Abstract

Methyl 2‐acetamido‐4‐amino‐2,4,6‐trideoxy‐α‐D‐galactopyranoside (10) was synthesized from D‐glucosamine hydrochloride in eight steps in an overall yield of 31%. Key steps include the selective benzoylation at O‐3 of methyl 2‐acetamido‐2,6‐dideoxy‐α‐D‐glucopyranoside in 89% yield and the subsequent Mitsunobu reaction using diphenylphosphoryl azide as the azide source which proceeded in 92% yield. Di‐ and mono‐benzyloxycarbonyl derivatives of 10 were also prepared.  相似文献

The hexopyranosid‐2‐ylidenemalononitrile 1 reacted with phenyl isothiocyanate in the presence of triethylamine to furnish (2R,4aR,6S,10bS)‐8‐amino‐4a,6,10,10b‐tetrahydro‐6‐methoxy‐2‐phenyl‐10‐phenylimino‐4H‐thiopyrano[3′,4′:4,5]pyrano[3,2‐d][1,3]dioxine‐7‐carbonitrile (2). Starting from 1, cyclization with sulphur and diethylamine yielded (2R,4aR,6S,9bR)‐8‐amino‐4,4a,6,9b‐tetrahydro‐6‐methoxy‐2‐phenylthieno[2′,3′:4,5]pyrano[3,2‐d][1,3]dioxine‐7‐carbonitrile (3), which could be transformed into the corresponding aminomethylenamino derivative 4 by treatment with triethyl orthoformate and ammonia. Intramolecular cyclization of 4 to yield (2R,4aR,6S,11bR)‐4,4a,6,11b‐tetrahydro‐6‐methoxy‐2‐phenyl[1,3]dioxino[4″,5″:5′,6′]pyrano[3′,4′:4,5]thieno [2,3‐d]pyrimidin‐7‐amine (5) was achieved by using NaH as base. (2R,4aR,6S,9bS)‐8‐Amino‐4a,6,9,9b‐tetrahydro‐6‐methoxy‐9‐(4‐methylphenyl‐sulfonyl)‐2‐phenyl‐4H‐[1,3]dioxino[4′,5′:5,6]pyrano[4,3‐b]pyrrole‐7‐carbonitrile (6) was prepared by treatment of compound 1 with tosylazide and triethylamine.  相似文献

The C‐glycoside of methyl α‐d‐altropyranosyl‐(1→4)‐α‐d‐glucopyranoside 2 was prepared in a convergent fashion, from readily available precursors, 4‐Otert‐butyldiphenylsilyl‐1,2‐O‐isopropylidene‐derythroS‐phenyl monothiohemiacetal 13 (five steps from D‐ribose) and the known acid, methyl 2,3,6‐tri‐O‐benzyl‐4‐C‐(carboxymethyl)‐4‐deoxy‐α‐d‐glucopyranoside 17 (seven steps from methyl α‐d‐glucopyranoside). The key reactions in the synthesis are the oxocarbenium ion cyclization of thioacetal‐enol ether 19 to a C1 substituted glycal 20, and the stereoselective hydroboration of 20 to the α‐C‐altroside 21.  相似文献

Abstract

A simple method for the preparation of 7‐alkoxy/hydroxy‐α‐methyl‐DL‐tryptamines is reported. The key steps of the synthesis are the Japp–Klingemann coupling of 2‐piperidone‐3‐carboxylic acid 3 with diazonium salts 4, the Fischer‐type cyclization of hydrazones 5 to β‐carboline derivatives 6 and their hydrolysis to title compounds 8.  相似文献

Treatment of 2‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)ethanal (3) with malononitrile in the presence of aluminium oxide provided 2‐cyano‐4‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)crotononitrile (4). Starting from 4, cyclization with sulphur and triethylamine yielded 2‐amino‐5‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)thiophene‐3‐carbonitrile (5). Further cyclization could be achieved with triethyl orthoformate/ammonia to furnish 4‐amino‐6‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐d‐altropyranosid‐2‐yl)thieno[2.3‐d]pyrimidine (8).  相似文献

Abstract

1‐(Methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐D‐altropyranosid‐2‐yl)‐4‐phenyl‐but‐3‐yn‐2‐one (4) was synthesized by the reaction of (methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐D‐altropyranosid‐2‐yl)ethanal (2) with lithium phenylethynide and following oxidation. Compound 4 and hydrazine hydrate provided the 3(5)‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐D‐altropyranosid‐2‐yl‐methyl)‐5(3)‐phenyl‐1H‐pyrazole (5). The reactions of 4 with amidinium salts and a S‐methyl‐isothiouronium salt, respectively, furnished the pyrimidine C‐nucleoside analogues 6a6c. Treatment of 4 with 2‐aminobenzimidazole afforded 2‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐D‐altropyranosid‐2‐ylmethyl)‐4‐phenyl‐benzo [4,5]imidazo[1,2‐a]pyrimidine (7a). Compound 4 and sodium azide yielded 2‐(methyl 3‐O‐benzyl‐4,6‐O‐benzylidene‐2‐deoxy‐α‐D‐altropyranosid‐2‐yl)‐1‐[5(4)‐phenyl‐1H(2H)‐1,2,3‐triazole‐4(5)‐yl]ethanone (8).  相似文献

