Poly(anhydride-ester)s and poly(anhydride-amide)s derived from both 4- and 5-aminosalicylate acids (4- and 5-ASA) were synthesized and characterized by physicochemical methods. Thermal and solubility characteristics directly correlated to the polymer backbone composition; polymers based on 5-ASA had greater solubilities in organic solvents than polymers based on 4-ASA, and the poly(anhydride-ester)s thermally decomposed at temperatures nearly 100 °C higher than the corresponding poly(anhydride-amide)s. The polymers were self-contained, controlled-release systems that combine the drug and controlled-release mechanism into the polymer backbone. The erosion and degradation characteristics of the polymers were measured in physiologically relevant media. All polymer matrices fully degraded in media buffered to pH 7.4, whereas in acidic media (pH 1.2), all polymer matrices maintained greater than 50% mass over a 90-day time period. © 2003 Wiley Periodicals, Inc. Fast‐degrading, salicylate‐based poly(anhydride‐esters) were designed to degrade and release the active component, salicylic acid (SA), within 1 week. The polymer degradation was enhanced by using shorter or oxygen‐containing aliphatic chains. A copolymer of diglycolic acid was also made with a salicylate‐based diacid for comparison of polymer properties, including SA release. Both methods resulted in polyanhydrides with molecular weights ranging from 14 500 to 27 800 Da and displayed glass transition temperatures near physiological conditions, namely 33–40 °C. The homo‐ and copolymers completely degraded within one week releasing the chemically incorporated SA.

A new graft copolymer, poly(2‐hydroxyethyl methacrylate‐co‐styrene) ‐graft‐poly(?‐caprolactone), was prepared by combination of reversible addition‐fragmentation chain transfer polymerization (RAFT) with coordination‐insertion ring‐opening polymerization (ROP). The copolymerization of styrene (St) and 2‐hydroxyethyl methacrylate (HEMA) was carried out at 60 °C in the presence of 2‐phenylprop‐2‐yl dithiobenzoate (PPDTB) using AIBN as initiator. The molecular weight of poly (2‐hydroxyethyl methacrylate‐co‐styrene) [poly(HEMA‐co‐St)] increased with the monomer conversion, and the molecular weight distribution was in the range of 1.09 ~ 1.39. The ring‐opening polymerization (ROP) of ?‐caprolactone was then initiated by the hydroxyl groups of the poly(HEMA‐co‐St) precursors in the presence of stannous octoate (Sn(Oct)2). GPC and 1H‐NMR data demonstrated the polymerization courses are under control, and nearly all hydroxyl groups took part in the initiation. Six new triorganotin complexes ( 1a – 1c and 2a – 2c ) of 5‐(salicylideneamino)salicylic acid, [5‐(3‐X‐2‐HOC6H3CH═N)‐2‐HOC6H3COO]SnR3 (X = H, 1 ; CH3O, 2 ; R = Ph, a ; Cy, b ; CH2C(CH3)2Ph, c ), have been synthesized by one‐pot reaction of 5‐aminosalicylic acid, salicylaldehyde and triorganotin hydroxide and characterized using elemental analysis and infrared and NMR (1H, 13C and 119Sn) spectra. The crystal structures of 1a , 1b , 2a ·CH3OH, 2b ·CH3OH and 2c ·CHCl3 have been determined using single‐crystal X‐ray diffraction. In non‐coordinated solvent CDCl3, the tin atoms in the complexes are all four‐coordinated. In the crystalline state, these compounds adopt a four‐ or five‐coordination mode. Complex 1a exhibits a 44‐membered macrocyclic tetrameric structure with trigonal bipyramidal geometry around the tin atoms in which the axial positions are occupied by the oxygen atom of carboxylate group of the ligand and the phenolic oxygen atom from the adjacent ligand. The coordination geometry of tin atom in 1b and 2c ·CHCl3 is a distorted tetrahedron shaped by three carbon atoms of alkyl groups and a carboxylate oxygen atom of the ligand. In 2a ·CH3OH and 2b ·CH3OH, the tin atom has a distorted trans‐C3SnO2 trigonal bipyramidal geometry formed by three alkyl groups, a monodentate carboxylate group and a coordinated methanol molecule. The molecules of 2a ·CH3OH and 2b ·CH3OH are linked via O─H···O hydrogen bonds into a one‐dimensional supramolecular chain and a centrosymmetric R44(22) macrocycle, respectively. Bioassay results against two human tumor cell types (A549 and HeLa) show the complexes are efficient cytostatic agents and may be explored as potential antitumor drugs.  相似文献

