A new RP-HPLC method has been developed for the simultaneous determination of 11 nucleosides and bases, including adenosine, cordycepin, cytidine, guanosine, inosine, thymidine, uridine, cytosine, guanine, thymine, and uracil in Cordyceps. Determination was achieved on a Zorbax 300SB C18 analytical column (4.6 x 250 mm id, 5 mm) using gradient elution with diode-array detection. All calibration curves showed good linearity (r2 > 0.9995) within the test ranges. The developed method was simple, rapid, and accurate, and showed good reproducibility for the quantification of 11 nucleosides and bases in natural and cultured Cordyceps with both intra- and inter-day variations of less than 1.8%. Furthermore, hierarchical clustering analysis based on the typical peaks of adenosine, cordycepin, and inosine in HPLC profiles from the 11 tested samples showed that natural and cultured Cordyceps were in different clusters, which could provide a means of discriminating between Cordyceps of different origins. Thus, adenosine, cordycepin, and inosine could be used as markers for quality control of Cordyceps.  相似文献

A CEC method is described for the simultaneous determination of 11 nucleosides and nucleobases including cytosine, uracil, uridine, hypoxanthine, 2'-deoxyuridine, inosine, guanosine, thymidine, adenine, adenosine, and cordycepin in Cordyceps using 5-chlorocytosine arabinoside as internal standard (IS). Chemometric optimization based on central composite design was employed to find the optimum conditions. The factors for optimization were defined as three parameters: voltage, pH, and concentration of ACN as organic modifier. The resolution (R(s)) between inosine and guanosine, as well as the entire run time were employed to evaluate the response function. A running buffer composed of 4 mM ammonium acetate and 2 mM triethylamine (TEA) adjusted to pH 5.3 using acetic acid, and containing 3% ACN as modifier, with gradient voltage (0-4 min: 20 kV, 4-12 min: linear gradient from 20 to 30 kV; 12-16 min: 30 kV) were found to be the optimum conditions for the separation. Separation of the 11 investigated compounds and 5-chlorocytosine arabinoside was achieved within 16 min. The contents of the 11 compounds in natural and cultured Cordyceps sinensis, and cultured Cordyceps militaris were also compared. The result showed that CEC is an efficient method for analysis of nucleosides and nucleobases in Cordyceps, which is helpful to control the quality of this valued traditional Chinese medicine.  相似文献

建立了以氰基正相色谱柱反相条件下快速分析腺苷和虫草素的方法。以微波辅助提取蛹虫草及其培养残基样品,每次提取1.5 min,共提取2次。采用Eclipse XDB-CN色谱柱测定提取液中腺苷和虫草素的含量,以甲醇-水(7:93, v/v)为流动相进行等度洗脱,检测波长260 nm,并考察和分析了影响分离性能的流动相组成和pH值。结果腺苷和虫草素在4.5 min内实现完全分离且无基质干扰。腺苷和虫草素在线性范围内线性关系良好,线性相关系数r2分别为0.9998和0.9995,定量限(以10倍信噪比计)分别为0.21 mg/L(腺苷)和0.083 mg/L(虫草素)。该方法日内和日间精密度的相对标准偏差(RSD)小于2%;腺苷和虫草素的平均加标回收率为93.8%~102.9%, RSD值不大于3.62%(n=5)。本方法简便、快速、准确、成本低,可用于冬虫夏草、蛹虫草子实体、虫草培养残基及虫草制剂中腺苷和虫草素含量的快速测定。  相似文献

