Matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) coupled with CID (collision‐induced dissociation) has been used for the detailed characterization of two poly(2‐ethyl‐2‐oxazoline)s as part of a continuing study of synthetic polymers by MALDI‐TOF MS/MS. These experiments provided information about the variety of fragmentation pathways for poly(oxazoline)s. It was possible to show that, in addition to the eliminations of small molecules, like ethene and hydrogen, the McLafferty rearrangement is also a possible fragmentation route. A library of fragmentation pathways for synthetic polymers was also constructed and such a library should enable the fast and automated data analysis of polymers in the future. The molar mass determination of block copolymers, in particular amphiphilic block copolymers, has been challenging with chromatographic techniques. Therefore, methoxy poly(ethylene glycol)‐b‐poly(styrene) (mPEG‐b‐PS) was synthesized by atom transfer radical polymerization (ATRP) and characterized in detail not only by conventional chromatographic techniques, such as size exclusion chromatography (SEC), but also by matrix‐assisted laser/desorption ionization tandem mass spectrometry (MALDI‐TOF MS/MS). As expected, different molar mass values were obtained in the SEC measurements depending on the calibration standards (either PEG or PS). In contrast, MALDI‐TOF MS/MS analysis allowed the molar mass determination of each block, by the scission of the weakest point between the PEG and PS block. Thus, fragments of the individual blocks could be obtained. The PEG block showed a depolymerization reaction, while for the PS block fragments were obtained in the monomeric, dimeric, and trimeric regions as a result of multiple chain scissions. The block length of PEG and PS could be calculated from the fragments recorded in the MALDI‐TOF MS/MS spectrum. Furthermore, the assignment of the substructures of the individual blocks acquired by MALDI‐TOF MS/MS was accomplished with the help of the fragments that were obtained from the corresponding homopolymers. © 2010 Wiley Periodicals, Inc. New amphiphilic triblock copoly(2‐oxazoline)s, containing hydrophobic domains with fluorine‐containing blocks, were synthesized. Using microwave radiation as heating source, triblock copolymers with narrow molar mass distributions were obtained by the sequential addition of 2‐ethyl‐2‐oxazoline, 2‐(1‐ethylheptyl)‐2‐oxazoline, and 2‐(2,6‐difluorophenyl)‐2‐oxazoline. The polymers obtained were characterized by size exclusion chromatography, 1H NMR spectroscopy and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). To investigate the incorporation of all three monomers into the triblock copolymers, a model polymer was prepared with shorter blocks exhibiting a suitable length to be measured in the reflector mode of a MALDI‐TOF MS. In addition, kinetic investigations on the homopolymerizations of all monomers were performed in nitromethane at 140 °C, yielding the polymerization rates under these conditions. DSC measurements of poly(2‐(1‐ethylheptyl)‐2‐oxazoline) and poly(2‐(2,6‐difluorophenyl)‐2‐oxazoline)) revealing glass transitions at about 33 and 120 °C, respectively. The thermal analysis of a blend of the two polymers showed two glass transitions revealing demixing, which could be an indicating for the immiscibility of the two components in the block copolymers. © 2010 Wiley Periodicals, Inc. In this contribution, linear poly(ethylene imine) (PEI) polymers, which are of importance in gene delivery, are investigated in detail by using electrospray ionization‐quadrupole‐time of flight (ESI‐Q‐TOF) and matrix‐assisted laser desorption/ionization‐time of flight (MALDI‐TOF) mass spectrometry (MS). The analyzed PEIs with different end groups were synthesized using the polymerization of substituted 2‐oxazoline via a living cationic ring‐opening polymerization (CROP) and a subsequent hydrolysis