The spermatozoa of Chrysomya megacephala are similar to those described for other Brachycera. In this species, the spermatozoa are long and thin, measuring about 590 μm in length, of which the head region measures approximately 60 μm. The head includes a monolayered acrosome with electron-lucid material, and the shape of the nucleus, in cross-sections, varies from circular to oval with completely condensed chromatin. The centriole was observed in the zone of flagellar implantation, below the “peg” region. In the region of overlap, the followings structures are observed: nucleus, centriolar adjunct, mitochondrial derivatives and axoneme. The two mitochondrial derivatives are of different lengths but similar diameter. The axoneme is of a conventional insectan type with a 9 + 9 + 2 microtubular arrangement, with accessory tubules flanked by the electron-dense intertubular material. The male internal reproductive tract consists of testis, vas deferens, seminal vesicle, accessory glands and ejaculatory duct.  相似文献

A penetrating view of the three-dimensional nanostructure of female spermatheca and male flagellum in the species Aleochara verna is obtained with 100-nm resolution using a hard X-ray microscope, which provides a fast noninvasive imaging technology for insect morphology. Through introducing Zernike phase contrast and heavy metal staining, images taken at 8 keV displayed sufficient contrast for observing nanoscale fine structures, such as the spermatheca cochleate duct and the subapex of the flagellum, which have some implications for the study of the sperm transfer process and genital evolution in insects. This work shows that both the spatial resolution and the contrast characteristic of hard X-ray microscopy are quite promising for insect morphology studies and, particularly, provide an attractive alternative to the destructive techniques used for investigating internal soft tissues.  相似文献

The three Pegoscapus species present the same internal reproductive tract features comprising testes with a single testicular tubule, seminal vesicles, vasa deferentia, accessory glands and an ejaculatory duct. The seminal vesicle shows two morphologically distinct portions although they do not resemble the separate chambers found in other Chalcidoidea. The anterior portion of the seminal vesicle shows a prominent epithelium and stores the mature spermatozoa, while the posterior region is formed by a thicker muscular sheath that participates on ejaculation. The sexual maturation in Pegoscapus is achieved at emergence, when the testicular degeneration occurs. The spermatozoa of Pegoscapus reveal a basic structure similar to that of other Chalcidoidea. In Pegoscapus sp1. and Pegoscapus sp2. they present the same features, whereas Pegoscapus tonduzi comprises some different characteristics. It measures approximately 160 μm in Pegoscapus sp1. and Pegoscapus sp2., while in P. tonduzi the spermatozoa measure about 360 μm. The extracellular sheath thickness is another difference among the species. While Pegoscapus sp1. and Pegoscapus sp2. show a thick extracellular sheath, in P. tonduzi this sheath is very thin resulting in a large space intervening between the extracellular sheath and the nucleus. Despite these differences, the three species analyzed share some characteristics that allow the establishment of an identity to the spermatozoon of the genus Pegoscapus: the seminal vesicle not divided in chambers; the absence of acrosomal structures in the spermatozoa; the length of the extracellular sheath; the central microtubules being the firsts to terminate in the sequence of microtubular cutoff at the final axonemal portion.  相似文献

The histology and ultrastructure of the seminal vesicle, male accessory gland and the epididymis in Panorpa and Sinopanorpa were observed using light microscopy and scanning and transmission electron microscopy. The seminal vesicle consists of a mono-layered elongated columnar epithelium, which contains abundant electron-dense granules and secretory vesicles, and a small central lumen. In the apical region of the epithelium of the seminal vesicle, the intense secretory activity seems to be effected by means of merocrine mechanisms. The epithelium of the accessory gland is rich in rough endoplasmic reticulum and Golgi complex, and secretes seminal fluid into the lumen via both apocrine and merocrine mechanisms. The seminal vesicle is similar to the accessory gland in the epithelium structure and their secretory activity, mainly serving a secretory function rather than storing sperms. Instead, the sperms are stored in the epididymis, whose epithelium secretes nutrients into the large lumen by merocrine mechanisms. The secretory activity and function of the seminal vesicle are briefly discussed.  相似文献

