋅kg−1) + NaHEPES (0.05 mol⋅kg−1) + NaCl (0.11 mol⋅kg−1); (f) HEPES (0.06 mol⋅kg−1) + NaHEPES (0.06 mol⋅kg−1) + NaCl (0.10 mol⋅kg−1); (g) HEPES (0.07 mol⋅kg−1) + NaHEPES (0.07 mol⋅kg−1) + NaCl (0.09 mol⋅kg−1); and (h) HEPES (0.08 mol⋅kg−1) + NaHEPES (0.08 mol⋅kg−1) + NaCl (0.08 mol⋅kg−1). Conventional pa H values, for all eight buffer solutions from 5 to 55 °C, have been calculated. The operational pH values with liquid junction corrections, at 25 and 37 °C have been determined based on the NBS/NIST standard between the physiological phosphate standard and four buffer solutions. These are recommended as pH standards for physiological fluids in the range of pH = 7.3 to 7.5 at I=0.16 mol⋅kg−1.  相似文献

Seasonal changes in petroleum hydrocarbons in water and streambed sediment from selected oil-related areas of Ondo State, Nigeria have been examined using gravimetric and infrared methods. The highest and lowest total petroleum hydrocarbon concentrations (TPH) in water (sediments in brackets) gravimetrically were 3.49 mg L−1 (199.3) mg kg−1 and 0.003 mg L−1 (81.0) mg kg−1 while the concentrations found by IR were 24.0 mg L−1 (135.0 mg kg−1) and 14.0 mg L−1 (33.0 mg kg−1) respectively. The two seasons were positively correlated (α = 0.01) by both methods. The TPH level was well correlated with the sediment organic carbon (OC) during both seasons. The characteristic carbonyl (C=O) vibrations at 1650 cm−1 and 1700 cm−1 indicate oxidation of the oil residue. The study recommends further investigation into the type of organics present to evaluate their toxicity and appropriate remediation.   相似文献

A method was described for simultaneous determination of nine organic heterocyclic pesticide residues by gas chromatography-mass spectrometry-selected ion monitoring. Atrazine, vinclozolin, procymidone, triflumizole, imazalil, buprofezin, propiconazole, fenarimol, and pyridaben were clearly separated from each other, extracted with acetone—hexane mixture, purified with graphitized carbon black cartridge and neutral Al2O3 cartridge, eluted with acetone—hexane mixture, simultaneously determined by GC-MS, and then quantified with an external standard method. Recoveries of pesticides ranged from 73 % to 116 % at the spiked level of 0.01–30 mg kg−1, while the relative standard deviation was between 3 % and 27 %. In addition, the limits of determination (0.01 mg kg−1 to 5.0 mg kg−1) and linearity (0.02–40 μg mL−1) revealed that simultaneous determination of multi-residues in Chinese teas (like Oolong tea, green tea, red tea, etc.) was possible. Furthermore, an interlaboratory study among 5 labs was conducted to further validate the method, and the results were satisfactory.  相似文献

An approach for rapid quantitation of 5-hydroxymethylfurfural (HMF) in honey using planar chromatography is suggested for the first time. In high-performance thin-layer chromatography (HPTLC) the migration time is approximately 5 min. Detection is performed by absorbance measurement at 290 nm. Polynomial calibration in the matrix over a range of 1:80 showed correlation coefficients, r, of ≥ 0.9997 for peak areas and ≥ 0.9996 for peak heights. Repeatability in the matrix confirmed the suitability of HPTLC–UV for quantitation of HMF in honey. The relative standard deviation (RSD, %, n = 6) of HMF at 10 ng/band was 2.9% (peak height) and 5.2% (peak area); it was 0.6% and 1.0%, respectively, at 100 ng/band. Other possible detection modes, for example fluorescence measurement after post-chromatographic derivatization and mass spectrometric detection, were also evaluated and can coupling can be used as an additional tool when it is necessary to confirm the results of prior quantitation by HPTLC–UV. The confirmation is provided by monitoring the HMF sodium adduct [M + Na]+ at m/z 149 followed by quantitation in TIC or SIM mode. Detection limits for HPTLC–UV, HPTLC–MS (TIC), and HPTLC–MS (SIM) were 0.8 ng/band, 4 ng/band, and 0.9 ng/band, respectively. If 12 μL honey solution was applied to an HPTLC plate, the respective detection limits for HMF in honey corresponded to 0.6 mg kg−1. Thus, the developed method was highly suitable for quantitation of HMF in honey at the strictest regulated level of 15 mg kg−1. Comparison of HPTLC–UV detection with HPTLC–MS showed findings were comparable, with a mean deviation of 5.1 mg kg−1 for quantitation in SIM mode and 6.1 mg kg−1 for quantitation in TIC mode. The mean deviation of the HPTLC method compared with the HPLC method was 0.9 mg kg-1 HMF in honey. Re-evaluation of the same HPTLC plate after one month showed a deviation of 0.5 mg kg−1 HMF in honey. It was demonstrated that the proposed HPTLC method is an effective method for HMF quantitation in honey.   相似文献

To determine sulphamethazine (SMZ) residues in edible animal foods (pig muscle, chicken muscle, egg, fish, milk and liver), a competitive direct enzyme-linked immunosorbent assay (ELISA) and a colloidal gold immunoassay were established. The limits of detection of the ELISA and the colloidal gold immunoassay were 0.02 and 0.5 μg kg−1, respectively. The specificity of the ELISA developed to the SMZ was high according to the results of cross-reactivity testing with 14 kinds of sulphonamides. To obtain a more sensitive immunoassay, buffer solution (30 mmol L−1 phosphate-buffered saline with 0.05% Tween 20, pH 8.5) was optimized through the whole test procedure. A simple and efficient extraction method for the rapid detection of SMZ residues in foods was developed, with recoveries between 74 and 117.5%. Matrix effects can be avoided by 1:10 dilution of pig muscle, chicken muscle, egg, fish, milk and liver with optimal buffer. The detection limit of SMZ was 5 μg kg−1 in liver and 2 μg kg−1 in the other five samples. For the validation of the ELISA tests, sample extracts were analysed by ELISA and high-performance liquid chromatography. The results obtained by these two methods showed a good correlation (r 2) which was greater than 0.9. The colloidal gold immunoassay presented in this assay was successfully applied to determine SMZ in pig muscle, milk and fish below or equal to the maximum residue level (20 μg kg−1).  相似文献

Hollow fibre based liquid phase microextraction (HF-LPME) with high performance liquid chromatography (HPLC) was developed for the determination of alkaloids in Corydalis yanhusuo. Three alkaloids (protopine, tetrahydropalmatine, tetrahydroberberine) were extracted from a 10 mL alkaline sample (donor phase) to an organic phase impregnated in the pores of the hollow fibre, and then, they were extracted to an acidic solution (acceptor phase) in the lumen of the fibre. The extract was determined directly by HPLC. Parameters affecting the HF-LPME include the organic solvent, pH of the donor and the acceptor phase, the extraction time and the stirring speed were investigated systematically. To minimize the error of the injection, palmatine was added as an internal standard (I.S.). Under optimal conditions, calibration curves were obtained in the range of 0.1–1.0 mg L−1 with a reasonable linearity (r 2 > 0.993) and the limits of detection (LODs) ranged between 10.0 × 10−3 mg L−1 and 13.7 × 10−3 mg L−1. Additionally, enrichment factors with 100 to 184-fold were obtained. The method was then applied to the crude extract of Corydalis yanhusuo successfully.  相似文献