Cordyceps sinensis is a well-known traditional Chinese medicine, and some of the active components are nucleosides. The analysis of nucleosides in Cordyceps material has been performed by reversed-phase high-performance liquid chromatography (HPLC) with gradient elution or by spectrometry. Here, we have explored the possibility of using capillary electrophoresis to determine the content of three major nucleosides (adenosine, guanosine and uridine) in Cordyceps. Capillary electrophoresis needs no gradients, and it provides a better separation due to its higher efficiency. In order to optimize the resolution, the separation of adenosine, guanosine and uridine was determined in Cordyceps with respect to the variation of buffer concentration, pH, temperature, and voltage. By using the calibrated electrophoresis system, the separation was achieved for the three nucleosides in less than 10 min with a background electrolyte consisting of 0.2 M boric acid-sodium hydroxide buffer, pH 8.5. The nucleoside contents of various types of natural Cordyceps and cultured Cordyceps mycelia were determined and compared. There was a great variation of nucleoside content in different sources of Cordyceps; the cultured Cordyceps mycelia, however, contains a much higher concentration than the natural Cordyceps.  相似文献

Nine nucleosides and nucleobases, including uracil, adenine, thymine, uridine, adenosine, thymidine, cytidine, guanosine, and cordycepin in natural Cordyceps sinensis, cultured Cordyceps mycelia, and Cordyceps fruiting bodies were extracted by matrix solid‐phase dispersion (MSPD) and determined by HPLC. The experimental conditions for the MSPD extraction were optimized. Florisil was used as dispersant, petroleum ether as washing solvent, and methanol as elution solvent. The Florisil‐to‐sample ratio was selected to be 4:1 and no additional clean‐up sorbent was needed. The calibration curves had good linear relationships (r > 0.9997). The LOD and LOQ were in the range of 12 ~ 79 and 41 ~ 265 ng/mL, respectively. The intra‐ and interday precision were lower than 8.3%. The recoveries were between 61.5 and 93.2%. The present method consumed less sample compared with ultrasonic extraction and heating reflux extraction (HRE). The extraction yields obtained by using the present method are much higher than those obtained by UE and comparable to those obtained by HRE.  相似文献

A simple method is described for simultaneous determination of six main nucleosides and bases including adenine, uracil, adenosine, guanosine, uridine and inosine in Cordyceps by capillary electrophoresis (CE). Chemometric optimization based on central composite design was employed to find the optimum resolution. The optimum factor space was defined by three parameters: buffer concentration, pH and concentration of acetonitrile as organic modifier. Resolution (Rs) was employed to evaluate the response function. A running buffer composed of 500 mM boric acid, adjusted pH to 8.6 with sodium hydroxide and 12.2% acetonitrile as modifier was found to be the most appropriate for the separation. The contents of the six components were determined by using adenosine monophosphate as an internal standard. Furthermore, hierarchical clustering analysis based on characteristics of 32 peaks in CE profiles from the tested 12 samples showed that natural and cultured Cordyceps were in different clusters. Adenosine and inosine were extracted as markers for discrimination of natural Cordyceps. The result of clustering based on the two peaks characteristics was in excellent agreement with that based on 32 peaks'. Thus, adenosine and inosine could be used as markers for quality control of natural and cultured Cordyceps.  相似文献

《中国药典》收载的发酵虫草菌粉产品的质量标准中,规定以鸟苷、腺苷、尿苷的含量作为评价相关产品质量的标准。但除此之外,还有许多其他的核苷类成分对发酵虫草菌粉质量控制的影响尚未被探讨。为探究发酵虫草菌粉及产品质控指标选择的合理性,采用超高效液相色谱-紫外检测法对19批发酵虫草菌粉及产品中9种核苷成分(尿嘧啶、胞苷、鸟嘌呤、尿苷、腺嘌呤、肌苷、鸟苷、胸苷、腺苷)进行了定量分析,建立了发酵虫草菌粉样品的指纹图谱,并结合统计学提供了一种分析指标性成分的方法。通过优化样品的提取方法,选择超声提取法制备19批发酵虫草菌粉及产品的供试液;采用Agilent Eclipse Plus C18色谱柱(150 mm×4.6 mm, 3.5 μm)进行色谱分析,以甲醇-水为流动相梯度洗脱,对方法的校正曲线、准确度、精密度、重复性和回收率进行了验证。结合对照品指认了指纹图谱中的9个核苷峰,并采用外标一点法测得了各核苷成分的含量。使用化学模式识别对指纹图谱中的共有峰进行分析,聚类分析和主成分分析得到了同样的分类结果:19批样品共分为5类,其中同一发酵虫草菌粉因工艺差异可分为2类,而心肝宝胶囊、百令胶囊、宁心宝胶囊则各单独分为1类。同时,使用主成分分析获得了各样品中的指标性成分,分别为尿苷、鸟苷、腺苷、腺嘌呤、尿嘧啶,并使用聚类分析再次进行确证,验证了指标性成分的合理性。  相似文献