The genera Centris and Epicharis constitute the Centridini and are widely distributed in the Neotropical region. Centris is also found in the southern portion of the Neartic region, although both genera are more abundant in the humid tropical regions. To describe the structure of the male reproductive system and spermatozoa, light and transmission electron microscopy were used. The male reproductive system of Centris sp. is formed by a pair of testes, a pair of deferent ducts, a pair of seminal vesicles, a pair of accessory glands and an ejaculatory duct connected to the external genitalia, the aedeagus. In this species, testes and the pre-vesicular deferens ducts as well as the seminal vesicles are encapsulated in a single conjunctive capsule, the scrotal membrane. Each testis consists of four testicular follicles, made up of cysts with up to 64 germinative cells. Histologically, the seminal vesicles are formed by a simple cylindrical epithelium, basal membrane and muscular tunic. The spermatozoa of Centris analis, C. fuscata, C. tarsata and Centris sp. are morphologically similar. They have two easily distinguishable regions: the head and flagellum. The head is formed by the two-layer acrosome, the linear nucleus and the flagellum, the centriole adjunct, the axoneme of pattern 9+9+2 microtubules, two asymmetric mitochondrial derivatives and two accessory bodies. These Centris species share various morphological characteristics of the male reproductive system and spermatozoa with the other bees previously described, indicating that several characteristics are synapomorphic for the family Apidae. Studies on the morphology of the male reproductive system and spermatozoa in Hymenoptera have demonstrated the diversity of the information provided by these reproductive structures, which can be used in taxonomy studies and the phylogeny of this important group of insects.  相似文献

The male reproductive system of Mischocyttarus cassununga consists of two testes, each one with three follicles, as occurs in most Vespidae. The seminal vesicle is divided in two chambers, separated by a constriction, so that the anterior locus is a little larger. In the testicular follicles, the spermatozoa are organized in cysts, with approximately 128 per cyst, where the nuclei are oriented toward the follicle center. The spermatozoa of M. cassununga is about 97 μm in length, which makes them the shortest sperm described for Vespidae. Sperm ultrastructure of M. cassununga is very similar of the others Vespidae. But, despite these similarities, the bilobated mitochondrial derivative represents an autapomorphy for M. cassununga. The subdivision of the seminal vesicle has never been observed in any other Vespidae. Thus, this study supports the validity of insect sperm morphology as a tool for phylogenetic analysis within Hymenoptera.  相似文献

Experimental results on mean inclination angle and its fluctuation due to thermal noise in tank-treading motion of a vesicle in shear flow as a function of vesicle excess area, normalized shear rate, viscosity, and viscosity contrast between inner and outer fluids, , are presented. Good quantitative agreement with theory made for was found. At the dependence is altered significantly. Dependence of the vesicle shape on shear rate is consistent with theory. A tank-treading velocity of the vesicle membrane is found to be a periodic function close to that predicted by theory.  相似文献

The RARE technique is an extremely useful tool for clinical diagnosis, since it delivers images comparable to those produced by X-ray myelography and X-ray urography. Contrary to these methods, RARE does not use the application of contrast agents. A low-flip-angle variant called FLARE (fast low angle refocused echo imaging) makes this technique accessible for high-field systems.  相似文献

Photodynamic therapy (PDT) is a new treatment for solid tumors utilizing the combined action of light and a photosensitizing drug. Laser-fiber optic delivery systems make it practical to treat superficial and interstitial cancers, including malignancies of the skin, head and neck, esophagus, endobronchial tract, stomach, urinary bladder, female genital tract, and other sites. The putative action mechanism in PDT involves photochemical destruction of tumor tissue membranes mediated by singlet molecular oxygen. Light dosimetry modeling based on tissue optics is applicable for treatment planning. Related research areas include non-invasive optical diagnosis and imaging, development of new PDT drugs and light sources, and the extension of PDT methods to treatment of atherosclerotic vascular disease and blood detoxification.  相似文献