under acidic conditions. The main goal of this study was to identify linear PEI polymers in a detailed way to gain information about their fragmentation pathways. For this purpose, a detailed characterization of three different linear PEIs was performed by using ESI‐Q‐TOF and MALDI‐TOF MS in combination with collision‐induced dissociation (CID) experiments. In ESI‐MS as well as MALDI‐MS analysis, the obtained spectra of PEIs resulted in fitting mass distributions for the investigated PEIs. In the tandem MS analysis, a 1,2‐hydride shift with a charge‐remote rearrangement via a four‐membered cyclic transition state, as well as charge‐induced fragmentation reactions, was proposed as the main fragmentation mechanisms according to the obtained fragmentation products from the protonated parent peaks. In addition, heterolytic and homolytic cleavages were proposed as alternative fragmentation pathways. Molecular weights of seven poly(phenylene ethynylene)‐based water‐soluble conjugated polyelectrolytes (CPEs) obtained through Sonogashira coupling are determined by matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). A standard sample preparation protocol is developed to characterize the seven CPEs using 2,5‐dihydroxybenzoic acid as the matrix (M) and AgTFA as the cationization reagent (CR). High‐quality MALDI mass spectra are obtained at volume mixing ratios (CPE/M/CR) of 5/5/1 for anionic polymers (P1–P4) and 5/50/1 for cationic polymers (P5–P7). Molecular weight, molecular weight distribution, and end‐group information are analyzed. High‐resolution matrix‐assisted laser desorption/ionization (MALDI) time‐of‐flight mass spectrometry (TOF MS) was used for the analysis of the low‐molecular‐weight products from the photo‐oxidation of poly(3‐hexylthiophene) (P3HT) in solution and thin film. Eight new peak series were observed in the low‐mass range of the mass spectra of the products degraded in solution, and the formulas of the eight components were determined from the accurate mass. From SEC/MALDI‐TOF MS, two components were identified as the degraded products, and the other six components were derived from the fragmentation of the degraded products during the MALDI process. A mechanism for the formation of these components was proposed on the basis of the results of MALDI‐TOF MS. For the thin film degradation, a part of products in the solution degradation were observed, which supports that the oxidation of P3HT in solution and thin film proceeded in the same mechanism. This study shows that high‐resolution MALDI‐TOF MS is effective for the analysis of the low‐molecular‐weight products from P3HT photo‐oxidation and expected to be feasible for the degradation analyses of other polymers. Matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) was chosen for an in‐detail analysis of poly(methyl methacrylate) (PMMA) in order to determine the possible fragmentation mechanism with the help of collision‐induced dissociation (CID). All experiments were performed on a well‐defined PMMA standard and were optimized for sample preparation and measurement conditions of both MS and MS/MS. In order to investigate the fragmentation pathways, two parent peaks—both charged with sodium (m/z = 1 625.9 and 2 226.2 Da, respectively)—were selected, thus permitting the examination of possible cleavages, and reaction pathways. For both chosen peaks, the MALDI‐TOF MS/MS spectra revealed four fragmentation series that could be explained by single or multiple main chain scissions and secondary reactions of the PMMA side groups. According to the molar mass of the fragments, a loss or migration of the side group to the end of the free radical, followed by a β‐scission, was favored. These insights are the first steps toward the construction of a library with fragments and fragmentation pathways, complementary to proteomics libraries, in order to obtain fast and automated identification of substances.

