Self‐assembled thermo‐ and pH‐responsive poly(acrylic acid)‐b‐poly(N‐isopropylacrylamide) (PAA‐b‐PNIPAM) micelles for entrapment and release of doxorubicin (DOX) was described. Block copolymer PAA‐b‐PNIPAM associated into core‐shell micelles in aqueous solution with collapsed PNIPAM block or protonated PAA block as the core on changing temperature or pH. Complexation of DOX with PAA‐b‐PNIPAM triggered by the electrostatic interaction and release of DOX from the complexes due to the changing of pH or temperature were studied. Complex micelles incorporated with DOX exhibited pH‐responsive and thermoresponsive drug release profile. The release of DOX from micelles was suppressed at pH 7.2 and accelerated at pH 4.0 due to the protonation of carboxyl groups. Furthermore, the cumulative release of DOX from complex micelles was enhanced around LCST ascribed to the structure deformation of the micelles. © 2008 Wiley Periodicals, Inc. The development of novel thermo‐ and photo‐dual‐responsive biodegradable polymeric micelles based on amphiphilic polyaspartamide derivatives (NB‐g‐PHPA‐g‐mPEG) for anticancer drug delivery is reported. The obtained polymers containing hydrophobic photocleavable o‐nitrobenzyl groups exhibit thermo‐ and photosensitivity. The micelles and paclitaxel‐loaded micelles based on the thermo‐ and photo‐dual‐sensitive polymers were prepared by a quick heating method without using toxic organic solvent. The paclitaxel release from the drug‐loaded micelles can be triggered under photoirradiation. Enhancement of the anticancer activity against HeLa cells was observed for paclitaxel‐loaded NB‐g‐PHPA‐g‐mPEG micelles after light irradiation, while the empty NB‐g‐PHPA‐g‐mPEG micelles with or without irradiation did not show any toxicity. Therefore, the thermo‐ and photo‐dual‐responsive NB‐g‐PHPA‐g‐mPEG micelles have a promising future applied as a light controlled drug delivery system for anticancer drugs. © 2016 Wiley Periodicals, Inc. For the preparation of core‐shell nanoparticles containing functional nanomaterials, a photo‐cross‐linkable amphiphilic ABC triblock copolymer, poly(ethylene glycol)‐b‐poly(2‐cinnamoyloxyethyl methacrylate)‐b‐poly(methyl methacrylate) (PEG‐PCEMA‐PMMA), was synthesized. This triblock copolymer was then used to encapsulate Au nanoparticles or pyrene. The triblock copolymer of PEG‐b‐poly(2‐hydroxyethyl methacrylate)‐b‐PMMA (PEG‐PHEMA‐PMMA) (Mn = 15,800 g/mol, Mw/Mn = 1.58) was first synthesized by activators generated by electron transfer atom transfer radical polymerization. Its middle block was then functionalized with cinnamoyl chloride. The degrees of polymerization of the PEG, PHEMA, and PMMA blocks were 45, 13, and 98, respectively. PMMA‐tethered Au nanoparticles (with an average diameter of 3.0 nm) or pyrene was successfully encapsulated within the PEG‐PCEMA‐PMMA micelles. The intermediary layers of the micelles were then cross‐linked by UV irradiation. The spherical structures of the PEG‐PCEMA‐PMMA micelles containing Au nanoparticles or pyrene were not changed by the photo‐cross‐linking process and they showed excellent colloidal stability. © 2009 Wiley Periodicals, Inc. The multi‐thermo‐responsive block copolymer of poly[2‐(2‐methoxyethoxy)ethyl methacrylate]‐block‐poly[N‐(4‐vinylbenzyl)‐N,N‐diethylamine] (PMEO2MA‐b‐PVEA) displaying phase transition at both the lower critical solution temperature (LCST) and the upper critical solution temperature (UCST) in the alcohol/water mixture is synthesized by reversible addition‐fragmentation chain transfer polymerization. The poly[2‐(2‐methoxyethoxy)ethyl methacrylate] (PMEO2MA) block exhibits the UCST phase transition in alcohol and the LCST phase transition in water, while the poly[N‐(4‐vinylbenzyl)‐N,N‐diethylamine] (PVEA) block shows the