In this research, thermo‐ and pH‐responsive chitosan‐based porous nanoparticles were prepared by the temperature‐dependent self assembly method. The chitosan‐graft‐poly(N‐isopropylacrylamide) (CS‐g‐PNIPAAm) copolymer solution was prepared through polymerization of N‐isopropylacrylamide (NIPAAm) monomer in the presence of chitosan (CS) solution using cerium ammounium nitrate as the initiator. Then, CS‐g‐PNIPAAm solution was diluted by deionized water and heated to 40 °C for CS‐g‐PNIPAAm self‐assembly. After that, CS‐g‐PNIPAAm assembled to form micelles in which shell layer was CS. Crosslinking agent was used to reinforce the micelle structure to form nanoparticle. The molar ratio of CS/NIPAAm in the feed mixture was changed to investigate its effect on structure, morphology, thermal‐ and pH‐responsive properties of the nanoparticles. TEM images showed that a porous structure of nanoparticles was developed. The synthesized nanoparticles carried positive charges on the surface and exhibited stimuli‐responsive properties, and their mean diameter thus could be manipulated by changing pH value and temperature of the environment. The nanoparticles showed a continuous release of the encapsulated doxycycline hyclate up to 10 days during an in‐vitro release experiment. These porous particles with environmentally sensitive properties are expected to be utilized in hydrophilic drug delivery system. © 2009 Wiley Periodicals, Inc. Polysulfone‐g‐poly(N‐isopropylacrylamide) (PSf‐g‐PNIPAAm) graft copolymers were prepared from atom transfer radical polymerization of NIPAAm using chloromethylated PSf as a macro‐initiator. The chain lengths of PNIPAAm of the graft copolymers were controllable with polymerization reaction time. The chemical structures of the graft copolymers were characterized with FTIR, NMR, and elemental analysis and their amphiphilic characteristics were examined and discussed. The PSf‐g‐PNIPAAm graft copolymers and the nanoparticles made from the graft copolymers exhibited repeatable temperature‐responsive properties in heating–cooling cycles. © 2008 Wiley Periodicals, Inc. In this research, thermo‐ and pH‐responsive nanoparticles with an average diameter of about 50–200 nm were synthesized via the surfactant‐free emulsion polymerization. The thermal/pH dual responsive properties of these nanoparticles were designed by the addition of a pH sensitive monomer, acrylic acid (AA), to be copolymerized with N‐isopropylacrylamide (NIPAAm) in a chitosan (CS) solution. The molar ratio of CS/AA/NIPAAm in the feed was changed to investigate its effect on structure, morphology, thermal‐ and pH‐responsive properties of the nanoparticles. It was found that CS‐PAA‐PNIPAAm nanoparticles could be well dispersed in the aqueous solution and carried positive charges on the surface. The addition of thermal‐sensitive NIPAAm monomer affected the polymerization mechanism and interactions between CS and AA. The particle size of the nanoparticles was found to be varied with the composition of NIPAAm monomer in the feed. The synthesized nanoparticles exhibited stimuli‐responsive properties, and their mean diameter thus could be manipulated by changing pH value and temperature of the environment. The nanoparticles showed a continuous release of the encapsulated doxycycline hyclate up to 10 days during an in vitro release experiment. Novel thermo‐responsive poly(N‐isopropylacrylamide)‐block‐poly(l ‐lactide)‐block‐poly(N‐isopropylacylamide) (PNIPAAm‐b‐PLLA‐b‐PNIPAAm) triblock copolymers were successfully prepared by atom transfer radical polymerization of NIPAAm with Br‐PLLA‐Br macroinitiator, using a CuCl/tris(2‐dimethylaminoethyl) amine (Me6TREN) complex as catalyst at 25 °C in a N,N‐dimethylformamide/water mixture. The molecular weight of the copolymers ranges from 18,000 to 38,000 g mol?1, and the dispersity from 1.10 to 1.28. Micelles are formed by self‐assembly of copolymers in aqueous medium at room temperature, as evidenced by 1H NMR, dynamic light scattering (DLS) and transmission electron microscopy (TEM). The critical micelle concentration determined by fluorescence spectroscopy ranges from 0.0077 to 0.016 mg mL?1. 