The purpose of this study was to evaluate the alterations of diluted molarity of contrast media to emit the maximum signal intensity by changing the parameters of pulse sequences. The phantom was developed by diluting the magnetic resonance imaging (MRI) T1 contrast medium. The phantom images were obtained by 1.5 and 3.0 T MRI systems. We conducted Pearson’s analysis to reveal the correlation of the signal-to-noise ratio (SNR)90%, the change of the concentration range of the contrast media which shows over 90% SNR, with changing the parameters of T1 effect pulse sequences in both 1.5 and 3.0 T imaging. As the flip angle increased, the SNR increased for all contrast media in magnetization-prepared rapid gradient echo and two-dimensional fast low angle shot pulse sequences at 1.5 and 3.0 T. Although the SNR increased until 30°, the SNR was almost the same over 30° in volumetric interpolated breath-hold examination at 1.5 and 3.0 T. The minimum contrast molarity of the representing SNR90% was decreased according to the increasing time to repeat in spin echo. The present study revealed that the high concentration technique of contrast media on three pulse sequences (VIBE, MPRAGE, and 2D FLASH) could be useful to obtain images with better SNR.  相似文献

A three-dimensional examination of blood vessels is provided using MR data from seven cases. The vascular surfaces are constructed with an algorithm that automatically follows the selected artery or vein and generates a projected three-dimensional gradient shaded image. Fast 3DFT pulse sequences were optimized to enhance the time-of-flight contrast of the intravascular region. By increasing the surface threshold value in a three-dimensional head study, the flesh of a patient's face was peeled away to demonstrate the superfacial temporal artery. Gated cardiac images show the great vessels and cardiac chambers. A three-dimensional view of the aorta shows an irregular surface in the vicinity of an adrenal tumor. 3D MR exams provide a non-invasive technique for assessing vascular morphology in a clinical setting.  相似文献

We present high resolution three dimensional (3D) connectivity, surface construction and display algorithms that detect, extract, and display the surface of a brain from contiguous magnetic resonance (MR) images. The algorithms identify the external brain surface and create a 3D image, showing the fissures and surface convolutions of the cerebral hemispheres, cerebellum, and brain stem. Images produced by these algorithms also show the morphology of other soft tissue boundaries such as the cerebral ventricular system and the skin of the patient. For the purposes of 3D reconstruction, our experiments show that T1 weighted images give better contrast between the surface of the brain and the cerebral spinal fluid than T2 weighted images. 3D reconstruction of MR data provides a non-invasive procedure for examination of the brain surface and other anatomical features.  相似文献

The purpose of this study is to describe the appearance of bowel-related abscesses on magnetic resonance (MR) images. Sixteen consecutive patients who had bowel-related abscesses underwent MR examination at 1.5T. MR sequences included T1-weighted fat-suppressed imaging pre- and post-intravenous gadolinium chelate administration (all patients) and breathing-independent single-shot T2-weighted half Fourier turbo (fast) spin echo (6 patients). Patients with pelvic abscesses also underwent sagittal imaging with post-gadolinium T1-weighted images (9 patients) and T2-weighted turbo (fast) spin echo (8 patients). Abscesses were confirmed by open surgery or surgical drainage (6 patients), percutaneous drainage (8 patients), or combined physical examination, fluoroscopic fistulogram, and clinical follow-up (2 patients). Oval-shaped fluid collections were identified in all of the patients, which ranged in diameter from 2 cm to 18 cm, mean: 8 cm. Abscesses were low to intermediate in signal on T1-weighted images, heterogenous and moderately high signal on T2-weighted images, and low signal on post-gadolinium images. A layering effect of lower signal material in the dependent portion of the abscess was noted in abscesses in 6 of 14 patients on T2-weighted images. Post-gadolinium images demonstrated a definable 3- to 7-mm thick abscess wall, which enhanced substantially with contrast. Definition of the wall was best shown on fat-suppressed images post-gadolinium. Substantial enhancement of surrounding periabscess tissues was demonstrated in all cases and was most clearly defined on fat-suppressed images. Image acquisition in two orthogonal planes was of value to demonstrate that fluid collections were oval, and separate from bowel. Image acquisition in the sagittal plane was useful in the evaluation of pelvic abscesses. The results from this preliminary study show that bowel-related abscesses are demonstrable on MR images using gadolinium-enhanced fat-suppressed T1-weighted and turbo (fast) spin-echo T2-weighted sequences. The presence of a thickened, enhancing lesion wall and enhancement of perilesional tissues on T1-weighted fat-suppressed images were observed in all abscesses. A layering effect of low signal intensity material in the dependent portion of the abscess was an important ancillary feature.  相似文献