UCST phase transition in isopropanol and the LCST phase transition in the alcohol/water mixture. Both the polymer molecular weight and the co‐solvent/nonsolvent exert great influence on the LCST or UCST of the block copolymer. By adjusting the solvent character including the water content and the temperature, the block copolymer undergoes multiphase transition at LCST or UCST, and various block copolymer morphologies including inverted micelles, core‐corona micelles, and corona‐collapsed micelles are prepared. © 2013 Wiley Periodicals, Inc. A double‐responsive amphiphilic random copolymer (P(OEtOxA)‐ran‐PNBA) composed of thermoresponsive poly(oligo(2‐ethyl‐2‐oxazoline)acrylate) (P(OEtOxA)) segments and photocleavable poly(2‐nitrobenzyl acrylate) (PNBA) segments is synthesized via combination of cationic ring‐opening polymerization (CROP) and reversible addition‐fragmentation chain transfer (RAFT) polymerization techniques. The P(OEtOxA)‐ran‐PNBA copolymer exhibits lower critical solution (LCST)‐type soluble‐to‐turbid phase transition in water with tunable cloud point (Tcp) with respect to chain length of P(OEtOxA) segment present. The photocleavage of PNBA segments by UV irradiation transforms amphiphilic P(OEtOxA)‐ran‐PNBA to fully hydrophilic P(OEtOxA)‐ran‐poly(acrylic acid) resulting in the appreciable increase of Tcp of copolymer in aqueous solution. Owing to the amphiphilic nature, the P(OEtOxA)‐ran‐PNBA copolymer molecules self‐assemble into well‐dispersed spherical micelles in water. There is a disruption of the copolymer micelles with UV light irradiation as well as shrinkage of micellar size with increasing temperature above the LCST of copolymer in solution. Finally, the encapsulation of hydrophobic guest molecule (nile red) into P(OEtOxA)‐ran‐PNBA copolymer micelles and thermo‐ and photo‐triggered release of nile red are demonstrated. © 2017 Wiley Periodicals, Inc. A novel POSS‐containing methacrylate monomer (HEMAPOSS) was fabricated by extending the side chain between polyhedral oligomeric silsesquioxane (POSS) unit and methacrylate group, which can efficiently decrease the steric hindrance in free‐radical polymerization of POSS‐methacrylate monomer. POSS‐containing homopolymers (PHEMAPOSS) with a higher degree of polymerization (DP) can be prepared using HEMAPOSS monomer via reversible addition–fragmentation chain transfer (RAFT) polymerization. PHEMAPOSS was further used as the macro‐RAFT agent to construct a series of amphiphilic POSS‐containing poly(N, N‐dimethylaminoethyl methacrylate) diblock copolymers, PHEMAPOSS‐b‐PDMAEMA. PHEMAPOSS‐b‐PDMAEMA block copolymers can self‐assemble into a plethora of morphologies ranging from irregular assembled aggregates to core‐shell spheres and further from complex spheres (pearl‐necklace‐liked structure) to large compound vesicles. The thermo‐ and pH‐responsive behaviors of the micelles were also investigated by dynamic laser scattering, UV spectroscopy, SEM, and TEM. Water‐soluble crosslinked hollow nanoparticles were prepared using pH‐responsive anionic polymer micelles as templates. The template micelles were formed from pH‐responsive diblock copolymers (PAMPS‐PAaH) composed of the poly(sodium 2‐(acrylamido)‐2‐methylpropanesulfonate) and poly(6‐(acrylamido)hexanoic acid) blocks in an aqueous acidic solution. The PAMPS and PAaH blocks form a hydrophilic anionic shell and hydrophobic core of the core‐shell polymer micelle, respectively. A cationic diblock copolymer (PEG‐P(APTAC/CEA)) with the poly(ethylene glycol) block and random copolymer block composed of poly((3‐acrylamidopropyl)trimethylammonium chloride) containing a small amount of the 2‐(cinnamoyl)ethylacrylate photo‐crosslinkable unit can be adsorbed to the anionic shell of the template micelle due to electrostatic