1H NMR analysis in selective solvents confirmed the core‐shell structure of micelles. The copolymers exhibit a lower critical solution temperature (LCST) between 32.1 and 32.8 °C. The micelles are spherical in shape with a mean diameter between 31.4 and 83.3 nm, as determined by TEM and DLS. When the temperature is raised above the LCST, micelle size increases at high copolymer concentrations due to aggregation. A series of environmentally sensitive ABA triblock copolymers with different block lengths were prepared by reversible addition‐fragmentation chain transfer (RAFT) polymerization from acrylic acid (AA) and N‐isopropylacrylamide (NIPAAm). The GPC and 1H NMR analyses demonstrated the narrow molecular weight distribution and precise chemical structure of the prepared P(AA‐b‐NIPAAm‐b‐AA) triblock copolymers owing to the controlled/living characteristics of RAFT polymerization. The lower critical solution temperature (LCST) of the triblock copolymers could be tailored by adjusting the length of PAA block and controlled by the pH value. Under heating, the triblock copolymers underwent self‐assemble in dilute aqueous solution and formed nanoparticles revealed via TEM images. Physically crosslinked nanogels induced by inter‐/intra‐hydrogen bonding or core‐shell micelle particles thus could be obtained by changing environmental conditions. With a well‐defined structure and stimuli‐responsive properties, the P(AA‐b‐NIPAAm‐b‐AA) copolymer is expected to be employed as a nanocarrier for biomedical applications in controlled‐drug delivery and targeting therapy. © 2015 Wiley Periodicals, Inc. Carbon black nanoparticle grafted with poly(N‐isopropylacrylamide) (CB‐g‐PNIPAAm) was synthesized by surface‐initiated atom transfer radical polymerization (SI‐ATRP). The temperature‐responsive behavior of CB‐g‐PNIPAAm was proved by temperature‐variable 1H NMR. A temperature‐dependent conductive composite was prepared by blending CB‐g‐PNIPAAm with epoxy resin. The relationship between temperature and resistivity of the composite was studied: the composite exhibited a negative temperature coefficient (NTC) phenomenon. Possible mechanism for the NTC phenomenon was suggested. The results showed that resultant composites can be used in intelligent temperature‐switching. © 2008 Wiley Periodicals, Inc. We report here a reversible self‐assembly formation system using block copolymers with thermo‐tunable properties. A series of double‐responsive block copolymers, poly(N‐isopropylacrylamide (NIPAAm))‐block‐poly(NIPAAm‐coN‐(isobutoxymethyl)acrylamide (BMAAm)) with two lower critical solution temperatures were synthesized by one‐pot atom transfer radical polymerization via sequential monomer addition. When dissolved in aqueous solution at room temperature, the block copolymers remained unimeric. Upon heating above room temperature, the block copolymers self‐assembled into micellar structures. The micelle formation temperature and the resulting diameter were controlled by varying the BMAAm content. 1H Nuclear Magnetic Resonance, dynamic light scattering, field‐emission scanning electron microscopy, and fluorescence spectra revealed the presence of a monodisperse nanoassembly, and demonstrated the assembly formation/inversion process was fully reversible. Moreover, a model hydrophobic molecule, pyrene, was successfully loaded into the micelle core by including pyrene in the original polymer solution. Further heating resulted in mesoscopic micelle aggregation and precipitation. This dual micelle and aggregation system will find utility in drug delivery applications as a thermal trigger permits both aqueous loading of hydrophobic drugs and their subsequent release. © 2010 Wiley Periodicals, Inc. A series of poly(N‐isopropylacrylamide‐coN‐hydroxymethylacrylamide) P(NIPAM‐co‐NHMA) copolymers were firstly synthesized via free radical polymerization. Then, the hydrophobic, photosensitive 2‐diazo‐1,2‐naphthoquinone (DNQ) molecules were partially and randomly grafted onto P(NIPAM‐co‐NHMA) backbone through esterification to obtain a triple‐stimuli (photo/pH/thermo) responsive copolymers of P(NIPAM‐co‐NHMA‐co‐DNQMA). UV‐vis spectra showed that the lower critical solution temperature (LCST) of P(NIPAM‐co‐NHMA) ascended with increasing hydrophilic comonomer NHMA molar fraction and can be tailored by pH variation as well. The LCST of the P(NIPAM‐co‐NHMA) went down firstly after DNQ modification and subsequently shifted to higher value after UV irradiation. Meanwhile, the phase transition profile of P(NIPAM‐co‐NHMA‐co‐DNQMA) could be triggered by pH and UV light as expected. Thus, a triple‐stimuli responsive copolymer whose solution properties could be, respectively, modulated by temperature, light, and pH, has been achieved. The synthesis of multi‐arm poly([R]‐3‐hydroxybutyrate) (PHB)‐based triblock copolymers (poly([R]‐3‐hydroxybutyrate)‐b‐poly(N‐isopropylacrylamide)‐b‐[[poly(methyl ether methacrylate)‐g‐poly(ethylene glycol)]‐co‐[poly(methacrylate)‐g‐poly(propylene glycol)]], PHB‐b‐PNIPAAM‐b‐(PPEGMEMA‐co‐PPPGMA), and their subsequent self‐assembly into thermo‐responsive hydrogels is described. Atom transfer radical polymerization (ATRP) of N‐isopropylacrylamide (NIPAAM) followed by poly(ethylene glycol) methyl ether methacrylate (PEGMEMA) and poly(propylene glycol) methacrylate (PPGMA) was achieved from bromoesterified multi‐arm PHB macroinitiators. The composition of the resulting copolymers was investigated by 1H and 13C J‐MOD NMR spectroscopy as well as size‐exclusion chromatography (SEC), thermogravimetric analysis (TGA), and differential scanning calorimetry (DSC). The copolymers featuring different architectures and distinct hydrophilic/hydrophobic contents were found to self‐assemble into thermo‐responsive gels in aqueous solution. Rheological studies indicated that the linear one‐arm PHB‐based copolymer tend to form a micellar solution, whereas the two‐ and four‐arm PHB‐based copolymers afforded gels with enhanced mechanical properties and solid‐like behavior. These investigations are the first to correlate the gelation properties to the arm number of a PHB‐based copolymer. All copolymers revealed a double thermo‐responsive behavior due to the NIPAAM and PPGMA blocks, thus allowing first the copolymer self‐assembly at room temperature, and then the delivery of a drug at body temperature (37 °C). The non‐significant toxic response of the gels, as assessed by the cell viability of the CCD‐112CoN human fibroblast cell line with different concentrations of the triblock copolymers ranging from 0.03 to 1 mg mL?1, suggest that these PHB‐based thermo‐responsive gels are promising candidate biomaterials for drug‐delivery applications.  相似文献

Facile strategy was developed for the fabrication of the monodisperse superparamagnetic pH‐sensitive single‐layer chitosan (CS) hollow microspheres with controllable structure. The carboxyl group‐functionalized polystyrene microspheres prepared by soap‐free emulsion polymerization were used as the templates. After the Fe3O4 nanoparticles were in situ formed onto the surface of the templates, the single‐layer CS was self‐assembled and cross‐linked with glutaraldehyde subsequently. Then, the magnetic single‐layer CS hollow microspheres were obtained after the templates were removed. It was found that the feeding ratio of the monomer acrylic acid in the soap‐free emulsion polymerization had played an important role on the particle size and surface carboxyl group content of the templates, which determined the particle size and shell thickness of the magnetic single‐layer CS hollow microspheres in the proposed strategy. © 2010 Wiley Periodicals, Inc. Fluorescence‐incorporated, crosslinker‐free, pH‐ and thermoresponsive nanocarriers were prepared by the incorporation of drug molecules into the thermoresponsive nanocapsules, which composed of poly(N‐isopropylacrylamide) (PNIPAAm) with carboxylic acid end groups via temperature induced self‐assembling