A well‐defined amphiphilic copolymer of ‐poly(ethylene oxide) (PEO) linked with comb‐shaped [poly(styrene‐co‐2‐hydeoxyethyl methacrylate)‐graft‐poly(ε‐caprolactone)] (PEO‐b‐P(St‐co‐HEMA)‐g‐PCL) was successfully synthesized by combination of reversible addition‐fragmentation chain transfer polymerization (RAFT) with ring‐opening anionic polymerization and coordination–insertion ring‐opening polymerization (ROP). The α‐methoxy poly(ethylene oxide) (mPEO) with ω,3‐benzylsulfanylthiocarbonylsufanylpropionic acid (BSPA) end group (mPEO‐BSPA) was prepared by the reaction of mPEO with 3‐benzylsulfanylthiocarbonylsufanyl propionic acid chloride (BSPAC), and the reaction efficiency was close to 100%; then the mPEO‐BSPA was used as a macro‐RAFT agent for the copolymerization of styrene (St) and 2‐hydroxyethyl methacrylate (HEMA) using 2,2‐azobisisobutyronitrile as initiator. The molecular weight of copolymer PEO‐b‐P(St‐co‐HEMA) increased with the monomer conversion, but the molecular weight distribution was a little wide. The influence of molecular weight of macro‐RAFT agent on the polymerization procedure was discussed. The ROP of ε‐caprolactone was then completed by initiation of hydroxyl groups of the PEO‐b‐P(St‐co‐HEMA) precursors in the presence of stannous octoate (Sn(Oct)2). Thus, the amphiphilic copolymer of linear PEO linked with comb‐like P(St‐co‐HEMA)‐g‐PCL was obtained. An amphiphilic block copolymer of poly(N‐vinyl pyrrolidone)‐b‐poly(ε‐caprolactone) (PVP‐b‐PCL) was synthesized by a combination of cobalt‐mediated radical polymerization (CMRP) and ring‐opening polymerization (ROP). The micellar characteristics of this copolymer were subsequently investigated. PVP (Mn = 11,400, Mw/Mn = 1.32) was synthesized at 20 °C via CMRP using a molar ratio of [VP]0/[V‐70]0/[Co]0 = 150/8/1. The PVP was then reacted with 2,2′‐azobis[2‐methyl‐N‐(2‐hydroxyethyl)propionamide] (VA‐086) to modify its cobalt complex chain end to a hydroxyl group. The cobalt (Co) content in the resulting PVP‐OH was 1.2 ppm, indicating that all of the covalent Co? C bonds were cleaved and reacted with VA‐086, and that the separated cobalt complexes were successfully removed. The ROP of CL was subsequently carried out using the produced PVP‐OH as a macroinitiator at 110 °C. The GPC trace of PVP‐b‐PCL was monomodal without any tailing caused by the residual PVP‐OH, indicating that the initiation efficiency was very high. The critical micelle concentration (CMC) of PVP‐b‐PCL (Mn = 18,000, Mw/Mn = 1.35) was 0.015 mg/mL. The PVP‐b‐PCL micelles were spherical in shape with an average diameter of 105 nm. The nanosized PVP‐b‐PCL micelles show promise as novel drug carriers in biomedical and pharmaceutical applications. © 2009 Wiley Periodicals, Inc. Poly(N‐phenylitaconimide) (polyPhII) was prepared using initiators for continuous activator regeneration atom transfer radical polymerization of PhII using FeBr3 complexes as catalysts. Conversion reached 69% in 24 h, yielding polyPhII with a number average molecular weight Mn = 11,900 and a molecular weight distribution Mw/Mn = 1.52. Copolymerizations of PhII with styrene at various molar ratios were performed providing a range of polyPhII‐copolySt polymers. When the copolymerization was carried out with higher [St]0 > [PhII]0 ratio, a one‐pot synthesis of poly(St‐alt‐PhII)‐b‐polySt was achieved. The thermal properties of the obtained copolymers were studied by differential scanning calorimetry. PolyPhII prepared by ATRP showed high glass transition temperature (Tg) of 216 °C and the poly(St‐alt‐PhII)‐b‐polySt exhibited two Tgs, at 162 and 104 °C, corresponding to a poly(St‐alt‐PhII) and polySt segments, respectively. © 2014 Wiley Periodicals, Inc. A series of well‐defined ABC 3‐Miktoarm star‐shaped terpolymers [Poly(styrene)‐Poly(ethylene oxide)‐Poly(ε‐caprolactone)](PS‐PEO‐PCL) with different molecular weight was synthesized by combination of the “living” anionic polymerization with the ring‐opening polymerization (ROP) using macro‐initiator strategy. Firstly, the “living” poly(styryl)lithium (PS?Li+) species were capped by 1‐ethoxyethyl glycidyl ether(EEGE) quantitatively and the PS‐EEGE with an active and an