interaction, which form a core‐shell‐corona three‐layered micelle. The shell of the core‐shell‐corona micelle is formed from a polyion complex with anionic PAMPS and cationic P(APTAC/CEA) chains. The P(APTAC/CEA) chains in the shell of the core‐shell‐corona micelle can be photo‐crosslinked with UV irradiation. The template micelle can be dissociated using NaOH, because the PAaH blocks are ionized. Furthermore, electrostatic interactions between PAMPS and PAPTAC in the shell are screened by adding excess NaCl in water. The template micelles can be completely removed by dialysis against water containing NaOH and NaCl to prepare the crosslinked hollow nanoparticles. Bioinspired core‐bound polymeric micelles, based on hydrogen bonding and photo‐crosslinking, of thymine have been prepared from poly(vinylbenzylthymine)‐b‐poly(vinylbenzyltriethylammonium chloride). The amphiphilic block copolymer was synthesized by 2,2‐tetramethylpiperidin‐1‐oxyl‐mediated living radical polymerization in water/ethylene glycol solution. Micelle characterization and critical micelle concentration measurements demonstrated that the hydrogen bonding of the attached thymine units stabilizes the micelles. Further, core‐crosslinked polymeric micelles were formed by ultraviolet (UV) radiation showing that the stability of the micelle could be controlled by the UV crosslinking of the attached thymines. © 2011 Wiley Periodicals, Inc. Multistimuli‐responsive precise morphological control over self‐assembled polymers is of great importance for applications in nanoscience as drug delivery system. A novel pH, photoresponsive, and cyclodextrin‐responsive block copolymer were developed to investigate the reversible morphological transition from micelles to vesicles. The azobenzene‐containing block copolymer poly(ethylene oxide)‐b‐poly(2‐(diethylamino)ethyl methacrylate‐co‐6‐(4‐phenylazo phenoxy)hexyl methacrylate) [PEO‐b‐P(DEAEMA‐co‐PPHMA)] was synthesized by atom transfer radical polymerization. This system can self‐assemble into vesicles in aqueous solution at pH 8. On adjusting the solution pH to 3, there was a transition from vesicles to micelles. The same behavior, that is, transition from vesicles to micelles was also realizable on addition of β‐cyclodextrin (β‐CD) to the PEO‐b‐P(DEAEMA‐co‐PPHMA) solution at pH 8. Furthermore, after β‐CD was added, alternating irradiation of the solution with UV and visible light can also induce the reversible micelle‐to‐vesicle transition because of the photoinduced trans‐to‐cis isomerization of azobenzene units. Thermo‐responsive block‐graft fluoropolymer is synthesized and investigated the self‐assembly morphology and the tunable wettability surface on cotton fabric by dip‐coating into the micelles with different temperatures. Well‐defined block‐graft copolymer is prepared by click chemistry with poly(hexafluorobutyl methacrylate)‐block‐poly(glycidyl methacrylate) (PHFBMA‐b‐PGMA) and homopolymer poly(N‐isopropylacrylate) with alkyne on main chain (Alkynyl‐PNIPAM) to synthesize final block‐graft polymer PHFBMA‐b‐(PGMA‐g‐PNIPAM). The thermo‐responsive behaviors of block‐graft polymer prove that the diameter for fluoropolymer micelle is about 50–70 nm with uniform sphere shape at room temperature and bigger and broader at 40 °C. The surface of cotton fabric processed in micelle solution at room temperature is smooth and has good hydrophobic property, while it has the hydrophilic property dipped in high temperature micelle solution. This work may give valuable guidance for fabricating a facile strategy to establish controllable wettability surfaces on different substrates, which is a promising candidate for the coating materials and industrial fields. © 2015 Wiley Periodicals, Inc. Self‐assembly of amphiphilic ABA random