method. Well‐defined, pH‐responsive carboxylic acid group‐ended PNIPAAm homopolymer (HOOC? PNIPAAm? COOH) was synthesized by reversible addition fragmentation chain transfer polymerization with S,S′‐bis(α,α′‐dimethyl‐α″‐acetic acid)trithiocarbonate (CMP) as a chain transfer agent. Rhodamine 6G (R6G), the model drug, was used for three kinds of application: First, the nanostructure fixing; second, the fluorescence‐labeling; and last, the controlled release modeling. The transmission electron microscope images showed the solution type dosing led to the encapsulation of drug molecules into the nanocarriers, while the powder‐type drug‐loading process significantly contributed to the structure preservation of nanocarriers. The controlled release behaviors with various pH values and temperatures were evaluated. Poly(N‐isopropylacrylamide) (PNIPAAm) gels are temperature‐responsive polymer gels; and were prepared by redox polymerization of N‐isopropylacrylamide in the presence of N,N′‐methylenebisacrylamide as a crosslinking reagent and core‐shell type bioconjugates, which were core‐crosslinked polyion complex micelles formed from the mixture of bovine pancreas trypsin and poly(ethylene glycol)‐block‐poly(α,β‐aspartic acid). The phase transition temperature of PNIPAAm gels was no change with physically immobilization of bioconjugates. Also, the enzymatic activity of bioconjugates was essentially maintained even in PNIPAAm gels, although enzymatic reaction rate was apparently controlled by temperature, i.e., by the degree of swelling of PNIPAAm gels. Further, the control of enzymatic reaction synchronizing the phase transition of PNIPAAm gels immobilized bioconjugates. PNIPAAm gels could immobilize core‐shell type bioconjugates, and were successfully prepared without interfering with the properties of temperature‐responsive polymer gels and the bionanoreactor. © 2007 Wiley Periodicals, Inc. 4‐Hydroxythiophenol (HTP) grafted poly(styrene‐b‐acrylic acid) (PS‐b‐PAA) block copolymers (BCPs) (PS‐b‐PAA‐g‐HTP) was synthesized using the esterification reaction between the carboxyl groups and hydroxyl groups. Self‐assembly behavior of the graft copolymer in 1,4‐dioxane/water was investigated. Assemblies of different morphologies, porous, and bowl‐shaped structures, could be easily prepared. A possible mechanism for the formation of the porous and bowl‐shaped structures was discussed. The present study showed a facile method for the preparation of functionalized PS‐b‐PAA BCPs, which could easily self‐assembly into novel structures in aqueous solution. Polymers with multiple tunable responses were achieved by incorporating boronic acid functionality along the backbone of a thermoresponsive polymer. The inherent Lewis acidity and diol‐sensitivity of boronic acid moieties allowed these polymers to respond to changes in pH and glucose concentration. Through reversible addition‐fragmentation chain transfer copolymerization of boronic acid‐containing monomers with N‐isopropylacrylamide, well‐defined block copolymers were synthesized containing a hydrophilic N,N‐dimethylacrylamide block and a second, responsive block with temperature‐dependent water solubility, making the resulting polymers capable of self‐assembly into nanostructures upon heating. By incorporating boronic acids within the thermoresponsive block, the cloud point of the polymer depended on the solution conditions, including pH and diol concentration, allowing tunable cloud point ranges. © 2017 Wiley Periodicals, Inc. A three‐step process, combining nitroxide‐mediated polymerization (NMP) and reversible addition‐fragmentation chain transfer (RAFT) polymerization techniques, for synthesizing well‐defined amphiphilic and thermosensitive graft copolymers with fluorescence poly(styrene‐co‐(p‐chloromethylstyrene))‐g‐poly(N‐isopropylacrylamide) (P(St‐co‐(p‐CMS))‐g‐PNIPAAM), was conducted. Firstly, the NMP of styrene (St) and p‐chloromethylstyrene (p‐CMS) were carried out using benzoyl peroxide (BPO) as the initiator to obtain the random copolymers of