ethoxyethyl‐protected hydroxyl group at the same end was obtained. Then, using PS‐EEGE and diphenylmethylpotassium (DPMK) as coinitiator, the diblock copolymers of (PS‐b‐PEO)p with the ethoxyethyl‐protected hydroxyl group at the junction point were achieved by the ROP of EO and the subsequent termination with bromoethane. The diblock copolymers of (PS‐b‐PEO)d with the active hydroxyl group at the junction point were recovered via the cleavage of ethoxyethyl group on (PS‐b‐PEO)p by acidolysis and saponification successively. Finally, the copolymers (PS‐b‐PEO)d served as the macro‐initiator for ROP of ε‐CL in the presence of tin(II)‐bis(2‐ethylhexanoate)(Sn(Oct)2) and the star(PS‐PEO‐PCL) terpolymers were obtained. (2004) 42(22) 5845 New multiblock copolymers derived from poly(L‐lactic acid) (PLLA) and poly(ε‐caprolactone) (PCL) were prepared with the coupling reaction between PLLA and PCL oligomers with ? NCO terminals. Fourier transform infrared (FTIR), 13C NMR, and differential scanning calorimetry (DSC) were used to characterize the copolymers and the results showed that PLLA and PCL were coupled by the reaction between ? NCO groups at the end of the PCL and ? OH (or ? COOH) groups at the end of the PLLA. DSC data indicated that the different compositions of PLLA and PCL had an influence on the thermal and crystallization properties including the glass‐transition temperature (Tg), melting temperature (TM), crystallizing temperature (Tc), melting enthalpy (ΔHm), crystallizing enthalpy (ΔHc), and crystallinity. Gel permeation chromatography (GPC) was employed to study the effect of the composition of PLLA and PCL and reaction time on the molecular weight and the molecular weight distribution of the copolymers. The weight‐average molecular weight of PLLA–PCL multiblock copolymers was up to 180,000 at a composition of 60% PLLA and 40% PCL, whereas that of the homopolymer of PLLA was only 14,000. A polarized optical microscope was used to observe the crystalline morphology of copolymers; the results showed that all polymers exhibited a spherulitic morphology. © 2004 Wiley Periodicals, Inc. A novel template monomer with multiple methacryloyl groups was synthesized with β‐cyclodextrin by the acetylation of primary hydroxyl groups and the esterification of secondary hydroxyl groups with methacrylic acid anhydride. The average number of methacryloyl groups in the monomer was 11. The radical polymerization of the monomer was carried out with the following initiators: α,α′‐azobisisobutylonitrile, H2O2? Fe2+ redox initiator, p‐xylyl‐N,N‐dimethyldithiocarbamate (XDC), and α‐bromo‐p‐xylyl‐N,N‐dimethyldithiocarbamate (BXDC). When the concentration of the monomer was less than 4.12 × 10?3 M, polymerization was limited inside the molecule, and gelation of the system was hindered. For controlled radical photopolymerization with XDC and BXDC, the methacryloyl groups of the monomer were homogeneously polymerized, and poly(methacrylic acid) with a narrow molecular weight distribution was obtained by the hydrolysis of the polymerized products. © 2001 John Wiley & Sons, Inc. Here, we report on the synthesis and different crystallization behavior of linear‐ and star‐ PCL's containing a photocleavable linker (5‐hydroxy‐2‐nitro benzaldehyde), modulated by photochemical switching. Basis is the attachment of a photocleavable moiety close to the star‐core of a three‐arm star poly(caprolactone), so that the crystallization behavior can be controlled via a photochemical stimulus. The polymerization of ε‐caprolactone using a trivalent photocleavable initiator and stannous octanoate catalyst resulted in the synthesis of different molecular weights of star‐shaped photocleavable polymers. Various techniques like 1H NMR and ESI‐TOF‐MS confirmed the successful synthesis of the star‐shaped polymers. Complete photocleavage is ensured via GPC, HPLC, and ESI‐TOF‐MS. DSC studies clearly indicated the enhancement in crystallinity after photocleavage of the star‐shaped poly(ε‐caprolactone)s. Hence, for the first time phototriggered crystallization behavior of PCL polymers is reported, where the confinement exerted by the star architecture is removed by photoirradiation. © 2014 Wiley Periodicals, Inc. The quasi‐living cationic