triblock copolymers in water serves as a novel approach to create unique structure micelles connected with flexible linkages. The ABA triblock copolymers consist of amphiphilic random copolymers bearing hydrophilic poly(ethylene glycol) and hydrophobic dodecyl pendants as the A segments and a hydrophilic poly(ethylene oxide) (PEO) as the middle B segment. The A block is varied in dodecyl methacrylate content of 20%–50% and degree of polymerization (DP) of 100‐200. By controlling the composition and DP of the A block, various architectures can be tailor‐made as micelles in water: PEO‐linked double core unimer micelles, PEO‐looped unimer or dimer micelles, and multichain micelles. Amphiphilic block copolymers composed of a hydrophilic poly(ethylene glycol) (PEG) block and a hydrophobic poly(glycidyl methacrylate) (PGMA) block were synthesized through cationic ring‐opening polymerization with PEG as the precursor. The model reactions indicated that the reactivity of the epoxy groups was higher than that of the double bonds in the bifunctional monomer glycidyl methacrylate (GMA) under the cationic polymerization conditions. Through the control of the reaction time in the synthesis of block copolymer PEG‐b‐PGMA, a linear GMA block was obtained through the ring‐opening polymerization of epoxy groups, whereas the double bond in GMA remained unreacted. The results showed that the molecular weight of the PEG precursor had little influence on the grafting of GMA, and the PGMA blocks almost kept the same length, despite the difference of the PEG blocks. In addition, the PGMA blocks only consisted of several GMA units. The obtained amphiphilic PEG‐b‐PGMA block copolymers could form polymeric core–shell micelles by direct molecular self‐assembly in water. The crosslinking of the PGMA core of the PEG‐b‐PGMA micelles, induced by ultraviolet radiation and heat instead of crosslinking agents, greatly increased the stability of the micelles. © 2005 Wiley Periodicals, Inc. In this article, the synthesis and self‐assembly of a novel well‐defined biocompatible amphiphilic POEGMA‐PDMS‐POEGMA triblock copolymer were studied. The copolymer was synthesized by atom transfer radical polymerization of oligo(ethylene glycol) methyl ether methacrylate (OEGMA) using α,ω‐dibromo polydimethylsiloxane macroinitiator (Br‐PDMS‐Br). Br‐PDMS‐Br was synthesized through the esterification of α,ω‐hydroxypropyl polydimethylsiloxane and 2‐bromoisobutyryl bromide. The structures of the copolymers were confirmed by proton nuclear magnetic resonance spectroscopy, and gel permeation chromatography. The copolymers showed reversible aggregation in response to temperature cycles with a lower critical solution temperature (LCST) between 61 and 66 °C, as determined by ultraviolet‐visible spectrophotometry and dynamic light scattering. The LCST values increased in proportion to the length of the hydrophilic block and were lower than that of the POEGMA homopolymer. The self‐assembly behavior of the copolymers in aqueous solution was investigated by fluorescence spectroscopy and transmission electron microscopy. The critical micelle concentration value (1.08–0.26 10?6 mol L?1) decreased as the length of the POEGMA chain increased. The POEGMA‐PDMS‐POEGMA copolymers can easily self‐assemble into spherical micelles in aqueous solution. A double hydrophilic block copolymer composed of poly(acrylic acid) (PAA) and poly(4‐vinyl pyridine) (P4VP) was obtained through hydrolysis of diblock copolymer of poly(tert‐butyl acrylate) (PtBA) and P4VP synthesized using atom transfer radical polymerization. Water‐soluble micelles with PAA core and P4VP corona were observed at low (acidic) pH, while micelles with P4VP core and PAA corona were formed at high (basic) pH. Two metalloporphyrins, zinc