A series of peptides of general primary structure (VSSLESK)n (n = 2, 3, 4, 5 and 6) were designed and synthesized by fluorenylmethyloxycarbonyl solid-phase synthesis using a convergent approach. Peptides containing 21, 28, 35 and 42 residues were modified with α-methoxy poly(ethylene glycol) (mPEG; mol. wt. 2000) by reaction of mPEG–succinimidyl carbonate with the α-amino group of the resin-attached protected peptides. The conformation and thermal stability of the peptides and of their AB block copolymers (A is the mPEG block, B the (VSSLESK)n block) in aqueous medium were investigated by circular dichroism, size-exclusion chromatography and by analytical ultracentrifugation. The helicity of peptides increased with increasing length in a cooperative manner. The peptides and mPEG–peptides with 35 and 42 amino acid residues (block copolymers) adopted a two-stranded α-helical coiled-coil conformation in aqueous solution. The presence of the polymer chain in the diblock hybrid copolymers had no disturbing effect with respect to the stability of the α-helical peptide part in these constructs. Moreover, the thermal stability of mPEG-modified 42-peptide was substantially higher than that of the native 42-peptide. Analytical ultracentrifugation data revealed that in phosphate-buffered saline solution (25–200 μM ) the block copolymer mPEG-block-(VSSLESK)6 ( PEG42 ) associated into stable intermolecular coiled-coil dimers.

The crystallization behaviors and morphology of asymmetric crystalline–crystalline diblock copolymers poly(ethylene oxide‐lactide) (PEO‐b‐PLLA) were investigated using differential scanning calorimetry (DSC), wide angle X‐ray diffraction (WAXD), and microscopic techniques (polarized optical microscopy (POM) and atomic force microscopy (AFM)). Both blocks of PEO5b‐PLLA16 can be crystallized, which was confirmed by WAXD, while PEO block in PEO5b‐PLLA30 is difficult to crystallize because of the confinement induced by the high glass transition temperature and crystallization of PLLA block with the microphase separation of the block copolymer. Comparing with the crystallization and morphology of PLLA homopolymer and differences between the two copolymers, we studied the influence of PEO block and microphase separation on the crystallization and morphology of PLLA block. The boundary temperature (Tb) was observed, which distinguishes the crystallization into high‐ and low‐temperature ranges, the growth rate and morphology were quite different between the ranges. Crystalline morphologies including banded spherulite, dendritic crystal, and dense branching in PEO5b‐PLLA16 copolymer were formed. The typical morphology of dendritic crystals including two different sectors were observed in PEO5b‐PLLA30 copolymer, which can be explained by secondary nucleation, chain growth direction, and phase separation between the two blocks during the crystallization process. Lozenge‐shaped crystals of PLLA with screw dislocation were also observed employing AFM, but the crystalline morphology of PEO block was not observed using microscopy techniques because of its small size. © 2008 Wiley Periodicals, Inc. Semi-interpenetrating polymer networks (semi-IPNs) of poly(ethylene glycol) (PEG) in poly(trimethylolpropane triacrylate) (TMPTA) were synthesized from PEG melts in neat TMPTA monomer, using PEG of molecular weights from 4000 to 100,000 g/mol. Differential scanning calorimetry and transmission electron microscopy were used to examine phase separation occurring during network formation. The degree of phase separation was observed to depend upon the rate of PEG aggregation relative to the rate of network formation during TMPTA polymerization. Higher molecular weight PEG and acrylate-functionalized PEG formed more phase-mixed networks compared to lower molecular weight PEG; acetatefunctionalized PEG showed no difference from unmodified PEG in the extent of phase mixing. For networks that demonstrated phase separation, the PEG was observed to be in two states: some being phase mixed and solvent inextractable, and some being phase separated and solvent extractable. Phase-mixed networks could be obtained from this thermodynamically incompatible polymer pair utilizing rapid photopolymerization systems to overcome PEG phase aggregation and kinetically entrap the PEG in a nonequilibrium phase-mixed state. These mixed-phase semi-IPNs of PEG and TMPTA may be useful in biological applications where the presence of PEG is desired throughout the bulk matrix rather than as a surface graft to reduce biological interactions. © 1994 John Wiley & Sons, Inc.  相似文献