copolymerization of 3,3‐bis(chloromethyl)oxetane (BCMO) and ε‐caprolactone (ε‐CL), using boron trifluoride etherate as catalyst and 1,4‐butanediol as coinitiator, was investigated in methylene chloride at 0°C. The resulting hydroxyl‐ended copolymers exhibit a narrow molecular weight polydispersity and a functionality of about 2. The reactivity ratios of BCMO (0.26) and ε‐CL (0.47), and the Tg of the copolymers, indicate their statistical character. The synthesis of poly(3,3‐bis(azidomethyl)oxetane‐co‐ε‐caprolactone) from poly(BCMO‐co‐ε‐CL) via the substitution of the chlorine atoms by azide groups, using sodium azide in DMSO at 110°C, occurs without any degradation, but the copolymers decompose at about 240°C. Novel amphiphilic A14B7 multimiktoarm star copolymers composed of 14 poly(ε‐caprolactone) (PCL) arms and 7 poly(acrylic acid) (PAA) arms with β‐cyclodextrin (β‐CD) as core moiety were synthesized by the combination of controlled ring‐opening polymerization (CROP) and atom transfer radical polymerization (ATRP). 14‐Arm star PCL homopolymers (CDSi‐SPCL) were first synthesized by the CROP of CL using per‐6‐(tert‐butyldimethylsilyl)‐β‐CD as the multifunctional initiator in the presence of Sn(Oct)2 at 125 °C. Subsequently, the hydroxyl end groups of CDSi‐SPCL were blocked by acetyl chloride. After desilylation of the tert‐butyldimethylsilyl ether groups from the β‐CD core, 7 ATRP initiating sites were introduced by treating with 2‐bromoisobutyryl bromide, which further initiated ATRP of tert‐butyl acrylate (tBA) to prepare well‐defined A14B7 multimiktoarm star copolymers [CDS(PCL‐PtBA)]. Their molecular structures and physical properties were in detail characterized by 1H NMR, SEC‐MALLS, and DSC. The selective hydrolysis of tert‐butyl ester groups of the PtBA block gave the amphiphilic A14B7 multimiktoarm star copolymers [CDS(PCL‐PAA)]. These amphiphilic copolymers could self‐assemble into multimorphological aggregates in aqueous solution, which were characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM) and atomic force microscopy (AFM). © 2010 Wiley Periodicals, Inc. A novel biodegradable amphiphilic copolymer with hydrophobic poly(ε‐caprolactone) branches containing cholic acid moiety and a hydrophilic poly(ethylene glycol) chain was synthesized. The copolymer was characterized by FTIR, 1H NMR, gel permeation chromatography (GPC), differential scanning calorimetry (DSC), polarizing light microscopy (PLM), and wide‐angle X‐ray diffraction (WAXD) analysis. The amphiphilic copolymer could self‐assemble into micelles in an aqueous solution. The critical micelle concentration of the amphiphilic copolymer was determined by fluorescence spectroscopy. A nanoparticle drug delivery system with a regularly spherical shape was prepared with high encapsulation efficiency. The in vitro drug release from the drug‐loaded polymeric nanoparticles was investigated. Because of the branched structure of the hydrophobic part of the copolymer and the relatively fast degradation rate of the copolymer, an improved release behavior was observed. © 2007 Wiley Periodicals, Inc. Poly(ethylene‐bε‐caprolactone) (PE‐b‐PCL) diblock copolymers were synthesized by ring‐opening polymerization (ROP) of ε‐caprolactone (CL) with α‐hydroxyl‐ω‐methyl polyethylene (PE‐OH) as a macroinitiator and ammonium decamolybdate (NH4)8[Mo10O34] as a catalyst. Polymerization was conducted in bulk (130–150°C) with high yield (87–97%). Block copolymers with different compositions were obtained and characterized by 1H and 13C NMR, MALDI‐TOF, SAXS, and DSC. End‐group analysis by NMR and MALDI‐TOF indicates the formation of α‐hydroxyl‐ω‐methyl PE‐b‐PCL. The PE‐b‐PCL degradation was studied using thermogravimetric analysis (TGA) and alkaline hydrolysis. A novel amphiphilic biodegradable triblock copolymer (PGL‐PLA‐PGL) with polylactide (PLA) as hydrophobic middle block and poly(glutamic acid) (PGL) as hydrophilic lateral blocks was successfully synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and N‐carboxy anhydride (NCA) consecutively and by subsequent catalytic hydrogenation. The results of cell experiment of PGL‐PLA‐PGL suggested that PGL could improve biocompatibility of polyester obviously. The copolymer could form