P(St‐co‐(p‐CMS)). Secondly, the random copolymers were converted into macro‐RAFT agents with fluorescent carbazole as Z‐group through a simple method. Then the macro‐RAFT agents were used in the RAFT polymerization of N‐isopropylacrylamide (NIPAAM) to prepare fluorescent amphiphilic graft copolymers P(St‐co‐(p‐CMS))‐g‐PNIPAAM with controlled molecular weights and well‐defined structures. The copolymers obtained were characterized by gel permeation chromatography (GPC), 1H nuclear magnetic resonance (NMR) spectroscopy, and FT‐IR spectroscopy. The size of self‐assembly micelles of the resulting graft copolymers in deionized water was studied by high performance particle sizer (HPPS), the results showed that the Z‐average size of the micelles increased with the increase of molecular weights of PNIPAAM in side chains. The aqueous solution of the micelles prepared from P(St‐co‐(p‐CMS))‐g‐PNIPAAM using a dialysis method showed a lower critical solution temperature (LCST) at ~ 27.5 °C, which was below the value of NIPAAM homopolymer (32 °C). © 2007 Wiley Periodicals, Inc. Poly(N‐isopropylacrylamide)‐block‐poly(ethylene oxide)‐block‐poly(N‐isopropylacrylamide) (PNIPAAm‐b‐PEO‐b‐PNIPAAm) triblock copolymer was synthesized via the reversible addition‐fragmentation chain transfer/macromolecular design via the interchange of xanthate (RAFT/MADIX) process with xanthate‐terminated poly(ethylene oxide) (PEO) as the macromolecular chain transfer agent. The successful synthesis of the ABA triblock copolymer inspired the preparation of poly(N‐isopropylacrylamide)‐block‐poly(ethylene oxide) (PNIPAAm‐b‐PEO) copolymer networks with N,N′‐methylenebisacrylamide as the crosslinking agent with the similar approach. With the RAFT/MADIX process, PEO chains were successfully blocked into poly(N‐isopropylacrylamide) (PNIPAAm) networks. The unique architecture of PNIPAAm‐b‐PEO networks allows investigating the effect of the blocked PEO chains on the deswelling and reswelling behavior of PNIPAAm hydrogels. It was found that with the inclusion of PEO chains into the PNIPAAm networks as midblocks, the swelling ratios of the hydrogels were significantly enhanced. Furthermore, the PNIPAAm‐b‐PEO hydrogels displayed faster response to the external temperature changes than the control PNIPAAm hydrogel. The accelerated deswelling and reswelling behaviors have been interpreted based on the formation of PEO microdomains in the PNIPAAm networks, which could act as the hydrophilic tunnels to facilitate the diffusion of water molecules in the PNIPAAm networks. © 2012 Wiley Periodicals, Inc. Self‐immolative polymers (SIPs) undergo depolymerization in response to the cleavage of stimuli‐responsive end‐caps from their termini. Some classes of SIPs, including polycarbamates, have depolymerization rates that depend on environmental factors such as solvent and pH. In previous work, hydrophobic SIPs have been incorporated into amphiphilic block copolymers and used to prepare nanoassemblies. However, stimuli‐responsive hydrophilic blocks have not previously been incorporated. In this work, we synthesized amphiphilic copolymers composed of a hydrophobic polycarbamate SIP block and a hydrophilic poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) block connected by a UV light‐responsive linker end‐cap. It was hypothesized that after assembly of the block copolymers into nanoparticles, chain collapse of the PDMAEMA above its lower critical solution temperature (LCST) might change the environment of the SIP block, thereby altering its depolymerization rate. Self‐assembly of the block copolymers was performed, and the depolymerization of the resulting assemblies was studied by fluorescence spectroscopy, dynamic light scattering, and NMR spectroscopy. At 20 °C, the system exhibited a selective response to the UV light. At 65 °C, above the LCST of PDMAEMA, the systems underwent more rapid depolymerization, suggesting that the increase in rate arising from the higher temperature dominated over environmental effects arising from chain collapse. © 