A series of poly(ethylene glycol) (PEG)/poly(L-lactic acid) (PLLA) multiblock copolymers were facilely synthesized using triphosgene as coupling agent. With the resulting multiblock copolymers, 10-hydroxycamptothecin (HCPT)-loaded nanoparticles were successfully prepared by dialysis method. The results obtained from dynamic light scattering (DLS) measurements confirmed that HCPT-loaded nanoparticles had the size of less than 200 nm and the average diameter decreased with increasing PLLA content. TEM images demonstrated that most of the drug-loaded nanoparticles had a distinct spherical shape and smooth surface without any aggregation. Atomic force microscopy (AFM) images further indicated that the nanoparticles were in spherical shape with smooth surface, no drug crystal was visualized on their surface. To investigate the drug state in HCPT-loaded nanoparticles, differential scanning calorimetry (DSC) and X-ray powder diffraction (XRD) measurements were carried out. The results from these tests suggested that HCPT was molecularly dispersed in the amorphous polymer matrix. Drug loading content and in vitro drug release behavior of HCPT-loaded nanoparticles showed dependence on polymer composition. Cytotoxicity test indicated that HCPT-loaded nanoparticles exhibited greatly superior cytotoxicity compared to free HCPT due to its molecular dispersion in the polymer matrix. Furthermore, the nanoparticles significantly increased the duration of the drug in circulation. All these results demonstrated that PEG/PLLA nanoparticles have great potential as promising delivery system for poorly soluble antitumor drugs.  相似文献

Diblock copolymers consisting of methoxy poly(ethylene glycol) (MPEG) and poly(?‐caprolactone) (PCL), poly(δ‐valerolactone) (PVL), poly(L ‐lactic acid) (PLLA), or poly(lactic‐co‐glycolic acid) (PLGA) as biodegradable polyesters were prepared to examine the phase transition of diblock copolymer solutions. MPEG–PCL and MPEG–PVL diblock copolymers and MPEG–PLLA and MPEG–PLGA diblock copolymers were synthesized by the ring‐opening polymerization of ?‐caprolactone or δ‐valerolactone in the presence of HCl · Et2O as a monomer activator at room temperature and by the ring‐opening polymerization of L ‐lactide or a mixture of L ‐lactide and glycolide in the presence of stannous octoate at 130 °C, respectively. The synthesized diblock copolymers were characterized with 1H NMR, IR, and gel permeation chromatography. The phase transitions for diblock copolymer aqueous solutions of various concentrations were explored according to the temperature variation. The diblock copolymer solutions exhibited the phase transition from gel to sol with increasing temperature. As the polyester block length of the diblock copolymers increased, the gel‐to‐sol transition moved to a lower concentration region. The gel‐to‐sol transition showed a dependence on the length of the polyester block segment. According to X‐ray diffraction and differential scanning calorimetry thermal studies, the gel‐to‐sol transition of the diblock copolymer solutions depended on their degrees of crystallinity because water could easily diffuse into amorphous polymers in comparison with polymers with a crystalline structure. A new series of segmented copolymers were synthesized from poly(ethylene terephthalate) (PET) oligomers and poly(ethylene glycol) (PEG) by a two‐step solution polymerization reaction. PET oligomers were obtained by glycolysis depolymerization. Structural features were defined by infrared and nuclear magnetic resonance (NMR) spectroscopy. The copolymer composition was calculated via 1H NMR spectroscopy. The content of soft PEG segments was higher than that of hard PET segments. A single glass‐transition temperature was detected for all the synthesized segmented copolymers. This observation was found to be independent of the initial PET‐to‐PEG molar ratio. In this study, a series of liquid crystalline diblock copolymers, composed of a soft poly(dimethylsiloxane) (PDMS) block with a de?ned length and a side-on liquid crystalline poly(3??-acryloyloxypropyl 2,5-di(4?-butyloxybenzoyloxy) benzoate) (P3ADBB) block with different lengths, are synthesised by the atom transfer radical polymerisation. The macromolecular structures, liquid crystalline properties and the microphase-separated morphologies of the diblock copolymer are investigated by 1H NMR, FT-IR, GPC, POM, DSC and TEM. The results show that the well-de?ned diblock copolymers (PDMSn-b-P3ADBBm) possess four different soft/rigid ratios (n = 58, m = 10, 25, 42, 66) and relatively narrow molecular distributions (PDI ≤ 1.30). P3ADBB blocks of the copolymers show nematic sub-phases, which are identical to the mesomorphic behaviour of the homopolymer P3ADBB. After being annealed at 90°C in a vacuum oven for 48 h, the copolymers form a lamellar morphology when m = 10 and morphologies of PDMS spheres embedded in P3ADBB matrix when m = 25, 42 and 66.  相似文献