micelles of spindly shape easily in aqueous solution. The pendant carboxyl groups of the triblock copolymer were further activated with N‐hydroxysuccinimide and combined with a cell‐adhesive peptide GRGDSY. Incorporation of the oligopeptide further enhanced the hydrophilicity and led to formation of spherical micelles. PGL‐PLA‐PGL showed better cell adhesion and spreading ability than pure PLA and the GRGDSY‐containing copolymer exhibited even further improvement in cell adhesion and spreading ability, indicating that the copolymer could find a promising application in drug delivery or tissue engineering. © 2007 Wiley Periodicals, Inc. Biodegradable copolymers of poly(lactic acid)‐block‐poly(ε‐caprolactone) (PLA‐b‐PCL) were successfully prepared by two steps. In the first step, lactic acid monomer is oligomerized to low molecular weight prepolymer and copolymerized with the (ε‐caprolactone) diol to prepolymer, and then the molecular weight is raised by joining prepolymer chains together using 1,6‐hexamethylene diisocyanate (HDI) as the chain extender. The polymer was carefully characterized by using 1H‐NMR analysis, gel permeation chromatography (GPC), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and Fourier transform infrared spectroscopy (FTIR). The results of 1H‐NMR and TGA indicate PLA‐b‐PCL prepolymer with number average molecular weights (Mn) of 4000–6000 were obtained. When PCL‐diols are 10 wt%, copolymer is better for chain extension reaction to obtain the polymer with high molecular weight. After chain extension, the weight average molecular weight can reach 250,000 g/mol, as determined by GPC, when the molar ratio of –NCO to –OH was 3:1. DSC curve showed that the degree of crystallization of PLA–PCL copolymer was low, even became amorphous after chain extended reaction. The product exhibits superior mechanical properties with elongation at break above 297% that is much higher than that of PLA chain extended products. Poly(styrene‐maleic anhydride)‐montmorillonite nanocomposites were prepared by intercalation of layered montmorillonite with the polymer ions. Synthetic approaches including polymerization and phosphonium salt formation have been used for polymer intercalation and dispersion of the host layers in the polymer matrix. The ratio of the mineral in the composites ranged 30–50%. Wide‐angle X‐ray diffraction (WAXD) disclosed that the d(001) spacing between the internal lamellar surface were only expanding to about 13 and 15 Å according to the type of phosphonium salt suggesting packing of polymer molecules between the layers. Examination of these materials by scanning and transmission electron microscopy showed spherical nano size particles of average diameter, 350 nm. Copyright © 2002 John Wiley & Sons, Ltd.  相似文献

Biodegradable, amphiphilic, four‐armed poly(?‐caprolactone)‐block‐poly(ethylene oxide) (PCL‐b‐PEO) copolymers were synthesized by ring‐opening polymerization of ethylene oxide in the presence of four‐armed poly(?‐caprolactone) (PCL) with terminal OH groups with diethylzinc (ZnEt2) as a catalyst. The chemical structure of PCL‐b‐PEO copolymer was confirmed by 1H NMR and 13C NMR. The hydroxyl end groups of the four‐armed PCL were successfully substituted by PEO blocks in the copolymer. The monomodal profile of molecular weight distribution by gel permeation chromatography provided further evidence for the four‐armed architecture of the copolymer. Physicochemical properties of the four‐armed block copolymers differed from their starting four‐armed PCL precursor. The melting points were between those of PCL precursor and linear poly(ethylene glycol). The length of the outer PEO blocks exhibited an obvious effect on the crystallizability of the block copolymer. The degree of swelling of the four‐armed block copolymer increased with PEO length and PEO content. The micelle formation of the four‐armed block copolymer was examined by a fluorescent probe technique, and the existence of the critical micelle concentration (cmc) confirmed the amphiphilic nature of the resulting copolymer. The cmc value increased with increasing PEO length. The absolute cmc values were higher than those for linear amphiphilic block copolymers. © 2004 Wiley Periodicals, Inc.