The ketoesters 3 and 4, obtained by the condensation of 2‐cyclohexanone carboxylate and 1‐chloro‐3‐pentanone, were heated with 2,3‐dichloro‐5,6‐dicyanobenzoquinone (DDQ) to yield the dienones 5 and 6, which on hydrolysis with potassium t‐butoxide and dimethyl sulfoxide afforded tetralin 8. These were converted to tetralone 10 by methylation and oxidation respectively. Further methylation of 10 yielded tetralone 11.  相似文献

A copper‐catalyzed reaction of propargyl 4,6‐di‐O‐acetyl‐2,3‐dideoxy‐α‐Derythro‐hex‐2‐enopyranoside with 3(4‐azidophenyl)‐1,2,4‐oxadiazoles gave the corresponding hexenopyranosides bearing an 1,2,4‐oxadiazole subunit in the aglyconic part of the molecule. The same reaction between ethyl 4‐azido‐2,3,4‐trideoxy‐α‐Derythro‐hex‐2‐enopyranoside and acetylenic 1,2,4‐oxadiazoles afforded the corresponding hexenopyranosides carrying a triazole and a 1,2,4‐oxadiazole ring at C‐4 of the carbohydrate. Combination of the two sequences gave hexenopyranosides displaying two 1,2,4‐oxadiazole subunits, each one being embedded in the C‐1 and C‐4 frameworks, of the carbohydrate moiety. A simple dihydroxylation reaction of these unsaturated carbohydrates yielded a series of mannopyranosides bearing one or two 1,2,4‐oxadiazole subunits at C‐1 or C‐4. These new compounds were evaluated for their cytotoxic activities against two cell strains: NCI‐H292 (lung carcinoma) and Hep‐2 (larynx carcinoma), some of them presenting impressive cell growth inhibitions.  相似文献

The title compound was synthesized starting from methyl 3,4,6‐tri‐O‐acetyl‐2‐acetamido‐2‐deoxy‐β‐D‐glucopyranoside, oxalyl chloride, and methyl 3,4,6‐tri‐O‐acetyl‐2‐amino‐2‐deoxy‐β‐D‐glucopyranoside. Herein we report a facile synthesis of esters of bis‐α‐aminoalkylphosphinic acids obtained by an addition of Cbz‐protected phosphinic analogues of amino acid methyl esters to an appropriate imine in refluxing benzene. The formation of four differently protected 4‐deoxy‐D‐lyxo‐hexose derivatives 7, 8, 12, and 14 is described. In the first procedure, a nucleophilic displacement of the allylic mesylate 4 by hydride was combined with a highly stereoselective osmylation of olefin 6 to afford diol 7. In the second radical procedure, tributyl tin hydride was substituted by the cheap and environmentally friendly hypophosphorous acid as a hydrogen donor in the reduction of xanthate 13 to 4‐deoxy lyxo‐hexose 14.  相似文献

A reaction system was found to prepare tetra‐O‐acetyl‐1‐thio‐d‐glycopyranose in both α and β‐forms. Methanolysis of the adduct prepared from the reaction of tetra‐O‐acetyl‐α‐d‐glucopyranosyl bromide with N,N‐dimethylthioformamide afforded the corresponding tetra‐O‐acetyl‐1‐thio‐d‐glucopyranose with an anomer ratio α/β of 52:48 in 98% yield. The anomer mixture was easily separated by column chromatography to obtain the product of α‐form. This synthetic method is very convenient to proceed by one‐pot reaction under ordinary conditions.  相似文献

The reaction of the lithio derivative of novel polyethylene glycol (PEG)‐supported α‐phenylselenopropionate with aldehydes, followed by oxidation–elimination with 30% hydrogen peroxide and then treatment with sodium nitrite, formed PEG‐bound 2‐nitromethyl‐2‐alkenoates. Subsequent cleavage from the PEG efficiently afforded methyl (E)‐2‐nitromethyl‐2‐alkenoates in good yields and high purities.  相似文献

Synthesis of α‐C‐ethylene phosphate and phosphonate as well as α‐C‐methylene phosphate analogues of N‐acetyl‐α‐d‐glucosamine 1‐phosphate is reported starting from the common perbenzylated 2‐acetamido‐2‐deoxy‐α‐C‐allyl glucoside. Anomerisation of the corresponding amino α‐C‐glucosyl aldehyde to the β‐aldehyde was observed. Thus, both amino α‐ and β‐C‐glucosyl methanol were obtained after reduction.  相似文献