The RNA nucleosides, namely adenosine, cytidine, guanosine and uridine were γ-radioyzed in solid state and in vacuo at room temperature to a total dose of 3.2 MGy. Through electronic absorption spectroscopy, differential scanning calorimetry and through polarimetry and optical rotatory dispersion spectroscopy, it was found that the purine-based nucleosides (adenosine and guanosine) show a much higher radiolysis resistance than the pyrimidine-based nucleosides (cytidine and uridine). In an astrochemical/astrobiological context, these results may explain why purine nucleobases are found in practically all carbonaceous chondrite meteorites while the pyrimidine nucleobases are absent or below the detection limits of the current analytical techniques. In the hypothesis that both purines and pyrimidines nucleobases were present in certain bodies at the beginning of the solar system 4.6?×?109 years ago, the radiolysis due to radionuclides decay has destroyed more easily and completely the pyrimidine bases due to their much lower radiolysis resistance than the purine bases.  相似文献

Determination of nucleosides and their metabolic compounds is important for physiological and pharmacological studies. Herein, a rapid ultra-performance liquid chromatography (UPLC) method was developed for the simultaneous determination of 14 nucleosides and nucleobases, namely adenine, adenosine, cytosine, cytidine, uracil, uridine, guanine, guanosine, hypoxanthin, inosine, thymine, thymidine, 2′-deoxyuridine and cordycepin. The separation was performed on Waters Acquity UPLC system with Acquity UPLC BEH C18 column and gradient elution of 0.5 mM acetic acid and acetonitrile in 5 min. The correlation coefficients of 14 analytes were high (R2 > 0.9995) within the test ranges. The LOD and LOQ were lower to 11.9 and 47.0 ng/ml with 1 μl of injection volume, respectively. The overall R.S.D. for intra- and inter-day of 14 analytes were less than 1.8%. The developed method was applied for the analysis of nucleosides and nucleobases in cultured Cordyceps, which also could be used for the fast determination of the analytes in pharmaceutical products and biological fluids.  相似文献

A high-throughput phosphoramidite method for HepDirect prodrug synthesis was optimized on seven representative nucleosides, adenosine, inosine, guanosine, uridine, cytidine, AICA-riboside, and thymidine, each on a 5 mg scale. The variables optimized included (1) reaction time, (2) reaction temperature, (3) activating agent, (4) solvent, (5) purification method, and (6) stoichiometry. Preparative HPLC with mass-based fraction collection and yield determination from an ELSD standard curve enabled high-throughput. The optimized conditions for the representative nucleosides required 6 mol equiv of phosphoramidite to nucleoside and resulted in an average HPLC determined yield of 31 +/- 14% and HPLC purity of 93 +/- 3%.  相似文献