tetraphenylporphyrin (ZnTPP) and cobalt tetraphenylporphyrin (CoTPP), were used as model compounds to investigate the encapsulation of hydrophobic molecules by both types of micelles. UV–vis spectroscopic measurements indicate that micelles with P4VP core are able to entrap more ZnTPP and CoTPP as a result of the axial coordination between the transition metals and the pyridine groups. The study found that metalloporphyrins encapsulated by the micelles with PAA core could be released on pH increase, while those entrapped by the micelles with P4VP core could be released on pH decrease. We present here the synthesis of two kinds of amphiphilic block copolymers, a diblock copolymer MPEG‐b‐PTAm and a triblock copolymer MPEG‐b‐PLA‐b‐PTAm, which can self‐assemble into micelles with nitroxyl radicals‐containing PTAm segment in the core. The structure of the block copolymers was characterized by 1H NMR and GPC. Dynamic laser light scattering and transmission electron microscopy were used to study the micellar behavior of the two block copolymers in aqueous solution. The micelles carrying nitroxyl radicals in the core can generate electron paramagnetic resonance, which is stable for a period of time up to 8 min even in the presence of reducing reagent such as ascorbic acid. The enhanced stability against the reducing agent was ascribed to the inaccessibility of the nitroxyl radical core placed in the interior of the micelles. Glucose responsive block copolymer featuring boronic acid as a glucose responsive moiety and glycine are reported. The first block is polymerized through reversible addition–fragmentation chain transfer (RAFT) polymerization and the resulting poly(N‐acryloylmorpholine)113 (PAcM) is employed as a macro‐chain transfer agent for chain extension with pentafluorophenyl acrylate (PFPA) yielding a well‐defined PAcM113block‐poly(pentafluorophenyl acrylate)84 (PPFPA). The PPFPA block is then reacted with functional (3‐aminomethyl) phenyl boronic acid and glycine via post‐polymerization modification and the structure of the block copolymer is confirmed by proton nuclear magnetic resonance (NMR), 19F NMR, Fourier transform infrared, and gel permeation chromatography. By copolymerizing glycine into the polymer backbone, the relative pKa of the block copolymer is significantly lowered. The block copolymer can self‐assemble into core–shell micelles in aqueous solution and disassemble in response to glucose at the physiological pH. Furthermore, the encapsulation and release of Nile red (NR) as a hydrophobic model drug is studied under the physiological pH. The influence of the glucose concentration on the NR release from the polymeric micelles is demonstrated. These results suggested that the glucose‐responsive poly[(AcM)113b‐(3‐(aminomethyl)phenylboronic acid hydrochloride(‐co‐Gly)84] block copolymer has potential applications as a glucose‐responsive polymer for insulin delivery. © 2018 Wiley Periodicals, Inc. Triple stimuli (temperature/pH/photo)‐responsive amphiphilic glycopolymer, poly(2‐(dimethylamino)ethyl methacrylate‐co‐6‐O‐methacryloyl‐1,2,3,4‐di‐O‐isopropylidene‐D‐galactopyranose)‐b‐poly(4‐(4‐methoxyphenylazo)phenoxy methacrylate) [P(DMAEMA‐co‐MAIpGP)‐b‐PMAZO] was synthesized by atom transfer radical polymerization, followed by the hydrolysis of MAIpGP groups, resulting in the target product poly(2‐(dimethylamino)ethyl methacrylate‐co‐6‐O‐methacryloyl‐D‐galactopyranose)‐b‐poly(4‐(4‐methoxyphenylazo)phenoxy methacrylate) [P(DMAEMA‐co‐MAGP)‐b‐PMAZO]. The composition, moleculer weight, and moleculer weight distribution of the resultant polymers were characterized by 1H NMR and gel permeation chromatography. The micelles formed in aqueous solutions were simulated by various chemical and physical stimuli and characterized