AB‐type block copolymers with poly(trimethylene carbonate) [poly(TMC); A] and poly(ethylene oxide) [PEO; B; number‐average molecular weight (Mn) = 5000] blocks [poly(TMC)‐b‐PEO] were synthesized via the ring‐opening polymerization of trimethylene carbonate (TMC) in the presence of monohydroxy PEO with stannous octoate as a catalyst. Mn of the resulting copolymers increased with increasing TMC content in the feed at a constant molar ratio of the monomer to the catalyst (monomer/catalyst = 125). The thermal properties of the AB diblock copolymers were investigated with differential scanning calorimetry. The melting temperature of the PEO blocks was lower than that of the homopolymer, and the crystallinity of the PEO block decreased as the length of the poly(TMC) blocks increased. The glass‐transition temperature of the poly(TMC) blocks was dependent on the diblock copolymer composition upon first heating. The static contact angle decreased sharply with increasing PEO content in the diblock copolymers. Ring opening polymerization of ε‐caprolactone was realized in the presence of monomethoxy poly(ethylene glycol) with Mn = 1000 and 2000, using Zn(La)2 as catalyst. The resulting PCL‐PEG diblock copolymers with CL/EO repeat unit molar ratios from 0.2 to 3.0 were characterized by using DSC, WAXD, SEC, and 1H NMR. The crystal phase of PCL blocks exist in all polymers, and the crystallization ability of PCL blocks increases with CL/EO ratio. PEG blocks are able to crystallize for copolymers with CL/EO below 1.0 only. Melt crystallization results were analyzed with Avrami equation. The Averami exponent n is around 3.0 in most cases, in agreement with heterogeneous nucleation with three dimensional growth. The morphology of the crystals was observed by using POM. Rod‐like crystals were found to grow in 1, 3 or 2, 4 quadrants for samples with low molecular weights. In the case of a copolymer with Mn,PEG = 2000 and Mn,PCL = 800, PEG blocks could crystallize and grow on PCL crystals after PCL finished to form rod‐like crystals, leading to formation of poorly or well structured spherulites. The spherulite growth rate (G) was determined at different crystallization temperatures (Tc) ranging from 9 to 49 °C. All the copolymers present a steady G decrease with increasing crystallization temperature due to lower undercooling. A series of poly(styrene‐block‐ferrocenyldimethylsilane) copolymers (SF) with different relative molar masses of the blocks were prepared by sequential anionic polymerization. The bulk morphology of these polymers, studied by TEM and SAXS, showed well‐ordered lamellar and cylindrical domains as well as disordered micellar structures. Temperature‐dependent rheological measurements exhibited an order–disorder transition for SF 17/8 (the numbers refer to the relative molar masses in 103 g/mol) between 170 and 180°C, and an order–order transition for SF 9/19 between 190 and 200°C. The morphologies of binary blends of the diblocks with homopolymer were also investigated. In the blends the molar mass of the homopolymer was always less than the molar mass of the matching block. Ordered spheres on a bcc lattice and double‐gyroid morphology were observed for the blends. A series of poly(lactic acid) (PLA) films that including fully paraben substituted triazine derivatives having anti-bacterial properties have been prepared by utilizing the solvent-casting method. PLA as biodegradable polymer, poly(ethylene glycol) (PEG) as a plasticizing agent and s-triazine molecules (TA01, TA02, TA03, TA04, and TA05) behaving as an anti-bacterial component have been utilized in the experiments. The influence of TA compounds on the antibacterial performance of PLA/PEG films was investigated for the first time against Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus) bacteria via the contact active method. TA01-03-05 incorporated PLA/PEG films gave the best results against E.coli bacteria and log10 reductions of these films were 0.78, 0.64, and 0.65 respectively. The effect of TA compounds on the cell viability was investigated against cancer and non-cancerous cell lines using an MTS assay. The results showed that TA compounds had a positive effect on cell growth in non-cancerous