Characterization of block size in poly(ethylene oxide)‐b‐poly(styrene) (PEO‐b‐PS) block copolymers could be achieved by matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF‐MS) after scission of the macromolecules into their constituent blocks. The performed hydrolytic cleavage was demonstrated to specifically occur on the targeted ester function in the junction group, yielding two homopolymers consisting of the constitutive initial blocks. This approach allows the use of well‐established MALDI protocols for a complete copolymer characterization while circumventing difficulties inherent to amphiphilic macromolecule ionization. Although the labile end‐group in PS homopolymer was modified by the MALDI process, PS block size could be determined from MS data since polymer chains were shown to remain intact during ionization. This methodology has been validated for a PEO‐b‐PS sample series, with two PEO of number average molecular weight (Mn) of 2000 and 5000 g mol?1 and Mn(PS) ranging from 4000 to 21,000 g mol?1. Weight average molecular weight (Mw), and thus polydispersity index, could also be reached for each segment and were consistent with values obtained by size exclusion chromatography. This approach is particularly valuable in the case of amphiphilic copolymers for which Mn values as determined by liquid state nuclear magnetic resonance might be affected by micelle formation. © 2009 Wiley Periodicals, Inc. On‐line ultra‐performance liquid chromatography (UPLC) coupled with diode‐array detection (UPLC/DAD) and electrospray ionization quadrupole time‐of‐flight mass spectrometry (ESI‐Q‐TOF‐MS) were used for separation, identification and structural analyses of saponins in Rhizoma Paridis saponins (RPS) and rat plasma after oral administration of RPS. Thirty steroidal saponins in RPS were identified by comparing their retention time, accurate mass measurement and positive and negative mass spectrometry data with that of reference compounds. The UPLC/Q‐TOF‐MS method was proved to be rapid and efficient in that 30 steroidal saponins, including three kinds of saponins (prototype, pennogenyl and diosgenyl saponins) were tentatively characterized within 6 min. After oral administration of RPS, 21 original saponins were absorbed in RPS‐treated rat plasma. Our results indicated that UPLC/Q‐TOF‐MS is a rapid and effective tool for identification of a series of saponins at trace level. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Linear–hyperbranched hybrid poly(butylene adipate) (HPBA) copolymers were synthesized through a branching reaction between the linear tailored prepolymer terminated with methyl ester groups and different mol percents of the 1,1,1‐tris(hydroxymethyl) propane (TMP) as branching agent, using the titanium(IV) isopropoxide as catalyst, at 180 °C under vacuum for different times. All samples were characterized by NMR and matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI‐TOF MS). In particular, MALDI‐TOF mass spectra of the unfractionated and size exclusion chromatography (SEC)‐fractionated hyperbranched (HB) samples gave information on their composition, on the end groups as well as on the TMP units present in each family of HB macromolecules. HB chains containing cyclic branches and ether bonds formed by intermolecular transesterification and intramolecular and intermolecular transetherification side reactions, respectively, were also revealed by MALDI‐TOF MS analysis. All samples were also investigated by SEC. The average molar masses (MMs) evaluated by SEC calibrated with the polystyrene (PS) narrow standards were overestimated with respect to those calculated by the SEC/MALDI‐TOF MS self‐ calibration method, which gave reliable values. Moreover, it also showed that the hydrodynamic volume of the HPBA polymers was higher than that of the linear PSs with similar MMs. © 2011 Wiley Periodicals, Inc. Sequences and end groups of complex copolyesters were determined by fragmentation analysis by means of matrix‐assisted laser desorption/ionization collision‐induced dissociation tandem mass spectrometry (MALDI CID MS/MS). The complexity of the crude copolyester mixture was reduced by a chromatographic separation followed by a MALDI time‐of‐flight (TOF) investigation of fractions. Due to overlapping compositional and end‐group information a clear assignment of end groups was very difficult. However, the fragmentation of suitable precursor ions resulted in typical fragment ion patterns and, therefore, enabled a fast and unambiguous determination of the end groups and composition of this important class of polymers. A cyclic initiator for the nitroxide‐mediated controlled radical