The growing interest in Leptoglossus occidentalis, the conifer seed bug pest accidentally introduced into Europe in the 1990s, led us to investigate the female reproductive structures of the hymenopteran platygastrid Gryon pennsylvanicum, which is its candidate antagonist for biological control programmes. Our study revealed a genital apparatus with some characteristic features, such as an unusual length of the oviduct (divided into a long proximal and a short distal tract), the absence of accessory glands and the presence of a spermatheca provided with a small spermathecal gland. The ultrastructural investigation revealed that the shorter part of the common oviduct is involved in ion uptake whereas the longer part has two cell types with secretory function: the former with dense bodies and the latter with granular particles. The secretory contents of both are released into the oviduct lumen. The granular particles are formed in a complex of modified endoplasmic reticulum and appear as virus-like particles.  相似文献

Light field displays comprise three-dimensional (3D) visual information presentation devices capable of providing realistic and full parallax autostereoscopic images. In this letter, the recent advances in the light field displays based on integral imaging (II) and holographic techniques are presented. Several advanced approaches to demonstrate the light field displays including viewing angle enhancement techniques of the II display, a fast hologram generation method using graphics processing unit (GPU) and multiple WRPs, and a holographic microscopy to display the living cells are reported. These methods improve some important constraints of the light field displays and add new features.  相似文献

Optimal angle, fast repeat time, gradient field echo imaging techniques such as FISP (Fast Imaging with Steady Precession) and FLASH (Fast Low Angle Shot) often fail to discriminate disease from healthy tissue for two main reasons. First, T1 and T2 of the affected tissue may increase such that the ratio of T1 to T2 remains nearly unchanged, hence there is no contrast change with FISP. Second, T2 weighted gradient field echo images suffer severely from T2* signal and resolution loss leading to a reduction in C/N. Although FLASH imaging with two separate angles can, in principle, extract the longer T1 tumors, contrast is often not good. To overcome the inhomogeneity and contrast problems, we have implemented a FAst optimal angle spin-echo sequence with a short TE(FATE). For the first echo, FATE has the same contrast properties as FLASH with a slight decrease in signal intensity. The advantage is that the intensity of the signal does not suffer from T2* signal decay, hence improved contrast and disease detection via T2 weighted FATE images is possible. Contrast-to-noise in lesion detection is also considered for CE FAST (Contrast Enhanced Fast), a T2-weighted version of FISP, and HYBRID.  相似文献

Current fast imaging techniques are based on gradient echo sequences with reduced flip angle excitation pulses and very short repetition times TR. Practical T2 values may be of the order of TR or longer. In this situation, a different image contrast can be obtained, depending on details of the sequence. Four essentially different versions of the basic Fast Field Echo (FFE) sequence can be distinguished and are described systematically in this article. For these sequences, image contrast formulas are presented. Practical imaging should tolerate small field inhomogeneities. This requirement can be satisfied by only three of the four versions. Numerical simulations are used to study the influence of a modified phase alternation scheme on image contrasts of two of the remaining sequences. The results of the calculations are verified by phantom studies on a 1.5-T whole-body imager. Implications for contrast in clinical images are discussed in relation to head images obtained on the same machine.  相似文献

The purpose of this investigation was to quantitatively evaluate the practical impact of alteration of key imaging parameters on image quality and artifacts in fast multi-planar gradient echo (GRE) pulse sequences. These include multi-planar GRASS (MPGR) and fast multi-planar spoiled GRASS (FMPSPGR). We developed a composite phantom with different T(1) and T(2) values comprising the range of common biological tissues, which was also subjected to periodic motion in order to evaluate motion effects. Magnetic resonance imaging was performed on a GE Signa 1.5-T system. Experimental variations in key parameters included excitation flip angle (FL), echo time (TE), repetition time (TR), and receive bandwidth (BW). Quantitative analysis consisted of signal-to-noise-ratio (SNR) and contrast (CN), image nonuniformity (NU), full-width-at-half-maximum (FWHM) (i.e., blurring or geometric distortion), and ghosting ratio (GR). We found that flip angle, TE, and TR play particularly critical roles in determining image signal, homogeneity, and ghosting artifact with these sequences. Optimum clinical application of these pulse sequences requires careful attention to these imaging parameters and to their complex interactions.  相似文献