2018 Wiley Periodicals, Inc. Dipeptide diphenylalanine has attracted significant research interests because of its ability to self‐assemble into various nanostructures such as nanotubes, nanowires, and nanoribbons. In this article, we present the synthesis and self‐assembly of a novel diphenylalanine‐based homopolymer and block/random copolymers by the reversible addition–fragmentation chain transfer (RAFT) polymerization of an acrylamide having a dipeptide moiety. The RAFT polymerization of N‐acryloyl‐l ,l ‐diphenylalanine (A‐Phe‐Phe‐OH) afforded novel amino acid‐based polymers with predetermined molecular weights and relatively narrow‐molecular weight distributions. The hierarchical self‐assembled structures of poly(A‐Phe‐Phe‐OH), which involve nanorods, larger nanofiber‐like microcrystals, and fiber bundles, were characterized by atomic force microscopy (AFM), transmission electron microscopy, scanning electron microscopy, and dynamic light scattering measurements. The circular dichroic measurements of poly(A‐Phe‐Phe‐OH) revealed its characteristic chiroptical property, which is affected by the nature of the solvents and the addition of urea and salts via hydrophobic, hydrogen bonding, and electrostatic interactions. Thermo‐ and pH‐responsive block and random copolymers composed of A‐Phe‐Phe‐OH and N‐isopropylacrylamide were synthesized by RAFT polymerization, and the thermoresponsive properties and assembled structures of the resulting copolymers were investigated by AFM and turbidity measurements. © 2019 Wiley Periodicals, Inc. Pyrrolyl‐capped poly(N‐isopropylacrylamide) macromonomers (Py‐PNIPAM) were prepared through reversible addition‐fragmentation‐transfer polymerization with benzyl 1‐pyrrolylcarbodithioate as chain‐transfer agent. Polymerizations of Py‐PNIPAM with/without pyrrole using AgNO3 as oxidizing agent and dimethylforamide as solvent resulted in graft copolymers of polypyrrole‐graft‐poly(N‐isopropylacrylamide) (PPy‐g‐PNIPAM) as well as silver nanoparticles, leading to the formation of PPy‐g‐PNIPAM/silver nanocomposites. The resulting nanocomposites were soluble in water when the content of PPy was low, and when the molar ratio of Py/Py‐PNIPAM increased to 30, the resulting products became insoluble in water. The resulting nanocomposites had special optical properties because of PPy as well as the temperature‐responsible PNIPAM. The chemical structure and composition of nanocomposite were characterized by 1H nuclear magnetic resonance spectroscopy, gel permeation chromatograms, fourier transform infrared spectroscopy, and X‐ray diffraction. Thermoresponsive and pH‐responsive graft copolymers, poly(L ‐glutamate)‐g‐oligo(2‐(2‐(2‐methoxyethoxy)ethoxy)ethyl methacrylate) and poly(L ‐glutamic acid‐co‐(L ‐glutamate‐g‐oligo(2‐(2‐(2‐methoxyethoxy)ethoxy)ethyl methacrylate))), were synthesized by ring‐opening polymerization (ROP) of N‐carboxyanhydride (NCA) monomers and subsequent atom transfer radical polymerization of 2‐(2‐(2‐methoxyethoxy)ethoxy)ethyl methacrylate. The thermoresponsiveness of graft copolymers could be tuned by the molecular weight of oligo(2‐(2‐(2‐methoxyethoxy)ethoxy)ethyl methacrylate) (OMEO3MA), composition of poly(L ‐glutamic acid) (PLGA) backbone and pH of the aqueous solution. The α‐helical contents of graft copolymers could be influenced by OMEO3MA length and pH of the aqueous solution. In addition, the graft copolymers exhibited tunable self‐assembly behavior. The hydrodynamic radius (Rh) and critical micellization concentration values of micelles were relevant to the length of OMEO3MA and the composition of biodegradable PLGA backbone. The Rh could also be adjusted by the temperature and pH values. Lastly, in vitro methyl thiazolyl tetrazolium (MTT) assay revealed that the graft copolymers were biocompatible to HeLa cells. Therefore, with good biocompatibility, well‐defined secondary structure, and mono‐, dual‐responsiveness, these graft copolymers are promising stimuli‐responsive materials for biomedical applications. © 2011 Wiley Periodicals, Inc.