polymerization (NMP) is a powerful tool for the preparation of macrocyclic polymers via a ring‐expansion vinyl polymerization mechanism. For this purpose, we prepared a Hawker‐type NMP‐initiator that includes an azide and a terminal alkyne as an acyclic precursor, which is subsequently tethered via an intramolecular azide/alkyne‐“click”‐reaction, producing the final cyclic NMP‐initiator. The polymerization reactions of styrene with cyclic initiator were demonstrated and the resultant polymers were characterized by the gel permeation chromatography (GPC) and the matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS). These results prove that the ring‐expansion polymerization of styrene occurred together with the radical ring‐crossover reactions originating from the exchange of the inherent nitroxides generating macrocyclic polystyrenes with higher expanded rings. © 2010 Wiley Periodicals, Inc. Poly(2‐oxazoline) (POx) based materials have experienced renewed interest, due to their biocompatibility and broad functional diversity. Although research pertaining to cationic ring opening polymerization (CROP) of 2‐oxazoline derivatives spans upwards of 5 decades, modern characterization techniques, specifically Matrix Assisted Laser‐Desorption Ionization—Time of Flight Mass Spectrometry (MALDI‐TOF MS), were not available during early studies to aid in identifying polymer end‐groups. Through careful synthesis and analytical characterization, evidence supports an alternative mode of nucleophilic attack during ammonia termination, with nucleophilic attack occurring at a different site on the terminal oxazolinium than where polymer propagation occurs. Furthermore, investigations employing targeted end‐group modification, in conjunction with MALDI and NMR analysis, determined the structure of the resultant terminal group was a hydroxyethylamino end group. We report on the micellization behavior of tri‐ and tetrablock copoly(2‐oxazoline)s in water–ethanol mixtures. The copolymers are based on different combinations of 2‐methyl‐, 2‐ethyl‐, 2‐phenyl‐, and 2‐nonyl‐2‐oxazoline. The solvophilic/solvophobic balance of these copolymers can be tuned thanks to the solubility dependence of the poly(2‐phenyl‐2‐oxazoline) block on the solvent composition. Characterization of the obtained micelles by dynamic light scattering and transmission electron microscopy revealed that their size and morphology depend on the solvophobic content of the copolymers and on the block order. Spherical micelles are always obtained when poly(2‐nonyl‐2‐oxazoline) is the only solvophobic block. When the solvophobic fraction consists of both the poly(2‐phenyl‐2‐oxazoline) and poly(2‐nonyl‐2‐oxazoline) blocks, spherical and cylindrical micelles as well as vesicles have been observed. © 2010 Wiley Periodicals, Inc. Recent advances in the resolving power of matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) enable the detailed characterization of linear homopolymers, and in particular provide invaluable data for the determination of their end‐group functionalities. With the growing importance of macromolecular coupling reactions in building complex polymer architectures, the ability to accurately monitor end‐group transformations is becoming increasingly important for synthetic polymer chemists. This tutorial demonstrates the application of MALDI‐TOF MS in determining both end‐group functionalities and their transformations for linear homopolymers. The principle relating to the selection of a proper matrix, cationization reagent, and solvent for matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry (MALDI‐TOF MS) of synthetic polymers is still a topic of research. In this work we focused on the selection of a suitable MALDI solvent. Polystyrene PS7600 and poly(ethylene glycol) PEG4820 were analyzed by MALDI‐TOF MS using various solvents which were selected based on the Hansen solubility parameter system. For polystyrene (PS), dithranol was used as the matrix and silver trifluoroacetate as the cationization reagent whereas, for poly(ethylene glycol) (PEG), the combination of 2,5‐dihydroxybenzoic acid and sodium trifluoroacetate was used for all experiments. When employing solvents which dissolve PS and PEG, reliable MALDI mass spectra were obtained while samples in non‐solvents (solvents which are not able to dissolve the polymer) failed to provide spectra. Iridoid glycosides (IGs), the major constituents in Fructus Gardeniae, have demonstrated various pharmacological activities, but there is no systematic chemical profile of IGs in Fructus Gardeniae in the published literature until now. Therefore, it is imperative that a rapid and sensitive high‐performance