by dynamic light scattering, transmission electron microscopy, and UV‐vis spectroscopy. It was found that the glycopolymer is responsive to three different types of stimulus (light, temperature, and pH). The poly(2‐(dimethylamino) ethyl methacrylate) segments give thermo‐ and pH‐responsiveness. The presence of the azobenzene moiety endows the block copolymer to exhibit light‐responsiveness due to its reversible trans‐cis isomerization conversion. The triple stimuli‐responsive glycopolymer micelles can simulate biomacromolecues in vivo/in vitro environment and can be expected to open up new applications in various fields. © 2014 Wiley Periodicals, Inc. The pH‐sensitive tertiary amino groups were introduced to synthesize temperature and pH dual‐sensitive degradable polyaspartamide derivatives (phe/DEAE‐g‐PHPA) containing pendant aromatic structures and ionizable tertiary amino groups. The thermo/pH‐responsive behavior of phe/DEAE‐g‐PHPA polymer can be tuned by adjusting the graft copolymer composition. Due to the pH sensitivity of the phe/DEAE‐g‐PHPA‐g‐mPEG polymer with hydrophilic long PEG chain, the micelles and the anticancer drug‐loaded micelles were prepared by a quick pH‐changing method without using toxic organic solvent. The obtained polymeric micelles, paclitaxel‐loaded micelles and doxorubicin‐loaded micelles were stable under physiological conditions. Both the drug‐loaded micelles showed much faster release at pH 5 than at pH 7.4. The doxorubicin‐loaded micelles showed obvious and better anticancer activity against both HepG2 and HeLa cells than free doxorubicin. A hetero‐arm star polymer, polystyrene‐poly(N‐isopropylacrylamide)‐ poly(2‐(dimethylamino)ethylmethacrylate) (PSt‐PNIPAM‐PDMAEMA), was synthesized by “clicking” the alkyne group at the junction of PSt‐b‐PNIPAM diblock copolymer onto the azide end‐group of PDMAEMA homopolymer via 1,3‐dipolar cycloaddition. The resultant polymer was characterized by gel permeation chromatography, proton nuclear magnetic resonance spectroscopy and Fourier transform infrared spectroscopy. PSt‐PNIPAM‐PDMAEMA micelles with PSt block as core and PNIPAM and PDMAEMA blocks as shell were formed when adding the copolymer solution in THF into 10 folds of water. Lower critical solution temperature (LCST) of PNIPAM and PDMAEMA homopolymer is 32 °C for PNIPAM and 40 to 50 °C for PDMAEMA, respectively. Upon continuous heating through their LCSTs, PSt‐PNIPAM‐PDMAEMA core‐shell micelles exhibited two‐stage thermally induced collapse. The first‐stage collapse, from 20 to 34 °C, is ascribed to the shrinkage of PNIPAM chains; and the second‐stage collapse, from 38 to 50 °C, is due to the shrinkage of PDMAEMA chains. A new class of temperature and pH dual‐responsive and injectable supramolecular hydrogel was developed, which was formed from block copolymer poly(ethylene glycol)‐block‐poly[(2‐dimethylamino)ethyl methacrylate] (PEG‐b‐PDMAEMA) and α‐cyclodextrin (α‐CD) inclusion complexes (ICs). The PEG‐b‐PDMAEMA diblock copolymers with different ratio of ethylene glycol (EG) to (2‐dimethylamino)ethyl methacrylate (DMAEMA) (102:46 and 102:96, respectively) were prepared by atom transfer radical polymerization (ATRP). 1H NMR measurement indicated that the ratio of EG unit to α‐CD in the resulted ICs was higher than 2:1. Thermal analysis showed that thermal stability of ICs was improved. The rheology studies showed that the hydrogels were temperature and pH sensitive. Moreover, the hydrogels were thixotropic and reversible. The self‐assembly morphologies of the ICs in different pH and ionic strength environment were studied by transmission electron microscopy. The formed biocompatible micelles have potential applications as biomedical and stimulus‐responsive material. © 2010 Wiley Periodicals, Inc.