建立了高效液相色谱同时测定中药材虎掌南星中核苷类活性成分(腺嘌呤、次黄嘌呤、黄嘌呤、尿苷、胸腺嘧啶、腺苷、鸟苷)含量的方法。以Lichrospher C18色谱柱(150 mm×4.6 mm, 5 μm)分离,以乙腈-水(含0.1%甲酸)为流动相,梯度洗脱,在254 nm下检测,腺嘌呤、次黄嘌呤、黄嘌呤、尿苷及鸟苷分别在1.6~50 mg/L范围内、胸腺嘧啶和腺苷分别在1.2~40 mg/L范围内的线性关系良好,相关系数均大于0.9995,加标回收率为98.9%~101.2%,相对标准偏差均小于3%。方法学考察结果显示符合含量测定要求,并应用于不同产地虎掌南星的测定。该方法操作简便、快速,结果可靠,重现性好,可作为虎掌南星质量评价的参考依据。  相似文献

Abstraet—2'-Acetylformanilide has been found to be an effective, near-ultraviolet (300–380 nm) sensitizer for the photooxidation of nucleosides and nucleotides in aqueous solution, with hydrogen peroxide being formed in high yield. The decreasing order of hydrogen peroxide formation and substrate destruction was found to be: guanosine. adenosine, thymidine, uridine and cytidine. The process was highly pH dependent, low pH being most favorable for photooxidation. Experiments using deuterium oxide and superoxide dismutase indicate that both singlet oxygen and superoxide ion can be involved in hydrogen peroxide formation.  相似文献

Cross-linked xylose isomerase (EC 5.3.1.5., from Streptomyces rubiginosus) crystals (CLXIC) packed into a 7.8 x 300 mm steel column showed specific affinity towards uridine (Urd), cytidine (Cyd), adenosine (Ado), guanosine (Guo), and thymidine. These nucleosides eluted out of the CLXIC column in the same order as the corresponding nucleoside bases, indicating that the retention depends mainly on the base component of the molecule. The interaction of nucleosides with the CLXIC material was not based merely on ion exchange or hydrophobic interactions but also on the unique properties of the CLXIC column. Decrease in temperature increased the retention but not the resolution factors of the adjacent nucleosides. The CLXIC column maintained its separation capacity even when 100 mg of ribonucleosides in equimass amounts were injected into the column in a volume of 1 mL corresponding to 10% of the total column volume. Analysis of sugar beet molasses, a side stream from sucrose production, showed it to contain 1-2.5 mg mL(-1) of Urd, Cyd, Ado, and Guo. The CLXIC column was able to separate and enrich these nucleosides also from highly viscous sugar beet molasses. The CLXIC column was especially efficient in the purification of guanosine. Other commercially interesting sugar beet molasses components such as the acidic compounds betaine, gamma-amino butyric acid, and D- and L-pyroglutamic acids or neutral sucrose did not interact with the CLXIC material.  相似文献

In an attempt to separate a number of methylated nucleoside constituents reportedly present in various mammalian RNA preparations, a comparison of the migration of selected nucleoside standards was assessed via thin-layer chromatographic techniques with respect to three solvent systems and an adsorbent of varying proportions of silica gel and microcrystalline cellulose. The results of these studies revealed that a mixture of seventeen methylated constituents could be separated following two-dimensional development on an adsorbent containing 60% cellulose and 40% silica gel. Constituents separated included the 2'-O-methylated derivatives of adenosine, guanosine, cytidine and uridine, 1-, N6- and N6,N6-methyladenosines, 1-methylinosine, 1-, 7-, N2- and N2,N2-methylguanosines, 3-, 4- and 5-methylcytidines and 3- and 5-methyluridines. In addition, all of the above constituents were separated from their respective non-methylated nucleosides and bases.  相似文献

本文利用液相色谱质谱联用技术研究了N-磷酰化丙氨酸和四种核苷(腺苷,尿苷,胞苷和鸟苷)的模板反应产物。结果表明生成了不同类型的单核苷酸和二核苷酸,并且生成的二核苷酸序列也得到了确证。研究结果揭示,二核苷酸骨架裂解形成的c离子可以作为确证二核苷酸序列的诊断离子。本文首次证明不论是在正离子模式还是在负离子模式下,c离子都可以用来确定此反应体系中生成的二核苷酸产物的序列。  相似文献