liquid chromatography coupled with quadrupole time‐of‐flight tandem mass spectrometry (HPLC‐Q/TOF‐MS/MS) method is established for comprehensive characterization of IGs in Fructus Gardeniae. Firstly, the fragmentation patterns of six known IGs were investigated and proposed and further concluded the diagnostic fragment ions and characteristic fragmentation pathways. Then, based on the summarized fragmentation patterns and the known compounds in the literatures, the other IGs in Fructus Gardeniae were identified successively. As a result, a total of 20 IGs were identified, of which three pairs of epimers were structurally characterized and differentiated. More importantly, one compound, the isoshanzhiside methyl ester, was tentatively identified as a new compound. The results of this study demonstrate the superiority of HPLC‐MS with a high‐resolution mass spectrometer for the rapid and sensitive structural elucidation of the multiple groups of constituents in Fructus Gardeniae. Accurate and rapid determination of trypanosomatids is essential in epidemiological surveillance and therapeutic studies. Matrix‐assisted laser desorption ionization/time of flight mass spectrometry (MALDI‐TOF MS) has been shown to be a useful and powerful technique to identify bacteria, fungi, metazoa and human intact cells with applications in clinical settings. Here, we developed and optimized a MALDI‐TOF MS method to profile trypanosomatids. trypanosomatid cells were deposited on a MALDI target plate followed by addition of matrix solution. The plate was then subjected to MALDI‐TOF MS measurement to create reference mass spectra library and unknown samples were identified by pattern matching using the BioTyper software tool. Several m/z peaks reproducibly and uniquely identified trypanosomatids species showing the potentials of direct identification of trypanosomatids by MALDI‐TOF MS. Moreover, this method discriminated different life stages of Trypanosoma cruzi, epimastigote and bloodstream trypomastigote and Trypanosoma brucei, procyclic and bloodstream. T. cruzi Discrete Typing Units (DTUs) were also discriminated in three clades. However, it was not possible to achieve enough resolution and software‐assisted identification at the strain level. Overall, this study shows the importance of MALDI‐TOF MS for the direct identification of trypanosomatids and opens new avenues for mass spectrometry‐based detection of parasites in biofluids. The microalga Haematococcus pluvialis produces the pigment astaxanthin mainly in esterified form with a multitude of fatty acids, which results in a complex mixture of carotenol mono‐ and diesters. For rapid fingerprinting of these esters, matrix‐assisted laser desorption ionization time of flight mass spectrometry (MALDI‐TOF/TOF‐MS) might be an alternative to traditional chromatographic separation combined with MS. Investigation of ionization and fragmentation of astaxanthin mono‐ and diester palmitate standards in MALDI‐TOF/TOF‐MS showed that sodium adduct parent masses [M + Na]+ gave much simpler MS2 spectra than radical / protonated [M]+● / [M + H]+ parents. [M + Na]+ fragments yielded diagnostic polyene‐specific eliminations and fatty acid neutral losses, whereas [M]+● / [M + H]+ fragmentation resulted in a multitude of non‐diagnostic daughters. For diesters, a benzonium fragment, formed by polyene elimination, was required for identification of the second fatty acid attached to the astaxanthin backbone. Parents were forced into [M + Na]+ ionization by addition of sodium acetate, and best signal‐to‐noise ratios were obtained in the 0.1 to 1.0 mM range. This method was applied to fingerprinting astaxanthin esters in a crude H. pluvialis extract. Prior to MALDI‐TOF/TOF‐MS, the extract was fractionated by normal phase Flash chromatography to obtain fractions enriched in mono‐ and diesters and to remove pheophytin a, which compromised monoester signals. All 12 types of all‐trans esterified esters found in LC were identified with MALDI‐TOF/TOF‐MS, with the exception of two minor monoesters. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

Summary: The feasibility of automated, high‐throughput contact‐angle measurements is demonstrated using a modified commercial contact‐angle measuring apparatus. Droplets are automatically dispensed, analyzed and aspirated. The system automatically measures more than 100 contact angles per hour using two test liquids, and calculates surface energies using the equation of state method. A library of poly(2‐oxazoline) homo‐ and block copolymers was investigated. Polymers with a 2‐nonyl‐2‐oxazoline block were found to have considerably lower surface energies.