cells, while they had a negative effect on cell growth in cancer cells. Furthermore, the addition of TA considerably increased the decomposition temperatures from 349° to 361° and char yield from 0.65 for PLA/PEG film to 2.3 for PLA/PEG/TA05. All of the films had good transparency and low opacity which was 7.2 for pure PLA used for control and the maximum opacity value was 11.2 observed for PLA/PEG/01. TA03 and TA04 caused a decrement of water vapor permission when compared to PLA/PEG films from 1439 to 749 and 664. It was also observed that pure PLA/PEG film lost weight rapidly over time during degradation tests. On the other hand, weight loss wasn't observed in PLA/PEG/TA films. This study focused on demonstrating the use of our new, flexible PLA/PEG derivatives in food and medical packaging.  相似文献

Block copolymerization by using isocyanates is an effective method for incorporating PHB and PEG because it can prepare copolymers with good properties, such as toughness, strength, and so on. In this study, we adopted soil suspension system to estimate the biodegradability of a series of PHB/PEG multiblock copolymers with different compositions and block lengths. In the degradation process, the changes in weight loss, molecular weight, and tensile strength were periodically measured to determine the biodegradability, and the surface morphology was also observed by SEM. In contrast to pure PHB, the weight loss of the copolymer was relatively lower. On the other hand, the tensile strength and molecular weight experienced apparent decrease, and for BHG1000-3-1, they reached 46.7% and 77.7% of the initial value, respectively. SEM observation showed that the surface was covered with numerous erosion pits. All these indicate that the degradation indeed took place and long-chain molecules have been hydrolyzed into shorter ones. The crystallization behavior was also investigated by DSC and WAXD. The results showed that both the segments, PEG and PHB, can form crystalline phases at lower PHB contents ranging from 29% to 44%, and when PHB component was more than 60%, only PHB phase can crystallize.  相似文献

The confined crystallization behavior, melting behavior, and nonisothermal crystallization kinetics of the poly(ethylene glycol) block (PEG) in poly(L ‐lactide)–poly(ethylene glycol) (PLLA–PEG) diblock copolymers were investigated with wide‐angle X‐ray diffraction and differential scanning calorimetry. The analysis showed that the nonisothermal crystallization behavior changed from fitting the Ozawa equation and the Avrami equation modified by Jeziorny to deviating from them with the molecular weight of the poly(L ‐lactide) (PLLA) block increasing. This resulted from the gradual strengthening of the confined effect, which was imposed by the crystallization of the PLLA block. The nucleation mechanism of the PEG block of PLLA15000–PEG5000 at a larger degree of supercooling was different from that of PLLA2500–PEG5000, PLLA5000–PEG5000, and PEG5000 (the numbers after PEG and PLLA denote the molecular weights of the PEG and PLLA blocks, respectively). They were homogeneous nucleation and heterogeneous nucleation, respectively. The PLLA block bonded chemically with the PEG block and increased the crystallization activation energy, but it provided nucleating sites for the crystallization of the PEG block, and the crystallization rate rose when it was heterogeneous nucleation. The number of melting peaks was three and one for the PEG homopolymer and the PEG block of the diblock copolymers, respectively. © 2006 Wiley Periodicals, Inc. We investigated the segmental and terminal relaxation dynamics of a well‐characterized disordered diblock copolymer, poly(isoprene‐b‐vinyl ethylene) (PI‐PVE), and miscible blends of polyisoprene (PI)/poly(vinyl ethylene) (PVE), using dielectric and viscoelastic spectroscopies. Generally, the concentration fluctuation (CF) amplitude of a disordered diblock copolymer is smaller than that of the miscible blend, especially in a length scale longer than the size of the whole block chain. To test whether the difference in the CF amplitudes causes the difference in the segmental relaxation spectra, we compared the shape of the dielectric loss curves between PI‐PVE and PI/PVE with the same composition (PI/PVE ratio = 17:83). However, no appreciable difference was observed, indicating that the CF amplitudes in PI‐PVE and PI/PVE are not so different in the length scale of the segmental motions. We also examined the effect of distinct friction coefficients of the PI and PVE chains on the terminal relaxation dynamics by comparisons of the viscoelastic and dielectric normal mode relaxations in PI‐PVE. The former probes the whole chain motion and the latter probes motions of the PI block. Shift factors (aT) for the viscoelastic and dielectric relaxations were compared. The dielectric normal mode aT was found to have weaker temperature dependence than the viscoelastic aT, which indicates that the friction for the PI block chain is lower than the average friction for the PI‐PVE chain. © 2004 Wiley Periodicals, Inc. A series of novel amphiphilic diblock copolymers composed of hydrophilic linear poly(ethylene glycol) (PEG) and linear brush hydrophobic polydimethylsiloxane (PDMS) were synthesized. Three different molecular weights of monomethyl ether PEG were initially functionalized with 2‐bromoisobutyryl bromide to afford macroinitiators suitable for atom‐transfer radical polymerization. The macroinitiators were characterized by gel permeation chromatography, 1H and 13C nuclear magnetic resonance spectroscopic analysis and matrix‐assisted laser desorption ionization time‐of‐flight mass spectroscopy. The three different molecular weight macroinitiators were then chain extended with monomethacryloxypropyl‐terminated PDMS and photoactive 2‐(methylacyloyloxy)ethyl anthracene‐9‐carboxylate in different molar ratios to afford a series of photoresponsive amphiphilic diblock copolymers with high conversions. Self‐assembly of these linear–linear brush diblock copolymers in N,N‐dimethylformamide afforded nanoparticles with hydrodynamic diameters (dH) ranging from 41 to 268 nm, as determined by dynamic light scattering analysis. Crosslinking and stabilization of the nanoparticles was achieved via [4+4] photodimerization of the anthracene moieties upon exposure to UV radiation at 365 nm with the reverse reaction studied at a wavelength of 254 nm. Thermo-responsive block copolymers poly(ethylene glycol)-block-poly(N-acryloyl-2,2-dimethyl-1,3-oxazolidine), PEG-b-PADMO,based on linear PEG were prepared via a versatile reversible addition-fragmentation chain transfer(RAFT) polymerization.PEG22(Mw = 1000) was used as the hydrophilic component,whose dehydration was the main driving force for the phase transition of these copolymers,as demonstrated by the 1H-NMR spectra.Their lower critical solution temperatures(LCSTs) could be tuned in the range of 20℃to 35℃,by adjusting the degree of polymerization(DP) of PADMO between 14-27.Furthermore,a sharp phase transition at ca.33℃,close to the physiological temperature with minimal hysteresis,was observed for the PEG22-b-PADMO14 copolymer.Moreover,excellent reversibility and reproducibility were displayed for the same copolymer over 10 cycles of repeated temperature change between 25℃(below the LCST) and 40℃(above the LCST).  相似文献

Copolymers of methacrylic acid (MAA) and poly(ethylene glycol) methyl ether methacrylate (PEGMA) were prepared and their cloud points in aqueous solution were studied as a function of comonomer ratio, solution pH, and presence of hydrophobic comonomers. Under acidic conditions, the cloud point falls below 0 °C for copolymers with between 25% to 60% ether content, because of the formation of hydrophobic H‐bonded ether–acid complexes. The cloud point also decreases with solution pH. For equivalent ether to acid ratios, the cloud point decreases with decreasing PEG chain length, because of the presence of a larger number of hydrophobic methyl and methacrylate groups. Similarly, the cloud point decreases upon incorporation of hydrophobic comonomers such as butyl, lauryl, or glycidyl methacrylates. © 2005 Wiley Periodicals, Inc. The mechanical, thermal and biodegradable properties of poly(d,l-lactide) (PDLLA), poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) and poly(ethylene glycol) (PEG) blends were studied. The influence of PEG on the tensile and impact strengths of the blends was investigated. The results showed that the toughness and elongation at break of the PDLLA/PHBV (70/30) blends were greatly improved by the addition of PEG, and the notched Izod impact strength increased about 400% and the elongation at break increased from 2.1% to 237.0%. The thermal and degradation properties of the blends were investigated by differential scanning calorimeter (DSC) and thermogravimetric analyzer (TGA), it was found that the thermal stability of PHBV in the presence of PDLLA was improved. The degradation test showed that the addition of PEG could notably accelerate the biodegradation of the blends in the soil at room temperature, and the mass loss is about 20% after 30 days of the storage.  相似文献
